Introduction: Widespread opportunistic pathogens pose a serious threat to global health, particularly in susceptible hospital populations. The escalating crisis of antibiotic resistance highlights the urgent need for novel antibacterial agents and alternative treatment approaches. Traditional Chinese Medicine (TCM) and its compounds have deep roots in the treatment of infectious diseases. It has a variety of active ingredients and multi-target properties, opening up new avenues for the discovery and development of antimicrobial drugs. Methods: This study focuses on assessing the efficacy of the Shensheng-Piwen changed medicinal powder (SPC) extracts against opportunistic pathogen infections by broth microdilution and agar disc diffusion methods. Additionally, biofilm inhibition and eradication assays were performed to evaluate the antibiofilm effects of SPC extracts. Results: Metabolite profiles were analyzed by LC-MS. Furthermore, the potential synergistic effect between SPC and Metal-Organic Framework (MOF) was investigated by bacterial growth curve analysis. The results indicated that the SPC extracts exhibited antibacterial activity against S. aureus, with a minimum inhibitory concentration (MIC) of 7.8 mg/mL (crude drug concentration). Notably, at 1/2 MIC, the SPC extracts significantly inhibited biofilm formation, with over 80% inhibition, which was critical in tackling chronic and hospital-acquired infections. Metabolomic analysis of S. aureus revealed that SPC extracts induced a notable reduction in the levels of various metabolites, including L-proline, L-asparagine. This suggested that the SPC extracts could interfere with the metabolism of S. aureus. Meanwhile, the growth curve experiment proved that SPC extracts and MOFs had a synergistic antibacterial effect. Discussion: In conclusion, the present study highlights the potential of SPC extracts as a novel antibacterial agent against S. aureus infections, with promising biofilm inhibition properties. The observed synergistic effect between SPC extracts and MOFs further supports the exploration of this combination as an alternative treatment approach.

RESUMO

The syndrome of post-traumatic hydrocephalus (PTH) has been recognized since Dandy's report in 1914. The pathogenesis of PTH has not been fully clarified. At present, it is believed that the obstacles of cerebrospinal fluid (CSF) secretion, absorption and circulation pathways are the reasons for the development of PTH. However, recent studies have also suggested that the osmotic pressure load of CSF and the pathological changes of CSF dynamics are caused by the development of hydrocephalus. Therefore, a better understanding of the definition, classification, diagnostic criteria, treatment, and evaluation of post-treatment effects of PTH is critical for the effective prevention and treatment of PTH. In this paper, we reviewed the classification and diagnosis of PTH and focused on the treatment and the imaging evaluation of post-treatment effects of PTH. This review might provide a judgment criterion for diagnosis of PTH and a basis for the effective prevention and treatment of PTH in the future.

Recent rapid warming has caused uneven impacts on the composition, structure, and functioning of northern ecosystems. It remains unknown how climatic drivers control linear and non-linear trends in ecosystem productivity. Based on a plant phenology index (PPI) product at a spatial resolution of 0.05° over 2000-2018, we used an automated polynomial fitting scheme to detect and characterize trend types (i.e., polynomial trends and no-trends) in the yearly-integrated PPI (PPIINT) for northern (> 30°N) ecosystems and their dependence on climatic drivers and ecosystem types. The averaged slope for the linear trends (p < 0.05) of PPIINT was positive across all the ecosystems, among which deciduous broadleaved forests and evergreen needle-leaved forests (ENF) showed the highest and lowest mean slopes, respectively. More than 50% of the pixels in ENF, arctic and boreal shrublands, and permanent wetlands (PW) had linear trends. A large fraction of PW also showed quadratic and cubic trends. These trend patterns agreed well with estimates of global vegetation productivity based on solar-induced chlorophyll fluorescence. Across all the biomes, PPIINT in pixels with linear trends showed lower mean values and higher partial correlation coefficients with temperature or precipitation than in pixels without linear trends. Overall, our study revealed the emergence of latitudinal convergence and divergence in climatic controls on the linear and non-linear trends of PPIINT, implying that northern shifts of vegetation and climate change may potentially increase the non-linear nature of climatic controls on ecosystem productivity. These results can improve our understanding and prediction of climate-induced changes in plant phenology and productivity and facilitate sustainable management of ecosystems by accounting for their resilience and vulnerability to future climate change.

