RESUMO

Despite significant efforts to control cancer progression and to improve oncology treatment outcomes, recurrence and tumor resistance are frequently observed in cancer patients. These problems are partly related to the presence of cancer stem cells (CSCs). Photodynamic therapy (PDT) has been developed as a therapeutic approach for solid tumors; however, it remains unclear how this therapy can affect CSCs. In this review, we focus on the effects of PDT on CSCs and the possible changes in the CSC population after PDT exposure. Tumor response to PDT varies according to the photosensitizer and light parameters employed, but most studies have reported the successful elimination of CSCs after PDT. However, some studies have reported that CSCs were more resistant to PDT than non-CSCs due to the increased efflux of photosensitizer molecules and the action of autophagy. Additionally, using different PDT approaches to target the CSCs resulted in increased sensitivity, reduction of sphere formation, invasiveness, stem cell phenotype, and improved response to chemotherapy. Lastly, although mainly limited to in vitro studies, PDT, combined with targeted therapies and/or chemotherapy, could successfully target CSCs in different solid tumors and promote the reduction of stemness, suggesting a promising therapeutic approach requiring evaluation in robust pre-clinical studies.

Background: Oral squamous cell carcinoma (OSCC) accounts for 90% of oral malignancies, which may be preceded by oral potentially malignant disorders (OPMDs). Cancer progression involves the downregulation of epithelial markers (E-cadherin) and the upregulation of mesenchymal markers (N-cadherin), which together characterise the epithelial-mesenchymal transition (EMT). Furthermore, caveolin can act on cell adhesion and migration events that regulate the expression of the E-cadherin/α-ß-catenin complex, thus favouring aggressive biological behaviour. This study aimed to analyse the immunoexpression of E-cadherin, N-cadherin and caveolin-2 at different stages of oral carcinogenesis to identify reliable biomarkers to predict malignant potential. Methods: Expressions of E-cadherin and N-cadherin in 14 normal oral mucosae (NOM), 14 OPMD and 33 OSCC specimens were evaluated using immunohistochemistry. Clinicopathological parameters were also assessed. Results: E-cadherin immunoexpression was significantly reduced during the progression of oral carcinogenesis (P = 0.0018). N-cadherin immunoexpression did not show any statistical differences between these groups. However, a representative number of N-cadherin-positive OSCC cases did not express E-cadherin. The expression of caveolin-2 increased significantly with the progression of the disease, from NOM to OSCC (P value: 0.0028). Conclusion: These findings indicate that cadherin switch and caveolin-2 immunoexpression may be regulatory events in oral carcinogenesis.

ABSTRACT Introduction: Vertebral fracture is the main complication of osteoporosis and is common among the elderly. Conservative treatment is the first choice for osteoporotic vertebral compression fractures (OVCF) but for persistent painful cases, percutaneous vertebral cement augmentation techniques, such as vertebroplasty and kyphoplasty, are indicated. We performed a systematic review to compare clinical and radiological outcomes of both methods. Methods: A systematic review was performed according to the PRISMA and Cochrane Handbook for Systematic Reviews of Interventions. The PICO search strategy consisted of the following terms: Population- Patients with OVCFs; Intervention- Kyphoplasty; Control- Vertebroplasty; Outcomes- Pain, Cement Leakage, Vertebral Body Height, Adjacent level fractures, Oswestry (ODI) and SF36. Results: Seven articles were included in the qualitative analysis, selecting only randomized controlled trials. Four hundred and fifty patients were treated with vertebroplasty (VP) and 469 with kyphoplasty (KP). The leakage rate of the VP group was 63% versus 14% for the KP group. However, these results were without statistical significance. The Visual Analogue Scale (VAS), ODI and SF-36 outcomes were evaluated based on the 6-month and 1-year follow-up results, and we were unable to find any significant differences between treatments. For restoration of vertebral height, the values of the KP group were, on average, 0.71 cm higher than those of the VP group, with 95% CI. Conclusion: Based on this systematic review, kyphoplasty is superior to vertebroplasty for achieving gains in vertebral body height. As regards cement leakage and other clinical outcomes, neither method showed statistically significant superiority. Level of Evidence I; Systematic review.

