RESUMO

Primary hyperoxaluria type 2 (PH2) is a rare hereditary disease that causes nephrolithiasis, nephrocalcinosis and kidney failure. This study aimed to investigate the clinical features and mutational spectrum of Chinese patients with PH2. A retrospective cohort study was performed on PH2 patients admitted to our center over seven years. We also systematically reviewed all the articles on Chinese PH2 patients published from January 2000 to May 2023 and conducted a meta-analysis. A total of 25 PH2 patients (10 from our center and 15 from published studies) were included in this study. The median age of onset in patients from our center was 8.50 (1.00, 24.00) years, and 50% were male. Among the full cohort of 25 Chinese patients, the median age of onset was 8.00 (0.40, 26.00) years, and 64% of them were male. Seven patients progressed to end-stage kidney disease, with a median age of 27.50 (12, 31) years. The cumulative renal survival rates were 100%, 91.67%, 45.83% and 30.56% at 10, 20, 30 and 40 years of age, respectively. A total of 18 different variants were identified, and c.864_865del was the dominant variant, accounting for 57.69% of the total alleles. Patients who were heterozygous for c.864_865del were more susceptible to nephrocalcinosis than those who were homozygous for c.864_865del and those harboring other mutations (83.33% versus 33.3% and 0%, respectively) (p = 0.025). The clinical features and mutational spectrum of Chinese PH2 patients were described. This study helps to expand awareness of the phenotypes and genotypes of Chinese PH2 patients and contributes to the improvement of diagnostic and treatment strategies for PH2 patients.

Objective: To investigate the correlation between programmed death ligand 1(PD-L1), tumor mutation burden (TMB) and the short-term efficacy and clinical characteristics of anti-PD-1 immune checkpoint inhibitor combination chemotherapy in NSCLC patients. The efficacy of the prediction model was evaluated. Methods: A total of 220 NSCLC patients receiving first-line treatment with anti-PD-1 immune checkpoint inhibitor combined with chemotherapy were retrospectively collected. The primary endpoint was short-term efficacy ORR. The correlation between short-term efficacy, PD-L1, TMB, and clinical characteristics using χ2 test or t-test was evaluated. Screen the independent prognostic factors using univariate and multivariate logistic regression analyses, and construct a nomogram prediction model using the "rms" package in R software. Using receiver operating characteristic (ROC) curve analysis to evaluate the independent Prognostic factors and the prediction model. Using decision curve analysis (DCA) to verify the superiority of the prediction model. Results: The mean values of PD-L1, TMB, neutrophils, lymphocytes, neutrophil-to-lymphocyte ratio, and albumin were the highest in the ORR group, PD-L1 expression and TMB correlated with epidermal growth factor receptor expression. Multivariate analyses showed that PD-L1, TMB, and neutrophil were independent prognostic factors for ORR. The area under the ROC curve (AUC) values of the ROC constructed based on these three indicators were 0.7104, 0.7139, and 0.7131, respectively. The AUC value under the ROC of the nomogram model was 0.813. The DCA of the model showed that all three indicators used together to build the prediction model of the net return were higher than those of the single indicator prediction model. Conclusion: PD-L1, TMB, and neutrophils are independent prognostic factors for short-term efficacy. The nomogram prediction model constructed using these three indicators can further improve predictive efficacy of ICIs in patients with NSCLC.