Background: Long-term differential expression of disease-associated genes is a crucial driver of pathological changes in mucinous gastric carcinoma. Therefore, there should be a correlation between depth features extracted from pathology-based full-scan images using deep learning and disease-associated gene expression. This study tried to provides preliminary evidence that long-term differentially expressed (disease-associated) genes lead to subtle changes in disease pathology by exploring their correlation, and offer a new ideas for precise analysis of pathomics and combined analysis of pathomics and genomics. Methods: Full pathological scans, gene sequencing data, and clinical data of patients with mucinous gastric carcinoma were downloaded from TCGA data. The VGG-16 network architecture was used to construct a binary classification model to explore the potential of VGG-16 applications and extract the deep features of the pathology-based full-scan map. Differential gene expression analysis was performed and a protein-protein interaction network was constructed to screen disease-related core genes. Differential, Lasso regression, and extensive correlation analyses were used to screen for valuable deep features. Finally, a correlation analysis was used to determine whether there was a correlation between valuable deep features and disease-related core genes. Result: The accuracy of the binary classification model was 0.775 ± 0.129. A total of 24 disease-related core genes were screened, including ASPM, AURKA, AURKB, BUB1, BUB1B, CCNA2, CCNB1, CCNB2, CDCA8, CDK1, CENPF, DLGAP5, KIF11, KIF20A, KIF2C, KIF4A, MELK, PBK, RRM2, TOP2A, TPX2, TTK, UBE2C, and ZWINT. In addition, differential, Lasso regression, and extensive correlation analyses were used to screen eight valuable deep features, including features 51, 106, 109, 118, 257, 282, 326, and 487. Finally, the results of the correlation analysis suggested that valuable deep features were either positively or negatively correlated with core gene expression. Conclusion: The preliminary results of this study support our hypotheses. Deep learning may be an important bridge for the joint analysis of pathomics and genomics and provides preliminary evidence for long-term abnormal expression of genes leading to subtle changes in pathology.

Wind erosion can cause desertification and sandstorms in arid and semiarid areas. However, quantitative studies of the dynamic changes in wind erosion over long time periods are relatively rare, and this knowledge gap hinders our understanding of desertification under the conditions of a changing climate. Here, we selected the Mongolian Plateau as the study area. Using the revised wind erosion equation (RWEQ) model, we assessed the spatial and temporal dynamics of wind erosion on the Mongolian Plateau from 1982 to 2018. Our results showed that the wind erosion intensity on the Mongolian Plateau increased from northeast to southwest. The annual mean wind erosion modulus was 46.5 t·ha-1 in 1982-2008, with a significant decline at a rate of -5.1 t·ha-1·10 yr-1. The intensity of wind erosion was the strongest in spring, followed by autumn and summer, and was weakest in winter. During 1982-2018, wind erosion showed a significant decreasing trend in all seasons except winter. The wind erosion contribution of spring to the total annual wind erosion significantly increased, while that of summer significantly decreased. These results can help decision-makers identify high-risk areas of soil erosion on the Mongolian Plateau and take effective measures to adapt to climate change.

