RESUMO

Electrospun fabrics have unique properties due to their uniform morphology and high surface area to volume ratio. Ultrathin nonwoven fabrics are produced for many applications: biomedical, nanosensors, tissue engineering and filtration systems. In this work, nonwoven polylactide, polylactide/natural rubber, poly-3-hydroxybutyrate, and poly-3-hydroxybutyrate/nitrile butadiene rubber fabrics were prepared by electrospinning methods. The obtained fabric samples were used as substrates for the growth of winter wheat seeds "Yubileinaya 100" (Triticum aestivum L.). The stimulating effect of polymer substrates on seed germination and plant growth was shown. The structure and properties of nonwoven agromaterials were controlled by differential scanning calorimetry, IR-spectroscopy, and optical microscopy. The mechanical properties of the obtained fabrics before and after their utilization as substrates were studied. After the wheat growing experiment, the degree of crystallinity of PHB and PHB/NBR samples decreased by 12% and they completely lost their mechanical properties. It is shown that the main factors providing the efficiency of seed growth technology on polymer substrates are the chemical nature and structure of the biodegradable matrix.

In this study, the mechanical and thermophysical properties of carbon fiber-reinforced polyethersulfone are investigated. To enhance the interfacial interaction between carbon fibers and the polymer matrix, the surface modification of carbon fibers by thermal oxidation is conducted. By means of AFM and X-ray spectroscopy, it is determined that surface modification changes the morphology and chemical composition of carbon fibers. It is shown that surface modification dramatically increases the mechanical properties of the composites. Thus, flexural strength and the E-modulus of the composites reinforced with modified fibers reached approximately 962 MPa and 60 GPa, respectively, compared with approximately 600 MPa and 50 GPa for the composites reinforced with the initial ones. The heat deflection temperatures of the composites reinforced with the initial and modified fibers were measured. It is shown that composites reinforced with modified fibers lose their stability at temperatures of about 211 °C, which correlates with the glass transition temperature of the PES matrix. The thermal conductivity of the composites with different fiber content is investigated in two directions: in-plane and transverse to layers of carbon fibers. The obtained composites had a relatively high realization of the thermal conductive properties of carbon fibers, up to 55-60%.

Background: Sleep-disordered breathing (SDB) is a widespread comorbidity in patients with chronic heart failure (HF) and may have a deleterious effect on the pathogenesis of HF. We aimed to evaluate the prognostic role of polysomnography parameters in HF patients with previous decompensation. Methods: 123 patients were included in the prospective cohort study. In addition to the standard examination, all patients underwent polysomnography (PSG). Results: The Kaplan−Meier analysis showed the incidence of the combined endpoint differs between LVEF categories ≤25.5% vs. >25.5% (χ2 = 9.6, log rank p = 0.002), NTpro-BNP > 680 vs. ≤680 pg/mL (χ2 = 12.7, log rank p = 0.001), VO2peak categories <16 vs. ≥16 mL/min/kg (χ2 = 14.2, log rank p = 0.001), VE/VCO2 slope ≥38.5 vs. <38.5 (χ2 = 14.5, log rank p = 0.001), wake after sleep onset >40 min vs. ≤40 min (χ2 = 9.7, log rank p = 0.03), and sleep stage 2 (S2) <44% vs. ≥44% (χ2 = 12.4, log rank p = 0.001). Conclusion: Among the PSG parameters, WASO > 40 min and S2 < 44% were associated with a combined endpoint in patients with previous decompensation of HF. Moreover, higher NT-proBNP and VE/VCO2 slope, lower LVEF, and VO2peak were also independent factors of a poor prognosis.

BACKGROUND: Nurses often face professional risks in their work. Most research in Russia dedicated to professional risks of nurses are quantitative studies and deal with a limited range of issues. There is an apparent lack of qualitative studies on this problem. OBJECTIVE: The aim of this research is to study opinions of oncology nurses concerning their working conditions from the point of view of their professional risks assessment and their minimization. METHOD: The research was conducted by an independent researcher using a method of semi-structured interview. The content-analysis with inductive approach was used for the data analysis. RESULTS: According to the data analysis there was singled out five main topics, in particular, professional risks that are appreciated by nurses, assessment of professional risks as employment obstacles, permitted ways of psychoemotional risks mastering, permitted ways of physical exertion mastering, effective external mechanisms of risks mastering. In general, nurses do not consider their professional risks as employment obstacles. They lay special emphasis on psychosocial risks, which are the core of all other professional risks. CONCLUSION: This research shows the opinion of the nursing service heads of oncology departments about professional risks arising in the process of their staff performing the duties. The results of the research demonstrate that the main perceived dangers are psychosocial, biological and chemical risks specific to the work with oncology patients. Possible ways to reduce the impact of professional risks on nurses can be both external institutional mechanisms and internal personal resources. Determining the working conditions of oncological nurses allows to manage occupational risks, to form a valuable attitude to health, as well as to identify indicators that form a social perception of the profession. The study of the working conditions of oncological nurses allows to manage professional risks, form a value-based attitude to health, and also identify indicators that form a social idea of the profession.

