RESUMO

Pueraria lobata (Willd.) Ohwi, known as kudzu and used as a "longevity powder" in China, is an edible plant which is rich in flavonoids and believed to be useful for regulating blood sugar and treating diabetes, although the modes of action are unknown. Here, a total of 53 flavonoids including 6 novel compounds were isolated from kudzu using multidimensional preparative liquid chromatography. The flavonoid components were found to lower blood sugar levels, promote urine sugar levels in mice, and reduce the urine volume. Molecular docking and in vitro assays suggested that the antidiabetic effect of kudzu was attributed to at least three targets: sodium-dependent glucose transporter 2 (SGLT2), protein tyrosine phosphatase-1B (PTP1B), and alpha-glucosidase (AG). This study suggests a possible mechanism for the antidiabetic effect that may involve the synergistic action of multiple active compounds from kudzu.

RESUMO

RNA N6-methyladenosine is a prevalent and abundant type of RNA modification that exerts significant influence on diverse biological processes. To date, numerous computational approaches have been developed for predicting methylation, with most of them ignoring the correlations of different encoding strategies and failing to explore the adaptability of various attention mechanisms for methylation identification. To solve the above issues, we proposed an innovative framework for predicting RNA m6A modification site, termed BLAM6A-Merge. Specifically, it utilized a multimodal feature fusion strategy to combine the classification results of four features and Blastn tool. Apart from this, different attention mechanisms were employed for extracting higher-level features on specific features after the screening process. Extensive experiments on 12 benchmarking datasets demonstrated that BLAM6A-Merge achieved superior performance (average AUC: 0.849 for the full transcript mode and 0.784 for the mature mRNA mode). Notably, the Blastn tool was employed for the first time in the identification of methylation sites. The data and code can be accessed at https://github.com/DoraemonXia/BLAM6A-Merge.

N4-acetylcytidine (ac4C) is a post-transcriptional modification in mRNA that is critical in mRNA translation in terms of stability and regulation. In the past few years, numerous approaches employing convolutional neural networks (CNN) and Transformer have been proposed for the identification of ac4C sites, with each variety of approaches processing distinct characteristics. CNN-based methods excels at extracting local features and positional information, whereas Transformer-based ones stands out in establishing long-range dependencies and generating global representations. Given the importance of both local and global features in mRNA ac4C sites identification, we propose a novel method termed TransC-ac4C which combines CNN and Transformer together for enhancing the feature extraction capability and improving the identification accuracy. Five different feature encoding strategies (One-hot, NCP, ND, EIIP, and K-mer) are employed to generate the mRNA sequence representations, in which way the sequence attributes and physical and chemical properties of the sequences can be embedded. To strengthen the relevance of features, we construct a novel feature fusion method. Firstly, the CNN is employed to process five single features, stitch them together and feed them to the Transformer layer. Then, our approach employs CNN to extract local features and Transformer subsequently to establish global long-range dependencies among extracted features. We use 5-fold cross-validation to evaluate the model, and the evaluation indicators are significantly improved. The prediction accuracy of the two datasets is as high as 81.42.

Crop disease detection and crop baking stage judgement require large image data to improve accuracy. However, the existing crop disease image datasets have high asymmetry, and the poor baking environment leads to image acquisition difficulties and colour distortion. Therefore, we explore the potential of the self-attention mechanism on crop image datasets and propose an innovative crop image data-enhancement method for recurrent generative adversarial networks (GANs) fused with the self-attention mechanism to significantly enhance the perception and information capture capabilities of recurrent GANs. By introducing the self-attention mechanism module, the cycle-consistent GAN (CycleGAN) is more adept at capturing the internal correlations and dependencies of image data, thus more effectively capturing the critical information among image data. Furthermore, we propose a new enhanced loss function for crop image data to optimise the model performance and meet specific task requirements. We further investigate crop image data enhancement in different contexts to validate the performance and stability of the model. The experimental results show that, the peak signal-to-noise ratio of the SM-CycleGAN for tobacco images and tea leaf disease images are improved by 2.13% and 3.55%, and the structural similarity index measure is improved by 1.16% and 2.48% compared to CycleGAN, respectively.