Large stocks of soil organic carbon (SOC) accumulated in the Northern Hemisphere permafrost regions may be vulnerable to climatic warming, but global estimates of SOC distribution and magnitude in permafrost regions still have large uncertainties. Based on multiple high-resolution environmental variables and a compiled soil sample dataset (>3000 soil profiles), we used machine-learning methods to estimate the size and spatial distribution of SOC for the top 3 m soils in the Northern Hemisphere permafrost regions. We also identified key environmental predictors of SOC. The results showed that the SOC storage for the top 3 m soil was 1079 ± 174 Pg C across the Northern Hemisphere permafrost regions (20.8 × 106 km2), including 1057 ± 167 Pg C in the northern permafrost regions and 22 ± 7 Pg C in the Third Pole permafrost regions. The mean annual air temperature and NDVI are the main controlling factors for the spatial distribution of SOC stocks in the northern and the Third Pole permafrost regions. Our estimations were more accurate than the existing global SOC stock maps. The results improve our understanding of the regional and global permafrost carbon cycle and their feedback to the climate system.

Most lakes on the Qinghai-Tibet Plateau have expanded in recent years. Zonag Lake, a critical habitat for Tibetan antelopes in the continuous permafrost zone, burst and overflowed after several years of expansion, resulting in a reduction of approximately 100 km2 in the lake area. Observations have revealed new permafrost is forming on the exposed bottom, accompanied by various periglacial landscapes. The permafrost aggradation on the exposed bottom is rapid, and the permafrost base reached 4.9 m, 5.4 m, and 5.7 m in the first three years, respectively. In this study, the future changes and influencing factors of recently formed permafrost are simulated using a one-dimensional finite element model of heat flow. The simulated results indicate that the permafrost on the exposed bottom is likely to continue to develop, appearing first quick back slow trend. Besides the surface temperature, the annual amplitude is also an important factor in affecting the aggradation of permafrost. The unidirectional permafrost aggradation in the study area is different from the bidirectional permafrost aggradation on the closed taliks around the Arctic. Additionally, snow cover and vegetation are two important factors influencing the future development of permafrost on the exposed lake bottom.

Introduction: Parkinson's disease is a neurodegenerative disorder involving loss of dopaminergic neurons. Multiple studies implicate the microbiota-gut-brain axis in Parkinson's disease pathophysiology. Ping-wei-san plus Herbal Decoction, a traditional Chinese medicine composition with beneficial effects in Parkinson's disease, may have a complex array of actions. Here we sought to determine whether gut microbiota and metabolic pathways are involved in Ping-wei-san plus herbal therapy for Parkinson's disease and to identify functional pathways to guide research. Methods and results: The model of Parkinson's disease were induced with the rotenone. The Ping-wei-san plus group received the PWP herbal decoction for 90 days, after which all groups were analyzed experimentally. PWP herbal treatment improved motor behavior and emotional performance, balanced gut microbiota, and benefited dietary metabolism. Tandem Mass Tags mass spectrometry identified many differentially expressed proteins (DEPs) in the substantia nigra and duodenum in the PWP group, and these DEPs were enriched in pathways such as those involving cAMP signaling, glutamatergic synapses, dopaminergic synapses, and ribosome-rich functions in the gut. The PWP group showed increases in recombinant tissue inhibitors of metalloproteinase 3, and nucleotide-binding oligomerization domain, leucine rich repeat, and pyrin domain containing proteins 6 in the substantia nigra and decreased parkin, gasdermin D, recombinant tissue inhibitors of metalloproteinase 3, and nucleotide-binding oligomerization domain, leucine rich repeat and pyrin domain containing proteins 6 in the duodenum. Discussion: In conclusion, this study combined gut microbiota, metabolomics, and proteomics to evaluate the mechanism of action of Ping-wei-san plus on Parkinson's disease and revealed that PWP herbal treatment modulated gut microbiota, altered metabolite biological pathways, and affected functional pathway protein expression in Parkinson's disease mice, resulting in therapeutic effects.

Micropipe, a "killer" defect in SiC crystals, severely hampers the outstanding performance of SiC-based devices. In this paper, the etching behavior of micropipes in 4H-SiC and 6H-SiC wafers was studied using the molten KOH etching method. The spectra of 4H-SiC and 6H-SiC crystals containing micropipes were examined using Raman scattering. A new Raman peak accompanying micropipes located near -784 cm-1 was observed, which may have been induced by polymorphic transformation during the etching process in the area of micropipe etch pits. This feature may provide a new way to distinguish micropipes from other defects. In addition, the preferable etching conditions for distinguishing micropipes from threading screw dislocations (TSDs) was determined using laser confocal microscopy, scanning electron microscopy (SEM) and optical microscopy. Meanwhile, the micropipe etching pits were classified into two types based on their morphology and formation mechanism.

