RESUMO

The dataset focuses on evaluating the performance of 17 sweet potato varieties (G) released by the Bangladesh Agricultural Research Institute (BARI) in terms of storage root yield and stability across five locations (E) in Bangladesh-Gazipur, Bogura, Jamalpur, Jashore, and Chattogram. The result revealed that BARI Mistialu-12 exhibited the highest average storage root yield at 45.35 t/ha, closely followed by BARI Mistialu-16 at 44.64 t/ha. Conversely, BARI Mistialu-1 had the lowest mean yield of 25.99 t/ha. Among the locations, Bogura recorded the highest mean root yield at 37.05 t/ha, while Chattogram exhibited the lowest at 31.27 t/ha. A combined analysis of variance revealed the presence of variability in storage root yield attributed to the genotype-location (environment) interaction (GEI). To delve deeper into this interaction, additive and multiplicative interaction effect models (AMMI) along with a linear mixed model (LMM) were employed for further investigations to confirm the significant contribution of GEI variance to root yield. The LMM results showed genetic variance (%), heritability (%), selection accuracy (%), and GEI correlation coefficients of 52.27, 54, 94, and 30, respectively. The AMMI analysis indicated that the first two principal components accounted for 74.60 % of GEI, with 20.16 % attributed to it. Assessing significant Interaction Principal Component Analyses (IPCAs) through the Weighted Average of Absolute Scores (WAAS) indicated that BARI Mistialu-12 is the most stable genotype, followed by BARI Mistialu-16 and BARI Mistialu-8, all displaying above-average root yield. The mega-environment analysis associated the highest root production of BARI Mistialu-11 and BARI Mistialu-2 with the Jamalpur location, while Gazipur, Bogura, and Jashore were linked with the superior performance of BARI Mistialu-12 and BARI Mistialu-16 genotypes. These findings are crucial for future breeding programs and the rapidly growing sweet potato industry, given the stable high-yield potential across diverse agro-ecological conditions. However, it is imperative to repeat the study to ensure reliable outcomes.

RESUMO

The dataset primarily focused on selecting genotypes of sweet oranges based on their phenotypic performances. The dataset resulted significant variations in the best linear unbiased predictions (BLUPs) of 20 out of 21 traits, including leaves, flowers, fruits, and seeds. A strong positive correlation (r= 0.73 to 0.95) was observed among the majority of morphological traits. The sweet orange genotypes demonstrated considerable genetic variance, surpassing 65% for almost all traits, with a selection accuracy exceeding 92%. Using the multi-trait genotype-ideotype distance index (MGIDI), CS Jain-001 emerged as the top-ranked genotype, followed by BAU Malta-3 and CS Jain-002 in order of desirability. The broad sense heritability of selected traits was above 75.60%, and the selection gain reached a maximum of 12.60. These identified genotypes show promise as potential parent donors in breeding programs, leveraging their strengths and weaknesses to develop promising varieties in Bangladesh.

RESUMO

The dataset explores the impact of waterlogging stress on sesame plants during the pre-flowering stage, recognizing its global impact on crop yield and the identification of tolerant genotypes using the MGIDI index. Carried out in Bangladesh, the research assesses the survival status, grain yield, and stress tolerance indices of 40 sesame genotypes, revealing that twelve of them demonstrated resilience under 72 h of waterlogging stress at the pre-flowering stage. There were variations in genotypic grain yield, and G15 exhibited the highest yields, recording 5.22 g/plant under normal conditions and 4.10 g/plant under waterlogging stress. The MGIDI index, evaluating waterlogging tolerance, identified G4 as the most favorable genotype, followed by G5 and G12. Factor analysis within the MGIDI index uncovered distinct tolerance and susceptibility indices, highlighting strengths and weaknesses in the selected genotypes. The selection gain percentages of these genotypes ranged from 12.9 to 37.4, indicating high broad-sense heritability (≥0.97). These results underscore the potential of genotype selection based on waterlogging stress indices, providing valuable insights for breeders addressing stress-related crop challenges in the face of changing climatic conditions.