Background: The role of Ferroptosis in the course of sepsis-induced myopathy is yet unclear. The objective of our work is to identify key genes connected with Ferroptosis in sepsis-induced myopathy and investigate possible pharmaceutical targets related to this process. This research aims to provide new insights into the management of sepsis-induced myopathy. Methods: We got the GSE13205 dataset from the Gene Expression Omnibus (GEO) and extracted Ferroptosis-associated genes from the FerrDb database. After conducting a functional annotation analysis of these genes, we created a protein-protein interaction network using Cytoscape software to identify important genes. Subsequently, we employed CMap to investigate prospective pharmaceuticals that could target these crucial genes. Results: A total of 61 genes that are expressed differently (DEGs) have been found concerning Ferroptosis. These genes are involved in a wide range of biological functions, including reacting to signals from outside the cell and the availability of nutrients, programmed cell death, controlling apoptosis, and responding to peptides, chemical stressors, and hormones. The KEGG pathway study revealed that these pathways are involved in Ferroptosis, autophagy, P53 signaling, PI3K-Akt signaling, mTOR signaling, HIF-1 signaling, endocrine resistance, and different tumorigenic processes. In addition, we created a network that shows the simultaneous expression of important genes and determined the top 10 medications that have the potential to treat sepsis-induced myopathy. Conclusion: The bioinformatics research undertaken sheds insight into the probable role of Ferroptosis-associated genes in sepsis-induced myopathy. The identified critical genes show potential as therapeutic targets for treating sepsis-induced myopathy, offering opportunities for the development of tailored medicines.

Engaging in behaviors that take advantage of one's personal strengths at work can promote employee flourishing in the workplace and mental health. Personal strengths use has thus gained increasing attention within occupational psychology and positive organizational scholarship. In this article, we first integrate work on personal strengths use with the latest developments in the job demands-resources theory (and its extensions) to develop a conceptual model explaining how and why personal strengths use on the job increases work engagement. Specifically, we propose that feelings of inspiration and meaningfulness explain the relationship between personal strengths use and work engagement. Second, we identify two mechanisms through which employees can amplify the benefits associated with personal strengths use at work; that is, we propose that the increased engagement associated with strengths use makes employees more likely to capitalize on the positive aspects of their work by engaging in work-family interpersonal capitalization and positive work reflection. Further, our model predicts that employees' psychological capital moderates the effects of personal strengths use. We tested our theoretical predictions in a sample of 160 full-time employees who provided ratings that comprise a three-level data set (person, week, and day) comprising 943 matched weekly ratings and 2,787 daily ratings. Synthesized allophane was employed in anaerobic digestion of chicken manure to improve the stability and methane production under ammonia inhibition. Adding 0.5 %, 1.0 % and 1.5 % (w/w) allophane increased the methane production by 261 âˆ¼ 350 % compared with the group without allophane addition. Further investigation indicated that the maximum adsorption capacity of allophane for NH4+-N achieved at 261.9 mg/g; it suggested that allophane adsorption potentially alleviated the ammonia inhibition, which also was reflected by the increase in the activity of the related enzyme, such as coenzyme F420. Moreover, allophane addition also intensified the direct interspecies electron transfer (DIET) in anaerobic digestion; it can be well supported by the increased relative abundance of Methanosaeta and Methanosarcina involved in the DIET. Overall, the improved anaerobic digestion via alleviating ammonia inhibition and intensifying DIET by allophane was elucidated comprehensively, which can contribute to the development of a functional additive for efficient anaerobic digestion in practical application.

High-entropy semiconductors are now an important class of materials widely investigated for thermoelectric applications. Understanding the impact of chemical and structural heterogeneity on transport properties in these compositionally complex systems is essential for thermoelectric design. In this work, we uncover the polar domain structures in the high-entropy PbGeSnSe1.5Te1.5 system and assess their impact on thermoelectric properties. We found that polar domains induced by crystal symmetry breaking give rise to well-structured alternating strain fields. These fields effectively disrupt phonon propagation and suppress the thermal conductivity. We demonstrate that the polar domain structures can be modulated by tuning crystal symmetry through entropy engineering in PbGeSnAgxSbxSe1.5+xTe1.5+x. Incremental increases in the entropy enhance the crystal symmetry of the system, which suppresses domain formation and loses its efficacy in suppressing phonon propagation. As a result, the room-temperature lattice thermal conductivity increases from κL = 0.63 Wm-1 K-1 (x = 0) to 0.79 Wm-1 K-1 (x = 0.10). In the meantime, the increase in crystal symmetry, however, leads to enhanced valley degeneracy and improves the weighted mobility from µw = 29.6 cm2 V-1 s-1 (x = 0) to 35.8 cm2 V-1 s-1 (x = 0.10). As such, optimal thermoelectric performance can be achieved through entropy engineering by balancing weighted mobility and lattice thermal conductivity. This work, for the first time, studies the impact of polar domain structures on thermoelectric properties, and the developed understanding of the intricate interplay between crystal symmetry, polar domains, and transport properties, along with the impact of entropy control, provides valuable insights into designing GeTe-based high-entropy thermoelectrics.

