Bovine pericardium (BP) has been used as a biomaterial for several decades in many medical applications particularly due to its mechanical properties and the high collagen content. In the acellular form it favors faster tissue repair, providing a three-dimensional support for cellular and vascular events observed during tissue repair and due, to a low elastin content, may favor its use as a breast implant cover, resulting in a low possibility of contracture of the biomaterial, preventing the appearance of irregularities during the reconstruction process. Thus, the aim of this study was to evaluate, histomorphologically, the behavior of acellularized bovine pericardium (ABP) as a mammary implant cover in rats. For this purpose, 16 animals were divided into two groups, with eight animals at each biological point: 7 and 15 days after surgery. Of the 16 animals, 32 specimens were obtained: 16 in the experimental group (EG) and 16 in the control group (CG). Throughout this study, none of the studied groups had postoperative complications. Results: The histomorphological results showed, in the two biological points, both in the EG and in the CG, chronic inflammatory infiltrate, leukocyte fibrin exudate, formation of granulation tissue and deposition of collagen fibers, more evident in the EG, regressive along the biological points. At 15 days, the implanted ABP showed initial biointegration with the fibrous capsule and surrounding tissues of the recipient bed. Conclusion: These results indicate that the due to the observed favorable tissue response ABP may be of potential use as a breast implant cover.

Los inhibidores de checkpoint (ICP) son anticuerpos usados en inmunoterapia contra el cáncer. Uno de sus blancos de acción es el receptor de muerte celular programada-1 (PD-1), el cual es importante para mantener la tolerancia inmunitaria. Sin embargo, este mecanismo se asocia a riesgo de eventos adversos relacionados a la inmunidad que pueden afectar a múltiples órganos incluyendo el sistema endocrino. Se describe el caso inhabitual de un paciente que a los 18 meses de terapia con ICP debutó con cetoacidosis diabética (CAD).


Immune checkpoint inhibitors consist in antibodies used in immunotherapy against cancer. One of their targets is the programmed cell death-1 (PD-1) receptor, which is important in maintaining self-tolerance. However, this mechanism is associated with a risk for immune-related adverse events potentially affecting multiple organs, including the endocrine system. We describe the unusual case of a patient who, after 18 months of treatment with an immune checkpoint inhibitor, debuted with diabetic ketoacidosis

Objective. Recent technological advances are revealing the complex physiology of the axon and challenging long-standing assumptions. Namely, while most action potential (AP) initiation occurs at the axon initial segment in central nervous system neurons, initiation in distal parts of the axon has been reported to occur in both physiological and pathological conditions. The functional role of these ectopic APs, if exists, is still not clear, nor its impact on network activity dynamics.Approach. Using an electrophysiology platform specifically designed for assessing axonal conduction we show here for the first time regular and effective bidirectional axonal conduction in hippocampal and dorsal root ganglia cultures. We investigate and characterize this bidirectional propagation both in physiological conditions and after distal axotomy.Main results.A significant fraction of APs are not coming from the canonical synapse-dendrite-soma signal flow, but instead from signals originating at the distal axon. Importantly, antidromic APs may carry information and can have a functional impact on the neuron, as they consistently depolarize the soma. Thus, plasticity or gene transduction mechanisms triggered by soma depolarization can also be affected by these antidromic APs. Conduction velocity is asymmetrical, with antidromic conduction being slower than orthodromic.Significance.Altogether these findings have important implications for the study of neuronal functionin vitro, reshaping our understanding on how information flows in neuronal cultures.