RESUMO

Monoamniotic twins develop when a blastocyst spontaneously splits its progenitor cells, and each group of progenitor cells independently grows to become an individual. It is the rarest type of twin pregnancy and usually has significant developmental or congenital abnormalities, a higher rate of abortion, perinatal morbidity, and mortality. There is no information regarding monoamniotic twins in livestock species. Here, we reported a spontaneous abortion of monoamniotic twins in a dromedary camel at 278 days of gestation. Gonadorelin acetate (100 µg) was injected intramuscularly to induce ovulation in the recipient. A 7 days-old embryo produced by somatic cell nuclear transfer was transferred transcervically to the recipient. Early pregnancy was confirmed by an elevated level of serum progesterone followed by ultrasonography at 22 and 44 days after embryo transfer. A single sac was observed on 22 days while twins were evident 44 days after embryo transfer. Pregnancy was periodically monitored by the tail-up phenomenon. A ruptured fetal sac was observed on the ground having two fetuses. On autopsy, full-grown fetuses were found. Their bodies were separated. There was no congenital anomaly or any malformation in the fetuses. According to the reported chronology in human twins, we hypothesized that the blastocyst splitted before 13 days as it was monoamniotic and not conjoined. If the embryo splits within 4 to 8 days, it develops two amniotic sacs, and splitting after 13 days develops conjoined fetuses. To the authors' knowledge, this is the first reported case of monoamniotic twin abortion in dromedary camels. This report will increase awareness among practicing veterinarians and camel breeders about twin abortions.

RESUMO

In recent times, high-performance wearable electronic devices that can transform mechanical force into electrical energy for biomedical monitoring applications are receiving an increasing amount of attention. In the present study, we focused on a flexible, self-powered and wearable triboelectric nanogenerator (TENG) based on electrospun polyvinylidene fluoride (PVDF)/aromatic hyperbranched polyester of 4th generation (Ar.HBP-G4, 0-40 wt.-% w.r.t. PVDF content) blend nanoweb as tribo-negative layer and melt-blown thermoplastic polyurethane (TPU) as tribo-positive layer for energy harvesting and human health monitoring applications. Among the varying Ar.HBP-G4 content used, incorporation of Ar.HBP-G4 (10 wt.-%) in PVDF (P-Ar.HBP-G4-10) showed higher increase in the triboelectric output voltage when compared to pristine PVDF and other Ar.HBP-G4 weight ratios. The optimized P-Ar.HBP-G4-10/TPU based TENG exhibited a peak-to-peak voltage (Vp-p) of 124.4 V under an applied load of 9.8 N and frequency 1 Hz which is superior to many other TENGs reported elsewhere. Higher triboelectric performance of P-Ar.HBP-G4 blend based TENG compared to that of neat PVDF is attributed to the effect of Ar.HBP-G4-10 in enhancing the degree of crystallinity and polar ß-crystalline phase content (98.3%) in PVDF. The ability of the TENG to power up portable electronic devices is demonstrated when it is powered for 750 s while connected through a capacitor and a rectifier, and the TENG was able to operate 45 light-emitting diodes directly. Evaluation of the triboelectric output of the TENG device attached to different parts of the human body reveal significantly better output voltage and sensitivity for human health monitoring. The results of this work pave a new way to develop TENG based on P-Ar.HBP-G4 nanowebs for sustainable energy generation and wearable healthcare monitoring systems.