l-threonine dehydrogenase (Tdh) is an enzyme that links threonine metabolism to epigenetic modifications and mitochondria biogenesis. In vitro studies show that it is critical for the regulation of trimethylation of histone H3 lysine 4 (H3K4me3) levels and cell fate determination of mouse embryonic stem cells (mESCs). However, whether Tdh regulates a developmental process in vivo and, if it does, whether it also primarily regulates H3K4me3 levels in this process as it does in mESCs, remains elusive. Here, we revealed that, in zebrafish hematopoiesis, tdh is preferentially expressed in neutrophils. Knockout of tdh causes a decrease in neutrophil number and slightly suppresses their acute injury-induced migration, but, unlike the mESCs, the level of H3K4me3 is not evidently reduced in neutrophils sorted from the kidney marrow of adult tdh-null zebrafish. These phenotypes are dependent on the enzymatic activity of Tdh. Importantly, a soluble supplement of nutrients that are able to fuel the acetyl-CoA pool, such as pyruvate, glucose and branched-chain amino acids, is sufficient to rescue the reduction in neutrophils caused by tdh deletion. In summary, our study presents evidence for the functional requirement of Tdh-mediated threonine metabolism in a developmental process in vivo. It also provides an animal model for investigating the nutritional regulation of myelopoiesis and immune response, as well as a useful tool for high-throughput drug/nutrition screening.

BACKGROUND: To establish a nomogram to predict the probability of survival of patients with stage II/III gastric cancer (GC) who received incomplete peri-operative adjuvant chemotherapy (PAC). METHODS: The medical records of stage II/III GC patients who received curative resection and 1 to 5 cycles of PAC from two tertiary hospitals were retrospectively reviewed. Patients were randomly classified into either a training group or validation group at a ratio of 7:3. The nomogram was constructed based on various prognostic factors using Cox regression analysis in the training cohort, and was validated by the validation group. Concordance index and calibration curves were used to evaluate the discrimination and calibration of the nomogram. Additionally, decision curve analysis (DCA) was used to compare the net clinical benefits of the nomogram and eighth version of TNM staging system. RESULTS: A total of 1,070 consecutive patients were included and 749 patients were enrolled into the training group. Lower body mass index (< 18.5 kg/m2), total gastrectomy, stage III disease and fewer cycles of PAC were identified to be independent predictors for poorer survival. The area under the curve (AUC) values of receiver operating characteristics (ROC) curve predicting 5-year survival probabilities and C-index were 0.768 and 0.742, 0.700 (95%CI: 0.674-0.726) and 0.689 (95%CI: 0.646-0.732) in the training and validation groups, respectively. The calibration curves in the validation cohort showed good agreement between the prediction and observation of 1-, 3- and 5-year survival probabilities. Furthermore, DCA showed that our model has a better net benefit than that of TNM staging system. CONCLUSIONS: The findings emphasize the value of completing PAC. The nomogram which was established to predict survival probability in patients with stage II/III GC receiving radical gastrectomy and incomplete PAC had good accuracy and was verified through both internal and external validation.

Shahe Reservoir is a key node in the upstream of the North Canal, and the water quality has gradually improved after the implementation of low water operation in 2018. The organic matter(OM) in the sediment decreased from 16.66 to 14.22%.In this study, the FI index and parallel factor method were used to investigate the traceability of OM and dissolved organic matter (DOM) in the Shahe Reservoir before and during low water level operation(LWLO), and the results showed that the terrestrial source fraction of OM in sediments was mainly related to organic-rich terrestrial plant residues carried by tributaries and overflows/outfalls during the rainy season, and the FI index indicated that the organic matter (OM) in the Shahe Reservoir before and during LWLO in each DOM in the area is derived from authigenic sources of autotrophic microorganisms, algae, etc. The parallel factor method shows that more than most of the pollutants in the DOM are input from endogenous sources and a small proportion of pollutants are input from exogenous sources. Nutrients in both sediment and interstitial water increased during the LWLO, with TN and TP levels increasing by 262.38 and 204.45 mg·kg-1 in sediment, NH4+-N, PO43--P, TN and TP in interstitial water increasing by 0.98, 1.36, 2.07 and 4.33 mg·L-1, respectively. Pearson correlation analysis and principal component analysis showed that OM was significantly correlated with nutrients: OM and TN (p < 0.01) and OM and TP (p < 0.05) in the pre-LWLO; OM and TN and TP (p < 0.01) in the LWLO.The results suggested that organic matter pollution control should be mainly carried out from the perspective of endogenous input, focusing on controlling the release of nutrients in sediments.

