We aimed to determine the prevalence of Demodex spp. and bacterial infection in patients with blepharitis and also to investigate the relationship between culturable microorganisms and Demodex spp. in this study. The study included patients diagnosed with blepharitis (n = 128) and volunteers without ocular problems (n = 113). Eyelash sampling was performed by epilating eight lashes, which were then tested for Demodex spp. using a light microscope. The examination consisted of assessing the patient's vision with and without ocular correction and tonus in both eyes and a careful examination of the anterior segment of both eyes. Bacterial identification was performed based on morphological, physiological, and biochemical methods. The prevalence of Demodex spp. was 8.0% in patients from the control group and all patients with blepharitis. Isolated forms of Demodex spp. were detected in all infested patients in the control group and in 58% of patients with blepharitis. A total of 35% of patients with blepharitis had from three to nine forms of Demodex spp., and 7% of patients with blepharitis had more than 10 mites in every field of vision. We found a statistically significant relationship between Demodex spp. infestation and the occurrence of eye dryness and sensations of burning and tearing, redness of the conjunctiva, feeling of a foreign body, loss of eyelashes, Meibomian gland dysfunction, and cylindrical dandruff. There were statistically significant relationships between Demodex sp. infestation and the presence of hyperopia, Meibomian cysts, chronic eyelid inflammation, and the use of eyeglasses. There was also a statistically significant relationship between the occurrence of Demodex spp. and seborrheic dermatitis and diabetes mellitus. Culturable microorganisms of the ocular surface were found in 8.7% of participants who were uninfested and in all patients infested with D. folliculorum. We isolated Staphylococcus aureus, Acinetobacter baumannii, Streptococcus pneumoniae, Klebsiella oxytoca, and Bacillus spp. from the conjunctival sac only in patients infested with D. folliculorum. This indicates an increased probability of colonization by pathogenic bacteria in patients with demodicosis. Therefore, patients infested with D. folliculorum should undergo a microbiological examination of conjunctival swabs.

The aim of the study was to investigate the relationships between the concentrations of calcium (Ca), magnesium (Mg), zinc (Zn), and lead (Pb) in cartilage, anterior cruciate ligament, and meniscus samples obtained following knee joint surgery in patients with osteoarthritis in northwestern Poland.‬ Furthermore, we examined the relationships between the concentrations of these metals in the studied parts of the knee joint and the influences of gender, age, BMI and hypertension. We found significantly higher concentrations of Ca, Mg, and Zn in the cartilage of men than in women and a significantly higher Pb concentration in the meniscus of the men. We also found a higher concentration of Pb in the cartilage of patients over 65 years of age.‬ There were no differences in the concentrations of the studied metals between patients with and without hypertension.‬ There was no relationship between Ca, Mg, Zn, and Pb levels in analyzed materials and BMI. Furthermore, we noted some new interactions between metals in the studied structures of the knee joint. The results reported in the study shows the influence of age, gender and BMI on the Ca, Mg, Zn and Pb in the studied structures of the knee joint.

Amoebas from the genus Acanthamoeba are cosmopolitan organisms, which can exist as free-living organisms and as parasites within host tissue. Acanthamoeba infection present a serious risk to human health and are characterized by high mortality, especially in immunocompromised individuals. These protozoa are the etiological factors of granulomatous amoebic encephalitis (GAE) and Acanthamoeba keratitis (AK). They can also live in the lungs, adrenals glands, nose, throat, and bones of the host. Furthermore, the amoebas can be vectors of pathogenic bacteria. Acanthamoeba infection caused is a serious clinical problem mainly due to limited progress in diagnostics and treatment of this infection, which is associated with insufficient knowledge of pathogenesis, pathophysiology and the host immune response against Acanthamoeba antigens. This review study presents the biology of Acanthamoeba sp. as well as pathogenicity, diagnostics, and treatment of amoebas infections. It also presents data, including experimental results, concerning pathogenic properties and the host's immunology response against Acanthamoeba sp.

