RESUMO

A major limitation of tumor antiangiogenic therapy is the pronounced off-target effect, which can lead to unavoidable injury in multiple organs. Ensuring sufficient delivery and controlled release of these antiangiogenic agents at tumor sites is crucial for realizing their clinical application. Here, we develop a smart DNA-based nanodrug, termed Endo-rDFN, by precisely assembling the antiangiogenic agent, endostar (Endo), into a reconfigurable DNA framework nanotube (rDFN) that could recognize tumor-overexpressed nucleolin to achieve the targeted delivery and controllable release of Endo. Endo-rDFN can not only effectively enhance the tumor-targeting capability of Endo and maintain its efficient accumulation in tumor tissues but also achieve on-demand release of Endo at tumor sites via the specific DNA aptamer for tumor-overexpressed nucleolin, named AS1411. We also found that Endo-rDFN exhibited significant inhibition of angiogenesis and tumor growth, while also providing effective protection against multiorgan injury (heart, liver, spleen, kidney, lung, etc.) to some extent, without compromising the function of these organs. Our study demonstrates that rDFN represents a promising vector for reducing antiangiogenic therapy-induced multiorgan injury, highlighting its potential for promoting the clinical application of antiangiogenic agents.

RESUMO

Photodynamic therapy (PDT), a promising treatment modality, employs photosensitizers to generate cytotoxic reactive oxygen species (ROS) within localized tumor regions. This technique involves administering a photosensitizer followed by light activation in the presence of oxygen (O2), resulting in cytotoxic ROS production. PDT's spatiotemporal selectivity, minimally invasive nature, and compatibility with other treatment modalities make it a compelling therapeutic approach. However, hypoxic tumor microenvironment (TME) poses a significant challenge to conventional PDT. To overcome this hurdle, various strategies have been devised, including in-situ O2 generation, targeted O2 delivery, tumor vasculature normalization, modulation of mitochondrial respiration, and photocatalytic O2 generation. This review aims to provide a comprehensive overview of recent developments in designing tumor-oxygenated nanomaterials to enhance PDT efficacy. Furthermore, we delineate ongoing challenges and propose strategies to improve PDT's clinical impact in cancer treatment.

Rational design of high-performance electrode materials is crucial for enhancing desalination performance of capacitive deionization (CDI). Here, ultrathin nitrogen-doped carbon/Ti3C2Tx-TiN (NC/MX-TiN) heterostructure was developed by pyrolyzing zeolite imidazolate framework-8 (ZIF-8) nanoparticles sandwiched MXene (ZSM), which were formed by assembling ultrafine ZIF-8 nanoparticles with size of 20 nm on both sides of MXene nanosheets. The introduction of ultrasmall ZIF-8 particles allowed for in situ nitridation of the MXene during pyrolysis, forming consecutive TiN layers tightly connected to the internal MXene. The two-dimensional (2D) heterostructure exhibited remarkable properties, including high specific surface area and excellent conductivity. Additionally, the resulting TiN demonstrated exceptional redox capability, which significantly enhanced the performance of CDI and ensured cycling stability. Benefiting from these advantages, the NC/MX-TiN exhibited a maximum adsorption capacity of 45.6 mg g-1 and a steady cycling performance in oxygenated saline water over 50 cycles. This work explores the rational design and construction of MXene-based 2D heterostructure and broadens new horizons for the development of novel CDI electrode materials.

Immunogenic cell death (ICD) is associated with the release of damage-associated molecular patterns, including ATP, to promote an effective immune cycle against tumors. However, tumors have evolved an effective strategy for degrading extracellular immunostimulatory ATP via the ATP-adenosine axis, allowing the sequential action of the ectonucleotidases CD39 to degrade accumulated immunostimulatory ATP into pleiotropic immunosuppressive adenosine. Here, an ingenious dissolving microneedle patch (DMNs) is designed for the intralesional delivery of CD39 inhibitor (sodium polyoxotungstate, POM-1) and ICD inducer (IR780) co-encapsulated solid lipid nanoparticles (P/I SLNs) for antitumor therapy. Upon insertion into the tumor site, IR780 induces ICD modalities with the release of damage-associated molecular patterns from endogenous tissues, which activates the antitumor immune cycle. Simultaneously, POM-1 promotes the liberation of immunostimulatory ATP and lowers the level of immunosuppressive extracellular adenosine, which supported immune control of tumors via recruiting CD39-expressing immune cells. In vivo antitumor studies prove that this platform can effectively eliminate mice melanoma (tumor growth inhibitory rate of 96.5%) and colorectal adenocarcinoma (tumor growth inhibitory rate of 93.5%). Our results shed light on the immunological aspects of combinatorial phototherapy and ATP-adenosine regulation, which will broaden the scope of synergistic antitumor immunotherapy.