BACKGROUND: Prognosis assessment of pulmonary hypertension (PH) is multifactorial and placement of patients on the lung transplantation (LT) waiting list requires the weighing of a complex set of criteria. The aim of this retrospective cohort study was to analyze a series of patients treated in our unit at the moment of their inclusion on the LT waiting list and long-term survival after LT. MATERIAL AND METHODS: Baseline characteristics, LT outcomes, and survival were evaluated in all patients diagnosed with pulmonary arterial hypertension (PAH) and pulmonary veno-occlusive disease (PVOD) who were included on the LT waiting list in 2011-2016. RESULTS: Thirty-three patients were listed with a diagnosis of PAH or PVOD. Patients had an average age of 43 ± 12 years and 71% were female. The median time between PAH diagnosis and inclusion on the LT waiting list was 62.5 months (interquartile range [IQR], 6-93.3 months). Twenty-eight patients (84%) underwent double LT. The difference between the waiting time in urgent cases (1.5 months; IQR, 0.4-4.2 months) and in elective cases (7.4 months; IQR, 2.7-16.2 months) was significant (P < .049). The 28 patients with PAH/PVOD in our hospital had a 95% short-term survival after LT both at 1 and at 3 months, without variance between urgent and elective LT. Longer-term survival rate was 84% both at 12 and 36 months. CONCLUSIONS: There is great complexity in determining the appropriate time for transplantation referral and inclusion on the waiting list for patients with PAH/PVOD so that LT can be more realistically incorporated into the treatment algorithm for PAH. LT offers a good short- and long-term survival in patients with PAH/PVOD.

The phythotherapy is an alternative to use of chemotherapeutical agents against monogenean infection. This study evaluated the anthelmintic activity of essential oil Ocimum gratissimum against monogenean Cichlidogyrus tilapiae as well as its acute toxicity in tilapia juveniles. The mean lethal concentration (LC50) and different concentrations of the essential oil, both in vitro and in vivo assays (short and long-term baths) were assessed. The LC50 was 40.70mg.L-1 and in the in vitro assay this concentration showed 80% efficacy at the last two hours and in the in vivo assay 65.87% efficacy in long-term bath. However, it provoked morphological alterations on the gills such as hyperplasia and edema. The parasites exposure at the highest concentration (320mg.L-1) showed 100% mortality after 2h exposure in the in vitro assay, whereas in the in vivo assay, short-term baths of 5min for 3 consecutive days showed an efficacy of 87.71% without gills damage. These results demonstrate the anthelminthic activity of essential oil O. gratissimum and the safety concentration to use in Nile tilapia.(AU)


A fitoterapia é uma alternativa ao uso de agentes químicos contra infecções por parasitos monogenéticos. Este estudo avaliou a atividade anti-helmíntica do óleo essencial Ocimum gratissimum contra o monogenea Cichlidogyrus tilapiae , bem como sua toxicidade aguda e histopatologia em juvenis de tilápias. Foram avaliadas a concentração letal média (CL50) e diferentes concentrações de óleo essencial, tanto in vitro como in vivo (banho de curta e longa duração). A CL50 foi de 40,70mg.L-1; no ensaio in vitro, essa concentração apresentou 80% de eficácia, e no ensaio in vivo 65,87% de eficácia em banho de exposição crônica. No entanto, provocou alterações morfológicas nas brânquias, como hiperplasia e edema. A exposição dos parasitas na concentração mais elevada (320mg.L-1) mostrou 100% de mortalidade após duas horas de exposição no ensaio in vitro, enquanto no ensaio in vivo, em banho de curta duração de cinco minutos, durante três dias consecutivos, apresentou uma eficácia de 87,71%, sem danos às brânquias. Esses resultados demonstraram a atividade anti-helmíntica do óleo essencial de O. gratissimum e a concentração de segurança para uso na tilápia-do-nilo em banhos de curta duração.(AU)

Abstract The present study evaluates the influence of anesthesia on the parasitic fauna of monogenea fish parasites, as its intensity and viability. Two experiments were conducted: Evaluation of an anesthetic method by sprinkling eugenol directly on gills and evaluation of monogenea motility and viability; Comparison of immersion and directly sprinkling on the gills with benzocaine and eugenol followed by evaluation on parasite intensity. The results suggest that the anesthetic sprinkling didn't interfere in the parasite motility, morphology and body surface integrity analyzed by fluorescence method. The monogenean intensity in the gills was lower in fish anesthetized by immersion method compared to the sprinkling method and the control group. This method of anesthesia can be used in parasitological studies.


