RESUMO

Thyroid storm (TS) leading to acute liver failure is rare but fatal in clinical practice and hepatic failure can remarkably limit medication options for TS. We successfully cured a patient with TS complicated with acute hepatic failure using therapeutic plasma exchange (TPE) and a double plasma molecular absorption system (DPMAS) and summarized the case characteristics of 10 similar critical patients reported worldwide. We recommend that patients with TS complicated with liver failure disuse propylthiouracil or methimazole. TPE should be utilized to rapidly decrease thyroid hormone levels, and DPMAS should be considered for supportive treatment in the presence of hepatic encephalopathy or dramatic bilirubin elevations.

The motivation-direction model has served as the primary framework for understanding frontal cortical activity. However, research on the link between approach/avoidance motivation and left/right frontal cortical activity has produced inconsistent findings. Recent studies suggest that regulatory systems may offer a more accurate explanation than the motivational direction model. Despite being regulatory systems, the relationship between regulatory focus and frontal cortical activity has received limited attention. Only one experimental study has explored this connection through correlational analysis, yet it lacks causal evidence. The present study aimed to address this gap by manipulating regulatory focus and measuring frontal cortical activity in 36 college students. Our results revealed that induced promotion focus led to increased left frontal cortical activity, whereas induced prevention focus led to increased right frontal cortical activity. These findings enhance our physiological understanding of regulatory focus and offer a deeper explanation of how regulatory focus influences alterations in psychology and behavior.

The aim of this research was to explore the therapeutic capabilities of extracellular vesicles (EVs) derived from human umbilical cord mesenchymal stem cells (hUC-MSCs) that had been subjected to heat shock pretreatment, in treating psychiatric disorders induced by sleep deprivation in mice. The EVs were isolated and characterized, while western blotting was utilized to assess the expression of exosomal markers and heat shock protein 70 (HSP70). To evaluate the impact of EV treatment on anxiety-like behavior and cognitive impairment in sleep-deprived (SD) mice, the open field test, plus maze test, and Y-maze task were conducted. Heat shock pretreatment significantly increased the expression of HSP70 in EVs. Administration of EVs from heat shock-pretreated hUC-MSCs improved anxiety-like behavior and cognitive function in SD mice. Furthermore, EV treatment promoted synaptic protein expression, HSP70 expression and inhibited neuroinflammation in the hippocampus of SD mice. Western blotting analysis also revealed that EV treatment reduced the levels of TLR4 and p65 in the hippocampus. EVs from heat shock-pretreated hUC-MSCs have therapeutic potential for sleep deprivation-induced psychiatric disorders by regulating neuroinflammation and synaptic function in mice.

White tea contains the highest flavonoids compared to other teas. While there have been numerous studies on the components of different tea varieties, research explicitly focusing on the flavonoid content of white tea remains scarce, making the need for a good flavonoid purification process for white tea even more important. This study compared the adsorption and desorption performance of five types of macroporous resins: D101, HP20, HPD500, DM301, and AB-8. Among the tested resins, AB-8 was selected based on its best adsorption and desorption performance to investigate the static adsorption kinetics and dynamic adsorption-desorption purification of white tea flavonoids. The optimal purification process was determined: adsorption temperature 25 °C, crude tea flavonoid extract pH 3, ethanol concentration 80 %, sample loading flow rate and eluent flow rate 1.5 BV/min, and eluent dosage 40 BV. The results indicated that the adsorption process followed pseudo-second-order kinetics. Under the above purification conditions, the purity of the total flavonoids in the purified white tea flavonoid increased from approximately 17.69 to 46.23 %, achieving a 2.61-fold improvement, indicating good purification results. The purified white tea flavonoid can be further used for nutraceutical and pharmaceutical applications.

Ultrasound, due to its noninvasive nature, has the potential to enhance or suppress neural activity, making it highly promising for regulating intractable brain disorders. Precise ultrasound stimulation is crucial for improving the efficiency of neural modulation and studying its mechanisms. However, the presence of the skull can cause distortion in the ultrasound field, thereby affecting the accuracy of stimulation. Existing correction methods primarily rely on magnetic resonance guidance and numerical simulation. Due to the large size and high cost, the MR-guided transcranial ultrasound is difficult to be widely applied in small animals. The numerical simulation usually requires further validation and optimization before application, and the most effective method is to visualize the excited ultrasound field. However, the ultrasound field correction methods based on acoustic field visualization are still lacking. Therefore, a shadowgraph-based transient ultrasonic field visualization system is developed, and an ex vivo transcranial ultrasound field correction is performed. By visualizing the ultrasound field with or without a rat skull and then calculating the time difference of each element's ultrasound wavefront, the parameters for ultrasound field correction can be achieved. The experimental results show that this method can improve both the shape and the size of the focal spot, as well as enhance the acoustic pressure at the focus. Overall, the results demonstrate that the ultrasonic field visualization technology can effectively improve the transcranial ultrasound focusing effect and provide a new tool for achieving precise ultrasonic neural modulation.