OBJECTIVE: To explore the potential mechanism of Huanglian Jiedu Decoction (HJD) treatment and prevention of metastatic Cutaneous Melanoma (CM) occurrence and metastasis based on network pharmacological methods and immune infiltration analysis. METHODS: The GEO database was used to obtain metastatic CM disease targets, the TCMSP database and the HERB database were used to obtain HJD action targets, core genes were screened by protein interaction network, and the potential mechanism of HJD in the treatment of metastatic CM was explored by enrichment analysis, prognostic analysis and immune infiltration analysis. RESULTS: HJD treatment of metastatic CM involved 60 targets, enrichment analysis showed that HJD treatment of metastatic CM involved Chemokine signaling pathway, NF-kappa B signaling pathway, and Fluid shear stress and atherosclerosis, etc. Prognostic analysis revealed that HJD had a certain ability to improve the prognosis of metastatic CM patients. Immune infiltration analysis showed that HJD could inhibit the immune cell infiltration of metastatic CM patients by acting on related targets. CONCLUSIONS: Our study identified the potential mechanism of HJD in the treatment of metastatic CM through network pharmacology, and revealed the mechanism of HJD in the prevention of Skin Cutaneous Melanoma metastasis through immune infiltration analysis and prognostic analysis.

Surface air temperature is an important factor for the permafrost thermal state in the Northern Hemisphere. It is therefore necessary to understand the variations and regional differences in air temperature to determine the interactions between permafrost degradation and climate change. In this study, we used observational data from the National Centers for Environmental Information, the China Meteorological Administration, and the World Data Centre for Meteorology to quantitatively analyze the variations and regional differences in air temperature from 1980 to 2018. The results demonstrated that the annual mean air temperatures were low in continuous permafrost regions and high in sporadic and isolated permafrost regions, with a significant warming rate of 0.371 ± 0.086 °C/decade. Air temperatures warmed the slowest during the winter and fastest during the spring, and no "warming hiatus" was observed in the permafrost regions of the Northern Hemisphere. The spatial patterns of freezing degree-days (FDDs) and thawing degree-days (TDDs) had different spatial characteristics. The decreasing rate of FDDs was -6.97 °C·days/year, while the increasing rate of TDDs was 6.4 °C·days/year. The air temperatures and warming trends had largely regional differences with respect to high latitude, transitional, and high altitude permafrost regions. Air temperature and its warming trend was the highest in high altitude regions. In addition, air temperature warming trends gradually decreased from the continuous permafrost zone to the island permafrost zone. The FDDs had a significant decreasing trend from the continuous permafrost zone to the island permafrost zone, whereas TDDs exhibited the opposite trend. The results indicate that the air temperature warming rate in the permafrost regions was approximately 2.0 times that of the global warming rate, and 1.3 times the global land warming rate from 1980 to 2018. These findings offer a perspective on the differences in permafrost and its thermal state across different regions under climate change.