Objective: To explore the prognostic value and correlation between the risk of lymph node metastasis (LNM) and Guanylate-binding Protein 1 (GBP1) in breast cancer (BC) patients. Methods: In this retrospective study, the clinical data of 150 patients with BC who were surgically resected in The Affiliated Qingdao Central Hospital of Qingdao University from January 2019 to December 2021 were included. Patients were divided into metastasis group (n=110) or non-metastasis group (n=40) according to whether there was LNM post-surgery. Logistic regression was used to analyze the risk factors for LNM in BC, and Kaplan-Meier was used to assess the risk of disease progression 12 months post-operation in both groups. Patients were divided into a GBP1 low expression-group (n=75) or a GBP1 high expression-group (n=75). The risk of disease progression, one-year post-surgery was analyzed, and the predictive value of GBP1 in BC tissue was assessed by the receiver operating characteristics (ROC) curve. Results: Independent risk factors for BC with LNM were GBP1, CEA and TNM stage (P<0.05). There is a linear relationship between GBP1 expression and LNM risk in BC (χ2=0.88, P<0.05). Patients with high expression of GBP1 had a higher risk of LNM (χ2=3.204, P<0.001) and early postoperative progression (χ2=7.412, P<0.05). The AUC of GBP1 in predicting the risk of LNM was 0.840. Conclusions: Patients with BC and a higher expression of GBP1 could be at an increased risk of LNM. Elevations in GBP1 expression can also suggest a poor prognosis for patients with BC.

Utilization of in situ/operando methods with broad beams and localized probes has accelerated our understanding of fluid-surface interactions in recent decades. The closed-cell microchips based on silicon nitride (SiNx) are widely used as "nanoscale reactors" inside the high-vacuum electron microscopes. However, the field has been stalled by the high background scattering from encapsulation (typically ~100 nanometers) that severely limits the figures of merit for in situ performance. This adverse effect is particularly notorious for gas cell as the sealing membranes dominate the overall scattering, thereby blurring any meaningful signals and limiting the resolution. Herein, we show that by adopting the back-supporting strategy, encapsulating membrane can be reduced substantially, down to ~10 nanometers while maintaining structural resiliency. The systematic gas cell work demonstrates advantages in figures of merit for hitherto the highest spatial resolution and spectral visibility. Furthermore, this strategy can be broadly adopted into other types of microchips, thus having broader impact beyond the in situ/operando fields.

Absorptive hypercalciuria (AH) is a prevalent cause of kidney stones, and the adenylate cyclase 10 (ADCY10) gene is a rare causative gene of AH. This study aims to investigate the genotypic and phenotypic characteristics of patients with AH caused by ADCY10 gene mutations. Whole-exome sequencing and Sanger sequencing were performed on the probands and their family members, respectively. Clinical and genetic data of patients with AH caused by ADCY10 gene mutations were collected and analysed retrospectively from the present study and published literature. Two female patients (6 years old and 1 year old) with multiple bilateral kidney stones were found to have a heterozygous c.3304T>C mutation and a heterozygous c.1726C>T mutation in the ADCY10 gene. Urinary metabolite analysis revealed that urine calcium / creatinine ratios were 0.95 mmol/mmol and 1.61 mmol/mmol, respectively. Both patients underwent thiazide intake postoperatively, and upon reexamination, urine calcium decreased to within the normal range. A total of 61 patients with AH were reported from previous and present studies. The sex ratio was 7:5 for males to females, and the mean age of onset was 23.61±20.08 years. A total of 16 ADCY10 gene mutations were identified, including seven missense (43.75%), five splicing (31.25%), two frameshift (12.50%) and two nonsense mutations (12.50%). Only two cases were identified as homozygous mutations (c.1205_1206del), and the others were heterozygous mutations. In summary, we identified two novel ADCY10 gene candidate pathogenic variants in Chinese pediatric patients, which expands the mutational spectrum of the ADCY10 gene and provides a potential diagnostic and therapeutic target.