Due the long-term nature of joint tissue remodeling processes, knee structures including meniscus and anterior cruciate ligament (ACL) can be a model for studying the bioaccumulation of mercury (Hg) and selenium (Se). The accumulation and retention of Hg in human tissues can have a negative effect on the proper functioning of homeostasis-affecting organisms. A factor of chronic poisoning with Hg forms is probably the Se:Hg ratio in tissues. Se:Hg molar ratios below one may increase Hg toxicity potentials, while molar ratios that approach or exceed one effectively may protect against Hg toxicity. Therefore, the aim of the study was to determine total mercury (THg), Se, and Se:THg molar ratios in the cartilage, meniscus and ACL of patients with osteoarthritis (OA) from northwestern Poland. In all studied samples (n=95), we observed higher Se than THg concentration. Taking into consideration the biological factors, we found significantly higher THg levels in the cartilage of women, patients under 65 years of age, patients without hypertension and in the ACL of patients with spinal degenerative disease. We found higher Se levels in the meniscus in women than in men. In all studied parts of the knee joint, we found the Se:THg molar ratio higher than one, which suggests that the joint forming structures are not much exposed to THg. Moreover the results reported here may provide a basis for establishing reference values for the meniscus and ACL in patients with OA who had undergone knee replacement surgery.

Heavy metals, including cadmium (Cd), lead (Pb) and mercury (Hg) act as nephrotoxic agents, particularly in the renal cortex. The aim of the study was to determine the concentrations of Cd, Pb and Hg in kidneys removed from patients due to lesions of various etiologies and from patients after the rejection of transplanted kidneys. Additionally, we determined the influence of selected biological and environmental factors on the concentrations of toxic metals. The study material consisted of kidneys with tumor lesions (n = 27), without tumors (n = 7) and its extracted grafts (n = 10) obtained from patients belongs to the north-western areas of Poland. The determined metal concentrations in the renal cortex and medulla may be arranged in the following descending order: Cd > Pb > Hg. The highest concentrations of Cd and Hg were found in the cortex, while the maximum content Pb was observed in the medulla. Significant correlations were found in the concentrations of the same metals between cortex and medulla and between Pb and Hg in the renal medulla. Pb content was higher in the renal medulla of men than in the cortex of the elderly (above 60 years of age). The highest concentrations of Pb and Hg were found in the cortex and medulla, of the kidneys had not neoplastic changes, and lower content of these metals were found in the extracted kidney grafts. In summary, renal grafts accumulate less heavy metals than cancerous kidneys, what could have been caused by immunosuppressors taken by the graft recipients. Moreover, sex, age and smoking are key factors responsible for xenobiotics concentrations.

The aim of this study was to investigate the impact of environmental factors on the concentrations of vanadium (V), chromium (Cr), and calcium (Ca) and to examine the synergistic or antagonistic relationships between these metals, in cartilage (C), cortical bone (CB), and spongy bone (SB) samples obtained following hip joint surgery on patients with osteoarthritis in NW Poland. We found significantly higher concentrations of V and Cr in spongy bone in patients who consumed game meat and also those with prosthetic implants. Chromium levels were significantly lower in patients with kidney diseases. The greatest positive correlations were found between spongy bone V and (i) the amount of consumed beer and (ii) seafood diet. Correlation analysis also showed a significant correlation between Cr levels and seafood diet. To a certain extent these results indicate that the concentrations of V, Cr, and Ca in the human hip joint tissues are connected with occupational exposure, kidney diseases, diet containing game meat, sea food, beer, and the presence of implants. Furthermore, we noted new types of interactions in specific parts of the femoral head. Vanadium may contribute to the lower bone Ca levels, especially in the external parts (cartilage and cortical bone).

The aim of this study was to determine the effect of an extremely low-frequency magnetic field (ELFMF) on the production of liver fluke larvae in a parasite-host system: Fasciola hepatica--Galba truncatula. Both F. hepatica eggs and F. hepatica-infected snails were exposed to an ELFMF (50 Hz, 2.0 mT) for 14 days and 36 days, respectively. F. hepatica-infected snails were divided into 4 groups, 10 specimens each. The snails of groups I and II were infected with F. hepatica larvae--miracidia obtained from control cultures, while the snails of groups III and IV were infected with miracidia reared from eggs that had been incubated in an ELFMF. After infection, the snails of groups II and IV were placed in an ELFMF, while those of groups I (control) and III were housed outside the ELFMF. At 36 days post-infection (dpi) there were no statistically significant differences between the number of F. hepatica larvae--cercariae and metacercariae, obtained from G. truncatula snails in the control group (group I) and the snail groups exposed to ELFMF (groups II, III and IV). However, a statistically significant difference between the average number of F. hepatica larvae in snail groups III and IV may indicate that the duration of exposure to ELFMF, i.e. embryogenesis period vs. the entire larval development, played a role in the production of F. hepatica larvae, and resulted in a reduction of their number.