O tratamento conservador é a primeira escolha para fraturas compressivas vertebrais por osteoporose (FCVO), mas para casos dolorosos persistentes, as técnicas de cimentação vertebral, como vertebroplastia e cifoplastia, são indicadas. Realizamos uma revisão sistemática para comparar os resultados clínicos e radiológicos de ambos os métodos. Métodos: Uma revisão sistemática foi realizada de acordo com o PRISMA e o Manual Cochrane de Revisões Sistemáticas. A estratégia de busca PICO foi: População - Pacientes com FCVOs; Intervenção - Cifoplastia; Controle - Vertebroplastia; Resultados - Dor, Extravazamento de Cimento, Altura do Corpo Vertebral, Fraturas em Nível Adjacente, Oswestry (ODI) e SF36. Resultados: Sete artigos foram incluídos na análise qualitativa, somente ensaios clínicos randomizados. Quatrocentos e cinquenta pacientes foram tratados com vertebroplastia (VP) e 469 com cifoplastia (CP). A taxa de extravazamento de cimento do grupo VP foi de 63% contra 14% do CP, no entanto, não atingiu significância estatística. Os desfechos da Escala Visual Analógica (EVA), ODI e SF-36 foram avaliados considerando os resultados de seis meses e um ano de seguimento e não pudemos apontar diferenças entre os tratamentos. Por fim, a CP apresenta valores médios 0,71 cm maiores do que a VP para a restauração da altura do corpo vertebral, com IC de 95%. Conclusão: Nesta revisão sistemática a cifoplastia foi superior à vertebroplastia para ganho de altura do corpo vertebral. Não houve superioridade estatisticamente significativa entre os dois métodos para extravazamento de cimento e outros resultados clínicos. Nível de Evidência I; Revisão sistemática

Se realizó una revisión sistemática para comparar los resultados clínicos y radiológicos de ambos métodos. Métodos: Se llevó a cabo una revisión sistemática de acuerdo con la declaración PRISMA y el Manual Cochrane de Revisiones Sistemáticas. La estrategia de búsqueda PICO fue: Población: Pacientes con FCVO; Intervención: Cifoplastia; Control- Vertebroplastia; Resultados: Dolor, Extravasación del cemento, Altura del Cuerpo Vertebral, Fracturas de Nivel Adyacente, Oswestry (ODI) y SF36. Los resultados de la Escala Visual Analógica (EVA), ODI y SF-36 se evaluaron teniendo en cuenta los resultados de 6 meses y 1 año de seguimiento y no pudimos señalar diferencias entre los tratamientos.. Finalmente, el CP presenta valores promedios 0,71 cm superiores al VP para restaurar la altura del cuerpo vertebral, con un IC del 95%. Conclusión: En esta revisión sistemática, la cifoplastia fue superior a la vertebroplastia para el aumento de altura del cuerpo vertebral. ABSTRACT Objective To describe the epidemiological and clinical characteristics of patients with pyogenic spondylodiscitis treated in a Brazilian hospital. Methods This is a retrospective study of patients diagnosed with nonspecific spondylodiscitis. Patients of both sexes, above 18 years of age with a minimum follow-up time of 6 months were included. Epidemiological, laboratory, and clinical data were analyzed. Results Nine patients were included. The mean age was 64 years, with seven men (77.7%) and two women (22.2%). All patients evaluated had back pain. The most affected location was the lumbar spine (44.4%). Only three patients (33.3%) had fever and five (55%) had constitutional symptoms. The mean duration of symptoms before diagnosis was 2.5 (± 1.5) weeks. Only four patients (44.4%) had positive cultures. As for neurological status, five patients (55.5%) presented neurological change. At the end of treatment, two patients improved one level in the Frankel score and two patients improved two levels. The main indication for surgery was neurological deficit (55.5%). Two of the patients evaluated died as a result of an infectious condition. Conclusions Less than half of the patients with pyogenic spondylodiscitis had fever or constitutional symptoms. Back pain was present in all cases. In less than half of the patients it was possible to isolate the responsible microorganism. Most patients underwent surgical treatment, although not all improved from the neurological deficit. RESUMO Objetivo Descrever as características epidemiológicas e clínicas de pacientes com espondilodiscite piogênica tratados em um hospital brasileiro. Métodos Trata-se de estudo retrospectivo de pacientes com diagnóstico de espondilodiscite inespecífica. Foram incluídos pacientes de ambos os sexos, acima de 18 anos, com tempo de seguimento mínimo de seis meses. Foram analisados dados epidemiológicos, laboratoriais e clínicos. Resultados Nove pacientes foram incluídos. A média de idade foi de 64 anos, sendo sete homens (77,7%) e duas mulheres (22,2%). Todos os pacientes avaliados tinham dorsalgia. O local mais acometido foi a coluna lombar (44,4%). Apenas três pacientes (33,3%) apresentaram febre e cinco (55%), sintomas constitucionais. O tempo médio de sintomas antes do diagnóstico foi de 2,5 (± 1,5) semanas. Apenas quatro pacientes (44,4%) tiveram culturas positivas. Cinco pacientes (55,5%) apresentaram alteração neurológica. Ao término do tratamento, dois pacientes melhoraram um nível no escore de Frankel, dois pacientes melhoraram dois níveis. A principal indicação para cirurgia foi déficit neurológico (55,5%). Dois pacientes avaliados foram a óbito em decorrência do quadro infeccioso. Conclusões Menos da metade dos pacientes com espondilodiscite piogênica tiveram febre ou sintomas constitucionais. A dorsalgia estava presente em todos os casos. Em menos da metade dos pacientes foi possível isolar o microrganismo responsável. A maioria dos pacientes foi submetida a tratamento cirúrgico, embora nem todos tiveram melhora do déficit neurológico. Nível de evidência II; Estudo Retrospectivo.