Acholeplasma laidlawii is widespread hypermutable bacteria (class Mollicutes) capable of infecting humans, animals, plants, which is the main contaminant of cell cultures and vaccine preparations. The mechanisms of the development of antimicrobial resistance of this bacterium are associated with the secretion of extracellular vesicles, which can mediate the lateral transfer of antibiotic resistance determinants. We compared the genome profiles of ciprofloxacin-resistant A.laidlawii strains PG8r1 (MIC 10 µg/ml) and PG8r3 (MIC 10 µg/ml) selected under different in vitro conditions - when ciprofloxacin-sensitive (MIC 0.5 µg/ml) A.laidlawii PG8B strain was cultured at increasing concentrations of ciprofloxacin in a broth medium alone, and with vesicles derived from the ciprofloxacin-resistant (MIC 20 µg/ml) A.laidlawii PG8R10c-2 strain, respectively. Genome profiles of PG8c-3 (obtained from a single colony of the strain PG8B) and PG8R10c-2 were analyzed too. Patterns of the quinolone target genes (gyrA, gyrB, parE, parC) containing in extracellular vesicles of PG8c-3, PG8R10c-2, PG8r1 and PG8r3 were determined. Genome sequencing was performed on the NextSeq Illumina platform. Search and annotation of single nucleotide polymorphisms were performed using Samtools and SnpEff, respectively. We also compared cellular proteomes of PG8c-3, PG8r1 and PG8r3. The cellular proteome profiles of the A. laidlawii strains were determined by two-dimensional gel electrophoresis and MALDI-TOF/TOF MS. This work presents data on single nucleotide polymorphisms (SNPs) found in the genomes of the ciprofloxacin-resistant strains selected under different in vitro conditions and proteins that were differentially expressed in the cells of ciprofloxacin-resistant strains selected under different conditions in vitro.

Thermally stable composites obtained by the low-temperature carbonization of an elastomeric matrix filled with hard dispersed silicon carbide particles were obtained and investigated. Evolution of the microstructure and of mechanical and thermal characteristics of composites during thermal degradation and carbonization processes in a wide range of filling from 0 to 450 parts per hundred rubber was studied. For highly filled composites, the compressive strength values were found to be more than 200 MPa; Young's modulus was more than 15 GPa. The thermal conductivity coefficient of composites was up to 1.6 W/(m·K), and this magnitude varied slightly in the temperature range of 25-300 °C. Coupled with the high thermal stability of the composites, the observed properties make it possible to consider using such composites as strained friction units instead of reinforced polymers.

To elucidate the regularities of adaptation of the representatives of class Mollicutes to antimicrobials and to identify the promising targets for eradication of mycoplasma infections and contaminations the comparative analysis of the molecular basis of bacterial resistance to antibiotics of different classes is needed. Previously, we presented the data on the whole-genome sequences of Acholeplasma laidlawii strains with different susceptibility to ciprofloxacin (GenBank: LXYB00000000.1), tetracycline (GenBank: NELO00000000.2) and melittin (GenBank: NELN00000000.2) as well as the data on cell and extracellular vesicle proteomes of melittin-resistant A. laidlawii strain [1]. The lists of extracellular vesicle proteins secreted by A. laidlawii strains with the increased resistance to ciprofloxacin (PG8R10) and tetracycline (PG8RTet) are presented here. The vesicle proteome profiles were obtained by 1D SDS-PAGE and liquid chromatography-mass spectrometry.