Beyond 30 years of age, men experience a decline in the production of testosterone, yet only a few develop late-onset hypogonadism. This study was designed to determine the relationship between blood concentrations of metals, macro- and micronutrients and age-related testosterone deficiency and associated hormonal changes in aging men. The research involved 313 men aged 50-75 years. We used ELISA to determine the concentrations of total testosterone (TT), free testosterone (FT), estradiol (E2), dehydroepiandrosterone sulfate (DHEAS) and sex hormone-binding globulin (SHBG). We calculated free androgen index (FAI). With the use of emission spectrometry in inductively coupled argon plasma, we determined the whole-blood concentrations of lead (Pb), cadmium (Cd), mercury (Hg), arsenic (As) and tungsten (W), as well as serum concentrations of magnesium (Mg), iron (Fe), calcium (Ca), copper (Cu), zinc (Zn), selenium (Se), chromium (Cr), manganese (Mn) and molybdenum (Mo). The study showed no relationship between TT and FT and the concentrations of metals. Men with TT deficiency had significantly lower concentrations of Mg and Fe and increased Mn. Men with FT deficiency had higher W and Cr levels and lower Fe. Assessing the correlation between the concentrations of hormones, SHBG and FAI, and the concentration of metals and macro- and microelements in the blood of the men, we found positive correlations between the concentrations of TT-Mg, TT-Fe, TT-Mo, FT-Fe, E2-As, SHBG-Mn, FAI-W, FAI-As, FAI-Zn and FAI-Ca, and negative correlations between the concentrations of TT-Mn, FT-Cd, FT-Cr, E2-Hg, E2-Cr, SHBG-W, SHBG-As, SHBG-Zn, SHBG-Ca, FAI-Pb and FAI-Mn. Positive correlations between As and E2 and between As and FAI may suggest a lack of association between this metal and hypogonadism in people not exposed to excess As levels. Our research indicates a positive relationship between the concentrations of Mg, Fe and Zn and endocrine system in aging men, in contrast to Mn and Cr. Toxic metals (Cd, Pb) seemed to negatively affect the level of bioavailable testosterone. In persons not exposed to As, As does not contribute late-onset hypogonadism. Heavy metals (Pb, Cd, Hg and W) may contribute to a lower concentration of DHEAS. The role of W in men with LOH was found to be ambiguous, as on the one hand its concentration was higher in men with FT deficiency, and on the other hand it positively correlated with FAI, which in turn indirectly indicates testosterone availability. Copper and selenium do not seem to play any significant role in the occurrence of TT deficiency in aging men.

Toll-like receptors (TLRs) play a fundamental role in the rapid activation of innate immune responses to a variety of pathogen-associated molecular patterns (PAMPs). In a previous study we observed an increase in the level of expression of TLR2 and TLR4 mRNA in the jejunum and colon during experimental hymenolepidosis in rats. In this study, we performed a quantitative real-time polymerase chain reaction (qRT-PCR), Western blot analysis and immunohistochemical staining of TLR3 and TLR9 receptors during experimental hymenolepidosis in rats. The levels of mRNA and protein expression of TLR3 and TLR9 in the jejunum had increased at 16 days post Hymenolepis diminuta infection (dpi) in the case of TLR3 and at 16 and 25 dpi in the case of TLR9. In the colon the expression of TLR3 and TLR9 had increased at 16, 25 and 40 dpi. The results of the immunohistochemical reactions showed that H. diminuta infected rats (16, 25, 40 and 60 dpi) exhibited changes in TLR3 and TLR9 localization and intensity in the epithelial cells of the jejunum and colon. The changes in the level of TLR3 and TLR9 expression may confirm involvement of the innate immune system in the pathomechanism of hymenolepidosis.

