RESUMO

The friction of a solid contact typically shows a positive dependence on normal load according to classic friction laws. A few exceptions were recently observed for nanoscale single-asperity contacts. Here, we report the experimental observation of negative friction coefficient in microscale monocrystalline heterojunctions at different temperatures. The results for the interface between graphite and muscovite mica heterojunction demonstrate a robust negative friction coefficient both in loading and unloading processes. Molecular dynamics simulations reveal that the underlying mechanism is a synergetic and nontrivial redistribution of water molecules at the interface, leading to larger density and more ordered structure of the confined subnanometer-thick water film. Our results are expected to be applicable to other hydrophilic van der Waals heterojunctions.

RESUMO

Previous studies have shown that rambutan peel phenolic (RPP) extract has excellent biological activities due to its abundant phenolic content and profile. In this study, the potential anti-osteoporosis (OP) effects of RPP were evaluated by suppressing receptor activator nuclear factor-kappa B ligand (RANKL)-induced differentiation of RAW264.7 cells into osteoclasts and amelioratingretinoic acid-induced OP in rats. Our results showed that RPP efficiently decreased the formation of tartrate-resistant acid phosphatase (TRAP)-positive cells and reduced total TRAP activity in RAW264.7 cells under RANKL stimulation. RPP treatment significantlyameliorated retinoid acid-induced calcium loss in rats (p < 0.05). The serum phosphorus level of osteoporotic rats was increased by RPP treatment, and the serum levels of total alkaline phosphatase and osteocalcin in osteoporotic rats were further reduced. RPP treatment improved the qualities of the femur and tibia, such asbone mineral density, bone length, bone maximum load, cortical bone area ratio, and trabecularelative bone density in osteoporotic rats to some extent. Furthermore, histological analysis showed that RPP effectively improved the bone microstructure of osteoporotic rats by regulating the cortical bone thickness and trabecular bone separation. These results indicate that RPP could have potential applications as a newnutraceutical and functional food in the prevention of OP.

RESUMO

Rambutan peel phenolics (RPP) have high antioxidant and anti-inflammatory activities. Leu-Ser-Gly-Tyr-Gly-Pro (LSGYGP) possesses good radical scavenging activity and matrix metalloproteinase (MMPs) inhibitory ability. These underlying mechanisms indicated that RPP and LSGYGP may be used for antiphotoaging. Few data on the effects of RPP and LSGYGP on in vivo photoaging are available. We evaluated the effects of RPP and/or LSGYGP on ultraviolet (UV)-induced hairless mice skin photoaging. In particular, we analyzed the additive effect of RPP and LSGYGP. The biochemical indices of mice skin, including composition (collagen and hyaluronic acid [HA] contents), oxidant stress (antioxidant enzyme activities and glutathione and malondialdehyde contents), MMPs (MMP-1, MMP-3, and MMP-9 levels), inflammatory cytokines (interleukin (IL)-1α, tumor nuclear factor-α, and IL-6 levels) and the phosphorylation of the mitogen-activated protein kinase pathway, were determined. Results showed a protective effect of RPP and/or LSGYGP on photoaging skin. LSGYGP showed considerable effects on skin collagen and HA contents. RPP showed improved effects on the regulation of the oxidant stress and inflammatory cytokine levels. RPP and LSGYGP exerted an additive effect on the amelioration of the biochemical indices of UV-induced photoaging skin. The histological changes showed that RPP and LSGYGP recovered the changes in skin tissue and endogenous collagen.

RESUMO

The lateral force calibration is a key procedure for applications of atomic force microscopes. Among different calibration methods, the diamagnetic lateral force calibration (DLFC) method has been widely used due to its ease of use as well as being able to estimate the cross talk conversion factor and achieve very small stiffness. The lateral stiffness of the system is the only parameter in the DLFC method; however, its dependence on the properties and parameters of the DLFC system remains unexplored. In this paper, a theoretical formulation of such dependence is developed and experimentally verified. These results provide a guidance to design and optimize future DLFC systems with better applicability and precision in calibrations. As an example, we optimized a DLFC system such that it is robust against normal load, which is previously assumed impossible.

RESUMO

In this study, melanin (BgM) was obtained from Boletus griseus. The chemical composition and structure of BgM were characterized by UV-visible absorption spectrum, Fourier transform infrared spectrum, elemental analysis, nuclear magnetic resonance, pyrolysis gas chromatography mass spectrometry, and ultra-performance liquid chromatography⁻high resolution mass spectrometry. The percentage contents of C, H, N, S and O elements were 56.38%, 5.86%, 6.17%, 2.44%, and 28.04%, and the S/N and C/N ratios were 0.17 and 10.66, respectively. The UV-vis spectrum of BgM showed a maximum absorption peak at 214 nm. Characteristic absorption peaks were observed at 3426, 1600 and 1105 cm-1, and BgM contained phenolic hydroxyl, amidogen, carbonyl, methylene, and methyl groups. Moreover, BgM is an eumelanin, and its main skeleton has both a benzene ring and an indole, and the branched chain mainly consists of alkanes, alcohols, and fatty acids. BgM was hydrolyzed by H2O2 and four compounds were tentatively analyzed from the UPLC-MS/MS profile. The chemical structure of BgM was characterized as 5,6-dihydroxyindole eumelanin, and the condensed molecular formula is [C28(OR1)4(OR2)3H11O6N4]n.

RESUMO

Previous studies have shown that 5,2',4'-trihydroxy-6,7,5'-trimethoxyflavone (TTF1) is the primary anticancer constituent of the traditional Chinese medicinal plant Sorbaria sorbifolia (SS), which has been applied to treat cancer in China. In this study, we investigated the in vitro and in vivo antitumor effects and biological mechanisms of small-molecule TTF1 nanoparticles (TTF1-NPs). The effects of TTF1-NPs on cell growth and apoptosis were investigated using human hepatoma cells. The molecular changes associated with the effects of TTF1-NPs were analyzed by immunocytochemistry and Western blot analysis. The in vivo effect of TTF1-NPs was investigated using the HepG2 tumor xenograft model. We found that TTF1-NPs exhibited antitumor effects in vitro accompanied by induction of apoptosis in human hepatoma cells. Mechanistically, our data showed that TTF1-NPs induced apoptosis via endoplasmic reticulum stress (ERS) pathway in hepatoma cells. Moreover, inhibition of ERS activation blocked TTF1-NP-induced apoptosis in HepG2 cells. Finally, TTF1-NPs inhibited the growth of HepG2 xenograft tumors. Taken together, our results demonstrated that TTF1-NP-induced apoptosis was mediated at least in part by the ERS pathway and thus inhibited hepatoma tumor growth.
