RESUMO
The study focused on the extraction of free erythromycin from commercially manufactured tablets and the use of metal salts to synthesize erythromycin-metal complexes, specifically involving silver (Ag), nickel (Ni), cobalt (Co), and copper (Cu). The synthesis was confirmed through various methods, including elemental analysis, thermogravimetric analysis, Fourier-transform infrared (FTIR), and UV-visible spectroscopy. The microbiological investigation involved Salmonella typhi, Escherichia coli, Staphylococcus aureus, Bacillus cereus, Candida albicans, and Microsporum canis as test organisms. The NCCLS broth microdilution reference method was used to determine the minimum fungicidal concentration and minimum inhibitory concentration of the complexes. The synthesized complexes were highly effective against a variety of fungi and bacteria, with compound Ery-Cu having MIC as low as 1.56 mg/mL, Ery-Cu and Ery-Ni with MBCs of 6.25 mg/mL and Ery-Cu having MFC of 6.25 mg/mL. Dose-dependent inhibitory effects were found upon examination of the antimicrobial susceptibility of specific complexes (Cu, Ni, Co and Ag) at varying concentrations of 100, 50, 25 and 12.5 mm/mL. Antibiotic susceptibility testing revealed efficacy against the tested pathogens. The study suggests that the synthesis of erythromycin-metal complexes, coupled with their antibacterial effectiveness against a diverse spectrum of bacteria and fungi, as they showed promising inhibitory properties when tested against a range of test species (Bacillus cereus, Staphylococcus aureus, Escherichia coli, Salmonella typhi, Candida albicans, and Microsporum canis), could lead to the development of innovative antibacterial agents. Molecular docking simulations were used to examine the interactions between metal complexes with proteins filamentous temperature-sensitive protein Z and lanosterol 14α-demethylase. The study highlights the need for further exploration in pharmaceutical research.
RESUMO
Wastewater-based surveillance (WBS) has shown to be an effective tool in monitoring the spread of SARS-CoV-2 and has helped guide public health actions. Consequently, WBS has expanded to now include the monitoring of mpox virus (MPXV) to contribute to its mitigation efforts. In this study, we demonstrate a unique sample processing and a molecular diagnostic strategy for MPXV detection that can inform on the epidemiological situation of mpox outbreaks through WBS. We conducted WBS for MPXV in 22 Canadian wastewater treatment plants (WWTPs) for 14 weeks. Three MPXV qPCR assays were assessed in this study for the detection of MPXV which include the G2R assays (G2R_WA and G2R_G) developed by the Centers for Disease Control and Prevention (CDC) in 2010, and an in-house-developed assay that we have termed G2R_NML. The G2R_NML assay was designed using reference genomes from the 2022 MPXV outbreak and provides a larger qPCR amplicon size to facilitate Sanger sequencing. Results show that all three assays have similar limits of detection and are able to detect the presence of MPXV in wastewater. The G2R_NML assay produced a significantly greater number of Sanger sequence-confirmed MPXV results compared to the CDC G2R assays. Detection of MPXV was possible where provincial surveillance indicated overall low caseloads, and in some sites forewarning of up to several weeks was observed. Overall, this study proposes that WBS of MPXV provides additional information to help fill knowledge gaps in clinical case-surveillance and is potentially an essential component to the management of mpox.

Phosphite represents a reduced form of phosphate that belongs to a class of crop growth-promoting chemicals termed biostimulants. Previous research has shown that phosphite application can enhance root growth, but its underlying mechanism, especially during environmental stresses, remains elusive. To uncover this, we undertook a series of morphological and physiological analyses under nutrient, water and heat stresses following a foliar application in wheat. Non-invasive 3D imaging of root system architecture directly in soil using X-ray Computed Tomography revealed that phosphite treatment improves root architectural traits and increased root biomass. Biochemical and physiological assays identified that phosphite treatment significantly increases Nitrate Reductase (NR) activity, leaf photosynthesis and stomatal conductance, suggesting improved Nitrogen and Carbon assimilation, respectively. These differences were more pronounced under heat or drought treatment (photosynthesis and photosystem II stability) and nutrient deficiency (root traits and NR). Overall our results suggest that phosphite treatment improves the ability of plants to tolerate abiotic stresses through improved Nitrogen and Carbon assimilation, combined with improved root growth which may improve biomass and yield.
RESUMO
Throughout the coronavirus disease 2019 (COVID-19) pandemic, wastewater surveillance has been used to monitor trends in severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) prevalence in the community. A major challenge in establishing wastewater surveillance programs, especially in remote areas, is the need for a well-equipped laboratory for sample analysis. Currently, no options exist for rapid, sensitive, mobile, and easy-to-use wastewater tests for SARS-CoV-2. The performance of the GeneXpert system, which offers cartridge-based, rapid molecular clinical testing for SARS-CoV-2 in a portable platform, was evaluated using wastewater as the input. The GeneXpert demonstrated a SARS-CoV-2 limit of detection in wastewater below 32 copies/mL with a sample processing time of less than an hour. Using wastewater samples collected from multiple sites across Canada during February and March 2021, a high overall agreement (97.8%) was observed between the GeneXpert assay and laboratory-developed tests regarding the presence or absence of SARS-CoV-2. Additionally, with the use of centrifugal filters, the detection threshold of the GeneXpert system was improved to <10 copies/mL in wastewater. Finally, to support on-site wastewater surveillance, GeneXpert testing was implemented in Yellowknife, a remote community in Northern Canada, where its use successfully alerted public health authorities to undetected transmission of COVID-19. The identification of SARS-CoV-2 in wastewater triggered clinical testing of recent travelers and identification of new COVID-19 cases/clusters. Taken together, these results suggest that GeneXpert is a viable option for surveillance of SARS-CoV-2 in wastewater in locations that do not have access to established testing laboratories. IMPORTANCE Wastewater-based surveillance is a powerful tool that provides an unbiased measure of COVID-19 prevalence in a community. This work describes a sensitive wastewater rapid test for SARS-CoV-2 based on a widely distributed technology, the GeneXpert. The advantages of an easy-to-use wastewater test for SARS-CoV-2 are clear: it supports surveillance in remote communities, improves access to testing, and provides faster results allowing for an immediate public health response. The application of wastewater rapid testing in a remote community facilitated the detection of a COVID-19 cluster and triggered public health action, clearly demonstrating the utility of this technology. Wastewater surveillance will become increasingly important in the postvaccination pandemic landscape as individuals with asymptomatic/mild infections continue transmitting SARS-CoV-2 but are unlikely to be tested.