OBJECTIVES: A decline in language function is a common complication after glioma surgery, affecting patients' quality of life and survival. This study predicts the postoperative decline in language function and whether it can be recovered based on the preoperative white matter structural network. MATERIALS AND METHODS: Eighty-one right-handed patients with glioma involving the left hemisphere were retrospectively included. Their language function was assessed using the Western Aphasia Battery before and 1 week and 3 months after surgery. Structural connectome combining DTI features was selected to predict postoperative language decline and recovery. Nested cross-validation was used to optimize the models, evaluate the prediction performance of the models, and identify the most predictive features. RESULTS: Five, seven, and seven features were finally selected as the predictive features in each model and used to establish predictive models for postoperative language decline (1 week after surgery), long-term language decline (3 months after surgery), and language recovery, respectively. The overall accuracy of the three models in nested cross-validation and overall area under the receiver operating characteristic curve were 0.840, 0.790, and 0.867, and 0.841, 0.778, and 0.901, respectively. CONCLUSION: We used machine learning algorithms to establish models to predict whether the language function of glioma patients will decline after surgery and whether postoperative language deficit can recover, which may help improve the development of treatment strategies. The difference in features in the non-language decline or the language recovery group may reflect the structural basis for the protection and compensation of language function in gliomas. CLINICAL RELEVANCE STATEMENT: Models can predict the postoperative language decline and whether it can recover in glioma patients, possibly improving the development of treatment strategies. The difference in selected features may reflect the structural basis for the protection and compensation of language function. KEY POINTS: • Structural connectome combining diffusion tensor imaging features predicted glioma patients' language decline after surgery. • Structural connectome combining diffusion tensor imaging features predicted language recovery of glioma patients with postoperative language disorder. • Diffusion tensor imaging and connectome features related to language function changes imply plastic brain regions and connections.

Skin aging is characterized by wrinkle formation and increased frailty and laxity, leading to the risk of age-related skin diseases. Keratinocyte is an important component of the epidermis in skin structure, and keratinocyte senescence has been identified as a pivotal factor in skin aging development. Because epigenetic pathways play a vital role in the regulation of skin aging, we evaluated human skin samples for DNA hydroxymethylation (5-hydroxymethylcytosine; 5-hmC) and SIRT4 expressions. Results found that both 5-hmC and SIRT4 showed a significant decrease in aged human skin samples. To test the results in vitro, human keratinocytes were cultured in H2O2, which modulates skin aging in vivo. However, H2O2-induced keratinocytes showed senescence-associated protein expression and significant downregulation of 5-hmC and SIRT4 expressions. Moreover, 5-hmC-converting enzymes ten eleven translocation 2 (TET2) showed a decrease and enhanced TET2 acetylation level in H2O2-induced keratinocytes. However, the overexpression of SIRT4 in keratinocytes alleviates the senescence phenotype, such as senescence-associated protein expression, decreases the TET2 acetylation, but increases TET2 and 5-hmC expressions. Our results provide a novel relevant mechanism whereby the epigenetic regulation of keratinocytes in skin aging may be correlated with SIRT4 expression and TET2 acetylation in 5-hmC alteration. Our study may provide a potential strategy for antiskin aging, which targets the SIRT4/TET2 axis involving epigenetic modification in keratinocyte senescence.