Climate warming could exacerbate the occurrence of thaw settlement hazard in the permafrost regions of the Qinghai-Tibet Plateau (QTP), which would threaten the stability of engineering infrastructure in cold regions. The risk associated with permafrost settlement, valuable for the regional sustainable development, remains poorly assessed or understood on the QTP. In this study, three common Geo-hazard indices were used to assess the settlement risks in the permafrost regions of the QTP, including the settlement index, the risk zonation index, and the allowable bearing capacity index. However, large spatial differences existed in simulating the risk maps by using the abovementioned Geo-hazard indices. Hence, we developed a combined index (Ic) by integrating the three indices to reduce the uncertainty of the simulations. The results indicated that the ground ice is a critical factor for assessing the settlement risk in permafrost regions. We also applied the Ic to assess the settlement risk along the Qinghai-Tibet Railway (QTR). The proportion of low-risk area along the QTR would be the highest (45.38%) for the future periods 2061-2080 under Representative Concentration Pathway 4.5. The medium-risk area combined with the high-risk area would be accounted for more than 40%, which were located at the boundary of the present permafrost regions. Therefore, the corresponding adaptation measures should be taken to reduce the potential economic losses caused by the high-risk regions to the infrastructure. Overall, the results would present valuable references for engineering design, construction and maintenance, and provide insights for early warning and prevention of permafrost thaw settlement hazard on the QTP.

ETHNOPHARMACOLOGICAL RELEVANCE: Shenling Baizhu San (SBS) as a classic Chinese medicine prescription, has been extensively used in gastrointestinal diseases, such as ulcerative colitis and chronic diarrhea. In recent years, SBS has shown a beneficial effect on chronic obstructive pulmonary disease (COPD) patients. However, clinical trials had shown conflicting results of SBS on improving pulmonary function and other related indicators of patients with stable COPD. The efficacy of SBS on stable COPD patients has not been fully assessed. AIM OF THE STUDY: To determine whether the SBS used in the treatment of gastrointestinal disease was effective to treat COPD, we assessed the clinical evidence and efficacy of SBS supplemental treatment on stable COPD patients by a systematic review and meta-analysis of clinical trials. MATERIALS AND METHODS: Nine electronic databases were searched to include clinical trials (published until August 31, 2020) with SBS as a supplementation treatment on stable COPD. Mean difference (MD) was used to evaluate continuous variables, odds ratio (OR) was calculated to evaluate dichotomous. The Egger's test was applied for publication bias. RESULTS: A total of 770 COPD participants from 11 trials that met the inclusion criteria were included. The meta-analysis showed that modified SBS could improve the exercise endurance, life quality scores of stable COPD patients, and also showed the potential benefits to pulmonary function of COPD patients than original SBS. CONCLUSION: The methodological quality of included trials may limit the conclusions that indicate that modified SBS may have a promising treatment for improving FEV1/FVC and MVV, increasing exercise endurance and life quality scores on stable COPD patients.

A composite material of graphene (G) and polydopamine (PDA) on a copper (Cu) substrate (G/PDA@Cu) was fabricated successfully by sequential immersion deposition in a dopamine solution and an aqueous graphene oxide suspension before annealing. Optimum preparation conditions were explored by the orthogonal experimental method. The morphology and chemical composition of G/PDA@Cu were studied systematically by a series of characterization techniques. The thermal-conductive performance was evaluated by a laser flash thermal analyser. The thermal conductivity of G/PDA@Cu was 519.43 W m-1 K-1, which is ultrahigh and 30.50% higher than that of the Cu substrate. The adhesion force between G/PDA and the Cu substrate was 4.18 mN, which means that G bonds to the Cu substrate tightly. The model simulation also showed that G/PDA@Cu exhibits excellent thermal conductivity, allowing it to play a significant role in the thermal management of advanced electronic chips. The thermal-conductive devices using this material were prepared for practical applications.

The spatial and temporal variations of the seasonal freeze-thaw cycles are important in understanding the ecological and hydrological processes and biogeochemical cycle associated with permafrost degradation caused by climate change, although observational data on the soil hydrothermal dynamics within the active layer of the permafrost region at the central and northern Qinghai-Tibet Plateau (QTP) are extremely scarce. In this study, soil temperature and moisture date from 11 observational sites along the Qinghai-Tibet Highway from 2010 to 2014 were used to analyze the freeze-thaw cycles of the active layer. The results revealed that mean annual ground surface temperature (MAGST) and mean annual temperature at the top of permafrost (TTOP) were the most closely related to the onset dates of soil freezing and thawing. The onset dates of soil freezing from bottom to top did not occur earlier than those from top to bottom. The differences between the onset dates of the two freezing directions and the proportion of bottom-up freezing depth increased with decreasing TTOP. The unfrozen water content of the cooling process was always higher than that of the warming process during the freezing stage. The hysteresis effect of the unfrozen water content could also be observed in the field experiment, and the maximum hysteresis levels occurred at their corresponding soil freezing points. Soil organic matter and soil moisture associated with vegetation cover are essential for water-heat exchanges between atmosphere and permafrost beneath active layer. We suggest that a better protected plant ecosystem, helps preserving the underlying permafrost on the Qinghai-Tibet Plateau.