Resumo O presente estudo avalia a influência da anestesia sobre a fauna parasitária de monogeneas em peixes, sua intensidade e sua viabilidade. Dois experimentos foram realizados: Avaliação de um método anestésico por aspersão eugenol diretamente nas brânquias e avaliação da motilidade das monogeneas e sua viabilidade; e Comparação entre imersão e aspersão diretamente nas brânquias com benzocaína e eugenol, seguido de avaliação sobre a intensidade parasitária. Os resultados sugerem que a aspersão do anestésico não interferiu na motilidade, morfologia, superfície corporal e integridade do parasita, analisadas pelo método de fluorescência. A intensidade de monogenéticos nas brânquias foi menor nos peixes anestesiados pelo método de imersão em comparação com o método de aspersão e o grupo controle. O método de anestesia por aspersão nas brânquias pode ser utilizado em estudos parasitológicos.

RESUMO

We herein report the synthesis and catalytic application of a new family of [N,P] ligands based on the pyrrole ring with alpha-phosphine or phosphole units. Their palladium complexes (3a-d) were obtained in very good yields and their catalytic properties were evaluated in the direct intramolecular arylation to obtain both benzopyranones and phenanthridinones. The air stable complex 3a exhibited the best catalytic performance of this series of complexes, using 1 mol% of catalyst in combination with microwaves to promote this reaction.

The aim of this study was analyzed if the flour or flaxseed oil treatment contributes to body composition in male rats subjected to early weaning. Pups were weaned for separation from mother at 14 (early weaning, EW) and 21 days (control, C). At 21 days, part of the pups was evaluated (C21 v. EW21). After 21 days, control (C60) was fed with control diet. EW was divided in control (EWC60); flaxseed flour (EWFF60); flaxseed oil (EWFO60) diets until 60 days. Body mass, length and body composition by dual-energy X-ray absorptiometry were determined. EW21 (v. C21) and EWC60 (v. C60 and EWFF60) showed lower (P<0.05) mass, length and body composition. EWFO60 (v. C60 and EWFF60) showed lower (P<0.05) body mass and length, body and trunk lean mass, bone mineral density and content and bone area. Flaxseed flour, in comparison with flaxseed oil, contributes to recovery of body composition after early weaning.

The precocious interruption of lactation is a prime factor for developmental plasticity. Here we analyzed whether flour or flaxseed oil treatment contributes to body and brain mass in male rats subjected to early weaning. Pups were weaned for separation from their mother at 14 (early weaning, EW) and 21 days (control, C). At 21 days, some of the pups were evaluated (C21 v. EW21). After 21 days, control pups (C60) were fed a control diet. EW pups were divided into those fed a control diet (EWC60), those given flaxseed flour (EWFF60), and those given flaxseed oil (EWFO60) until 60 days. EW21 showed lower body and absolute brain mass and higher relative brain mass. At 60 days, EWC60 and EWFO60 had lower body mass. With regard to relative brain mass, EWC60 was heavier; EWFO60 had lower values compared with EWC60 and higher values compared with C60 and EWFF60. These results indicated that flaxseed flour, in comparison with flaxseed oil, contributes to brain development after EW.

Body weight in different mammalian species influences reproductive potential. The aim of the present study was to determine the relationship of body weight at the time of semen collection with libido, seminal characteristics and number of semen doses for artificial insemination (AI) in New Zealand White mature fertile male rabbits. Data came from 728 semen collections of 14 rabbits, 15-months of age that were sexually experienced with proven semen quality and fertility. Semen collection was performed twice a week with two ejaculates at each collection time and lasted 14 weeks. A second ejaculation was collected at 1-2h after the first. Data from each male from first and second ejaculates from 1 day of semen collection throughout the trial were averaged (n=324) and partial correlation coefficients and regression equations were estimated to describe the relationship of male body weight to ejaculation reaction time and 12 semen and sperm characteristics. As body weight increased there was a linear (P<0.05) increase in reaction time, abnormal sperm with an intact membrane and abnormal sperm with a damaged membrane and a linear (P<0.05) decrease in semen volume, sperm concentration per ejaculate, normal sperm with an intact membrane, number of normal motile sperm with an intact membrane and suitable semen doses for AI. Body weight of the mature male rabbit at semen collection had some influence on libido, semen and sperm characteristics, with a general trend toward a lesser reproduction potential as body weight increases.