BACKGROUND: Many neurobehavioral processes, including psychomotor, cognitive, and affection are negatively impacted by sleep deprivation (SD), which may be harmful to a person's physical and mental health. Heat shock proteins (Hsps) have been demonstrated to play a protective role in a number of neurodegenerative diseases and are essential for maintaining intracellular protein homeostasis, but their roles in SD remain elusive. METHODS: A mouse SD model was constructed using a modified multi-platform water environment method. The cognitive function was tested by novel object recognition test and Y-maze test, and anxiety-like behaviors were assessed by open field test (OFT). Protein expression was determined by Western blotting assay and ELISA assay. RESULTS: We found that SD could profoundly enhance anxiety levels and impair cognitive function in mice. SD also reduced the expression levels of p-cAMP-response element binding protein (CREB) and brain-derived neurotrophic factor (BDNF) and increased microglial activation and neuroinflammatory response in the hippocampus of mice. The intranasal injection of human recombinant Hsp70 protein could alleviate SD-induced anxiety and cognitive impairment, as well as restore pCREB and BDNF levels and reduce microglia-induced neuroinflammation in the hippocampus of SD mice. CONCLUSIONS: Hsp70 treatment might serve as a potential treatment for mitigating SD-related unfavorable symptoms.

During the COVID-19 pandemic, rumors were shared widely and quickly, leading to unfortunate consequences. To explore the dominant motivation underlying such rumor sharing behavior and the potential consequences for sharers' life satisfaction, two studies were conducted. Study 1 was based on representative popular rumors that circulated throughout Chinese society during the pandemic to examine the dominant motivation underlying rumor sharing behavior. Study 2 employed a longitudinal design to further test the dominant motivation underlying rumor sharing behavior and its effects on life satisfaction. The results of these two studies generally supported our hypotheses that people chose to share rumors during the pandemic mainly for the purpose of fact-finding. Regarding the effects of rumor sharing behavior on life satisfaction, although sharing wish rumors (i.e., rumors expressing hopes) had no effect on sharers' life satisfaction, sharing dread rumors (i.e., rumors reflecting fears) and aggression rumors (i.e., rumors implying aggression and hatred) reduced sharers' life satisfaction. This research lends support to the integrative model of rumor and provides practical implications for mitigating the spread of rumors.

Rumours circulated quickly online and offline during the COVID-19 pandemic, but empirical research on the subject is limited. Combining qualitative (Study 1, content analysis was conducted on 2344 actual rumours extracted from a rumour-refuting website) and quantitative methods (Study 2, a three-wave study with 10-day intervals), the current study suggests that (1) rumours during the pandemic can be categorised into three types, that is, wish, dread, and aggression rumours, and (2) exposure to different types of rumours is associated with coping consequences, subjective well-being (comprising positive affect, negative affect, and life satisfaction), and interpersonal trust in different ways. Generally, wish rumours seem benign, while dread and aggression rumours are malicious. Specifically, wish rumours are believed to assist coping and to be positively associated with positive affect and interpersonal trust. In contrast, dread rumours are believed not to assist coping and to be marginally significantly and positively associated with negative affect and negatively associated with interpersonal trust. Meanwhile, aggression rumours are believed not to assist coping and are marginally significantly and positively associated with negative affect. All other relationships are nonsignificant. The results of the current study will help national governments and international agencies design and evaluate rumour control strategies and policies.