Macrophages play a very important role in the conduction of several regenerative processes mainly due to their plasticity and multiple functions. In the muscle repair process, while M1 macrophages regulate the inflammatory and proliferative phases, M2 (anti-inflammatory) macrophages direct the differentiation and remodelling phases, leading to tissue regeneration. The aim of this study was to evaluate the effect of red and near infrared (NIR) photobiomodulation (PBM) on macrophage phenotypes and correlate these findings with the repair process following acute muscle injury. Wistar rats were divided into 4 groups: control; muscle injury; muscle injury + red PBM; and muscle injury + NIR PBM. After 2, 4 and 7 days, the tibialis anterior muscle was processed for analysis. Macrophages phenotypic profile was evaluated by immunohistochemistry and correlated with the different stages of the skeletal muscle repair by the qualitative and quantitative morphological analysis as well as by the evaluation of IL-6, TNF-α and TGF-ß mRNA expression. Photobiomodulation at both wavelengths was able to decrease the number of CD68+ (M1) macrophages 2 days after muscle injury and increase the number of CD163+ (M2) macrophages 7 days after injury. However, only NIR treatment was able to increase the number of CD206+ M2 macrophages (Day 2) and TGF-ß mRNA expression (Day 2, 4 and 7), favouring the repair process more expressivelly. Treatment with PBM was able to modulate the inflammation phase, optimize the transition from the inflammatory to the regeneration phase (mainly with NIR light) and improve the final step of regeneration, enhancing tissue repair.

The use of colorimetric methods for protein quantification in microalgae is hindered by their elevated amounts of membrane-embedded intracellular proteins. In this work, the protein content of three species of microalgae was determined by the Lowry method after the cells were dried, ball-milled, and treated with the detergent sodium dodecyl sulfate (SDS). Results demonstrated that the association of milling and SDS treatment resulted in a 3- to 7-fold increase in protein quantification. Milling promoted microalgal disaggregation and cell wall disruption enabling access of the SDS detergent to the microalgal intracellular membrane proteins and their efficient solubilization and quantification.

