RESUMO

TRIM proteins play a crucial antiviral effector role in the innate immune system of vertebrates. In this study, we found that TRIM proteins exhibited the highest expression levels in immune organs such as spleen and kidney during IHNV infection in rainbow trout, meanwhile, we successfully amplified TRIM23 and TRIM32 from diseased rainbow trout and analyzed their gene sequences, revealing that rainbow trout TRIM23 and TRIM32 proteins are closely related to Atlantic salmon and Chinook salmon; In this experiment, the TRIM23 and TRIM32 protein genes were resoundingly constructed as a recombinant plasmids and expressed in CHSE-214 cells. Upon transfected with the recombinant plasmid, followed by viral infection, significant decreasion in the copy numbers of the virus was observed, indicating that the TRIM23 and TRIM32 proteins of rainbow trout play an important role in inhibiting virus replication, with the TRIM32 role being the most pronounced. These results provide a basis for subsequent in-depth study of the antiviral effects of TRIM proteins, and provide new ideas for immune enhancers.

Along with the advancement of manipulation technologies, image modification is becoming increasingly convenient and imperceptible. To tackle the challenging image tampering detection problem, this article presents an attentional cross-domain deep architecture, which can be trained end-to-end. This architecture is composed of three convolutional neural network (CNN) streams to extract three types of features, including visual perception, resampling, and local inconsistency features, from spatial and frequency domains. The multitype and cross-domain features are then combined to formulate hybrid features to distinguish manipulated regions from nonmanipulated parts. Compared with other deep architectures, the proposed one spans a more complementary and discriminative feature space by integrating richer types of features from different domains in a unified end-to-end trainable framework and thus can better capture artifacts caused by different types of manipulations. In addition, we design and train a module called tampering discriminative attention network (TDA-Net) to highlight suspicious parts. These part-level representations are then integrated with the global ones to further enhance the discriminating capability of the hybrid features. To adequately train the proposed architecture, we synthesize a large dataset containing various types of manipulations based on DRESDEN and COCO. Experiments on four public datasets demonstrate that the proposed model can localize various manipulations and achieve the state-of-the-art performance. We also conduct ablation studies to verify the effectiveness of each stream and the TDA-Net module.

To investigate the effects of dietary carbohydrate/lipid (CHO: L) ratios on non-specific immune responses, antioxidant capacity, and expression of TLR-MAPK/NF-κB signaling pathway-related genes of red swamp crayfish (Procambarus clarkii). Four isonitrogenous and isoenergetic diets containing different CHO: L ratios were formulated. The results showed that the group with a CHO: L ratio of 5.94 had better growth performance (P < 0.05). The highest T-AOC, CAT, and SOD activities and the lowest MDA content in hemolymph and hepatopancreas were observed in the group with a CHO: L ratio of 5.94 (P < 0.05). The lowest activities of ALT, AST, ACP, AKP, and ALB in the hemolymph were observed in CHO: L ratio 5.94 group (P < 0.05), while the highest LZM activity, TP, and GLB content were observed in CHO: L 5.94 group (P < 0.05). The highest mRNA expression levels of tlr3, myd88, and mapk3, and the lowest mRNA expression levels of nf-kb α, nf-kb ß, nf-kb p105, and traf6 were observed in the CHO: L of 5.94 group (P < 0.05). The highest mRNA expression levels of immune-related genes were observed in the CHO: L of 5.94 group (P < 0.05). Overall, these results indicated that the optimum dietary CHO: L ratio is vital in promoting growth and enhancing antioxidants and immunity to maintain red swamp crayfish's intestinal and hepatopancreas health status. In conclusion, the diets with a CHO:L ratio of 5.94 (approximately 36.23% carbohydrate and 6.10% lipid) is optimal for juvenile red swamp crayfish's physiological condition and health status.
