RESUMO

Real-time imaging and monitoring of biothiols in living cells are essential for understanding pathophysiological processes. However, the design of the fluorescent probe that has accurate and repeatable real-time monitoring capabilities for these targets is highly challenging. In this study, we prepared a fluorescent sensor, Lc-NBD-Cu(II), which contains a N1, N1, N2-tris-(pyridin-2-ylmethyl) ethane-1,2-diamine as a Cu(II) chelating unit and a 7-nitrobenz-2-oxa-1,3-diazole fluorophore to detect Cysteine (Cys). Emission changes promoted by addition of Cys to this probe are distinctive and correspond to a range of processes including Cys induced loss of Cu(II) from Lc-NBD-Cu(II) to form Lc-NBD, Cu(I) oxidation to reform Cu(II), Cys oxidation to form Cys-Cys, Cu(II) binding to Lc-NBD to reform Lc-NBD-Cu(II), and competitive binding of Cu(II) to Cys-Cys. The study also shows that Lc-NBD-Cu(II) maintains high stability during the sensing process and that it can be utilized over a number of detection cycles. Finally, the findings show that Lc-NBD-Cu(II) can be utilized to repetitively sense Cys in living HeLa cells.

Biomarkers are essential in biology, physiology, and pharmacology; thus, their detection is of extensive importance. Fluorescent probes provide effective tools for detecting biomarkers exactly. Excited state intramolecular proton transfer (ESIPT), one of the significant photophysical processes that possesses specific photoisomerization between Keto and Enol forms, can effectively avoid annoying interference from the background with a large Stokes shift. Hence, ESIPT is an excellent choice for biomarker monitoring. Based on the ESIPT process, abundant probes were designed and synthesized using three major design methods. In this review, we conclude probes for 14 kinds of biomarkers based on ESIPT explored in the past five years, summarize these general design methods, and highlight their application for biomarker detection in vitro or in vivo.

Peak runoff in streams and rivers of the western United States is strongly influenced by melting of accumulated mountain snowpack. A significant decline in this resource has a direct connection to streamflow, with substantial economic and societal impacts. Observations and reanalyses indicate that between the 1980s and 2000s, there was a 10-20% loss in the annual maximum amount of water contained in the region's snowpack. Here we show that this loss is consistent with results from a large ensemble of climate simulations forced with natural and anthropogenic changes, but is inconsistent with simulations forced by natural changes alone. A further loss of up to 60% is projected within the next 30 years. Uncertainties in loss estimates depend on the size and the rate of response to continued anthropogenic forcing and the magnitude and phasing of internal decadal variability. The projected losses have serious implications for the hydropower, municipal and agricultural sectors in the region.

Bone marrow-derived mesenchymal stem cells (BMMSCs) from the patients suffering from age-related osteoporosis were found to have numerous degeneration, such as decreased growth rate, impaired capacity of differentiating into local tissue, and repressed telomerase activity. However, it is not clear whether post-menopausal osteoporotic bone is either subject to such decline in cellular function. In the present study, bone marrow cells were harvested from ovariectomized (OVX) and Sham rats and cultured in vitro at 3 months post-surgery. MTT assay indicated that the proliferation potential of (OVX)BMMSCs was always higher than that of (Sham)BMMSCs, no matter cultured in basic, osteoblastic or adipogenic medium. Alkaline phosphatase activity assay, Alizarin red S staining, Oil red O staining and real-time RT-PCR analysis further demonstrated that bilateral ovariectomization positively influenced the osteoblastic and adipogenic differentiation potential of BMMSCs, this action may be partly mediated through up-regulation of osteoblastic special markers core binding factor a1, collagen type I and low-density lipoprotein receptor-related protein 5, as well as adipogenic special markers peroxisome proliferators activated receptor gamma, CCAAT/enhancer binding protein alpha and adipocyte lipid-binding protein 2. These results may hold great promise for using post-menopausal osteoporotic bone as an attractive autologous marrow source for tissue engineering and cell-based therapies.

OBJECTIVE: To observe the expression of angiopoietins in oral squamous cell carcinoma and relationship between the expression of angiopoietins and pathologic classification. METHODS: The expression of Ang-1 and Ang-2 protein in samples from 42 oral squamous cell carcinoma and 16 oral normal mucosa were detected by immunohistochemical technique. RESULTS: The expression of Ang-1 and Ang-2 was found in both oral squamous cell carcinoma and normal control. It was found Ang-1 expression alike in oral squamous cell carcinoma and control (P > 0.05). Ang-2 expressed at low level in control while strongly positive in oral squamous cell caroinoma and the level of Ang-2 expression in oral squamous cell caroinoma was related to pathologic classification (P < 0.05). CONCLUSION: Ang-2 expression in oral squamous cell carcinoma is related to angiogenesis and pathological classification, which is probably involved in angiogenesis regulation, promotes the development and metastasis of oral squamous cell carcinoma.