Stable isotopic tracing has proven to be a useful tool for assessing surface water source dynamics and hydrological connectivity in permafrost regions. This study has investigated the contribution of precipitation to water within the active layer at three long-term observation sites, including Fenghuoshan (FHS), Hoh Xil (KKXL) and Wudaoliang (WDL), by using isotopic tracer technique and two-component mixing model. The results showed that precipitation was the predominant source for water within the active layer, permafrost and ground ice near permafrost table at the three sites. Precipitation in August was the predominant source for water within the active layer at FHS, and precipitation in September was the main source at KKXL and WDL. The variation of isotopic values at different levels indicated that the water sources within the active layer could vary as the depth increases. The evaporation fractionation of water within the active layer at WDL was noticeable at depths of 0-50 cm, and the evaporation intensity decrease gradually from late June to late September. The relationship of isotopic tracing values between precipitation and water within the active layer at depths of 0-50 cm becomes more significant as the amount of the recently-occurring precipitation increases. Moreover, the relatively higher d-excess in precipitation indicates that local recycled moisture has greater contribution to precipitation. The differences of d-excess in most water within the active layer, permafrost and ground ice near permafrost table revealed that there were isotopic fractionation when precipitation supplying to above-mentioned three water bodies. The precipitation event amounted to 8.1 mm at KKXL can exert 49% ±â€¯7.1% and 30.8% ±â€¯3.6% contribution to water within the active layer at depths of 0-10 cm and 10-20 cm, respectively. While the long-period contribution cannot be identified because of the impact of evaporation. The results would provide new insights into the contribution of precipitation to water within the active layer on the QTP, which is also helpful to improve process-based hydrological models in the permafrost regions.

There are several publications related to the soil organic carbon (SOC) on the Qinghai-Tibetan Plateau (QTP). However, most of these reports were from different parts of the plateau with various sampling depth. Here, we present the results from a systematic sampling and analysis of 200 soil pits. Most of the pits were deeper than 2 m from an east-west transect across the plateau. The SOC and total nitrogen (TN) pools of the 148 × 104 km2, the area of the permafrost zone, for the upper 2 m soils calculated from the vegetation map were estimated to be 17.07 Pg (interquartile range: 11.34-25.33 Pg) and 1.72 Pg (interquartile range: 1.08-2.06 Pg), respectively. We also predicted the distribution of land cover types in 2050 and 2070 using decision tree rules and climate scenarios, and then predicted SOC and TN pools of this region. The results suggested that the SOC and TN pools will decrease in the future. The results not only contribute to the carbon and nitrogen storage and stocks in the permafrost regions as a whole but most importantly, to our knowledge of the possible changes of C and N storage on the QTP in the future.

