RESUMO

Tiller number is a key component of wheat plant architecture having a direct impact on grain yield. Because of their viability, biotic resistance, and abiotic stress tolerance, wild relative species are a valuable gene source for increasing wheat genetic diversity, including yield potential. Agropyron glael, a perennial hybrid of Thinopyrum intermedium and Th. ponticum, was created in the 1930s. Recent genome analyses identified five evolutionarily distinct subgenomes (J, Jst, Jvs, Jr, and St), making A. glael an important gene source for transferring useful agronomical traits into wheat. During a bread wheat × A. glael crossing program, a genetically stable translocation line, WT153397, was developed. Sequential in situ hybridizations (McGISH) with J-, St-, and D-genomic DNA probes and pSc119.2, Afa family, pTa71, and (GAA)7 DNA repeats, as well as molecular markers specific for the wheat 6D chromosome, revealed the presence of a 6DS.6Jvs Robertsonian translocation in the genetic line. Field trials in low-input and high-input breeding nurseries over four growing seasons demonstrated the Agropyron chromosome arm's high compensating ability for the missing 6DL, as spike morphology and fertility of WT153397 did not differ significantly from those of wheat parents, Mv9kr1 and 'Mv Karizma.' Moreover, the introgressed 6Jvs chromosome arm significantly increased the number of productive tillers, resulting in a significantly higher grain yield potential compared to the parental wheat cultivars. The translocated chromosome could be highly purified by flow cytometric sorting due to the intense fluorescent labeling of (GAA)7 clusters on the Thinopyrum chromosome arm, providing an opportunity to use chromosome genomics to identify Agropyron gene variant(s) responsible for the tillering capacity. The translocation line WT153397 is an important genetic stock for functional genetic studies of tiller formation and useful breeding material for increasing wheat yield potential. The annual goatgrass, Aegilops biuncialis is a rich source of genes with considerable agronomic value. This genetic potential can be exploited for wheat improvement through interspecific hybridization to increase stress resistance, grain quality and adaptability. However, the low throughput of cytogenetic selection hampers the development of alien introgressions. Using the sequence of flow-sorted chromosomes of diploid progenitors, the present study enabled the development of chromosome-specific markers. In total, 482 PCR markers were validated on wheat (Mv9kr1) and Ae. biuncialis (MvGB642) crossing partners, and 126 on wheat-Aegilops additions. Thirty-two markers specific for U- or M-chromosomes were used in combination with GISH and FISH for the screening of 44 Mv9kr1 × Ae. biuncialis BC3F3 genotypes. The predominance of chromosomes 4M and 5M, as well as the presence of chromosomal aberrations, may indicate that these chromosomes have a gametocidal effect. A new wheat-Ae. biuncialis disomic 4U addition, 4M(4D) and 5M(5D) substitutions, as well as several introgression lines were selected. Spike morphology and fertility indicated that the Aegilops 4M or 5M compensated well for the loss of 4D and 5D, respectively. The new cytogenetic stocks represent valuable genetic resources for the introgression of key genes alleles into wheat.

An increase in the production and consumption of spelt products can be associated with positive effects on human health, which are attributed to bioactive compounds present in the grain. The basic success of spelt wheat in organic farming might be explained by the fact that spelt wheat belongs to the group of hulled wheat where the presence of a husk protects the seed from abiotic and biotic stress factors, thus demanding less chemical protection. The goal of this study was to investigate the variations in the bioactive compound (alkylresorcinol, arabinoxylan, ß-glucan), protein, starch and fructan content of bread and spelt wheat under different farming systems (conventional and organic). The results showed higher protein and alkylresorcinol but lower fructan content in spelt wheat. Organic spelt had significantly higher starch, fiber and alkylresorcinol content but lower ß-glucan and protein content than conventionally grown spelt. The spelt variety 'Oberkulmer-Rotkorn' was characterized by the highest values for the majority of analyzed traits under both farming systems. Overall, the environmental conditions (Hungary and Serbia), farming systems (conventional and organic) and wheat species (bread and spelt) contributed to the variations of the compositional traits in different manners.