Radiation therapy (RT) has emerged as one of the most promising anti-tumor strategies for neuroblastoma. Nevertheless, the special tumor microenvironment (TME), including hypoxic and GSH-overexpressed TME, often greatly restricts the RT outcome. In this study, we demonstrated a dual-channel parallel radicals nanoamplifier (ATO@PAE-PEG-AS1411/Fe3+). The nanoamplifier was shaped into a bilayer shell-core structure, in which atovaquone-loaded poly (ß-amino esters)-poly (ethylene glycol) (ATO@PAE-PEG) served as the core while Fe3+-absorbed AS1411 aptamer (AS1411/Fe3+) served as the shell. Taking advantage of the targeting ability of AS1411, ATO@PAE-PEG-AS1411/Fe3+ specifically accumulated in tumor cells, and then released ATO as well as Fe3+ in response to the acidic TME. The released ATO dramatically inhibited the mitochondrial respiration of tumor cells, thus sparing vast amounts of oxygen for the generation of free radicals during RT process, which was the first free radicals-amplifying pathway Meanwhile, the released Fe3+ could consume the tumor-overexpressed GSH through the redox reaction, thus effectively preserving the generated free radicals in RT process, which was the second free radicals-amplifying pathway. Taken together, our study demonstrates a dual-channel parallel free radicals-amplifying RT strategy, and it is expected this work will promote the clinical application prospects of RT treatment against neuroblastoma.

Integrating dissolving microneedles (DMNs) and nanocarriers (NC) holds great potential in transdermal drug delivery because it can simultaneously overcome the stratum corneum barrier and achieve efficient and controlled drug delivery. However, different skin sites with different thicknesses and compositions can affect the transdermal diffusion of NC-loaded DMNs. There are few reports on the biological fate (especially transdermal diffusion) of NC-loaded DMNs, and inaccurate bioimaging information of intact NC limits the accurate understanding of the in vivo fate of NC-loaded DMNs. The aggregation-caused quenching (ACQ) probes P4 emitted intense fluorescence signals in intact NC while quenched after the degradation of NC, had been demonstrated the feasibility of label intact NC. In this study, P4 was loaded in solid lipid nanoparticles (SLNs), and further encapsulated into DMNs, to track the transdermal diffusion of SLNs delivered at different skin sites. The results showed that SLNs had excellent stability after being loaded into DMNs with no significant changes in morphology and fluorescence properties. The in vivo live and ex vivo imaging showed that the transdermal diffusion rate of NC-loaded DMNs was positively correlated with skin thickness, with the order ear > abdomen > back. In conclusion, this study confirmed the site-dependency of transdermal diffusion in NC-loaded DMNs.

Cognitive dysfunction is common in hypothyroid patients, even after undergoing sufficient levothyroxine (LT4) replacement therapy for euthyroid. Our previous studies indicated that cholinergic neurons might contribute to the decline of cognition in adult-onset hypothyroidism. Nevertheless, the role of the cellular and neural control of basal forebrain (BF) cholinergic neurons in hypothyroidism-induced cognitive impairments is unknown. Using transgenic mice that specifically expressed chemogenetic activators in their BF cholinergic neurons, we systematically investigated the role of BF cholinergic neurons in hypothyroidism-induced cognitive dysfunction by the combined approaches of patch clamp electrophysiology, behavioral testing, and immunohistochemistry. The results showed that LT4 treatment in the adult-onset hypothyroid mice reversed only 78 % of the BF cholinergic neurons to their normal values of electrophysiological properties. LT4 monotherapy did not rehabilitate cognitive function in the hypothyroid mice. Chemogenetic selective activation of the BF cholinergic neurons combined with LT4 treatment significantly improved learning and memory functions in the hypothyroid mice. In addition, chemogenetic activation of the cholinergic neurons induced the robust expression of c-Fos protein in the BF, prefrontal cortex (PFC), and hippocampus. This indicated that the BF cholinergic neurons improved learning and memory functions in the hypothyroid mice via the BF-PFC and BF-hippocampus pathways. In the hypothyroid C57BL/6 J mice, combined treatment via LT4 and donepezil, a cholinesterase inhibitor, significantly increased cognitive functions. The results suggested that the BF cholinergic neurons are critical for regulating learning and memory and reveal a novel pathophysiological mechanism for hypothyroidism-induced cognitive impairments.