Assessment of exposure to fluoride (F(-)) is increasingly focused on mineralized tissues, mainly bones. Their periodic growth and continuous reconstruction make them a good material for studying long-term F(-) accumulation. In this study, F(-)concentrations were determined in the bones of foxes and raccoon dogs from north-western Poland and relationships between bone F(-) and the age categories of the animals were attempted to be identified. Bone samples were collected from femurs of 32 foxes (15 males and 17 females) and 18 raccoon dogs (10 males and 8 females) from polluted, medium-polluted, and unpolluted by F(-) areas. Bone F(-) was determined by potentiometric method, and results were expressed per dry weight (dw); they ranged from 176 to 3,668 mg/kg dw in foxes and from 84 to 1,190 mg/kg dw in raccoon dogs. Foxes from north-western Poland accumulated much more F(-) in their bones than raccoon dogs. Our study shows that the assessment of hazards created by industrial emitters can be conducted conveniently by the measurements of fluorine content in hard tissues of wild animals. Due to availability of such type of material for studies, it seems that the analysis of fluoride content in bones can be a good tool in the development of ecotoxicology.

This is the first report on mercury (Hg) levels in the liver, kidney, skeletal muscle, and brain of raccoon in Europe. It studied Hg concentration in 24 raccoons from the Warta Mouth National Park, northwestern Poland by atomic absorption spectroscopy (AAS). The highest total Hg concentrations in the raccoon were found in the liver (maximum, 18.45 mg/kg dry weight), while the lowest in the brain (maximum, 0.49 mg/kg dw). In adult raccoons, Hg concentrations in the liver, kidney, and brain were higher than in immature individuals (p<0.001), while similar in skeletal muscle in both age groups. Our results are consistent with studies by other authors conducted in North America in areas with similar environmental conditions.

Although the relationship between mercury (Hg) and selenium (Se) has been studied in wild birds in areas with sufficient or excessive Se levels, little is known about this relationship in areas where the supply of Se is limited. As Hg detoxification is based on the production of biologically inactive Hg-Se complexes, the aim of this study was to investigate the relationships between the concentrations of total mercury (THg), methylmercury (MeHg), inorganic mercury (InHg=THg-MeHg), percent MeHg of THg, Se and molar ratios (THg:Se, MeHg:Se, InHg:Se) in the breast muscle (n=16) of the piscivorous common mergansers (Mergus merganser) from a Se-deficient and moderately Hg-polluted area in Poland. Mergansers were divided into two groups differing in condition (A-very good condition; C-moderate condition). Concentrations of THg, MeHg and Se were determined by atomic absorption spectrometry, modified gas chromatography atomic fluorescence spectroscopy, and spectrofluorometric methods, respectively. In all studied mergansers, mean concentrations of THg, MeHg, InHg, and Se in muscle were 2.63, 1.92, 0.46, and 0.54µgg(-1)dw, respectively. THg and MeHg concentrations in the muscle of group A mergansers were greater than in group C. The ratio of THg:Se was higher in group A than in group C (2.32 vs 1.36; p<0.01), as well as the molar ratio of MeHg:Se (A vs C: 1.98 vs 1.03; p<0.05). Comparisons between mergansers from Poland and Canada showed similar THg and percent MeHg in the muscle, but mergansers from Poland had several times lower Se levels and higher THg:Se ratios (>2.0) than the Canadian mergansers and other European and North American waterbirds. We found statistically significant positive correlations (MeHg-THg, percent MeHg/THg-MeHg, percent InHg/THg-InHg, THg:Se-THg, MeHg:Se-THg, THg:Se-MeHg, MeHg:Se-MeHg, InHg:Se-InHg, MeHg:Se-THg:Se) and some negative correlations (percent InHg/THg-MeHg, percent MeHg/THg-InHg, THg:Se-Se, MeHg:Se-Se). As THg and percent MeHg in the studied mergansers were similar to populations living in non-Se-deficient areas, it is likely that different mechanisms of muscle Hg detoxification have evolved in mergansers populations living in Se-deficient areas.