As root electrical capacitance (CR*) was assumed to depend on the stem properties, the efficiency of measuring CR* at flowering for whole-plant phenotyping was assessed in five wheat cultivars in three replicate plots over two years. Linear regression analysis was used to correlate CR* with plant-size parameters and flag-leaf traits (extension and SPAD chlorophyll content) at flowering, and with yield components at maturity. The plot-mean CR* was correlated with the plot leaf area index (LAI), the chlorophyll quantity (LAI×SPAD), and the grain yield across years. At plant scale, CR* was found to show the strongest positive regression with total chlorophyll in the flag leaf (flag leaf area × SPAD; R2: 0.65−0.74) and with grain mass (R2: 0.55−0.70) for each cultivar and year (p < 0.001). Likewise, at plot scale, the regression was strongest between CR* and the LAI×SPAD value (R2: 0.86−0.99; p < 0.01) for the cultivars. Consequently, CR* indicated the total plant nutrient and photosynthate supply at flowering, which depended on root uptake capacity, and strongly influenced the final yield. Our results suggested that the polarization of the active root membrane surfaces was the main contributor to CR*, and that the measurement could be suitable for evaluating root size and functional intensity. In conclusion, the capacitance method can be applied for nondestructive whole-plant phenotyping, with potential to estimate root and shoot traits linked to the nutrient supply, and to predict grain yield. CR* can be incorporated into allometric models of cereal development, contributing to optimal crop management and genetic improvement.

Spelt wheat (Triticum aestivum subsp. spelta L.) is an underexploited hexaploid wheat species that has become an increasingly fashionable raw material of bakery products in the last decades, partly because of its ability to grow under organic agricultural conditions and partly because of the growing number of people following the trend of having a healthy diet. However, due to its difficult threshing, most research on spelt seed is based on a very limited number of genotypes. Therefore, we determined the physical, compositional, and breadmaking quality traits of 90 spelt genotypes in order to highlight the variation of these properties and to identify possible genetic resources for spelt improvement. The thousand kernel weight of the spelt genotypes ranged between 23.2 and 49.7 g, the protein content between 12.1% and 22.2%, the gluten index between 0.7 and 98.8, the dough stability between 0.0 and 19.6 min, and the starch damage between 6.3 and 19.4 UCD value. The average values showed that spelt has higher protein and gluten contents but weaker dough strength and stability than common bread wheat. The starch pasting temperature was also higher in spelt, but the starch damage was lower, resulting in lower water absorption. Some genebank accessions (MVGB142, 145, 353, and 525) and internationally available cultivars (Bohemia, Bodensonne, Black-Bearded, and White-Beardless) were identified as good genetic resources for improving the breadmaking-quality traits of spelt.

This study evaluated the concurrent application and the results of the root electrical capacitance (CR) and minirhizotron (MR) methods in the same plant populations. The container experiment involved three winter wheat cultivars, grown as sole crops or intercropped with winter pea under well-watered or drought-stressed conditions. The wheat root activity (characterized by CR) and the MR-based root length (RL) and root surface area (RSA) were monitored during the vegetation period, the flag leaf chlorophyll content was measured at flowering, and the wheat shoot dry mass (SDM) and grain yield (GY) were determined at maturity. CR, RL and RSA exhibited similar seasonal patterns with peaks around the flowering. The presence of pea reduced the maximum CR, RL and RSA. Drought significantly decreased CR, but increased the MR-based root size. Both intercropping and drought reduced wheat chlorophyll content, SDM and GY. The relative decrease caused by pea or drought in the maximum CR was proportional to the rate of change in SDM or GY. Significant linear correlations (R2: 0.77-0.97) were found between CR and RSA, with significantly smaller specific root capacitance (per unit RSA) for the drought-stress treatments. CR measurements tend to predict root function and the accompanying effect on above-ground production and grain yield. The parallel application of the two in situ methods improves the evaluation of root dynamics and plant responses.

Understanding the genetic diversity of Aegilops biuncialis, a valuable source of agronomical useful genes, may significantly facilitate the introgression breeding of wheat. The genetic diversity and population structure of 86 Ae. biuncialis genotypes were investigated by 32700 DArT markers with the simultaneous application of three statistical methods- neighbor-joining clustering, Principal Coordinate Analysis, and the Bayesian approach to classification. The collection of Ae. biuncialis accessions was divided into five groups that correlated well with their eco-geographic habitat: A (North Africa), B (mainly from Balkans), C (Kosovo and Near East), D (Turkey, Crimea, and Peloponnese), and E (Azerbaijan and the Levant region). The diversity between the Ae. biuncialis accessions for a phenological trait (heading time), which is of decisive importance in the adaptation of plants to different eco-geographical environments, was studied over 3 years. A comparison of the intraspecific variation in the heading time trait by means of analysis of variance and principal component analysis revealed four phenotypic categories showing association with the genetic structure and geographic distribution, except for minor differences. The detailed exploration of genetic and phenologic divergence provides an insight into the adaptation capacity of Ae. biuncialis, identifying promising genotypes that could be utilized for wheat improvement.