Over the past years, persulfate (PS) is widely applied due to their high versatility and efficacy in decontamination and sterilization. While treatment of organic chemicals, remediation of soil and groundwater, sludge treatment, disinfection on pathogen microorganisms have been covered by most published reviews, there are no comprehensive and specific reviews on its application to address diverse sustainability challenges, including solid waste treatment, resources recovery and regeneration of ecomaterials. PS applications mainly rely on direct oxidation by PS itself or the reactive sulfate radical (SO4•-) or hydroxyl radical (•OH) from the activation of peroxodisulfate (PDS, S2O82-) or peroxymonosulfate (PMS, HSO5-) in SO4•--based advanced oxidation processes (SO4•--AOPs). From a broader perspective of environmental cleanup and sustainability, this review summarizes the various applications of PS except pollutant decontamination and elaborates the possible reaction mechanisms. Additionally, the differences between PS treatment and conventional technologies are highlighted. Challenges, research needs and future prospect are thus discussed to promote the development of the applications of PS-based oxidation processes in niche environmental fields. In all, this review is a call to pay more attention to the possibilities of PS application in practical resource reutilization and environmental protection except widely reported pollutant degradation.

Two-dimensional (2D) transition metal dichalcogenides (TMDCs) have been widely investigated for optoelectronic applications. Here, by employing the nanocasting method, molybdenum disulfide (MoS2) nanostructures, including supercrystals, nanoparticles and nanowires, are synthesized with curved features by changing the precursor concentration and template types. The Raman properties of different MoS2 nanostructures are investigated by varying the laser power under both resonant and non-resonant excitations. The defect disorder induced LA(M) mode and other silent Raman modes in planar 2D materials are clearly observed under the resonant excitation. We believe that the varying optical properties of TMDC nanostructures will greatly broaden the optoelectronic applications of 2D materials.

Interpersonal brain synchrony (IBS) during cooperation has not been systematically investigated. To address this research gap, this study assessed neural synchrony during a cooperative jigsaw puzzle solving task using functional near-infrared spectroscopy (fNIRS)-based hyperscanning. IBS was measured for successful and failed tasks in 31 dyads in which the partners were familiar or unknown to each other. No significant difference in IBS was observed between the different types of cooperative partnership; however, stronger IBS within regions of the pars triangularis Broca's area, right frontopolar cortex, and right temporoparietal junction was observed during task success. These results highlight the effect of better task performance on cooperative IBS for the first time and further extend understanding of the neural basis of cooperation.

Cordyceps sinensis (C. sinensis) is a widely used and highly valuable traditional Chinese medicine. Several dipeptides have been detected in C. sinensis, but current scientific knowledge of its chemical makeup remains limited. In this study, an improved approach that integrates offline two-dimensional liquid chromatography (2D LC) separation, precursor ion list, library screening, and diagnostic ion filtering was established to systematically screen and characterize dipeptides in C. sinensis. Offline 2D LC integrating hydrophilic interaction LC and reverse phase separations was established to eliminate interference and identify the target dipeptides. A library containing the potential 400 dipeptides was created, and a precursor ion list with all theoretical precursor ions was adopted to trigger the MS/MS scan with high sensitivity. To identify dipeptides, the type and connection sequence of amino acids were determined according to the product ions. Ile and Leu residues were differentiated for the first time according to the characteristic ion at m/z 69.07. Ultimately, 170 dipeptides were identified or tentatively characterized from C. sinensis, and most are reported for the first time in this species herein. In addition, the identified dipeptides were also applied for discrimination among the three Cordyceps species, and 11 markers were identified. The obtained results provide a deeper understanding of the chemical basis of C. sinensis.