Requirements of hazard analysis and critical control points (HACCP) are becoming essential for international trade in food commodities as a safety assurance component. This research reports the level of the adoption of ISO 9000 and the HACCP system by Federal Inspection Type (TIF) pork-exporting enterprises. Implementation and operating costs are reported as well as the benefits involved in this food industry process. In Mexico, there are 97 companies classified as TIF enterprises, and 22 are registered as exporters of processed pork with the National Services for Safety and Quality and Animal Health of the Secretariat of Agriculture, Livestock, Rural Development, Fisheries and Food. Surveys were administered to 22 companies, with a 95.2% response rate. Enterprise characteristics were evaluated, as well as their operating activities. Fieldwork consisted of administering structured questionnaires to TIF exporters. All the surveyed enterprises had implemented HACCP, whereas the ISO 9000 regulation was applied in only 30%. Of total production, 75% is exported to 13 countries, and 25% goes to the Mexican market niche. Results indicate that the main factors for adopting HACCP are related to accessibility to international markets, improving quality, and reducing product quality audits by customers. The results also indicated that staff training was the most important issue. Microbiological testing was the highest cost of the operation. The main benefits reported were related to better access to international markets and a considerable reduction in microbial counts. This study shows the willingness of Mexican pork processors to implement food safety protocols for producing safe and quality products to compete in the international food trade.

Acidentes por escorpião constituem problema de saúde pública em Belo Horizonte. Realizou-se um estudo epidemiológico observacional retrospectivo para analisar a frequência e distribuição espacial dos acidentes escorpiônicos em Belo Horizonte, entre 2005 e 2009, e associá-las às categorias de risco classificadas pelo Índice de Vulnerabilidade à Saúde (IVS), um índice socioeconômico regional composto. Foram utilizados dados de notificação do Sistema Nacional de Agravos de Notificação (SINAN) e Sistema de Vigilância Epidemiológica (SISVE), referentes aos anos de 2008 e 2009. Para o período de 2005 a 2007, foi necessário o resgate de dados das fichas clínicas do Hospital João XXIII (HPSJXXIII) correspondentes às variáveis encontradas no SINAN e SISVE, uma vez que os mesmos não estavam digitalizados nos referidos sistemas de informação. Para georreferenciamento e análise espacial, foi utilizada a base geográfica EndGeo, o aplicativo de mapeamento MapInfo versão 10.0 e os programas Hotspot Detective e SatScan. Entre 2005 e 2009, ocorreram em Belo Horizonte 2.769 casos de acidentes por escorpião, o que representa uma incidência média de 22,4 casos por 100.000 habitantes. Do total de casos, 1.924 (69,5 porcento) foram georreferenciados e houve predomínio de incidência em dois Distritos Sanitários (DS), com grande concentração de casos nas regiões de cemitérios do município. Foram detectados dois "clusters" no período, sendo um em 2005, nos DS Noroeste e Oeste, e outro entre 2006 e 2007, nos DS Noroeste e Nordeste. Não houve associação entre as áreas de maior incidência de escorpionismo e as áreas de maior risco à saúde classificadas pelo IVS. Com base nos resultados, conclui-se que há necessidade de melhorias no processo de notificação do agravo e que o mapeamento dos casos é uma ferramenta relevante capaz de embasar o direcionamento das ações educativas de controle para as áreas prioritárias de Belo Horizonte...


Scorpion sting accidents are a public health problem in Belo Horizonte and veterinarians play an important role in its control program when based on capture, mapping of cases and educational actions with the population. A retrospective epidemiological study was done to analyze the frequency and spatial distribution of scorpion sting cases in Belo Horizonte, between 2005 and 2009, and to associate them with the Health Vulnerability Index (IVS), a regional and composed socio-economic index. Notification data of scorpion sting accidents were used from the National System of Notifiable Diseases (SINAN) and Epidemiological Surveillance System (SISVE) for years 2008 and 2009. For the period between 2005 and 2007 it was necessary to retrieve data from the clinical records of João XXIII Hospital (HPSJ XXIII) which were not digitalized in the referred information systems. For georeference and spatial analysis, geographic base EndGeo and features of the programs MapInfo version 10.0, Hotspot Detective and SatScan were used. Between 2005 and 2009 2769 cases of scorpionism, occurred in Belo Horizonte which represents an average incidence of 22.4 cases per 100,000 inhabitants. Among 1924 (69.5 percent) georeferenced cases, the highest incidence was observed in two Sanitary District (DS). A large concentration of cases was also observed in the areas of the city cemeteries. Two clusters were detected during this period, one in 2005, located in the Northwest and West DS, and another between 2006 and 2007 in the Northwest and Northeast DS. There was no association between higher incidence areas of scorpionism in Belo Horizonte and the areas of higher health risk classified by IVS. It is necessary to improve the reporting process regarding scorpion sting accidents. Another conclusion is that the mapping of cases is a relevant tool to base the targeting of educative actions to the priority areas of Belo Horizonte...