RESUMO

Propagation of transgenic animals by germline transmission using assisted reproductive technologies such as in vitro fertilization (IVF) is the most efficient way to produce transgenic colonies for biomedical research. The objective of this study was to generate transgenic puppies from a founder dog expressing the mutated human amyloid precursor protein (mhAPP) gene. Experiment I assessed the characteristics of the semen prepared by freshly diluted, swim-up, and Percoll gradient methods using a computer-assisted semen analyzer (CASA). Motile and progressively motile sperm counts were higher in the Percoll gradient samples (p < 0.05) than in the swim-up and freshly diluted samples. In Experiment II, a total of 59, 70, and 65 presumptive zygotes produced by fresh, Percoll gradient, and swim-up methods, respectively, were transferred to surrogates (5 for each group); the Percoll gradient (27.27%) and swim-up samples (14.29%) showed the highest blastocyst formation rates, while fresh diluted semen did not produce any blastocyst. Experiment III examined the full-term developmental ability of embryos. Among the 5 surrogates in the Percoll gradient group, one (20.0%) became pregnant; it had 4 (6.15%) sacs and delivered 4 (6.15%; 2 males and 2 females) live puppies. Among the 4 puppies, 2 (50.0%) were found to transmit the transgene on their nail and toe under GFP fluorescence. Furthermore, the integration and expression of the mhAPP transgene were examined in the umbilical cords of all the IVF-derived puppies, and the presence of the transgene was only observed in the GFP-positive puppies. Thus, semen prepared by the Percoll method could generate transgenic puppies by male germline transmission using the IVF technique. Our result will help propagate transgenic dogs efficiently, which will foster human biomedical research.

RESUMO

This study aims to find base materials for dry electrode fabrication with high accuracy and without reducing electrode performance for long-term bioelectric potential monitoring after electroless silver plating. Most applications of dry electrodes that have been developed in the past few decades are restricted by low accuracy compared to commercial Ag/AgCl gel electrodes, as in our previous study of PVDF-based dry electrodes. In a recent study, however, nanoweb-based chlorinated polyisoprene (CPI) and poly(styrene-b-butadiene-b-styrene) (SBS) rubber were selected as promising candidates due to their excellent elastic properties, as well as their nanofibril nature, which may improve electrode durability and skin contact. The electroless silver plating technique was employed to coat the nanofiber web with silver, and silver nanoweb(AgNW)-based dry electrodes were fabricated. The key electrode properties (contact impedance, step response, and noise characteristics) for AgNW dry electrodes were investigated thoroughly using agar phantoms. The dry electrodes were subsequently tested on human subjects to establish their realistic performance in terms of ECG, EMG monitoring, and electrical impedance tomography (EIT) measurements. The experimental results demonstrated that the AgNW dry electrodes, particularly the SBS-AgNW dry electrodes, performed similarly to commercial Ag/AgCl gel electrodes and were outperformed in terms of long-term stability.

RESUMO

Hepatitis C virus (HCV) is a causative agent that causes chronic liver diseases worldwide. It is a little, enclosed, single-stranded ribonucleic acid (RNA) virus. The recognition of the pathogenic HCV genotype is critical for the remedy of its sufferers. The aim of this study was to identify the HCV RNA genotype to decide the correct treatment of hepatitis C positive sufferers in Bangladesh. Blood samples were collected from 390 individuals and isolated RNA (60 µg) from blood plasma. Extracted RNA was used for quantitative HCV RNA, and complementary DNA (cDNA) was prepared by polymerase chain reaction (PCR) conducted by reverse transcriptase enzyme. This cDNA amplified in multiplex by RT-PCR, which was performed with specific set of primers. The HCV RNA genotype was detected 297 of 390 patients. Of the 390 test samples, 200 (51.28%) samples were from males and 190 (48.71%) were from females, with age ranging from 5 to 78 years. In all, 166 of 200 male samples and 131/190 female samples were found positive for HCV. Of these 390 participants included in the study, 213 (54.61%) were identified as genotype 3 positive, 78 (20%) as genotype 1 positive, 6 (1.53%) as genotype 6 positive, and the remaining 93 (23.85%) samples were unclassified due to low/undetected viral load. In this study, we detected the highest percentage (30.89%) of genotype 3 HCV in patients aged 51 to 60 years. The results suggested that genotype 3 HCV is frequently present in Bangladesh and it is usually responses better to interferon therapy. However, genotype 1 and 6 HCV have also been found circulating in this country, which demands longer treatments and effective control measures.