Ecological network construction plays a key role in ecological restoration, which can effectively mitigate negative effects of habitat fragmentation on biodiversity. Here, we proposed an improved analytical framework for ecological network identification. Specifically, ecological sources were identified using a three-dimension indicator system in terms of form, quality and functions of habitats. Ecological resistance surfaces were determined based on the incorporation of points of interest data that could characterize human activities into habitat risk assessment (HRA) model, while ecological corridors were extracted using circuit theory approach. With Wuhan Metropolitan Area as a case, we explored the key points and structures of ecological network. Moreover, we compared the construction method of ecological resistance surface based on points of interest and HRA model with traditional methods that determined by land use types and by traditional HRA model, to validate the proposed framework. The results showed that the ecological source area of Wuhan Metropolitan Area was 15200 km2, the length of ecological corridor was 1956.68 km, and that there were 87 ecological "pinch points" and 67 ecological barrier points. Compared with traditional methods, the material circulation, network complexity, and ecological connectivity of the ecological network identified by the improved framework were significantly improved, with network closure, dot-line ratio, and network connectivity being increased by 61.5%, 28.1% and 28.7% on average. The identified ecological "pinch points" and barrier points could provide precise decision-making support for ecological restoration and conservation.

Checkpoint kinase 1 (CHK1), a key effector in the cellular response to DNA lesions, is a crucial component of all cell cycle checkpoints. Recent reports have revealed that CHK1 is highly expressed in numerous cancer types in the clinical settings. However, the mechanisms underlying the regulation of CHK1 expression in tumor cells remain unclear. Here, we report that CHK1 is negatively regulated by the bromodomain-containing protein 7 (BRD7). Specifically, BRD7 silencing increased CHK1 (but not CHK2) expression at both mRNA and protein levels, in a p53-independent manner in multiple tumor cell lines. Furthermore, BRD7 silencing stabilized CHK1 via reducing its ubiquitination. Mechanistically, BRD7 knockdown not only increased the levels of USP1, a deubiquitinase for CHK1, but also promoted the interaction between CHK1 and USP1, subsequently enhancing the de-ubiquitination of CHK1. USP1 knockdown abrogated BRD7 silencing-induced CHK1 induction. Biologically, the increased expression of CHK1 in tumor cells caused by BRD7 silencing significantly increased cell sensitivity to CHK1 inhibitors by enhancing tumor cell apoptosis, and this effect was reversed by the simultaneous knockdown of CHK1 or USP1. Taken together, our findings suggest that BRD7 is a potential genetic or drug target that may help to improve the efficacy of chemotherapeutic drugs targeting CHK1 in combinatorial therapy.

BACKGROUND: The aim of this study was to investigate the predictive value of procalcitonin (PCT) on post-operative day (POD) 3 and 5 for the prognosis of gastric adenocarcinoma (GA) patients who underwent radical gastrectomy surgery in extended cohort from a prospective bi-center study. METHODS: Consecutive GA patients who received surgery in the Hunan Cancer Hospital were enrolled as the training cohort, and those from Wuhan Union Hospital were included as external validation cohort. The optimal cutoff concentration of PCT for overall survival (OS) in the training cohort was determined by X-tile. The independent predictive factors for OS were identified using univariate and multivariate Cox regression analyses. Furthermore, the predictive value of elevated PCT was clarified in the validation cohort and propensity score matched cohort, respectively. RESULTS: The optimal cutoff concentrations of PCT for OS were 0.67 ng/mL at POD 3 and 0.39 ng/mL at POD 5 in the training cohort (n = 906). Patients with higher PCT concentrations (≥ 0.39 ng/mL) at POD 5 had a significantly worse prognosis whether developing post-operative infections or not. Moreover, a synergistic influence was confirmed in those with elevated PCT concentration and infections. Multivariate analyses confirmed that PCT concentration ≥ 0.39 ng/mL at POD 5 was significantly associated with poorer survival in training cohort (HR: 1.422, 95% CI 1.041-1.943, P = 0.027), validation cohort (n = 297, HR: 2.136, 95% CI 1.073-4.252, P = 0.031) and matched cohort (n = 901, HR: 1.454, 95% CI 1.104-1.914, P = 0.008), separately. CONCLUSIONS: PCT concentration ≥ 0.39 ng/mL at POD 5 was a reliable predictor for poorer prognosis in GA patients undergoing radical gastrectomy.