Broadening the genetic base of crops is crucial for developing varieties to respond to global agricultural challenges such as climate change. Here, we analysed a diverse panel of 371 domesticated lines of the model crop barley to explore the genetics of crop adaptation. We first collected exome sequence data and phenotypes of key life history traits from contrasting multi-environment common garden trials. Then we applied refined statistical methods, including some based on exomic haplotype states, for genotype-by-environment (G×E) modelling. Sub-populations defined from exomic profiles were coincident with barley's biology, geography and history, and explained a high proportion of trial phenotypic variance. Clear G×E interactions indicated adaptation profiles that varied for landraces and cultivars. Exploration of circadian clock-related genes, associated with the environmentally adaptive days to heading trait (crucial for the crop's spread from the Fertile Crescent), illustrated complexities in G×E effect directions, and the importance of latitudinally based genic context in the expression of large-effect alleles. Our analysis supports a gene-level scientific understanding of crop adaption and leads to practical opportunities for crop improvement, allowing the prioritisation of genomic regions and particular sets of lines for breeding efforts seeking to cope with climate change and other stresses.

The profiles of polar metabolites were determined in wholemeal flours of grain from the Broadbalk wheat experiment and from plants grown under organic and low-input systems to study the effects of nutrition on composition. The Broadbalk samples showed increased amino acids, acetate, and choline and decreased fructose and succinate with increasing nitrogen fertilization. Samples receiving farm yard manure had similar grain nitrogen to those receiving 96 kg of N/ha but had higher contents of amino acids, sugars, and organic acids. A comparison of the profiles of grain from organic and low-input systems showed only partial separation, with clear effects of climate and agronomy. However, supervised multivariate analysis showed that the low-input samples had higher contents of many amino acids, raffinose, glucose, organic acids, and choline and lower sucrose, fructose, and glycine. Consequently, although differences between organic and conventional grain occur, these cannot be used to confirm sample identity.

The root electrical capacitance (C R ) method is suitable for assessing root growth and activity, but soil water content (SWC) strongly influences the measurement results. This study aimed to adapt the method for field monitoring by evaluating the effect of SWC on root capacitance to ensure the comparability of C R detected at different SWC. First a pot experiment was conducted with maize and soybean to establish C R -SWC functions for the field soil. Ontogenetic changes in root activity were monitored under field conditions by simultaneously measuring C R and SWC around the roots. The C R values were normalized using SWC data and experimental C R -SWC functions to obtain C R*, the comparable indicator of root activity. The effect of arbuscular mycorrhizal fungi (AMF) inoculation on the C R* and biomass of field-grown soybean was investigated. The pot trial showed an exponential increase in C R with SWC. C R -SWC functions proved to be species-specific. C R showed strong correlation with root dry mass (R2 = 0.83-0.87). The root activity (C R*) of field-grown crops increased until flowering, then decreased during maturity. This was consistent with data obtained with other methods. AMF inoculation of soybean resulted in significantly higher C R* during the late vegetative and early flowering stages, when destructive sampling concurrently showed higher shoot biomass. The results demonstrated that the root capacitance method could be useful for time course studies on root activity under field conditions, and for comparing single-time capacitance data collected in areas with heterogeneous soil water status.

The cytomolecular discrimination of the Am- and A-genome chromosomes facilitates the selection of wheat-Triticum monococcum introgression lines. Fluorescence in situ hybridisation (FISH) with the commonly used DNA probes Afa family, 18S rDNA and pSc119.2 showed that the more complex hybridisation pattern obtained in T. monococcum relative to bread wheat made it possible to differentiate the Am and A chromosomes within homoeologous groups 1, 4 and 5. In order to provide additional chromosomal landmarks to discriminate the Am and A chromosomes, the microsatellite repeats (GAA)n, (CAG)n, (CAC)n, (AAC)n, (AGG)n and (ACT)n were tested as FISH probes. These showed that T. monococcum chromosomes have fewer, generally weaker, simple sequence repeat (SSR) signals than the A-genome chromosomes of hexaploid wheat. A differential hybridisation pattern was observed on 6Am and 6A chromosomes with all the SSR probes tested except for the (ACT)n probe. The 2Am and 2A chromosomes were differentiated by the signals given by the (GAA)n, (CAG)n and (AAC)n repeats, while only (GAA)n discriminated the chromosomes 3Am and 3A. Chromosomes 7Am and 7A could be differentiated by the lack of (GAA)n and (AGG)n signals on 7A. As potential landmarks for identifying the Am chromosomes, SSR repeats will facilitate the introgression of T. monococcum chromatin into wheat.