Permafrost degradation can stimulate the decomposition of organic soil matter and cause a large amount of greenhouse gas emissions into the atmosphere. The light fraction organic matter (LFOM) is a labile substrate for microbial decomposition and probably plays an important role in future permafrost carbon cycles. However, little is known about the distribution of LFOM and its relationship with permafrost and environmental factors. Here, we investigated the light fraction carbon (LFC) and nitrogen (LFN) contents and stocks under meadows and wet meadows with different permafrost conditions on the southern Qinghai-Tibetan Plateau. Our results showed that LFC and LFN were mainly distributed in the upper 30cm of soils, and the sites with permafrost had significantly higher contents of LFC and LFN than those from the sites without existing permafrost. The LFC and LFN decreased sharply with depth, suggesting that the soil organic matter (SOM) in this area was highly decomposed in deep soils. Soil moisture and bulk density explained approximately 50% of the variances in LFC and LFN for all the sampling sites, while soil moisture explained approximately 30% of the variance in permafrost sites. Both the C:N ratios and LFC:LFN ratios in the sites with permafrost were higher than those in the sites without permafrost. The results suggested that the permafrost and land cover types are the main factors controlling LFOM content and stock, and that permafrost degradation would lead to a decrease of LFOM and soil C:N ratios, thus accelerating the decomposition of SOM.

Soil nutrient stoichiometry and its environmental controllers play vital roles in understanding soil-plant interaction and nutrient cycling under a changing environment, while they remain poorly understood in alpine grassland due to lack of systematic field investigations. We examined the patterns and controls of soil nutrients stoichiometry for the top 10cm soils across the Tibetan ecosystems. Soil nutrient stoichiometry varied substantially among vegetation types. Alpine swamp meadow had larger topsoil C:N, C:P, N:P, and C:K ratios compared to the alpine meadow, alpine steppe, and alpine desert. In addition, the presence or absence of permafrost did not significantly impact soil nutrient stoichiometry in Tibetan grassland. Moreover, clay and silt contents explained approximately 32.5% of the total variation in soil C:N ratio. Climate, topography, soil properties, and vegetation combined to explain 10.3-13.2% for the stoichiometry of soil C:P, N:P, and C:K. Furthermore, soil C and N were weakly related to P and K in alpine grassland. These results indicated that the nutrient limitation in alpine ecosystem might shifts from N-limited to P-limited or K-limited due to the increase of N deposition and decrease of soil P and K contents under the changing climate conditions and weathering stages. Finally, we suggested that soil moisture and mud content could be good predictors of topsoil nutrient stoichiometry in Tibetan grassland.

OBJECTIVE Therapeutic neovascularization is a promising strategy for treating patients after an ischemic stroke; however, single-factor therapy has limitations. Stromal cell-derived factor 1 (SDF-1) and vascular endothelial growth factor (VEGF) proteins synergistically promote angiogenesis. In this study, the authors assessed the effect of combined gene therapy with VEGF165 and SDF-1 in a rat model of cerebral infarction. METHODS An adenoviral vector expressing VEGF165 and SDF-1 connected via an internal ribosome entry site was constructed (Ad- VEGF165-SDF-1). A rat model of middle cerebral artery occlusion (MCAO) was established; either Ad- VEGF165-SDF-1 or control adenovirus Ad- LacZ was stereotactically microinjected into the lateral ventricle of 80 rats 24 hours after MCAO. Coexpression and distribution of VEGF165 and SDF-1 were examined by reverse-transcription polymerase chain reaction, Western blotting, and immunofluorescence. The neurological severity score of each rat was measured on Days 3, 7, 14, 21, and 28 after MCAO. Angiogenesis and vascular remodeling were evaluated via bromodeoxyuridine and CD34 immunofluorescence labeling. Relative cerebral infarction volumes were determined by T2-weighted MRI and triphenyltetrazolium chloride staining. Cerebral blood flow, relative cerebral blood volume, and relative mean transmit time were assessed using perfusion-weighted MRI. RESULTS The Ad- VEGF165-SDF-1 vector mediated coexpression of VEGF165 and SDF-1 in multiple sites around the ischemic core, including the cortex, corpus striatum, and hippocampal granular layer. Coexpression of VEGF165 and SDF-1 improved neural function, reduced cerebral infarction volume, increased microvascular density and promoted angiogenesis in the ischemic penumbra, and improved cerebral blood flow and perfusion. CONCLUSIONS Combined VEGF165 and SDF-1 gene therapy represents a potential strategy for improving vascular remodeling and recovery of neural function after cerebral infarction.