Importance: More than 1 million US veterans are dually enrolled in a Medicare Advantage (MA) plan and in the Veterans Affairs (VA) health care system. The federal government prepays private MA plans to cover veterans; if a dually enrolled veteran receives an MA-covered service at the VA, the government is making 2 payments for the same service. It is not clear what proportion of veterans dually enrolled in VA and MA are undergoing coronary revascularization at VA vs non-VA hospitals. Objective: To describe where veterans who are enrolled in both VA and MA undergo coronary revascularization and the associated costs. Design, Settings, and Participants: This is a cohort study consisting of US veterans dually enrolled in VA and MA from January 1, 2010, to December 31, 2013, who had at least 1 VA encounter and underwent coronary revascularization during the study period. Data were analyzed from April 2019 to September 2019. Main Outcomes and Measures: Number of coronary artery bypass graft (CABG) operations and percutaneous coronary interventions (PCIs) performed through the VA and through MA during years 2010 to 2013, and the associated VA costs of coronary revascularization. In addition, multivariable logistic regression was performed to assess patient factors associated with receiving care through the VA. Results: A total of 18 874 VA users with concurrent MA enrollment who underwent coronary revascularization during 2010 to 2013 were identified (mean [SD] age, 75.3 [8.8] years; 18 739 men [99.0%]). Enrollees were predominantly white (17 457 patients [92.0%]). Among patients, 4115 (22.0%) underwent either CABG or PCI through the VA only, 14 281 (75.0%) did so through MA only, and 478 (2.5%) underwent coronary revascularization procedures through both payers. From 2010 to 2013, these veterans underwent 4764 coronary revascularization procedures (721 CABGs and 3043 PCIs) that cost the VA $214.7 million ($115.8 million for CABGs and $99.0 million for PCIs). In multivariable analysis, nonwhite patients were more likely than white patients to undergo coronary revascularization through the VA (odds ratio, 1.73; 95% CI, 1.52-1.96; P < .001), and for each year of age, veterans were less likely to undergo coronary revascularization through the VA (odds ratio, 0.95; 95% CI, 0.94-0.95; P < .001). There was no statistically significant association between undergoing coronary vascularization through the VA and distance in miles to the nearest VA hospital (odds ratio, 1.00; 95% CI, 0.99-1.00; P = .30). Conclusions and Relevance: A substantial share of VA users concurrently enrolled in an MA plan underwent coronary revascularization procedures through the VA, incurring significant duplicative federal health care spending. Given the financial pressures facing both Medicare and the VA, government officials should consider policy solutions to mitigate redundant spending.

Nanotechnologies involving physical methods of tumor destruction using functional oligonucleotides are promising for targeted cancer therapy. Our study presents magnetodynamic therapy for selective elimination of tumor cells in vivo using DNA aptamer-functionalized magnetic nanoparticles exposed to a low frequency alternating magnetic field. We developed an enhanced targeting approach of cancer cells with aptamers and arabinogalactan. Aptamers to fibronectin (AS-14) and heat shock cognate 71 kDa protein (AS-42) facilitated the delivery of the nanoparticles to Ehrlich carcinoma cells, and arabinogalactan (AG) promoted internalization through asialoglycoprotein receptors. Specific delivery of the aptamer-modified FeAG nanoparticles to the tumor site was confirmed by magnetic resonance imaging (MRI). After the following treatment with a low frequency alternating magnetic field, AS-FeAG caused cancer cell death in vitro and tumor reduction in vivo. Histological analyses showed mechanical disruption of tumor tissues, total necrosis, cell lysis, and disruption of the extracellular matrix. The enhanced targeted magnetic theranostics with the aptamer conjugated superparamagnetic ferroarabinogalactans opens up a new venue for making biocompatible contrasting agents for MRI imaging and performing non-invasive anti-cancer therapies with a deep penetrated magnetic field.

RESUMO

This review is devoted to the mechanisms of antibiotic resistance in mollicutes (class Bacilli, subclass Mollicutes), the smallest self-replicating bacteria, that can cause diseases in plants, animals and humans, and also contaminate cell cultures and vaccine preparations. Research in this area has been mainly based on the ubiquitous mollicute and the main contaminant of cell cultures, Acholeplasma laidlawii. The omics technologies applied to this and other bacteria have yielded a complex picture of responses to antimicrobials, including their removal from the cell, the acquisition of antibiotic resistance genes and mutations that potentially allow global reprogramming of many cellular processes. This review provides a brief summary of well-known resistance mechanisms that have been demonstrated in several mollicutes species and, in more detail, novel mechanisms revealed in A. laidlawii, including the least explored vesicle-mediated transfer of short RNAs with a regulatory potency. We hope that this review highlights new avenues for further studies on antimicrobial resistance in these bacteria for both a basic science and an application perspective of infection control and management in clinical and research/production settings.

BACKGROUND: Galectin-3 is a recently developed biomarker of fibrosis, which may play a role in cardiac remodeling and associated with both the progression and severity of heart failure (HF). METHODS: A prospective cohort study of 190 patients with documented prior myocardial infarction and chronic HF (NYHA class II-IV) was conducted. Patients were divided into 3 groups based on their NYHA functional class. Levels of galectin-3, NT-proBNP, CRP, IL-6, oxidized-LDL, extracellular superoxide dismutase (EC-SOD), 3-nitrotyrosine, SH-groups, cystatin-C were determined. Follow-up period was 26 months, and all-cause mortality was determined as the primary endpoint. Statistical analysis was performed and statistical significance was set at P<0.05. RESULTS: The cytokines hs-CRP, IL-6 and markers of oxidative stress had significant positive correlation with plasma galectin-3 levels in all groups of patients. The level of galectin-3 was significantly different between the groups (P<0.05). Galectin-3 was found to be the most sensitive and specific value in determination of 26 months mortality in patients with chronic HF. Logistic regression analysis showed that age, galectin-3 and cystatin-C were associated with death during the follow up period. CONCLUSIONS: Galectin-3 levels are elevated in patients with chronic HF across all NYHA functional classes. Galectin-3 shows positive correlation with markers of oxidative stress, inflammation and kidney dysfunction. Galectin-3 levels and cystatin-C levels are independent predictors of 26-month mortality in patients with chronic HF. Patients with cystatin-C level >2800 pg/mL carry a worse prognosis. Galectin-3 level >21 ng/mL associated with increased mortality.