Fertilization rates and subsequent embryo development rely on sperm factors related to semen quality and viability. Photobiomodulation therapy (PBMT) is based on emission of electromagnetic waves of a laser optical system that interact with cells and tissues resulting in biological effects. This interaction is mediated by photoacceptors that absorb the electromagnetic energy. Effects are dependent of irradiation parameters, target cell type, and species. In sperm, PBMT improves several features like motility and viability, affecting sperm aerobic metabolism and energy production. The aim of this study was to investigate, under same conditions, how different output powers (5, 7.5, and 10 mW) and time of irradiation (5 and 10 min) of laser (He-Ne laser, 633 nm) may affect frozen/thawed bovine sperm functions. Results showed significant effects depending on power while using 10 min of irradiation on motility parameters and mitochondrial potential. However, no effect was observed using 5 min of irradiation, regardless of power applied. In conclusion, PBMT is effective to modulate bovine sperm function. The effectiveness is dependent on the interaction between power applied and duration of irradiation, showing that these two parameters simultaneously influence sperm function. In this context, when using the same fluency and energy with different combinations of power and time of exposure, we observed distinct effects, revealing that biological effects should be also based on simple parameters rather than only composite parameters such as fluency, irradiance and energy. Laser irradiation of frozen/thawed bovine semen led to an increase on mitochondrial function and motility parameters that could potentially improve fertility rates.

Epigenetics have essential roles in development and human diseases. Compared to the complex histone modifications, epigenetic changes on mammalian DNA are as simple as methylation on cytosine. Guanine, however, can be oxidized as an epigenetic change which can undergo base-pair transversion, causing a genetic difference. Accumulating evidence indicates that reactive oxygen species (ROS) are important signaling molecules for embryonic stem cell (ESC) differentiation, possibly through transient changes on genomic DNA such as 7,8-dihydro-8-oxoguanine (8-oxoG). Technical limitations on detecting such DNA modifications, however, restrict the investigation of the role of 8-oxoG in ESC differentiation. Here, we developed a Hoogsteen base pairing-mediated PCR-sequencing assay to detect 8-oxoG lesions that can subsequently cause G to T transversions during PCR. We then used this assay to assess the epigenetic and transient 8-oxoG formation in the Tbx5 gene of R1 mouse ESCs subjected to oxidative stress by removing 2-mercaptoethanol (2ME) from the culture media. To our surprise, significantly higher numbers of 8-oxoG-mediated G∙C to C∙G transversion, not G∙C to T∙A, were detected at 7th and 9th base position from the transcription start site of exon 1 of Tbx5 in ESCs in the (-)2ME than (+)2ME group (p < 0.05). This was consistent with the decrease in the amount of amplifiable of DNA harboring the 8-oxoG lesions at the Tbx5 promoter region in the oxidative stressed ESCs. The ESCs responded to oxidative stress, possibly through the epigenetic effects of guanine oxidation with decreased proliferation (p < 0.05) and increased formation of beating embryoid bodies (EBs; p < 0.001). Additionally, the epigenetic changes of guanine induced up-regulation of Ogg1 and PolB, two base excision repairing genes for 8-oxoG, in ESCs treated with (-)2ME (p < 0.01). Together, we developed a gene-specific and direct quantification assay for guanine oxidation. Using oxidative stressed mouse ESCs, we validated this assay and assessed the epigenetic effects of 8-oxoG by studying expression of DNA repair genes, ESC proliferation, and EB formation.