An assessment was previously made of the effects of organic and low-input field management systems on the physical, grain compositional and processing quality of wheat and on the performance of varieties developed using different breeding methods ("Comparison of quality parameters of wheat varieties with different breeding origin under organic and low-input conventional conditions" [1]). Here, accompanying data are provided on the performance and stability analysis of the genotypes using the coefficient of variation and the 'ranking' and 'which-won-where' plots of GGE biplot analysis for the most important quality traits. Broad-sense heritability was also evaluated and is given for the most important physical and quality properties of the seed in organic and low-input management systems, while mean values and standard deviation of the studied properties are presented separately for organic and low-input fields.

A recently developed synthetic amphiploid, Triticum timococcum Kost., nom. nud. (2n = 6x = 42, AtAtGGAmAm) is described in the present study. This hexaploid taxon was developed by colchicine treatment in Martonvásár from the hybrid of a selected accession of Triticum timopheevii Zhuk. (2n = 4x = 28, AtAtGG) and a prebred semi-dwarf line of Triticum monococcum L. (2n = 2x = 14, AmAm). A detailed cytomolecular examination was carried out using the sequential multicolour fluorescence and genomic in situ hybridization techniques (FISH and mcGISH). It was proved that T. timococcum has 42 chromosomes originating from its parents. The chromosomes of the A genomes of T. monococcum and T. timopheevii could be distinguished in the amphiploid using FISH. The successful discrimination of the chromosomes was supported by the karyotypes of the three genomes and the successful optimization of the mcGISH technique for the A and G chromosomes achieved in the present study. A phenotypic evaluation was also carried out under natural and artificial growing conditions in 2012 and 2013. Based on the results, T. timococcum has intermediate characteristics in terms of spike (spikelet) shape and plant height, while it is similar to the female parent, T. timopheevii regarding pubescence. Like its parents, T. timococcum showed outstanding resistance to the main fungal diseases of wheat. T. timococcum headed later and developed longer and looser spikes, fewer tillers and only a third as many seeds than its parents. The third generation of T. timococcum was able to develop an acceptable number of seeds, even taking into account the reduced germination ability in the field.

INTRODUCTION: Oral potassium replacement is still inevitable. To reduce the irritation of the gastric and intestinal mucosa, pellet and matrix based formulations ensuring extended release of potassium chloride are used. The dissolution tests may help to understand the in vivo steps of the release of potassium chloride and the absorption of potassium. AIM: Using dissolution tests extended to 12 hours the authors evaluated potassium chloride release characteristics of pellet and matrix tablet based formulations used for potassium replacement. METHOD: The tests were performed in line with the CPMP/EWP/QWP/1401/98 guideline at nine time points (0, 1, 2, 3, 4, 5, 7, 9 and 12 hours) in three dissolution media (0.1 M hydrochloric acid, pH 1.2; acetate buffer, pH 4.5; phosphate buffer, pH 6.8). RESULTS: Similar results were found in all three dissolution media. CONCLUSIONS: It is conceivable, that the release of potassium chloride begins already in the stomach (pH = 1.2) and at an average speed of gastrointestinal transit - in about 6-7 hours - 80% of the potassium chloride content of both formulations is dissolved by the time of the entrance to the large bowel. It seems likely, that in vivo in the proximal section of the gastrointestinal tract more potassium chloride is dissolved out of the matrix based formulation, than from the pellet based one. Both formulations meet the clinical requirements of the effective potassium chloride release.

