RESUMO

Radiotherapy is a widely used cancer treatment that utilizes powerful radiation to destroy cancer cells and shrink tumors. While radiation can be beneficial, it can also harm the healthy tissues surrounding the tumor. Recent research indicates that the microbiota, the collection of microorganisms in our body, may play a role in influencing the effectiveness and side effects of radiation therapy. Studies have shown that specific species of bacteria living in the stomach can influence the immune system's response to radiation, potentially increasing the effectiveness of treatment. Additionally, the microbiota may contribute to adverse effects like radiation-induced diarrhea. A potential strategy to enhance radiotherapy outcomes and capitalize on the microbiome involves using probiotics. Probiotics are living microorganisms that offer health benefits when consumed in sufficient quantities. Several studies have indicated that probiotics have the potential to alter the composition of the gut microbiota, resulting in an enhanced immune response to radiation therapy and consequently improving the efficacy of the treatment. It is important to note that radiation can disrupt the natural balance of gut bacteria, resulting in increased intestinal permeability and inflammatory conditions. These disruptions can lead to adverse effects such as diarrhea and damage to the intestinal lining. The emerging field of radiotherapy microbiome research offers a promising avenue for optimizing cancer treatment outcomes. This paper aims to provide an overview of the human microbiome and its role in augmenting radiation effectiveness while minimizing damage.

Cycloicaritin (CICT), a bioactive flavonoid derived from the genus Epimedium, exhibits a variety of beneficial biological activities, including promising anticancer effects. However, its poor oral bioavailability is attributed to its extremely low aqueous solubility and rapid elimination via phase II conjugative metabolism. To overcome these limitations, we designed and synthesized a series of carbamate-bridged prodrugs, protecting the hydroxyl group at the 3-position of cycloicaritin by binding with the N-terminus of a natural amino acid. The optimal prodrug 4b demonstrated a significant increase in aqueous solubility as compared to CICT, as well as improved stability in phase II metabolism, while allowing for a rapid release of CICT in the blood upon gastrointestinal absorption. The prodrug 4b also facilitated oral absorption through organic anion-transporting polypeptide 2B1-mediated transport and exhibited moderate cytotoxicity. Importantly, the prodrug enhanced the oral bioavailability of CICT and displayed dose-dependent antitumor activity with superior safety. In summary, the prodrug 4b is a novel potential antitumor drug candidate, and the carbamate-bridged amino acid prodrug approach is a promising strategy for the oral delivery of CICT.

An ultra-performance liquid chromatography-tandem mass spectrometry was developed to assay the concentration for the quantification of cycloicaritin and its carbamate prodrug (3-O-L-valyl carbamate prodrug of cycloicaritin) in the plasma of Sprague-Dawley rats. Analytes were separated on an Acquity UPLC BEH C18 (2.1 × 50 mm, 1.7 µm) after liquid-liquid extraction with methyl tert-butyl ether. Acetonitrile and water containing 0.1 % formic acid were the mobile phases of the method. Using electrospray ionization in the positive ion mode, the method was performed with a total run time of 2.60 min. The response of the experiments was linear over the concentration ranges from 1 to 250 ng/mL for cycloicaritin and 1-250 ng/mL for prodrug. The intra- and inter-day precision and accuracy were within the recommended limits of the FDA. The matrix effect that we observed met the criteria. The method was successfully applied to the pharmacokinetics of cycloicaritin and its carbamate prodrug in Sprague-Dawley rats.

Student mental health is an integral part of the fight against disease, and health was an evolving concern during the spread of COVID-19. As the COVID-19 pandemic unfolds, physical distancing and social restrictions were introduced, and because of these, it was found a great impact on students' lives and their mental health. Adolescent mental health is focused on prevention, early detection, therapeutic innovation, and service development. In these circumstances, service providers can expand telemedicine Digital services that may help provide future mental health services to young people, particularly students. This study collects and analyzes data from students on the impact of these new online learning techniques, and by collecting and analyzing the challenges and issues faced by college students during the COVID-19 pandemic. The questionnaire was based on and extended the European Students' Union Survey and targeted higher education students concerning what student life looked like during the pandemic, including teaching and learning, their social contacts, habits/routines, as well as how they were coping with the situation emotionally and financially. The case of universities and distance learning education units in higher education during the COVID-19 pandemic process. This article highlighted the impact of COVID-19 on students of all ages and their time schedules such as online learning and reflection, study environment, sleep habits, routines, and outcomes. It was found that the students generally require greater self-discipline and motivation to complete online classes. It was also reflected that the pandemic adversely affected student mental health, leading to an increased prevalence of Major Depressive Disorder (MDD) and Generalized Anxiety Disorder (GAD). This may have a significant impact on their mental health issues such as frustration, stress, and sadness. This will significantly lead to cooperation between various local authorities and the private sector.

RESUMO

The magnetic and macro/mesoporous bioactive glasses scaffolds are synthesised successfully by the combination of coral and P123 as co-templates through an evaporation-induced self-assembly process. The prepared material can induce the precipitation of hydroxyapatite layers on their surface in SBF only within 12 h. At the same time, the material exhibited excellent super-paramagnetic and mechanical property. Furthermore, the biocompatible assessment confirmed that the obtained material presented the good biocompatibility and the enhanced adherence of HeLa cells. Herein, the novel materials are expected to have potential application for bone tissue engineering.

OBJECTIVE: To investigate the protective effects and its mechanisms of total epimedium flavonoids (TEF) against Quindinince acid (QA)-induced toxicity in SH-SY5Y cells. METHODS: The MT assay was used to detect the toxicity of QA and the cell viability of pretreatment of TEF and co-incubation with QA in SH-SY5Y cells; Cell apoptosis was observed by Hochest33258 staining; SOD and GSH-Px activities as well as MDA content were measured by colorimetric method; The mitochondrial membrane potential (delta psi m) and intracellular free calcium concentration ([Ca2+]i) were monitored by FCM. RESULTS: TEF increased the cell viability of SH-SY5Y cells, reduced apoptosis cells significantly, improved SOD and GSH-Px activities, decreased MDA content, increased the mitochondrial membrane potential and reduced intracellular free calcium concentration( [Ca2+]i ). CONCLUSION: TEF has significant protective effects against QA-induced toxicity in SH-SY5Y cells,the action mechanism may be associated with resisting oxidative stress, decreasing the [Ca2]i and up-regulating delta psi m.