Resistance (R) genes in the Triticeae tribe include not only genes encoding the canonical intracellular nucleotide-binding leucine-rich-repeat proteins (NLRs) but also genes encoding kinase fusion proteins (KFPs). Exploring these unconventional KFPs may expand the scope of effector-triggered immunity (ETI) and will have significant implications for crop improvement.

Herein, a novel oxygen- enriched melting process for fly ash, which uses the biogas produced from the leachate of municipal solid waste incineration (MSWI) plants, is proposed to reduce the high cost of conventional fly ash - melting technology. The fly ash composition was estimated via X-ray fluorescence analysis; the six constituent elements detected in fly ash in the decreasing order of their content were calcium, chlorine, silicon, sulfur, sodium, and potassium. Based on literature and actual production data, the average yield of the leachate was 15% of the total waste entering the MSWI plants and the COD of leachate was 30,000-75,000 mg/L. The amount of biogas that can be used per ton of fly ash was calculated to be 62.0-157.0 m3. The analysis of melting thermal equilibrium revealed the amount of biogas required per ton of fly ash as 57.8 m3. The aforementioned research findings indicate that the biogas produced by MSWI plants can successfully meet the demands of the oxygen- enriched melting of fly ash produced in these plants. By establishing an oxygen- enriched- melting pilot platform, the pilot tests of melting were conducted on fly ash; the results revealed the good melting effects of fly ash. The X-ray diffraction analysis of the slag demonstrated that the content of the vitreous body met the technical requirements for glassy substances. Furthermore, the leaching toxicity test results revealed that heavy metals were well solidified in the slag. This study presents a novel fly ash - melting scheme for MSWI fly ash, namely, biogas oxygen- enriched melting strategy, which has the advantages of technical feasibility and cost- effectiveness. The proposed technique exhibits considerable prospects for widespread application in MSWI plants in China and can play an important role in the safe disposal of fly ash.Implications: In this paper, a low- cost melting method of municipal solid waste incineration(MSWI) fly ash is proposed. This method uses the biogas generated by MSWI plant itself as fuel for melting. Through research, it has been found that the production of biogas can meet the demand for fly ash melting. Adopting biogas as a molten fuel can significantly reduce the cost of melting, thereby significantly reducing the cost of fly ash melting. This study established a pilot scale platform for the melting of biogas and conducted pilot scale experiments on fly ash and additives. The experimental results showed that the melting system operated well and achieved the vitrification of fly ash. The leaching test results of the molten slag showed that heavy metals were well solidified in the slag. The research results can be extended to the MSWI plant for application, which can significantly reduce the cost of fly ash melting disposal, and has broad application prospects.

In this case report, a 53-year-old woman was diagnosed with severe NE after receiving chemotherapy for breast cancer. The patient with breast cancer was treated with a single cycle of docetaxel (140 mg) + epirubicin (130 mg) + cyclophosphamide (0.9 g) chemotherapy. However, the woman presented with symptoms of fatigue and diarrhea 5 days later accompanied with severe neutropenia according to the routine blood test. The computed tomography examination displayed the thickening and swelling of the colorectal wall. After the diagnosis of NE, the woman received antibiotics and supportive treatment, but her symptoms were not improved. The Chinese herbal medicine (CHM) diagnostic pattern was then designed for the patient. The patient was administered with two CHM decoctions. One decoction contained 24 kinds of herbal materials, and the other one was called pure ginseng decoction. These two decoctions were administered to the patient 2 or 3 times per day to tonify the spleen, nourish Qi and blood, and remove phlegm and damp heat symptoms. After the CHM treatment lasting for 10 days, the symptoms of the patient were improved, and she was discharged. In conclusion, CHM treatment played an indispensable role in curing the woman with chemotherapy-induced NE.

