RESUMO

The quantitative analysis of multiple components with a single marker (QAMS) method was firstly established for simultaneous determination of 18 active components in Ilex kudingcha C. J. Tseng by HPLC. Using rutin, isochlorogenic acid A and kudinoside A as internal refererence substances (IRS), compatibility results showed that the relative correction factors (RCFs) of all compounds showed good reproducibility under different chromatographic conditions. On the basis of previous studies, the accuracy of the QAMS method was systematically evaluated by investigating the influences of curve intercept, analytes and IRS concentration. The results showed that the concentration (especially at low level) of analytes and curve intercept were the major influencing parameters for the LRG-QAMS method (LRG = linear regression), whereas the influence of IRS concentration seemed more apparent in terms of the AVG-QAMS method (AVG = average). The two approaches were complementary with each other. In addition, hierarchical clustering analysis (HCA), principal components analysis (PCA) and similarity analysis (SA) were performed to differentiate and classify the samples based on the contents of 18 marker compounds. The results of the different chemometric analyses were completely consistent with each other, and could be supported by the quantification results.

OBJECTIVE: To study the toxicokinetics and tissue distribution of total arsenic after giving different doses realgar to rats. METHOD: The concentration of arsenic in blood, urine and main organs were determined after single oral administration of 3.738, 1.869, 0.935 g x kg(-1) realgar in rats. RESULT: The distribution half life (t(1/2 alpha)) and elimination half life (t(1/2 beta)) of total arsenic were 7.73 h and 17.21 h respectively in high dose group. The apparent distribution volume (Vd) were 10.8, 5.6, 3.9 L x kg(-1) respectively, the total clearance (Cltot) were 0.437, 0.16, 0.111 L x h(-1) x kg(-1) respectively in the three groups. The excretion of total urine arsenic got rid of 5.5%-13.0% of realgar which given to rats in 96 hours. Arsenic could be detected distributing into rat's main organs and tissues after administration of a single dose. Suprarenal gland, bladder and ovary accumulated most of the arsenic, while liver, spleen, lung and kidney accumulated less. Heart, brain, stomach and testis accumulated the least amount. CONCLUSION: It has nonlinear dynamics in the body of rats in toxic doses as the kinetics of realgar was changed according to the dosage.

RESUMO

To prepare the oral self-microemulsifying drug delivery system (SMEDDS) of GBE50, balance solubility method was used to screen emulsifier and assistant emulsifier; a pseudo-tamary phase diagram was used to prepare microemulsion; and orthogonal design was used to optimize formulation. Self-microemulsifying efficiency, dissolution, stability and pharmacokinetics of the preparation were studied. As a result, GBE50-SMEDDS of IPM, Cremophor EL, 1,2-propanediol and GBE50 could be self emulsified to form stable microemulsion with particle diameter between 20 and 50 nm when emulsifying with water. Its self-microemulsifying efficiency and dissolution are quick with good stability and it has a higher bioavailability than market existing agents Xingling particles. GBE50-SMEDDS is stable and effective.

OBJECTIVE: To observe the effects of Rhizoma Zingiberis and Pericarpium Citri Reticulatae extracts on acute myocardial ischemia rats and explore the mechanism. METHODS: The model of myocardial ischemia in rats was established by ligating the front descending anterior branch of the coronary artery. With Fufang Danshen Pill as positive control drug,the effects of Rhizoma Zingiberis and Pericarpium Citri Reticulatae extracts on the electrocardiogram (ECG), the extension of myocardial infarction, the hemorheology indexes, lactic dehydrogenase (LDH), creatine kinase (CK), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and malondialdehyde (MDA) in rats were evaluated. RESULTS: Rhizoma Zingiberis and Pericarpium Citri Reticulatae extracts decreased the ST-segment of ECG (P < 0.01), reduced the extension of myocardial infarction (P < 0.05), decreased the contents of CK and LDH in serum (P < 0.01 or P < 0.05), improved hemorheology (P < 0.05), increased SOD and GSH-Px activity and decreased MDA content (P < 0.05). CONCLUSION: Rhizoma Zingiberis and Pericarpium Citri Reticulatae extracts have protective effect on myocardial ischemia in rats, and its mechanism may be related to inhibiting lipid peroxidation.

OBJECTIVE: To observe the therapeutic effect of Wuling Powder extract on rats with renal hypertension and to evaluate the influence of it on the volume of urine and the concentrations of Na(+), K(+), Cl(-). METHODS: Reformed Gold-blatt hypertension rat model (G-2K1C) was established. The rats were divided into 6 groups as follows: sham-operation group; model group, Wuling Powder high dosage group (80 g/kg), Wuling Powder middle dosage group (40 g/kg), Wuling Powder low dosage group (20 g/kg), and hydrochlorothiazide (HCT) group (25 mg/kg). Urine volume of the rats was measured during the experiment. Tail arterial pressure and [Na(+)], [K(+)], [Cl(-)] in serum of the rats were detected after 30 days of treatment. RESULTS: The blood pressure of the G-2K1C rats was decreased in the three Wuling Powder groups (P<0.05 or P<0.01), but higher than that of the false-operation group (P<0.01), and there was no difference between each of the Wuling Powder groups and the HCT group (P>0.05). Diuretic effect of the three dosages of Wuling Powder was weaker than that of the HCT (P<0.05 or P<0.01). The effects of the three dosages of Wuling Powder and HCT on [Na(+)] and [Cl(-)] in the serum were not obviously different (P>0.05), but [K(+)] of the HCT group was significantly decreased compared with that of the false-operation group and the three Wuling Powder groups (P<0.01). CONCLUSION: Wuling Powder extract had satisfying therapeutic effects in increasing the discharge of urine, decreasing the blood pressure and keeping the balance of the serum electrolyte contents in rats with renal hypertension.