RESUMO

The present study was conducted to evaluate the number and maturity of the recovered oocytes after two intervals of in-vivo maturation. In addition to evaluating the effect of the developmental stage, as well as the number of cloned transferred blastocysts on the pregnancy rate and early pregnancy loss (EPL) in dromedary camel. The donor animals (n = 52) were super-stimulated using a single injection of 3000 IU of eCG followed by GnRH administration for oocyte maturation. Cumulus oocyte complexes (COCs) were collected by transvaginal ultrasound-guided aspiration (OPU) either 24-26 h or 18-20 h after GnRH administration. A fewer number of COCs with a lower percentage of oocyte maturity was observed at 24-26 h in comparison to 18-20 h. The effect of the cloned blastocysts' transferred number and developmental stage on the pregnancy rate and EPL was investigated. The total pregnancy rates at 10 days post-ET, 1 and 2 months were 21.9, 12.4, and 8.6%, respectively. Transfer of two or 3-4 embryos per surrogate was accompanied with a higher pregnancy rate at 1 and 2 months than a single embryo transfer. Rates of EPL were 43.5 and 60.1% at 1 and 2 months of pregnancy, respectively. The transfer of two embryos per surrogate was associated with a lower rate of EPL than ET of a single embryo at 1 and 2 months of pregnancy. Also, the ET of 3-4 embryos per surrogate showed a higher rate of EPL than the ET of two embryos at 2 months of pregnancy. ET of hatching (HG) blastocysts showed higher pregnancy rates and fewer EPL than ET of unhatched (UH) or fully hatched (HD) cloned blastocysts at 1 and 2 months of pregnancy. In conclusion, a high number of in-vivo matured oocytes can be recovered by ultrasound-guided transvaginal OPU from super-stimulated females using 3000 IU eCG and an interval of 18-20 h after GnRH administration. The transfer of two hatching cloned blastocytes per surrogate increases the pregnancy rate and decreases EPL in dromedary camels.

RESUMO

A flexible and portable triboelectric nanogenerator (TENG) based on electrospun polyvinylidene fluoride (PVDF) doped with copper oxide (CuO) nanoparticles (NPs, 2, 4, 6, 8, and 10 wt.-% w.r.t. PVDF content) was fabricated. The structural and crystalline properties of the as-prepared PVDF-CuO composite membranes were characterized using SEM, FTIR, and XRD. To fabricate the TENG device, the PVDF-CuO was considered a tribo-negative film and the polyurethane (PU) a counter-positive film. The output voltage of the TENG was analyzed using a custom-made dynamic pressure setup, under a constant load of 1.0 kgf and 1.0 Hz frequency. The neat PVDF/PU showed only 1.7 V, which further increased up to 7.5 V when increasing the CuO contents from 2 to 8 wt.-%. A decrease in output voltage to 3.9 V was observed for 10 wt.-% CuO. Based on the above results, further measurements were carried out using the optimal sample (8 wt.-% CuO). Its output voltage performance was evaluated as a function of varying load (1 to 3 kgf) and frequency (0.1 to 1.0 Hz) conditions. Finally, the optimized device was demonstrated in real-time wearable sensor applications, such as human motion and health-monitoring applications (respiration and heart rate).

RESUMO

Since the last decade, hyperbranched polymers (HBPs) have gained wider theoretical interest and practical applications in sensor technology due to their ease of synthesis, highly branched structure but dimensions within nanoscale, a larger number of modified terminal groups and lowering of viscosity in polymer blends even at higher HBP concentrations. Many researchers have reported the synthesis of HBPs using different organic-based core-shell moieties. Interestingly, silanes, as organic-inorganic hybrid modifiers of HBP, are of great interest as they resulted in a tremendous improvement in HBP properties like increasing thermal, mechanical and electrical properties compared to that of organic-only moieties. This review focuses on the research progress in organofunctional silanes, silane-based HBPs and their applications since the last decade. The effect of silane type, its bi-functional nature, its influence on the final HBP structure and the resultant properties are covered in detail. Methods to enhance the HBP properties and challenges that need to be overcome in the near future are also discussed.