Mycoplasmas (class Mollicutes), the smallest prokaryotes capable of self-replication, as well as Archaea, Gram-positive and Gram-negative bacteria constitutively produce extracellular vesicles (EVs). However, little is known regarding the content and functions of mycoplasma vesicles. Here, we present for the first time a proteomics-based characterisation of extracellular membrane vesicles from Acholeplasma laidlawii PG8. The ubiquitous mycoplasma is widespread in nature, found in humans, animals and plants, and is the causative agent of phytomycoplasmoses and the predominant contaminant of cell cultures. Taking a proteomics approach using LC-ESI-MS/MS, we identified 97 proteins. Analysis of the identified proteins indicated that A. laidlawii-derived EVs are enriched in virulence proteins that may play critical roles in mycoplasma-induced pathogenesis. Our data will help to elucidate the functions of mycoplasma-derived EVs and to develop effective methods to control infections and contaminations of cell cultures by mycoplasmas. In the present study, we have documented for the first time the proteins in EVs secreted by mycoplasma vesicular proteins identified in this study are likely involved in the adaptation of bacteria to stressors, survival in microbial communities and pathogen-host interactions. These findings suggest that the secretion of EVs is an evolutionally conserved and universal process that occurs in organisms from the simplest wall-less bacteria to complex organisms and indicate the necessity of developing new approaches to control infects.

This study demonstrated that extracellular membrane vesicles are involved with the development of resistance to fluoroquinolones by mycoplasmas (class Mollicutes). This study assessed the differences in susceptibility to ciprofloxacin among strains of Acholeplasma laidlawii PG8. The mechanisms of mycoplasma resistance to antibiotics may be associated with a mutation in a gene related to the target of quinolones, which could modulate the vesiculation level. A. laidlawii extracellular vesicles mediated the export of the nucleotide sequences of the antibiotic target gene as well as the traffic of ciprofloxacin. These results may facilitate the development of effective approaches to control mycoplasma infections, as well as the contamination of cell cultures and vaccine preparations.

For the first time, we studied the phytopathogenicity toward Oryza sativa L. of unadapted and adapted to unfavorable environment (starvation) cells of Acholeplasma laidlawii PG8--ubiquitous mycoplasma found in the soil, waste waters, tissues of the highest eukaryotes and being the basic contaminant of cell cultures and a causative agent of phytomycoplasmoses. The features of morphology, ultrastructural organization and proteomes of unadapted and adapted cells of the mycoplasma and infected plants were presented. Using 2D-DIGE and MS, 43 proteins of O. sativa L. that were differentially expressed in the leaves of plants cultivated in media with A. laidlawii PG8 were identified. The qualitative and quantitative responses of the plant proteome toward adapted and unadapted mycoplasma cells differed. That may be explained by differences in the virulence of the corresponding bacterial cells. Using 2D-DIGE and MS, 82 proteins that were differentially expressed in adapted and unadapted mycoplasma cells were detected. In adapted cells of the mycoplasma, in comparison with unadapted ones, a significant increase in the expression of PNPase--a global regulator of virulence in phytopathogenic bacteria occurred; there was also decreased expression of 40 proteins including 14 involved in bacterial virulence and the expression of 31 proteins including 5 involved in virulence was not detected. We propose that differences in the phytopathogenicity of adapted and unadapted A. laidlawii PG8 cells may be related to features of their proteomes and membrane vesicles.

Extracellular vesicle production is believed to be a ubiquitous process in bacteria, but the data on such a process in Mollicutes are absent. We report the isolation of ultramicroforms - extracellular vesicles from supernatants of Acholeplasma laidlawii PG8 (ubiquitous mycoplasma; the main contaminant of cell culture). Considering sizes, morphology, and ultrastructural organization, the ultramicroforms of A. laidlawii PG8 are similar to membrane vesicles of Gram-positive and Gram-negative bacteria. We demonstrate that A. laidlawii PG8 vesicles contain genetic material and proteins, and are mutagenic to lymphocytes of human peripheral blood. We show that Mycoplasma gallisepticum S6, the other mycoplasma, also produce similar structures, which suggests that shedding of the vesicles might be the common phenomenon in Mollicutes. We found that the action of stress conditions results in the intensive formation of ultramicroforms in mycoplasmas. The role of vesicular formation in mycoplasmas remains to be studied.