Degenerative changes of the endometrium are directly related to age and fertility in mares. Chronic degenerative endometritis (CDE) is correlated with uterine fluid retention and reduced ability to clear uterine inflammation. Recent research in the areas of equine surgery and sports medicine has shown that platelet-rich plasma (PRP) treatment acts as an immunomodulator of the inflammatory response. Therefore, the aim of this study was to determine if the uterine infusion of PRP could modulate the local inflammatory response and modify the intrauterine NO concentrations after artificial insemination (AI) in both normal mares and those with CDE. Thirteen mares with endometrium classified as grade III on the histology (mares with CDE) and eight mares with endometrial histological classification I or II-a normal mares were selected to investigate the effect of PRP therapy. The mares were inseminated with fresh semen in two consecutive cycles in a crossover study design. Thereby, each mare served as its own control and the treatment was performed with intrauterine PRP infusion four hours after AI. The percentage of neutrophils in uterine cytology (CIT, %), uterine fluid accumulation observed on ultrasonography (FLU, mm) and nitric oxide concentration of uterine fluid (NO, µM) were analyzed before and 24 hours after AI. The results reported that mares with CDE (CIT, 68.3 ± 3.27, FLU, 10.7 ± 1.61) have a higher (P < 0.05) intrauterine inflammatory response after AI than normal mares (CIT, 24.4 ± 3.56, FLU, 0), but NO concentrations did not differ (P > 0.05) between categories of mares. In treated cycles with PRP, the intrauterine inflammatory response decrease (P < 0.05) in CDE mares (CDE: CIT, 31.4 ± 6.48, FLU, 5.5 ± 1.28; normal mares: CIT, 13.5 ± 4.31, FLU, 0) when compared with nontreated cycle (CDE: CIT, 68.3 ± 3.27, FLU, 10.7 ± 1.61; NM: CIT, 24.4 ± 3.56, FLU, 0), but did not modify NO concentrations in uterine fluid. Thus, we can conclude that PRP was effective in modulating the exacerbated uterine inflammatory response to semen in mares with CDE but did not reduce NO concentrations in intrauterine fluid.

Focused Clinical Question: In patients with normal crown dimensions and excessive gingival display, does lip-repositioning surgery improve long-term smile outcome and dental esthetics? Clinical Scenario: A 27-year-old systemically healthy female presents with the chief complaint of "excessive gingival display when smiling" (Fig. 1). She reports that orthodontic treatment was performed and that she was referred for orthognathic surgery to correct the maxillary vertical excess and altered passive eruption. Clinically, the patient is periodontally healthy with favorable long-term periodontal prognosis. She is unsatisfied with her smile and states that it is "compromising her self-esteem." However, she refuses to accept the orthognathic surgery because of the extensiveness and morbidity of the procedure. She inquires about alternative solutions for her esthetic problem and whether it is possible to solve her gummy smile with a less invasive procedure. Lip-repositioning and anatomic crown-lengthening surgeries are presented as treatment options. The patient decides to pursue these treatment options. Lip-repositioning surgery is performed as described by Rosenblatt and Simon1 and Simon et al.,2 and healing is uneventful. Anatomic crown lengthening is performed 6 months after the previous surgery. The patient is pleased with the reduction of the excessive gingival display after these procedures (Fig. 2). However, there is lack of clinical evidence regarding the long-term stability of this surgical procedure.

Tremor is the most common movement disorder characterized by repetitive and stereotyped movements. The analysis of hand-drawing movements is commonly used in the evaluation of patients with tremor. The data collection conducted under kinetic conditions and while performing a voluntary movement highlights the kinetic tremor. Most investigations on tremor attempt to understand its relation to neuromuscular dysfunctions. Therefore, there is a lack of studies that aim to investigate the complex relation between the physiological tremor and aging, especially in kinetic conditions. In this context, the main motivation of this research was to generalize the results obtained in Almeida et al., a study that quantified age-related changes in the kinetic tremor of clinically healthy individuals. For this, a number of features extracted from tremor activity, obtained from digitized drawings of Archimedes' spirals, were analyzed. The analyses followed the sequence: Linearization of the Archimedes' spiral; Estimate of tremor activity; Data pre-processing; Feature extraction from the tremor activity; and Application of Linear Discriminant Analysis (LDA) technique as described in Almeida et al.. The generalization of the obtained results showed that the method based on LDA allows for a linear correlation between physiological kinetic tremor and aging.

In this context, the main motivation of this research was to quantify age-related changes in the kinetic tremor of clinically healthy individuals. For this, a number of features extracted from tremor activity, obtained from digitized drawings of Archimedes' spirals, were analyzed. In total, 59 subjects participated in the experiments. These individuals were divided into seven groups according to their ages and two types of analysis were carried out. First, the tremor activity of young and elderly adults was investigated by means of traditional features commonly used in tremor analysis, and secondly, linear discriminant analysis (LDA) was employed for the study of the correlation between age and tremor. The results showed significant statistical differences between the kinetic tremor activity of the young and elderly groups. Furthermore, it was found that LDA allowed for the estimate of a unique feature, so-called LDA-value, which showed to be linearly correlated with age.