Although we are surrounded by various kinds of rumors during the coronavirus disease pandemic, little is known about their primary content, what effect they might have on our emotions, and the potential factors that may buffer their effect. Combining qualitative (study 1 extracted 1907 rumors from top rumor-refuting websites using the Python Web Crawler and conducted content analysis) and quantitative (study 2 conducted an online survey adopting a three-wave design, N = 444) research methods, the current study revealed that government-related rumors accounted for the largest proportion of rumors during the outbreak stage of the pandemic and were positively associated with the public's negative emotions. We also found that trust in government negatively moderated the relationship between government-related rumors and negative emotions. Specifically, when people had low trust in government, exposure to government-related rumors was positively associated with negative emotions. However, when people had high trust in government, the association was non-significant. For positive emotions, we found no significant effects of government-related rumors. The findings highlight the importance of rumor control during public emergencies and cultivating public trust in government in the long run. BACKGROUND: Inflammatory pain mediated by nuclear factor kappa-B (NF-κB) signal pathway has become an increasingly important clinical issue in the last decade. As a potent antioxidant, Nodakenetin has been shown to have a prominent inhibitory effect on inflammation. However, the therapeutic effects and underlying pharmacological mechanisms of Nodakenetin for inflammatory pain remain unclear. METHODS: Intraplanar injection of complete Freund's adjuvant (CFA) was used to establish a model of chronic inflammation pain in C57BL/6 mice. The chronic neuropathic pain model was conducted by the sciatic nerve ligation surgery. QRT-PCR was performed to estimate the RNA levels of tumor necrosis factor-α (TNF-α), interleukin-1ß (IL-1ß), and interleukin-6 (IL-6). Western blot was used to demonstrated the protein levels of phospho-IkappaBα (IκBα), p50, and p65 in HEK293T cells. RESULTS: The bioactive components of the traditional Chinese medicine Notopterygium forbesii boiss mainly include Nodakenetin, isoimperatorin, and pregnenolone. Nodakenetin significantly alleviated CFA-induced inflammatory pain but showed no significant therapeutic effect on surgically induced neuralgia in a mouse model. In contrast, isoimperatorin and pregnenolone did not relieve CFA-induced inflammatory pain. Mechanistically, Nodakenetin inhibited IL-1ß-induced activation of the NF-κB pathway and phosphorylation of IκBα in HEK293T cells. Furthermore, Nodakenetin treatment suppressed the expression of IL-6, TNF-α, and IL-1ß in mouse bone marrow-derived macrophages. CONCLUSION: Nodakenetin alleviates inflammatory pain induced by CFA injection in vivo and modulates NF-κB signal pathway in vitro.

The causal influence of the dorsolateral prefrontal cortex (DLPFC) in voluntary forgetting remains unclear. Here, we employed repetitive transcranial magnetic stimulation (rTMS) over the right DLPFC to temporarily disrupt function of this brain region and examined its influence on an item-method directed forgetting (DF) task with both neutral and negative emotional memories. Participants were assigned to either an active or a sham rTMS group, in which we administered stimulation for 20 min before the DF task. We then examined the explicit and implicit DF effects with an explicit recognition and an implicit word completion test. We found that while participants in the sham group showed the classic DF effects in both explicit and implicit memory tests, temporally disrupting activity of the right DLPFC selectively reduced the DF effect on explicit recognition, but not on implicit word completion test. Our findings provide novel evidence that the right DLPFC plays a causal role in voluntary forgetting and support the direct inhibition account of voluntary memory control. Intriguingly, preserved implicit DF effects in the active stimulation group suggest that unintentional expressions of unwanted memories may be more sensitive to DF and less dependent on the right DLPFC.

Generally, successful cooperation can only be established when the interacting persons believe that they would not be betrayed; this belief can be updated by observing the other persons' actual choices. Thus, the process of belief updating plays an important role in conditional cooperation. Using the Prisoner's Dilemma Game (PDG) with event-related potential (ERP) hyperscanning, this study investigated the dynamics of belief updating in a dyad. During the task, participants were asked if they believed that their opponent would cooperate in the next trial, and their answers functioned as a self-reported index of reciprocal belief. The results suggested that this index shows strong associations with participants' behavioral choices (cooperate/betray). At the individual level, the amplitudes of the ERP components frontal P3a and parietal P3b elicited by the decision outcome were sensitive to belief updating. At the interpersonal level, the between-subject synchronization in P3b was higher than those in the other conditions when the paired participants confirmed each other's reciprocal beliefs. Since previous studies have linked the P3b with memory updating, we suggest that a cooperative relationship is built up when the memory systems (which support belief updating) of two interacting persons reach a high level of coordination. These findings may help explain how conditional cooperation develops between strangers.

Emerging evidence has revealed significant roles for long noncoding RNA (lncRNA) in various biological processes, including cell proliferation, apoptosis and invasion. The lncRNA distal­less homeobox 6 antisense 1 (DLX6­AS1) has been reported to serve as a vital oncogene during tumorigenesis and progression. However, the expression levels and functional roles of DLX6­AS1 in cervical cancer are not yet well understood. In the present study, DLX6­AS1 expression was identified to be significantly upregulated in cervical cancer tissues and cell lines by reverse transcription­quantitative polymerase chain reactions. Knockdown of DLX6­AS1 inhibited cell proliferation and induced cell apoptosis. Bioinformatics analysis predicted that micro RNA (miR)­199a was a direct target of DLX6­AS1. Overexpression of miR­199a counteracted the role of DLX6­AS1 in facilitating proliferation and inhibiting apoptosis in in vitro rescue assays. The present results suggest that DLX6­AS1 acting as a sponge for miR­199a may serve a critical role in the development and progression of cervical cancer.