INTRODUCTION: The vitamin D3 formation in skin is the most important natural source of vitamin D demands of humans. The key step of the phototransformation of provitamin D into previtamin D from which the vitamin D3 is formed by thermal conversion. According to studies run at the latitudes of Hungary the UV-B radiation in winter time is not satisfactory to ensure the need of the vitamin D, which can result in vitamin D 3 deficiency and increases risk of osteoporosis. AIMS: The aim of this study was to verify whether in the Hungarian population the UV radiation from the Sun ensures the daily synthesis of 1000 IU vitamin D3, or Vitamin D deficiency could occur in the winter and spring months which are less favourable in terms of UV exposure. METHODS: The data of UV-B radiation reaching the Earth's surface were used for the evaluation. These data based on regular measurements in Budapest. According to the average of UV-B radiation of each months the "best case scenario" of vitamin D3 production was estimated by using the most optimal conditions of vitamin D synthesis. RESULTS: It was calculated, that the effective UV irradiance reaching the Earth's surface at noon in Budapest is the highest in July, while the lowest is in December. The difference between these two months is more than 35-fold for July. In the period between November and March more than 200 minutes have to be spent outdoor to ensure the production of satisfactory amount of vitamin D in skin. On one hand, it is irrealistically long time because it exceeds the duration of maximum irradiation around noon which was the basis of our calculation. On the other hand, if only the face and hands are uncovered then the required radiant exposure exceeds the 1 minimal erythemal dose, i.e. the skin would be burnt. CONCLUSIONS: Based on our calculations it was found, that in Budapest more than 95% of yearly effective UV-B radiation is measurable in the period between March and October. Therefore it can be assumed that the UV-B radiation would not be sufficient in the period between November and February, even if its efficacy would not be limited by clothing and less time spent outdoor which are anyway characteristic and necessary in that part of the year.

INTRODUCTION: Butamirate citrate containing medicines are marketed in Hungary since 1991. Every year several hundred thousands of children and adults are treated with these pharmaceutical products. Aim of the study was to compare the prescription habits with the directions provided in the Summary of Product Characteristics and to get an overview on the safety and efficacy of these products. METHODS: The study results are based on the evaluation of 3215 questionnaires provided by 276 general practitioners and pulmonologists having medical practice in many regions and the capital of Hungary. Responses were included into the evaluation only if the name of the physician, the address and type of the practice, the therapeutic indication could be precisely identified and if the patient returned for at least one control visit. RESULTS: The main therapeutic indication of butamirate was cough due to acute lower respiratory tract infections (tracheitis, laryngitis, bronchitis). The treatment interval was usually 1-2 weeks. Depot tablets were prescribed according to the Summary of Product Characteristics. The doses of syrup and drops were frequently and notoriously below the one proposed in the Summary of Product Characteristics. In 14% of the cases the syrup was prescribed for the treatment of children below 3 years of age. All three products were regarded as effective. Adverse events (nausea, vomiting, diarrhoea, skin exanthemas) were reported at 0.5-1% of the patients. CONCLUSIONS: The prescribed average daily doses of drops between the ages 1 month-12 years and the syrup between the ages 6-12 years were 20-30% below the recommended amounts. In Hungary the butamirate citrate containing medicines are regarded to be effective. The adverse events were rare and non-serious.

INTRODUCTION: Dimethindene maleate, the active substance of the extended-release Fenistil 24 capsule, is a first-generation histamine receptor antagonist, which is marketed in 10 European countries. In Hungary approximately 200,000 boxes are sold yearly from this product, which is mainly used for treatment of symptoms associated with allergic diseases (urticaria, itch, hay fever) The aim of the present study was to gather information with the help of medical doctors about the indications, efficacy and primarily the safety of use, observed side effects and frequency of adverse events associated with this product. METHOD: During the study 4574 questionnaires were evaluated out of the 5578 filled by 249 doctors. Responses were included into the evaluation only if the address and type of polyclinic of the doctor filling the questionnaire, indication of the use of the product could be precisely identified and the patient returned for control visit at least once. RESULTS: Based on the study results in Hungary in most cases the product is prescribed according to the indications and dosing schedule given in the SPC. In this study participating doctors found the drug to be effective in 95% of cases. Unexpected, new adverse events were not observed during the study. Approximately 22% of patients reported adverse events, tiredness and sleepiness being the most frequent ones. Occurrence of sedative adverse events was more frequent among those who were prone to tiredness (51 vs. 17%). The adverse events most frequently were reported 10-12 hours after intake. The average length of treatment was 4 weeks or less. The medicine is taken usually at night, before going to sleep. CONCLUSIONS: In Hungary in most cases the product is prescribed according to the indications and dosing schedule given in the current approved Summary of Product Characteristics.