RESUMO

Flexible pressure sensors have played an increasingly important role in the Internet of Things and human-machine interaction systems. For a sensor device to be commercially viable, it is essential to fabricate a sensor with higher sensitivity and lower power consumption. Polyvinylidene fluoride (PVDF)-based triboelectric nanogenerators (TENGs) prepared by electrospinning are widely used in self-powered electronics owing to their exceptional voltage generation performance and flexible nature. In the present study, aromatic hyperbranched polyester of the third generation (Ar.HBP-3) was added into PVDF as a filler (0, 10, 20, 30 and 40 wt.% w.r.t. PVDF content) to prepare nanofibers by electrospinning. The triboelectric performances (open-circuit voltage and short-circuit current) of PVDF-Ar.HBP-3/polyurethane (PU)-based TENG shows better performance than a PVDF/PU pair. Among the various wt.% of Ar.HBP-3, a 10 wt.% sample shows maximum output performances of 107 V which is almost 10 times that of neat PVDF (12 V); whereas, the current slightly increases from 0.5 µA to 1.3 µA. The self-powered TENG is also effective in measuring human motion. Overall, we have reported a simpler technique for producing high-performance TENG using morphological alteration of PVDF, which has the potential for use as mechanical energy harvesters and as effective power sources for wearable and portable electronic devices.

RESUMO

Development and validation of rapid and easy-to-perform diagnostics continue to be a high priority during the current COVID-19 pandemic. Although vaccines are now widely available, early detection and consistent transmission control provide ideal means to mitigate the spread of SARS-CoV-2. Nucleic acid-based real-time PCR tests are widely acknowledged as the gold standard for reliable diagnosis of COVID-19 infection. These tests are based on detecting viable or nonviable viral nucleic acids. SARS-CoV-2 spike protein is an alternative and ideal target for SARS-CoV-2 diagnosis in the early phase of infection, but point-of-care kits to detect the SARS-CoV-2 spike protein are limited. Here we describe a rapid and convenient method based on Lateral Flow Immunoassay (LFIA) to detect SARS-CoV-2 spike proteins, including SARS-CoV-2 variants (A.23.1, B.1.1.1, 1.617.2, B.1.1.7, B.1.351, P.1, N501Y, R.1, P681H, P3, UK, and South African) within 5 to 10 minutes. We generated highly specific monoclonal antibodies (mAbs) against rationally designed SARS-CoV-2 spike protein. Matched pair mAbs were selected by epitope mapping and employed as antigen capture reagents by spotting onto a nitrocellulose membrane and as detector reagents by conjugation with colloidal gold nanoparticles. We evaluated the performance of the LFIA using recombinant spike proteins of SARS-CoV-2 and several SARS-CoV-2 variants. The specificity of the LFIA was assessed using heat-inactivated SARS-CoV-2 and related human coronaviruses (HCoV-OC43, HCoV-229E, HCoV-HKU1, and HCoV-NL63) and an FDA-approved respiratory pathogens (RP) panel. The assay exhibited 98% specificity and acceptable performance with respect to the minimum limit of detection (25 ng/test) in validation tests. This new LFIA provides improved performance for the early diagnosis of SARS-CoV-2, particularly for home monitoring and in situations with limited access to molecular methods.

