RESUMO

The herbal formulation Sinpo-Tang (SPT) has long been used for various allergic diseases. We investigated the effect of SPT on the mast cell-mediated anaphylactic reactions in vivo and in vitro murine models. SPT dose dependently inhibited compound 48/80-induced ear swelling response and histamine release. SPT (0.001-0.1g/kg) significantly inhibited passive cutaneous anaphylaxis (PCA). The mast cell number was markedly decreased at the SPT-treated PCA site. In addition, SPT decreased intracellular calcium levels of activated mast cells. These results suggest that SPT inhibit the anaphylactic degranulation of mast cells through decrease of the intracellular calcium level.

Dodutang has been used for treatment of various allergic inflammation diseases in Korea. However, it is still unclear how Dodutang prevents these diseases in experimental animal models. Mast cells play an important roles in allergic and other inflammatory reactions by producing a spectrum of powerful mediators including preformed and de novo synthesized cytokines. In this study, we investigated the effect of Dodutang on mast cell-mediated allergic and inflammatory reactions. Dodutang (0.001-5 g/L) significantly inhibited the histamine release from rat peritoneal mast cells activated by compound 48/80. Dodutang (0.001-5 g/kg) dose-dependantly inhibited the passive cutaneous anaphylaxis reaction activated by anti-dinitrophenyl (DNP) IgE in a rat model, especially, by 78.96% at the concentration of 5 g/kg. In addition, Dodutang potently inhibited the secretion of tumor necrosis factor-alpha (TNF-alpha), interleukin (IL)-1beta in phorbol 12-myristate 13-acetate plus calcium ionophore A23187-stimulated human mast cells. However, IL-6 secretion was enhanced at the same conditions. Dodutang also inhibited the main inflammatory cytokine TNF-alpha protein expression in human mast cells. These results provide evidences that Dodutang may be beneficial in the treatment of acute and chronic allergic diseases.

Yangkyuk-Sanhwa-Tang (YS-Tang), a specific prescription composed of nine herbal mixtures, has been developed as a formula for the Soyangin cerebral infarction (CI) patients according to Sasang constitutional philosophy. However, the mechanisms by which this formula affects CI remain unknown. This study revealed changes in cytokine production in the acute stage of Soyangin constitution CI patients after YS-Tang administration. Clinical signs (vertigo, headache and slurred speech) of CI disappeared significantly in about 2 weeks after oral administration of YS-Tang (P < .05). The mean interleukin (IL)-2 plasma levels were lower by 15% in the patients with CI than in the normal groups, whereas the mean TNF-alpha, IL-4, IL-6 and IgE levels were significantly higher in the patients (P < .01). There were no significant differences in interferon-gamma (IFN-gamma) levels between the groups. Serum IFN-gamma and IL-2 levels were elevated significantly (P < .01) in the patients with CI by YS-Tang administration. Significant reduced plasma levels (P < .01) of TNF-alpha, IL-4, IL-6 and IgE were observed in the patients treated with YS-Tang. During the period of YS-Tang administration, there were no other adverse effects. The data indicate that YS-Tang has an enhancing effect on antiinflammatory cytokines and an inhibitory effect on inflammatory cytokines. These results may implicate a good CI treatment effect of YS-tang and that its action may be due to regulation of cytokine production.