BACKGROUND: The human body adopts a number of strategies to maintain an upright position. The analysis of the human balance allows for the understanding and identification of such strategies. The displacement of the centre of pressure (COP) is a measure that has been successfully employed in studies regarding the postural control. Most of these investigations are related to the analysis of individuals suffering from neuromuscular disorders. Recent studies have shown that the elderly population is growing very fast in many countries all over the world, and therefore, researches that try to understand changes in this group are required. In this context, this study proposes the analysis of the postural control, measured by the displacement of the COP, in groups of young and elderly adults. METHODS: In total 59 subjects participated of this study. They were divided into seven groups according to their age. The displacement of the COP was collected for each subject standing on a force plate. Two experimental conditions, of 30 seconds each, were investigated: opened eyes and closed eyes. Traditional and recent digital signal processing tools were employed for feature computation from the displacement of the COP. Statistical analyses were carried out in order to identify significant differences between the features computed from the distinct groups that could allow for their discrimination. RESULTS: Our results showed that Linear Discrimination Analysis (LDA), which is one of the most popular feature extraction and classifier design techniques, could be successfully employed as a linear transformation, based on the linear combination of standard features for COP analysis, capable of estimating a unique feature, so-called LDA-value, from which it was possible to discriminate the investigated groups and show a high correlation between this feature and age. CONCLUSION: These results show that the analysis of features computed from the displacement of the COP are of great importance in studies trying to understand the ageing process. In particular, the LDA-value showed to be an adequate feature for assessment of changes in the postural control which can be related to functional changes that occur over the ageing.

Intra-thoracic antigenic challenge (ovalbumin, 12.5 microg/cavity) led to increased numbers of gammadelta T lymphocytes in pleural cavities, blood and thoracic lymph nodes in sensitized mice within 48 h. Part of these cells expressed CD62L, which increased on gammadelta T cell surfaces obtained from lymph nodes after ovalbumin (OVA) challenge. Selectin blockade by fucoidan pre-treatment (10 mg/kg, i.v.) impaired in vivo increase in CD25(+) and c-fos(+) gammadelta T cell numbers in lymph nodes, indicating a role for selectins on gammadelta T lymphocyte activation and proliferation. In vivo selectin blockade by fucoidan or alpha-CD62L mAb (200 microg/mice, i.p.) also inhibited OVA-induced gammadelta T cell accumulation in pleural cavities. Confirming the direct effect of CD62L on gammadelta T cell transmigration, the migration of i.v. adoptively-transferred CFSE-labeled gammadelta T lymphocytes into pleural cavities of challenged recipient mice was impaired by fucoidan ex vivo treatment. It is noteworthy that eosinophil influx was also impaired in those mice, indicating that reduced eosinophil migration by CD62L in vivo blockade depended on gammadelta T cell migration via CD62L molecules. Accordingly, pleural gammadelta T lymphocytes from fucoidan-treated mice presented reduced OVA-induced IL-5 and CCL11 production. Supporting these data, the depletion of Vgamma4 T lymphocytes, which are pulmonary gammadelta T cells, decreased OVA-induced eosinophil influx into allergic site. Such results demonstrate that CD62L is crucial for the activation of gammadelta T cells in lymph nodes, for their migration into inflamed tissue and for the modulation of eosinophil influx during allergic response.