The subject of the present study was the piscivorous common mergansers (Mergus merganser). The total mercury (THg), methylmercury (MeHg), selenium (Se) inorganic mercury (InHg; THg - MeHg), percentage of THg that is MeHg (%MeHg), molar ratios (THg:Se, MeHg:Se, InHg:Se), and their mutual relations in livers and kidneys were determined in ducks from an Se-deficient area in Poland. The authors verified a hypothesis that, as a result of living in an Se-deficient region, mergansers from Poland should have higher THg:Se ratios than other waterbirds with similar THg tissue levels. Although a comparison of healthy mergansers from Poland and Canada showed similar THg tissue contents, the group in the present study had a few times lower Se levels and higher THg:Se ratios (overall means >1.7 in both livers and kidneys in all studied individuals) than the Canadian group and other European and North American waterbirds. The authors found significant correlations between various relations, including MeHg-THg, InHg-THg, Se-THg, %MeHg-THg, InHg/%THg-THg, %MeHg/THg-InHg, %InHg/THg-InHg, MeHg:Se-THg:Se, InHg:Se-THg:Se, InHg:Se-MeHg:Se in liver and InHg-THg, Se-THg, Se-InHg, %MeHg-THg, %MeHg-InHg, %InHg/THg-InHg, THg:Se-THg, InHg:Se-THg, MeHg:Se-MeHg, THg:Se-InHg, InHg:Se-InHg, and InHg:Se-THg:Se in kidney. It is likely that the main factor responsible for the high value of THG:Se ratio (>1) in mergansers from Poland is Se deficiency in central and northern Europe. Therefore, this element is unlikely to participate in the detoxification of Hg in these birds.

The aims of the study were to determine manganese (Mn), iron (Fe) and strontium (Sr) concentrations in fox bone samples from north-western Poland and to examine the relationships between the bone Mn, Fe and Sr concentrations and the sex and age of the foxes. In the studied samples of fox cartilage, cartilage with adjacent compact bone, compact bone and spongy bone, the concentrations of the analysed metals had the following descending order: Fe > Sr > Mn. The only exception was in compact bone, in which the concentrations were arranged in the order Sr > Fe > Mn. Manganese concentrations were significantly higher in cartilage, compact bone and cartilage with compact bone than in spongy bone. Iron concentrations were higher in cartilage and spongy bone compared with compact bone. Strontium concentrations were greater in compact bone than in cartilage and spongy bone. The manganese, iron and strontium concentrations in the same type of bone material in many cases correlated with each other, with the strongest correlation (r > 0.70) between Mn and Fe in almost all types of samples. In addition, concentrations of the same metals in different bone materials were closely correlated for Mn and Fe in cartilage and cartilage with adjacent compact bone, and for Sr in compact bone and cartilage with compact bone. In the fox from NW Poland, there were no statistically significant differences in Mn, Fe and Sr in any of the types of bone material between the sexes and immature and adult foxes.

The aim of this study was to assess the impact of environmental factors: cigarette smoking, dental amalgam fillings, eating habits and osteoporosis, on the concentrations of copper (Cu), zinc (Zn), lead (Pb), cadmium (Cd) and mercury (Hg) in the bone of hip joint in patients in the Orthopedics Clinic in Szczecin, Poland. Amalgam dental fillings had an effect on the Cu concentration in the cartilage with the adjacent compact bone, and on Hg concentration in the spongy bone. The highest concentrations of Cu and Hg were found in people with multiple amalgam fillings. Smoking appeared to influence Pb concentration in the cartilage with the adjacent compact bone, and Cd concentration in the spongy bone. Increased Pb was detected in smokers, and increased Cd in non-smokers. Diets rich in fish and seafood correlated with Cd concentration in the cartilage with adjacent compact bone. The greatest concentration was in people who ate fish or seafood at least once a month.

PURPOSE: To examine the proportion of Demodex folliculorum and D. brevis in the eyelashes of healthy and immunocompromised patients. METHODS: Eyelashes were taken from 95 patients at the Pomeranian Medical University, Clinic of Haematology, and from 1091 healthy controls. RESULTS: Demodex folliculorum and D. brevis were found in 22.9% of controls and in 20% of immunocompromised patients. In neither the control group nor the immunocompromised patients did we observe a correlation between the proportion of symptoms of blepharitis and the presence of Demodex spp. - symptoms including burning sensations, loss of eyelashes, sensitivity to light, smoke and dust, dacryorrhea (excessive flow of tears), and foreign body eye sensation. A correlation between itching eyes and Demodex spp. infection was found only in the control group. A correlation between the proportion of cylindrical dandruff in eyelashes and the occurrence of Demodex spp was found in both the control group and immunocompromised patients. CONCLUSION: Proportions of D. folliculorum and D. brevis, and also symptoms of blepharitis were similar in the control group and immunocompromised patients. Immunosuppression did not seem to increase the rate of Demodex spp. infection.

