RESUMO

The understanding of large biophysical systems at the systems level often depends on a precise knowledge of their microstructure. This is difficult to obtain, especially in vivo, because most imaging methods are either limited in terms of achievable field of view, or make use of penetrating ionizing radiations such as x-rays, in which case the resolution is severely limited by the deposited dose. Here, we describe a new method, x-ray vector radiography (XVR), which yields various types of information about the local orientation, anisotropy and average size of the sample microstructures. We demonstrate the feasibility by showing first experimental XVRs of human vertebra bone samples, giving information on the trabecular structures even with a pixel resolution of half a millimetre, much larger than the structures themselves. This last point is critical for the development of low-dose measurement methods which will allow for in vivo studies and potentially in the future for new medical diagnostics methods of bone metabolic disorder diseases such as osteoporosis.

RESUMO

X-ray computed tomography (CT) has recently received increased attention in the food science community. The aim of this paper is to demonstrate how grating based phase-contrast CT can provide contrast superior to standard absorption based CT. The method of phase-contrast CT is applied to two samples of porcine subcutaneous fat and rind. The additional contrast obtained may be used for quality testing, to investigate variations in fatty acid composition of the fat-fraction, and density variations in the meat-fraction. The possibility of integrating the method into an abattoir environment is discussed.

RESUMO

We demonstrate high-resolution small-angle x-ray scattering computed tomography (SAXS-CT) of soft matter and soft tissue samples. Complete SAXS patterns over extended ranges of momentum transfer are reconstructed spatially resolved from volumes inside an extended sample. Several SAXS standard samples are used to quantitatively validate the method and demonstrate its performance. Further results on biomedical tissue samples (rat brains) are presented that demonstrate the advantages of the method compared to existing biomedical x-ray imaging approaches. Functional areas of the brains as well as tumor morphology are imaged. By providing insights into the structural organization at the nano-level, SAXS-CT complements and extends results obtainable with standard methods such as x-ray absorption tomography and histology.

RESUMO

We introduce a novel x-ray imaging approach that yields information about the local texture of structures smaller than the image pixel resolution inside an object. The approach is based on a recently developed x-ray dark-field imaging technique, using scattering from sub-micron structures in the sample. We show that the method can be used to determine the local angle and degree of orientation of bone, and fibers in a leaf. As the method is based on the use of a conventional x-ray tube we believe that it can have a great impact on medical diagnostics and non-destructive testing applications.

RESUMO

Here we review our recent progress in the field of X-ray dark-field and phase-contrast imaging using a grating interferometer. We describe the basic imaging principles of grating-based phase-contrast and dark-field radiography and present some exemplary results obtained for simple test objects and biological specimens. Furthermore, we discuss how phase-contrast and dark-field radiography can be combined with the concept of computed tomography, and yield highly detailed three-dimensional insights into biomedical sample. Exemplary results obtained with standard X-ray tube sources and highly brilliant synchrotron sources are presented.

RESUMO

We report the first experimental soft-tissue phase-contrast tomography results using a conventional x-ray tube source, with a millimeter-sized focal spot. The setup is based on a Talbot-Lau grating interferometer operated at a mean energy of 28 keV. We present three-dimensional ex vivo images of a chicken heart sample, fixated in formalin. The results clearly demonstrate the advantageous contrast attainable through phase-contrast imaging over conventional attenuation-based approaches.

Heat shock genes respond to moderate heat stress by a wave of transcription. The induction phase is accompanied by the massive eviction of histones, which later reassemble with DNA during the ensuing phase of transcription downregulation. In this article, we identify determinants of this reassembly throughout the heat shock protein 104 gene (HSP104) transcription unit. The results show that, although histone H3 lacking amino acids 4-30 of its N-terminal tail (H3Delta4-30) is normally deposited, reassembly of H3Delta4-40 is obliterated with an accompanying sustained transcription. On mutation of the histone chaperones Spt6p and Spt16p, but not Asf1p, reassociation of H3 with DNA is compromised. However, despite a lasting open chromatin structure, transcription ceases normally in the spt6 mutant. Thus, transcriptional downregulation can be uncoupled from histone redeposition and ongoing transcription is not required to prevent chromatin reassembly.