Objetivo: Observar a evolução clínica e radiográfica dos pacientes submetidos ao tratamento não operatório das fraturas da coluna toracolombar tipo A3 explosão, conforme a classificação de Margerl, na Santa Casa de Misericórdia de São Paulo. Métodos: Foram analisados retrospectivamente os dados de 17 pacientes portadores de fratura toracolombar explosão internados entre 1994 e 2003 e tratados com o uso de colete de Jewett ou gesso em hiperextensão. Todos foram avaliados por meio de parâmetros clínicos (escala de Denis e Frankel) e radiográficos. Resultados: Com seguimento mínimo de 15 meses, foram constatados resultados clínicos excelentes e bons em 15 pacientes (88,2 por cento); regular e ruim em dois (11,7 por cento). Não houve deterioração da função neurológica em nenhum caso. Dos pacientes, dois apresentavam déficit neurológico inicial (Frankel D), sem melhora até o final do seguimento. Onze (64,7 por cento) pacientes retomaram às suas atividades prévias; quatro (23,5 por cento) mudaram de atividade e dois (11,7 por cento) apresentaram incapacidade para o trabalho. A avaliação radiográfica mostrou piora média da cifose de 1,8°. A cifose residual não foi estatisticamente significativa em relação à sintomatologia ou queixa atual do paciente (p = 0,4). Conclusão: No grupo de pacientes estudados o tratamento conservador mostrou resultados satisfatórios após seguimento médio de 34,7 meses, devendo ser considerado como boa opção para o tratamento da fraturas toracolombares tipo A3 explosão em pacientes com pouco ou nenhum comprometimento neurológico

Brown spider (Loxosceles genus) venom causes necrotic lesions often accompanied by fever, hemolysis, thrombocytopenia, and acute renal failure. Using mice exposed to Loxosceles intermedia venom, we aimed to show whether the venom directly induces renal damage. The experimental groups were composed of 50 mice as controls and 50 mice that received the venom. Light microscopic analysis of renal biopsy specimens showed alterations including hyalinization of proximal and distal tubules, erythrocytes in Bowman's space, glomerular collapse, tubule epithelial cell blebs and vacuoles, interstitial edema, and deposition of eosinophilic material in the tubule lumen. Electron microscopic findings indicated changes including glomerular epithelial and endothelial cell cytotoxicity as well as disorders of the basement membrane. Tubule alterations include epithelial cell cytotoxicity with cytoplasmic membrane blebs, mitochondrial changes, increase in smooth endoplasmic reticulum, presence of autophagosomes, and deposits of amorphous material in the tubules. We also found that the venom caused azotemia with elevation of blood urea levels but did not decrease C3 complement concentration or cause hemolysis in vivo. Confocal microscopy with antibodies against venom proteins showed direct binding of toxins to renal structures, confirmed by competition assays. Double-staining immunofluorescence reactions with antibodies against type IV collagen or laminin, antibodies to venom toxins, and fluorescent cytochemistry with DAPI revealed deposition of toxins in glomerular and tubule epithelial cells and in renal basement membranes. Two-dimensional electrophoresis showed venom rich in low molecular mass and cationic toxins. By immunoblotting with antibodies to venom toxins on renal extracts from venom-treated mice, we detected a renal binding toxin at 30 kD. The data provide experimental evidence that L. intermedia venom is directly involved in nephrotoxicity.

We measured the effects in vitro of pure and commercial pesticides on Ca(2+)-activated ATPase and cholinesterase (ChE) activities in the nervous system of the slug Phyllocaulis soleiformis. The pesticides used in this study included carbamate and organophosphates, which acts as reversible and irreversible anticholinesterases, respectively. Both enzymes were insensitive to pure carbofuran (1 mM), glyphosate (1 mM) and malathion (120 microM). However, the carbamate carbofuran, in the commercial formulation Furandan 350S, inhibited ATPase and ChE activities. The organophosphate glyphosate used in the commercial preparation of Gliz 480CS inhibited ATPase activity and increased cholinesterase activity. These effects are likely due to the action of adjuvant substances of the chemical formulation. The commercial formulation (Malatol 500CE) did not alter enzymes activities. Our results suggest that cholinesterase present in the slug nervous tissue has a different behavior to those identified in vertebrate nervous tissue, since it was insensitive to pure compounds, known as anticholinesterases in vertebrates. Considering the insensitivity of the Ca(2+)-activated ATPase, we suggested that the purinergic neurotransmission and other roles of ATP might not be affected by the pure pesticides tested.