The aim of this study was to compare zinc, copper, lead, cadmium, and mercury concentrations in the bones of long-living mammals-humans (Homo sapiens) and Canidae (dogs Canis familiaris and foxes Vulpes vulpes) from northwestern Poland and to determine the usefulness of Canidae as bioindicators of environmental exposure to metals in humans. Zinc concentrations in cartilage with adjacent compact bone and in spongy bone were highest in foxes (∼120 mg/kg dry weight (dw)) and lowest in dogs (80 mg/kg dw). Copper concentrations in cartilage with adjacent compact bone were greatest in foxes (1.17 mg/kg dw) and smallest in humans (∼0.8 mg/kg dw), while in spongy bone they were greatest in dogs (0.76 mg/kg dw) and lowest in foxes (0.45 mg/kg dw). Lead concentrations in both analyzed materials were highest in dogs (>3 mg/kg dw) and lowest in humans (>0.6 mg/kg dw). Cadmium concentration, also in both the analyzed materials, were highest in foxes (>0.15 mg/kg dw) and lowest in humans (>0.04 mg/kg dw). Mercury concentration in bones was low and did not exceed 0.004 mg/kg dw in all the examined species. The concentrations of essential metals in the bones of the examined long-living mammals were similar. The different concentrations of toxic metals were due to environmental factors. As bone tissues are used in the assessment of the long-term effects of environmental exposure to heavy metals on the human body, ecotoxicological studies on the bones of domesticated and wild long-living mammals, including Canidae, may constitute a significant supplement to this research.

The aim of this study was to determine the concentration of mercury (Hg) in liver (L), kidney (K), breast muscle [BM (musculus pectoralis major)], breast feathers (BF), and stomach contents (SC) of mallard (Anas platyrhynchos L. 1758). Among the edible parts of mallard, the greatest concentrations of Hg were observed in K and L, although they did not exceed 1.5 mg/kg dry weight (dw). Average concentrations in K, L, and BM were 0.27, 0.25, and 0.13 mg/kg dw, respectively. Significant correlations were observed between Hg concentrations in BM and K and in BM and L (r s = 0.92) as well as between Hg concentrations in these tissues and BF. In addition, we found significant correlations between Hg concentrations in SC and BM (r s = 0.72) and in L and K (r s = 0.55). In conclusion, mallard exhibits a measurable response to environmental Hg pollution and meets the requirements of a bioindicator.

In this study we determined the levels of trace elements (zinc, copper, lead, cadmium and mercury) in three layers of bones of the hip joint (cartilage, compact bone and spongy bone) of 30 red foxes (Vulpes vulpes) from north-western Poland. Concentrations of Cu, Zn, Pb and Cd were determined by atomic absorption spectrophotometry (ICP-AES) in inductively coupled argon plasma using a Perkin-Elmer Optima 2000 DV. Determination of Hg concentration was performed by atomic absorption spectroscopy. In cartilage, compact bone and spongy bone samples from the red fox, median concentrations of the metals studied could be arranged in the following descending series: Zn > Cu > Pb > Cd > Hg, the values ranging from 142 to 0.002 mg/kg dw. There was a significant difference in Cu concentrations, among all the materials analyzed, with much more Cu found in spongy bone than in compact bone. Significant differences were also noted in the case of Hg concentrations in cartilage with compact bone and the spongy bone, and between concentrations of this metal in compact bone and spongy bone. In males, the concentration of Hg in spongy bone was greater than in females. Younger foxes had a higher concentration of this metal in cartilage than adults. The strongest synergistic relationships were observed in spongy bone between the Zn and Cu, Zn and Cd, as well as between Cu and Cd. Statistically significant antagonistic relationships were detected between zinc and lead in compact bone. In addition to monitoring studies conducted on the abiotic environment, an urgent need exists for long-term monitoring of concentrations of heavy metals with long-term effects on living organisms. An important addition is provided by biomonitoring studies on domesticated and free-living mammals, including Canidae.