CONTEXT: Semen cuscutae is commonly used to treat male infertility (MI), and semen cuscutae flavonoid (SCF) is the main active component of semen cuscutae. The therapeutic mechanism of SCF on MI is still unclear. OBJECTIVE: To clarify the mechanisms of SCF against MI. MATERIALS AND METHODS: Network pharmacology and molecular docking were used to predict the potential pathways of SCF against MI. Primary Sertoli cells (SCs) were extracted from testis of 60-day-old rats and divided into Control, Model, and 3 treatment groups. The Control and Model groups were given normal medium, the treatment groups were treated with various concentrations of SCF-containing medium (200, 400, and 800 µg/mL). After 24 h, the Model and treatment groups were exposed to heat stress at 43 °C for 15 min. Western blotting and immunohistochemistry were used to detect the expression of targets. RESULT: Network pharmacology indicated that the treatment of SCF on MI was closely related to PI3K-AKT signaling pathway. The in vitro experiments showed that SCF could up-regulated the expression of AKT, AR, occludin, and Ki67, and down-regulated the expression of CK-18 in SCs after heat stress. The AKT inhibitor could block this process. CONCLUSIONS: SCF can treat MI by regulating the proliferation and differentiation of SCs and the integrity of the blood-testis barrier. The study could provide experimental basis for clinical research.

The stellate ganglion (SG), as a type of sympathetic ganglion, consists of the sixth and seventh cervical vertebrae and the first thoracic sympathetic ganglia. SG block (SGB) is a minimally invasive injection that aims to inject low-concentration local anesthetics to induce a broad sympathetic blocking effect near the SG. There have been no changes and progress in the clinical application of SGB since the 1830s due to several potential risks, including hematoma from blood vessel injury, hoarseness from recurrent laryngeal nerve injury, and cardiopulmonary arrest. The feasibility and safety of SGB have greatly improved since the appearance of ultrasound-guided SGB. In recent years, SGB has been widely applied in the field of non-anesthesiology sedation, with significant therapeutic effects on pain, immunological diseases, somnipathy, psychological disorders, arrhythmias, and endocrine diseases. The present study reviews the present application of SGB in clinical practice.

Euphorbia kansui is the dried root of Euphorbia kansui T. N. Liou ex T.P. Wang. Its main chemical components are diterpenoids, triterpenes, and volatile oil. In this study, an offline two-dimensional (2D) normal-phase × reversed-phase liquid chromatography method coupled with quadrupole time-of-flight mass spectrometry was established to comprehensively analyze the chemical constituents in E. kansui. A total of 240 compounds were identified from the E. kansui extract, including 218 diterpenoids (77 known, 141 new), 16 known volatile oils, and six known triterpenes. The relationship between the structural characteristics and tandem mass spectroscopy fragments of diterpenoids was further analyzed. Based on the characteristic fragment ions, 141 new diterpenoids were determined as 118 ingenane diterpenoids and 23 jatrophane diterpenoids. The newly identified diterpenoids may provide lead compounds for drug discovery, improving the medicinal value of E. kansui.

The encouraging effects of HER2-ADC in patients with HER2-low expression cancers indicated the classical classifications based on positive and negative HER2 might no longer be suitable. However, the biology and prognosis of colorectal cancer patients with different HER2 expression status were still not clear. This is a multi-center retrospective study that included patients with histologically confirmed colorectal cancer and determined HER2 status who received radical surgical resection. HER2 immunohistochemistry (IHC) 1+ and IHC 2+ groups were combined and defined as a HER2-low group because of the concordance of clinicopathological characteristics. As compared with the HER2-high group, both the HER2-zero and the HER2-low group had less tumor with perineural invasion (14.3%, 13.1% vs. 31.6%, p = 0.001 and p < 0.001), less stage III disease (41.8%, 39.9% vs. 56.1%, p = 0.044 and p = 0.022), more RAS/BRAF mutation (52.1%, 49.9% vs. 19.5%, p < 0.001 and p < 0.001) and better disease-free survival (DFS) (3y-DFS rate of 78.7%, 82.4% vs. 59.3%, p < 0.001 and p < 0.001). Multivariate analysis and propensity score matching also revealed that HER2-high expression was an independent prognostic factor of DFS. In conclusion, our study revealed that HER2-low colorectal cancer tumors are close to HER2-zero tumors, but different from HER2-high tumors. The routine examination of HER2 IHC is needed in early-stage colorectal cancer.