Tuberculosis is a disease caused by the Mycobacterium tuberculosis complex (MTb). In 2011, global mortality due to tuberculosis was 1·4 million individuals. The only available vaccine is the attenuated M. bovis [bacillus Calmette-Guérin (BCG)] strain, which confers variable protection against pulmonary tuberculosis. Some widely distributed non-tuberculous mycobacteria (NTM), such as M. avium and M. arupense, are also potential pathogens for humans. This work aimed to produce and characterize monoclonal antibodies against the M. bovis BCG Mexico strain of the MTb, M. avium subs. hominissuis and the M. arupense strain from NTM. Hybridomas were produced from splenocytes of BALB/c female mice immunized with radiation-inactivated mycobacteria, and the immunoglobulin (Ig)G2a antibody-producing clones with the highest antigenic recognition were selected. The selected clones, Mbv 2A10 for M. bovis BCG Mexico, Mav 3H1 for M. avium and Mar 2D10 for M. arupense, were used in further studies. Enzyme-linked immunosorbent assay (ELISA) and immune proteomics analyses characterized the clones as having the highest cross-reactivity with mycobacteria. Using mass spectrometry, a number of proteins recognized by the monoclonal antibody (mAb) clones were identified. These proteins had roles in metabolic processes, hypoxia, cell cycle and dormancy. In addition, a Clustal W and Immune Epitope Database (IEDB) in-silico analysis was performed in protein sequences that result in the conserved regions within probability epitopes that could be recognized for Mbv2A10 and Mav3H1 clones.

Our lung transplant unit began activity in October 2008. We have performed 37 lung transplants with a hospital mortality of 2.7% (n = 1). The need for a greater number of donors and the presence of an already existent non-heart-beating donor (NHBD) program for abdominal grafts and tissues encouraged us to consider assessing lung grafts from these donors. It was necessary to develop a new multiorgan preservation methodology, "bithermia preservation." The clinical experience with which during the first year June 2010 to July 2011, including 15 NHBDs is presented herein. The chest x-ray was normal in 6 donors (40%) and 7 had pulmonary infiltrates. Bronchoscopy was normal in 8 donors (53%) but 3 had abundant bleeding airway secretions and signs of bronchoaspiration. Preservation procedures were performed in 6 donors. Pulmonary functional evaluation in 4 donors showed gas measurements to be adequate in 75% of cases. Three double-lung grafts were judged to be valid for implantation, among which we performed 3 lung transplantations, 1 bilateral and 2 unilaterals, while 2 grafts were offered to the National Transplant Organization for other units. No transplant suffered primary graft dysfunction; all 3 showed excellent function allowing early extubation in 2 cases. There was no in-hospital mortality. All 3 patients are alive and leading normal lives; none has bronchiolitis obliterans syndrome. In conclusion, the "bithermia preservation" methodology achieved adequate lung preservation in NHBDs, allowing liver, kidneys, and lungs to be obtained from the same donor.

Os objetivos desta pesquisa foram caracterizar as recusas por parte da população em permitir a borrifação de seus imóveis, em quatro regiões de Belo Horizonte, no período de 2006 a 2008, e relacionar estas recusas com o surgimento de casos de leishmaniose visceral, no mesmo local e período. Utilizaram-se as localizações onde se encontraram 27 casos humanos de leishmaniose visceral obtidos do SINAN, todos referentes a quatro áreas de abrangência da região noroeste de Belo Horizonte, no período de 2006 a 2008. De um total de 33.579 residências visitadas pelo controle químico vetorial, 9636 (28,70%) aceitaram a borrifação interna e a externa do domicílio, 20741 (61,77%) concordaram com a borrifação apenas do peridomicílio, 141 (0,42%) aceitaram apenas a borrifação do intradomicílio, 909 (2,70%) se encontraram fechadas no momento da borrifação e 2152 (6,41%) recusaram qualquer tipo de borrifação no imóvel. Na maioria das residências, o controle químico foi permitido apenas no peridomicílio. A diferença entre o perfil de recusas dos quarteirões contidos em uma área de 200 metros ao redor de um caso humano de leishmaniose visceral e o mesmo perfil dos quarteirões fora desta área foi significativa (P<0,05) em sete (25,92%) dos 27 casos da doença.