In this study, a static railway track smoothness detection system based on laser reference, which can measure various track smoothness parameters by using multiple sensors, is proposed. Furthermore, in order to improve the measurement accuracy and stability of the system, this paper also conducted three key analyses based on the static track measurement system. By using a liquid double-wedge automatic compensation device to compensate the horizontal angle of the beam, a mathematical model of liquid double-wedge automatic compensation was established. Then, by using an optical ring grating system to ring-grate and characterize the laser spot, the collimation efficiency of the system was improved when measuring at long distances. For the special ring grating spot image, an adaptive image processing algorithm was proposed, which can achieve sub-pixel-level positioning accuracy. This study also conducted a field measurement experiment, comparing the experimental data obtained via the static track measurement system with the results of existing track measurement products, and verifying that the static track measurement system has high measurement accuracy and stability.

Covalent organic frameworks (COFs) create extended two-dimensional (2D) skeletons and aligned one-dimensional (1D) channels, constituting a class of novel π architectures with predesignable structural ordering. A distinct feature is that stacks of π building units in skeletons shape the pore walls, onto which a diversity of different units can be assembled to form various pore interfaces, opening a great potential to trigger a strong structural correlation between the skeleton and the pore. However, such a possibility has not yet been explored. Herein, we report reversible three-dimensional (3D) coalescence and interlocked actions between the skeleton and pore in COFs by controlling hydrogen-bonding networks in the pores. Introducing carboxylic acid units to the pore walls develops COFs that can confine water molecular networks, which are locked by the surface carboxylic acid units on the pore walls via multipoint, multichain, and multidirectional hydrogen-bonding interactions. As a result, the skeleton undergoes an interlocked action with pores to shrink over the x-y plane and to stack closer along the z direction upon water uptake. Remarkably, this interlocked action between the skeleton and pore is reversibly driven by water adsorption and desorption and triggers profound effects on π electronic structures and functions, including band gap, light absorption, and emission.

Extracellular vesicles (EVs) can mediate intercellular communication, including signaling between the placenta and maternal tissues. Human placental explant culture is a versatile in vitro model system to investigate placental function. We performed systematic studies in different tissue culture media types and oxygen tensions to identify a defined serum-free culture condition that supports high trophoblast viability and metabolism, as well as the release of similar populations of EVs, compared to traditional undefined conditions that contain media additives potentially contaminated with exogenous EVs. We also determined the time frame in which trophoblast viability and functionality remain optimal. Multiplex vesicle flow cytometry with classical EV and placenta-specific markers revealed three separate populations of explant-derived EVs: small CD63+ EVs; large PLAP+ EVs; and CD63-/PLAP- EVs. These culture and analytical approaches will enable in vitro modeling of short-term effects of environmental perturbations associated with pregnancy complications on placental function and EV release.

Hyphantria cunea (Drury), a destructive polyphagous pest, has been spreading southward after invading northern China, which indicates that this insect species is facing a huge thermal challenge. Small heat shock proteins (sHSPs) function as ATP-independent molecular chaperones that protect insects from heat stress damage. In order to explore the role of sHSPs in the thermotolerance of H. cunea, five novel sHSP genes of H. cunea were cloned, including an orthologous gene (HcHSP21.4) and four species-specific sHSP genes (HcHSP18.9, HcHSP20.1, HcHSP21.5, and HcHSP29.8). Bioinformatics analysis showed that the proteins encoded by these five HcHSPs contained typical α-crystallin domains. Quantitative real-time PCR analysis revealed the ubiquitous expression of all HcHSPs across all developmental stages of H. cunea, with the highest expression levels in pupae and adults. Four species-specific HcHSPs were sensitive to high temperatures. The expression levels of HcHSPs were significantly up-regulated under heat stress and increased with increasing temperature. The expression levels of HcHSPs in eggs exhibited an initial up-regulation in response to a temperature of 40 °C. In other developmental stages, the transcription of HcHSPs was immediately up-regulated at 30 °C or 35 °C. HcHSPs transcripts were abundant in the cuticle before and after heat shock. The expression of HcHSP21.4 showed weak responses to heat stress and constitutive expression in the tissues tested. These results suggest that most of the HcHSPs are involved in high-temperature response and may also have functions in the normal development and reproduction of H. cunea.