AIM AND OBJECTIVE: To assess the relationship between serum folate and schizophrenia (SZ) risk in the Chinese Han adult population in different papers, a systematic review and metaanalysis were conducted. MATERIALS AND METHODS: We searched for this meta-analysis on three English databases (PubMed, Embase, and Web of science) and four Chinese databases (CNKI, SinoMed, Wanfang, and CQVIP) on March 27, 2021. INCLUSION CRITERIA: studies provided folate levels in serum of cases and controls as mean and standard deviation. EXCLUSION CRITERIA: subjects were not Chinese Han adult population. The Newcastle-Ottawa Scale score was used to assess the risk of bias in the included studies. Standard mean difference (SMD) was used to measure the difference between SZ patients and healthy controls. Subgroup analyses by measurement time, duration, and age were performed, respectively. RESULTS: This meta-analysis included 19 publications involving 1571 SZ cases and 1283 healthy controls. In total studies, the pooled result showed that SZ patients had decreased serum folate levels compared with healthy controls (SMD [95%CI] = -1.37[-1.83,-0.90], PSMD<0.001), and in most of the subgroups, the associations reached decreased significantly; while in the subgroup of drugs use, the association was not reached significantly. CONCLUSION: Dose-response analysis and subgroup analyses by gender were not performed due to the lack of data. Folate deficiency is associated with the patients, and antipsychotic drugs might have positive effects on improving serum folate levels in Chinese Han adult SZ.

Titanium alloys are widely used in various structural materials due to their lightweight properties. However, the low wear resistance causes significant economic losses every year. Therefore, it is necessary to implement wear-resistant protection on the surface of titanium alloys. In this study, four types of in situ composite ceramic coatings with two-layer gradient structures were prepared on a Ti-6Al-4V (TC4) substrate using laser cladding. In order to reduce the dilution rate, a transition layer (Ti-40SiC (vol.%)) was first prepared on TC4 alloy. Then, a high-volume-fraction in situ composite ceramic working layer (Ti-xFe-80SiC (vol.%)) with different contents of Fe-based alloy powder (x = 0, 5, 10 and 15 vol.%) was prepared. The working surface of Ti-40SiC (TL) exhibited a typical XRD pattern of Ti, TiC, Ti5Si3, and Ti3SiC2. In comparison, both Ti-80SiC (WL-F0) and Ti-5Fe-80SiC (WL-F5) exhibited similar phase compositions to the TL coating, with no new phase identified in the coatings. However, the TiFeSi2 and SiC phases were presented in Ti-10Fe-80SiC (WL-F10) and Ti-15Fe-80SiC (WL-F15). It is proven that the addition of the Fe element could regulate the in situ reaction in the original Ti-Si-C ternary system to form the new phases with high hardness and good wear resistance. The hardness of the WL-F15 (1842.9 HV1) is five times higher than that of the matrix (350 HV1). Due to the existence of self-lubricating phases such as Ti5Si3 and Ti3SiC2, a lubricating film was presented in the WL-F0 and WL-F5 coatings, which could block the further damage of the friction pair and enhance the wear resistance. Furthermore, a wear-transition phenomenon was observed in the WL-F10 and WL-F15 coatings, which was similar to the friction behavior of structural ceramics. Under the load of 10 N and 20 N, the wear volume of WL-F15 coating is 5.2% and 63.7% of that in the substrate, and the depth of friction of WL-15 coating is only 14.4% and 80% of that in the substrate. The transition of wear volume and depth can be attributed to the wear mechanism changing from oxidation wear to adhesive wear.

The demands for model accuracy and computing efficiency in fault warning scenarios are increasing as high-speed railway train technology continues to advance. The black box model is difficult to interpret, making it impossible for this technology to be widely adopted in the railway industry, which has strict safety regulations. This paper proposes a fault early warning machine learning model based on feature contribution and causal inference. First, the contributions of the features are calculated through the Shapley additive explanations model. Then, causal relationships are discovered through causal inference models. Finally, data from causal and high-contribution time series are applied to the model. Ablation tests are conducted with the Naïve Bayes, Gradient Boosting Decision Tree, eXtreme Gradient Boosting, and other models in order to confirm the efficiency of the method based on early warning data regarding the on-site high-speed train traction equipment circuit board failure. The findings indicate that the strategy improves the evaluation markers, including the early warning accuracy, precision, recall, and F1 score, by an average of more than 10%. There is a 35% improvement in the computing efficiency, and the model can provide feature causal graph verification for expert product decision-making.