The purpose of this study was to characterize the population's refusal to allow their houses to be sprayed in four regions of Belo Horizonte, in the period from 2006 to 2008, and relate these denials with the emergence of cases of visceral leishmaniasis in the same location and time. We used the location of 27 human cases of visceral leishmaniasis obtained from SINAN, all data from four regions in the West Region of the city of Belo Horizonte, from 2006 to 2008. Of a total of 33,579 houses visited by the chemical vector control, 9636 (28.70%) accepted spraying inside and outside the home, 20741 (61.77%) agreed with spraying only the areas surrounding the property, 141 (0.42%) agreed only to spray indoors, 909 (2.70%) were found closed at the time of spraying, and 2152 (6.41%) refused any kind of spray at house. A chemical control vector was allowed only in surrounding areas of most of the houses studied. The difference between the profile of denials of the blocks contained in an area of 200 meters around a human case of VL, with the same profile of the blocks out of this area was significant (P<0,05) in 07 (25,92%) of 27 cases of the disease.

OBJECTIVES: To evaluate the effect of flaxseed upon body growth and brain of rats. METHODS: Experimental phase lasted 52 days, using 42 Wistar rats which were divided into four groups: Control (CG, n = 12), 10% casein diet; Flaxseed (GL, n = 12), 10% flaxseed diet plus casein; Modified Control (GCM, n = 12), 10% casein diet with changes in lipid and fiber comparable to GL; Non-protein (GA, n = 6), diet without protein. Considering food intake, protein intake and weight variation, the Protein Efficiency Ratio (PER) was calculated. Net Protein Retention (NPR) and the Food Efficiency Ratio (CEA) were also determined relative to brain weight at 30 days of life (M30) and at 52 days (M52). Analysis of Variance (ANOVA), Test of Sheffe and post-test of Bonferroni were used, p < 0.05. RESULTS: GL had lower food intake, protein and weight variation than GC, but had higher values than GCM. Concerning PER, GL was lower than GC and similar to the GCM, as well as CEA. As for NPR, GL had lower values than the CG and GCM. At M30, GL was superior to GC in relation to brain weight. Likewise, the same was observed at M52. CONCLUSION: Flaxseed promoted adequate growth and better brain development in animals, which might be explained by increased incorporation of omega-3 into these tissues.

Flaxseed has a high content of n-3 fatty acids and its intake associated with an environmental enrichment may promote distinct behavioral results upon habituation and animal behavior. This work aimed to evaluating animal behavior under the use of these two tools in the Open Field Test. Thirty-six male Wistar rats were divided into 6 groups (n = 6): FEEG, receiving chow made up of flaxseed and kept in enriched environment; FSEG, receiving flaxseed based diet and kept in a standard environment; CEEG, receiving casein based diet and kept in enriched environment; CSEG, receiving casein based chow and kept in standard environment; MCEEG, receiving chow made up of casein but modified so as to provide the same content of fibers and lipids found in flaxseed diet and kept in enriched environment; MCSEG, receiving modified casein based diet and kept in standard environment. All animals were kept under controlled temperature, collective cages and dark/light cycle, receiving chow and water ad libitum, except for MCEEG and MCSEG, which were pair fed with FEEG and FSEG, respectively. Chow intake and animal body weight were evaluated twice in a week. Animals were maintained in these groups from the first until the second month of life, by the time when 3 day tests in Open Field Test began. Finishing the tests, animals were sacrificed and their brains were obtained in order to calculate the relative brain weight. Our results show an interplay between flaxseed and environmental enrichment in habituation to a new environment, making the animals more manageable and less stressed.