BACKGROUND: Respiratory mechanics affects the effect of positive end-expiratory pressure (PEEP) on intracranial pressure (ICP). Respiratory mechanics of the lung and the chest wall was not differentiated in previous studies. In the present study, we investigated the influence of the following possible determinants of ICP responsiveness to PEEP: chest wall elastance (ECW), lung elastance (EL), and baseline ICP. METHODS: Eight healthy Bama miniature pigs were studied. The increase of EL was induced by instillation of hydrochloride, and the increase of ECW was induced by strapping the animals' chest wall and abdomen. A balloon-tipped catheter was placed intracranially for inducing intracranial hypertension. Six experimental conditions were investigated in sequence: 1) Normal; 2) Stiff Chest Wall; 3) Lung Injury; 4) Lung Injury + Stiff Chest Wall; 5) Lung Injury + Stiff Chest Wall + Intracranial Hypertension and 6) Lung Injury + Intracranial Hypertension. PEEP was gradually increased in a 5 cm H2O interval from 5 to 25 cm H2O in each condition. Blood pressure, central venous pressure, ICP, airway pressure and esophageal pressure were measured. RESULTS: Hydrochloride instillation significantly increased EL in conditions with lung injury. ECW significantly increased in the conditions with chest wall and abdomen strapping (all p <  0.05). ICP significantly increased with increments of PEEP in all non-intracranial hypertension conditions (p <  0.001). The greatest cumulative increase in ICP was observed in the Stiff Chest Wall condition (6 [5.3, 6.8] mm Hg), while the lowest cumulative increase in ICP was observed in the Lung Injury condition (2 [1.3, 3.8] mm Hg). ICP significantly decreased when PEEP was increased in the intracranial hypertension conditions (p <  0.001). There was no significant difference in cumulative ICP change between the two intracranial hypertension conditions (p = 0.924). CONCLUSIONS: Different respiratory mechanics models can be established via hydrochloride induced lung injury and chest wall and abdominal strapping. The effect of PEEP on ICP is determined by respiratory mechanics in pigs with normal ICP. However, the responsiveness of ICP to PEEP is independent of respiratory mechanics when there is intracranial hypertension.

There is abundant evidence suggesting that the right ventrolateral prefrontal cortex (rVLPFC) plays an important role in down-regulating the emotional response to social exclusion. However, a causal relationship between rVLPFC function and explicit emotional regulation is not clear in the context of social exclusion. This study employed anodal transcranial direct current stimulation (tDCS) to activate rVLPFC while participants used emotional regulation to reappraise pictures of social exclusion. Forty-four participants were randomly assigned to an active tDCS group or a sham group. Both groups viewed social exclusion images under two conditions: in the no-reappraisal condition, participants were instructed to passively view social exclusion images; in the reappraisal condition, they reappraised the images to down-regulate negative emotional responses. Compared to sham stimulation, anodal tDCS over the rVLPFC resulted in less negative emotion ratings, and produced significantly smaller pupil diameter in the reappraisal, compared to no-reappraisal block. The tDCS also led to longer fixation durations to rejectees and shorter fixation durations to rejecters. Taken together, these findings suggest a causal role for rVLPFC in down-regulation of negative emotions produced by social exclusion. This study has implications for clinical interventions targeting emotional regulation deficits.

OBJECTIVE: MicroRNAs (miRNAs) have become the focus of most recent efforts in cancer research. However, there have been inconsistencies in the literature regarding the suitability of circulating miRNAs for early detection of gastrointestinal cancers. This study aims to assess the diagnostic performance of circulating miRNAs in detection of gastrointestinal cancer through a meta-analysis. METHODS: Eligible studies were selected by conducting a systematic literature search of public databases. The sensitivity and specificity were used to plot the summary receiver operator characteristic (SROC) curve and calculate the area under the SROC curve (AUC). The between-study heterogeneity was evaluated by Q test and I2 statistics. Subgroup analyses and meta-regression were further performed to explore the potential sources of heterogeneity. All analyses were performed using the STATA 12.0 software. RESULTS: A total of 107 studies from 42 articles were included for the meta-analysis according to the inclusion criteria. The overall analysis of all gastrointestinal cancers showed that circulating miRNAs have a relatively good diagnostic performance in gastrointestinal cancers, with a sensitivity of 0.75, a specificity of 0.81 and an AUC of 0.85. In addition, subgroup analyses based on different type of miRNA assay suggested that single-miRNA assay displayed a relatively low diagnostic performance with the AUC values of 0.84 for gastric cancer (GC) and 0.79 for colorectal cancer (CRC), while multiple-miRNAs assay significantly improved the diagnosing accuracy with AUC rising to 0.92 for GC and 0.89 for CRC. Another interesting finding was that plasma-based miRNA assay reach a higher accuracy compared with serum-based one for GC, while opposite conclusion was drawn for CRC. CONCLUSIONS: In conclusion, circulating miRNAs, particularly the combination of multiple miRNAs, may present as promising biomarkers for the diagnosis of gastrointestinal cancers. Further large-scale prospective studies are necessary to validate their potential applicability in human cancer diagnosis.