RESUMO

OBJECTIVE: Cutaneous ulcerative skin lesions in a complex of invasive Gulf of Mexico lionfish (Red Lionfish Pterois volitans, Devil Firefish P. miles, and the hybrid Red Lionfish × Devil Firefish) became epizootic beginning in mid-August 2017. Herein, we provide the first pathological descriptions of these lesions and summarize our analyses to elucidate the etiology of the disease. METHODS: We examined ulcerated and normal fish through gross pathology and histopathology, bacterial sampling, and unbiased metagenomic next-generation sequencing. We tracked prevalence of the disease, and we used biological health indicators (condition factor, splenosomatic and hepatosomatic index) to evaluate impacts to health, while considering sex and age as potential risk factors. RESULT: Typical ulcerative lesions were deep, exposing skeletal muscle, and were bordered by pale or reddened areas often with some degree of scale loss. Only incidental parasites were found in our examinations. Most fish (86%; n = 50) exhibited wound healing grossly and histologically, confirmed by the presence of granulation tissues. A primary bacterial pathogen was not evident through bacterial culture or histopathology. Metagenomic next-generation sequencing did not reveal a viral pathogen (DNA or RNA) but did provide information about the microbiome of some ulcerated specimens. Compared with clinically healthy fish, ulcerated fish had a significantly lower condition factor and a higher splenosomatic index. Disease prevalence at monitored sites through July 2021 indicated that ulcerated fish were still present but at substantially lower prevalence than observed in 2017. CONCLUSION: Although some common findings in a number of specimens suggest a potential role for opportunistic bacteria, collectively our suite of diagnostics and analyses did not reveal an intralesional infectious agent, and we must consider the possibility that there was no communicable pathogen.

RESUMO

Ornamentals come in a variety of shapes, sizes, and colors to suit a wide range of climates, landscapes, and gardening needs. Compared to demand, a shortage of plant materials and diversity force the search for solutions for their constant acquisition and improvement to increase their commercial value, respectively. In vitro cultures are a suitable solution to meet expectations using callus culture, somatic embryogenesis, protoplast culture, and the organogenesis of protocorm-like bodies; many of these techniques are commercially practiced. Factors such as culture media, explants, carbohydrates, plant growth regulators, and light are associated with the success of in vitro propagation. Techniques, especially embryo rescue and somatic hybridization, are widely used to improve ornamentals. The development of synthetic seed allows season-independent seed production and preservation in the long term. Despite the advantages of propagation and the improvement of ornamentals, many barriers still need to be resolved. In contrast to propagation and crop developmental studies, there is also a high scope for molecular studies, especially epigenetic changes caused by plant tissue culture of ornamentals. In this review, we have accumulated and discussed an overall update on cultivation factors, propagation techniques in ornamental plant tissue culture, in vitro plant improvement techniques, and future perspectives.

RESUMO

The umbilical cord acts as the critical lifeline of the developing fetus by providing nutrients and oxygen to it. Umbilical cord abnormalities are considered the leading cause of stillbirth in humans, but information on stillbirths associated with umbilical cord abnormalities is very scant in the clinical practice of animals. Here, we described a case of fetal demise in camels indicated to be caused by fetal death from strangulation by its umbilical cord, which is commonly known as the nuchal cord. A pregnant camel at its 36 weeks of gestation spontaneously aborted a single fetus. The camel was 5 years old and nullipara. A 6-day-old cloned embryo was transferred transcervically to the recipient. Pregnancy was confirmed 50 days after embryo transfer by ultrasonography, and the pregnant camel was maintained under a standard nutritional plan. The neck of the aborted fetus was strangulated tightly by a double loop of the umbilical cord. There was no congenital anomaly or other malformation in the fetus. We concluded that the nuchal cord was tightly coiled around the neck of the fetus and interfered with the blood flow in the fetus by collapsing the umbilical vein and subsequently causing fetal death and abortion. To the authors' knowledge, this is the first reported case of a nuchal cord in camels.