BACKGROUND: Umbilical cord blood transplantation (UCBT) from unrelated donors is one of the successful treatments for acute leukemia in childhood. The most frequent side effect of UCBT is peri-engraftment syndrome (PES), which is directly associated with the greater prevalence of acute and chronic graft-versus-host-disease (aGvHD and cGvHD). In haploidentical stem cell transplantation, posttransplant cyclophosphamide (PTCY) has been demonstrated to be an effective method against GvHD. However, the effects of PTCY as a GvHD prophylactic in UCBT had not been investigated. This study aimed to evaluate the effects of PTCY on the outcomes of UCBT for pediatric acute leukemia. METHODS: This retrospective study included 52 children with acute leukemia who underwent unrelated single-unit UCBT after myeloablative conditioning regimens. The results from the PTCY and non-PTCY groups were compared. RESULTS: The incidence of transplantation-related mortality in non-PTCY and PTCY were 5% and 10% (p = 0.525), respectively. The incidence of relapse in non-PTCY and PTCY were 5% and 23% (p = 0.095), respectively. Second complete remission status (CR2) was an independent risk factor for relapse-free survival (hazard ratio = 9.782, p = 0.001). The odds ratio for sepsis or bacteremia incidence was significantly greater in the PTCY group (9.524, p = 0.017). PTCY group had increased rates of cytomegalovirus activity and fungal infection. The incidence of PES, aGvHD, cGvHD, and hemorrhagic cystitis in the PTCY group was lower than that in the non-PTCY group, although it was not significantly different. Additionally, higher doses of PTCY (29 mg/kg and 40 mg/kg) were associated with lower incidences of aGvHD and severe GvHD (65% and 29%, respectively) than lower doses (93% and 57%, respectively). Engraftment time and graft failure incidence were similar across groups. CONCLUSION: The results support the safety and efficiency of PTCY as part of PES controlling and GvHD prophylaxis in single-unit UCBT for children with acute leukemia. A PTCY dosage of 29 mg/kg to 40 mg/kg appears to be more effective in GvHD prophylaxis for UCBT patients.

Tripterygium wilfordii Hook F from the family Celastraceae is a traditional Chinese medicine (TCM) whose principal chemical constituents are terpenoids, including sesquiterpene alkaloids and diterpenoids, which have unique and diverse structures and remarkable biological activities. In order to advance pharmacological research and guide the preparation of monomer compounds derived from T. wilfordii, a systematic approach to efficiently discover new compounds or their derivatives is needed. Herein, compound separation and identification were performed by offline reversed-phase × supercritical fluid chromatography coupled mass spectrometry (RP × SFC-Q-TOF-MS/MS) and Global Natural Product Social (GNPS) molecular networking. The 2D chromatography system exhibited a high degree of orthogonality and significant peak capacity, and SFC has an advantage during the separation of sesquiterpene alkaloid isomers. Feature-based molecular networking offers the great advantage of quickly detecting and clustering unknown compounds, which greatly assists in intuitively judging the type of compound, and this networking technique has the potential to dramatically accelerate the identification and characterization of compounds from natural sources. A total of 324 compounds were identified and quantitated, including 284 alkaloids, 22 diterpenoids and 18 triterpenoids, which means that there are numerous potential new compounds with novel structures to be further explored. Overall, feature-based molecular networking provides an effective method for discovering and characterizing novel compounds and guides the separation and preparation of targeted natural products.

Quercetin and kaempferol are flavonoids widely present in fruits, vegetables, and medicinal plants. They have attracted much attention due to their antioxidant, anti-inflammatory, anticancer, antibacterial, and neuroprotective properties. As the guarantee cells in direct contact with germ cells, Sertoli cells exert the role of support, nutrition, and protection in spermatogenesis. In the current study, network pharmacology was used to explore the targets and signaling pathways of quercetin and kaempferol in treating spermatogenic disorders. In vitro experiments were integrated to verify the results of quercetin and kaempferol against heat stress-induced Sertoli cell injury. The online platform was used to analyze the GO biological pathway and KEGG pathway. The results of the network pharmacology showed that quercetin and kaempferol intervention in spermatogenesis disorders were mostly targeting the oxidative response to oxidative stress, the ROS metabolic process and the NFκB pathway. The results of the cell experiment showed that Quercetin and kaempferol can prevent the decline of cell viability induced by heat stress, reduce the expression levels of HSP70 and ROS in Sertoli cells, reduce p-NF-κB-p65 and p-IκB levels, up-regulate the expression of occludin, vimentin and F-actin in Sertoli cells, and protect cell structure. Our research is the first to demonstrate that quercetin and kaempferol may exert effects in resisting the injury of cell viability and structure under heat stress.