A novel type of molybdenum disulfide (MoS2) nanoparticles (NPs) was chemically synthesized, which possessed curved features with three-dimensional (3D) freedom compared with planar two-dimensional (2D) materials. Due to the introduction of curved features, the synthesized NPs exhibited a strongly enhanced nonlinear refractive index (n2 ∼ 10-5 cm-2 W-1) and third-order susceptibility (χ(3) ∼ 10-7 esu), which were experimentally verified by the spatial self-phase modulation effect in the visible wavelength range. Both the nonlinear parameters were two orders of magnitude higher than their planar MoS2 nanostructure counterparts. In addition, the relative change of the effective nonlinear refractive index Δn2/n2 was found to be distinctly dependent on the intensity of the applied electromagnetic field. Moreover, an all-optical modulation was experimentally realized based on the spatial cross-phase modulation effect. Our results demonstrate planar MoS2 materials with 3D features as potential candidates for next generation all-optical applications and open a substantial approach for the design of efficient nanomaterials with favorable optical nonlinearity.

A rapid start of the completely autotrophic nitrogen removal over nitrite (CANON) process based on granular sludge and efficient nitrogen removal under mixotrophic conditions are important steps in a continuous flow reactor for CANON engineering applications. In this study, an aged CANON granular sludge was mechanically crushed to 0.3 mm as inoculum in an airlift internal-loop reactor (AIR) to achieve simultaneous COD removal and mixotrophic denitrification of the single-stage granular sludge. The system achieved stable partial nitrification by controlling DO after 26 days of startup. Granulation and anaerobic ammonia oxidation were then promoted by shortening the HRT to increase the ammonia nitrogen load to 5.65 kg ·(m3 ·d)-1. The total nitrogen removal rate reached 58% on the 68th day. Subsequently, the C/N ratio of influent was increased from 0 to 0.25 and 0.5, which promoted the synergistic growth of AOB, AMX, and heterotrophic microorganisms. The removal rates of ammonia and total nitrogen were 95% and 85% respectively, and the removal of COD reached approximately 80%. The activity of NOB such as Nitrospira was effectively inhibited as the COD concentration was increased. q(NH4+-N) and q(TN) were stable at 0.4 g ·(g ·h)-1 and 0.34 g ·(g ·h)-1, respectively, while q(NO3--N) was approximately 0.02 g ·(g ·h)-1. Microbial diversity was observed using MiSeq high-throughput sequencing. It showed that organic carbon had no significant effect on the abundance of Nitrosomomas and Candidutus_Kuenenia, while increasing the abundance of Candidutus_Brocadia and Denitratisoma in the sludge. This study provides ideas for the rapid start of continuous flow CANON granular sludge process to treat wastewater with low C/N ratio.

Objective: As traditional techniques for microscopic identification of Chinese medicines currently lack objective and high-quality reference images, here we developed a systemic procedure to be used in microscopic identification of Chinese medicines, which would lead to more objective, effective and accurate identification process. Methods: Spatholobi Caulis (Jixueteng in Chinese) was used as the specimen in the development of such procedure. Jixueteng samples were microscopically examined in bright- and dark-field microscopy. Microscopic images were obtained by regular, EDF, and image stitching techniques. Results: The microscopic images of the characteristics in pulverized Jixueteng were captured, thanks to EDF imaging and image stitching techniques which allowed the detailed and full sighting of each characteristic to be obtained simultaneously. Different layers in anatomical transverse section, including cork, phelloderm, cortex, phloem, cambium, xylem and pith, were distinctively observed. Moreover, by comparing images of bright- and dark-field microscopy, birefringent and non- birefringent components could readily be distinguished. Conclusion: With application of the developed procedure, high-definition, panoramic and microscopic images were acquired, which could be used as the reference images for microscopic identification of Chinese medicines.

A new coumarin-based fluorescent probe, containing an allylic esters group, has been designed and synthesized for sensing cysteine in physiological pH. In this fluorescent probe, the coumarin was applied as the fluorophore and an allylic esters group was combined as both a fluorescence quencher and a recognition unit. The probe can selectively and sensitively detect cysteine (Cys) over homocysteine, glutathione, and other amino acids, and has a rapid response time of 30 min and a low detection limit of 47.7 nM. In addition, the probe could be applied for cell imaging with low cytotoxicity.