RESUMO

The present study investigated the effect of superstimulation to improve in vitro embryo production in the Gulf area, where the temperature is high. Holstein cows were classified into the control and superstimulation groups. Superstimulation was induced with a single intramuscular injection of pregnant mare serum gonadotropin (PMSG; 2500 IU) on day 14 of the estrus cycle (day 0; estrus). The development of follicles was evaluated by ultrasonography of the ovaries daily. At 40 h after the PMSG injection, oocytes were collected by the ovum pick-up (OPU) technique. OPU was performed at the same stage of the estrus cycle in the control group as in the superstimulation group. The number of follicles with a diameter of more than 6 mm and the number of retrieved cumulus-oocyte complexes were significantly higher in the superstimulation group than in the control group. Furthermore, the maturation rate was higher in the superstimulation group than in the control group. Cloned embryos were produced by somatic cell nuclear transfer using matured oocytes. The cleavage and blastocyst formation rates were significantly higher in the superstimulation group than in the control group. In conclusion, a single injection of PMSG can facilitate the efficient production of cloned cow embryos.

RESUMO

OBJECTIVE: The present study analyzed the influence of co-transferring embryos with high and low cloning efficiencies produced via somatic cell nuclear transfer (SCNT) on pregnancy outcomes in dogs. METHODS: Cloned dogs were produced by SCNT using donor cells derived from a Tibetan Mastiff (TM) and Toy Poodle (TP). The in vivo developmental capacity of cloned embryos was evaluated. The pregnancy and parturition rates were determined following single transfer of 284 fused oocytes into 21 surrogates and co-transfer of 47 fused oocytes into four surrogates. RESULTS: When cloned embryos produced using a single type of donor cell were transferred into surrogates, the pregnancy and live birth rates were significantly higher following transfer of embryos produced using TP donor cells than following transfer of embryos produced using TM donor cells. Next, pregnancy and live birth rates were compared following single and co-transfer of these cloned embryos. The pregnancy and live birth rates were similar upon co-transfer of embryos and single transfer of embryos produced using TP donor cells but were significantly lower upon single transfer of embryos produced using TM donor cells. Furthermore, the parturition rate for TM dogs and the percentage of these dogs that remained alive until weaning was significantly higher upon co-transfer than upon single transfer of embryos. However, there was no difference between the two embryo transfer methods for TP dogs. The mean birth weight of cloned TM dogs was significantly higher upon single transfer than upon co-transfer of embryos. However, the body weight of TM dogs did not significantly differ between the two embryo transfer methods after day 5. CONCLUSION: For cloned embryos with a lower developmental competence, the parturition rate and percentage of dogs that remain alive until weaning are increased when they are co-transferred with cloned embryos with a greater developmental competence.

RESUMO

We compared the pregnancy and live birth rates following transfer of early-stage embryos or blastocysts produced by somatic cell nuclear transfer using in vitro-matured oocytes. In total 102 ovaries were collected from dromedary camels at a local abattoir; from these 1048 cumulus-oocytes complexes (COCs) were aspirated and cultured for 42 h in a commercial maturation medium. Metaphase II oocytes were subjected to nuclear transfer. Somatic cell nuclear transfer-derived embryos were cultured in a commercial embryo medium for 2 or 7 days. Next, 71 early-stage embryos were surgically transferred to the left fallopian tube of 28 recipients and 47 blastocysts were transferred to the left uterine horn of 26 recipients. Early pregnancy was detected by serum progesterone (P4), and pregnancy was confirmed using ultrasonography on days 30 and 90 after embryo transfer. Pregnancy rate based on P4 level was 17.86% (5/28) and 11.54% (3/26) for early-stage embryo and blastocyst transfer, respectively. In the early-stage embryo group, out of five recipients, one recipient had lost the pregnancy by the first ultrasonography on day 30; two other recipients aborted at 14 and 24 weeks, and two recipients gave live births. In the blastocyst group, out of three recipients, one lost the pregnancy at an early stage and two recipients gave live births. Therefore, for dromedary camels, we recommend transvaginal blastocyst transfer from the standpoint of the pregnancy and live birth rate, ease of the transfer procedure, and comfort and safety of the recipients.