We report the case of a previously healthy 49-year-old woman who presented with upper gastrointestinal bleeding, which was found at laparotomy to be due to high-grade B cell gastric lymphoma. CT scans showed that this was partially adherent to the spleen, with erosion of the gastric wall and suggested impending perforation. Given the risk of perforation, further surgical intervention (gastrectomy and splenectomy) was considered; however, after multidisciplinary team discussion, we chose to offer chemotherapy and careful inpatient observation instead. Our patient made a full recovery with no perforation. The message from our experience and literature review is that medical management may lead to a more favourable outcome in gastric lymphoma than surgery, despite radiological appearances suggesting impending perforation. This approach avoids the risk of the lymphoma progressing at other anatomical sites secondary to delays in giving chemotherapy. If this approach is followed, the patient must be carefully monitored.

High proportion of copper has become a global challenge owing to its negative impact on the environment and public health complications. The present study focuses on the fabrication of a polyvinylidene fluoride (PVDF)-polyvinyl pyrrolidone (PVP) fiber membrane incorporated with varying loading (0, 0.5, 1.0, 1.5, and 2.0 wt%) of titanium dioxide (TiO2) nanoparticles via phase inversion technique to achieve hydrophilicity along with high selectivity for copper removal. The developed fibers were characterized based on scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDX), permeability, porosity, zeta potential, and contact angle. The improved membrane (with 1.0 wt% TiO2) concentration recorded the maximum flux (223 L/m2·h) and copper rejection (98.18%). Similarly, 1.0 wt% concentration of TiO2 nanoparticles made the membrane matrix more hydrophilic with the least contact angle of 50.01°. The maximum copper adsorption capacity of 69.68 mg/g was attained at 1.0 wt% TiO2 concentration. The experimental data of adsorption capacity were best fitted to the Freundlich isotherm model with R2 value of 0.99573. The hybrid membrane developed in this study has considerably eliminated copper from leachate and the concentration of copper in the permeate was substantially reduced to 0.044 mg/L, which is below standard discharge threshold.
RESUMO
The present study synthesized nano-magnetite (Fe3O4) from milled steel chips using the high energy ball milling (HEBM) method, characterized it, and then utilized it as a sorbent to remediate boron concentration at various pH (4-9), dosages (0.1-0.5 g), contact times (20-240 min), and initial concentrations (10-100 mg/L). The nano-sorbents were characterized based on SEM structure, elemental composition (EDX), surface area analysis (BET), crystallinity (XRD), and functional group analysis (FTIR). The highest adsorption capacity of 8.44 mg/g with removal efficiency of 84% was attained at pH 8, 0.5 g dosage, contact time of 180 min, and 50 mg/L initial concentration. The experimental data fit best with the pseudo-second-order kinetic model with R2 of 0.998, while the Freundlich adsorption isotherm describes the adsorption process with an R2 value of 0.9464. A regeneration efficiency of 47% was attained even after five cycles of reusability studies. This efficiency implies that the nano-magnetite has the potential for sustainable industrial application.

Globally, there are prevailing knowledge gaps in the epidemiology, clinical manifestations, and outcomes of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection among children and adolescents; and these gaps are especially wide in African countries. The availability of robust age-disaggregated data is a critical first step in improving knowledge on disease burden and manifestations of coronavirus disease 2019 (COVID-19) among children. Furthermore, it is essential to improve understanding of SARS-CoV-2 interactions with comorbidities and coinfections such as human immunodeficiency virus (HIV), tuberculosis, malaria, sickle cell disease, and malnutrition, which are highly prevalent among children in sub-Saharan Africa. The African Forum for Research and Education in Health (AFREhealth) COVID-19 Research Collaboration on Children and Adolescents is conducting studies across Western, Central, Eastern, and Southern Africa to address existing knowledge gaps. This consortium is expected to generate key evidence to inform clinical practice and public health policy-making for COVID-19 while concurrently addressing other major diseases affecting children in African countries.

The continuous increase in anthropogenic activities resulting in an increase in boron concentration in the environment is becoming a serious threat to public health and the ecosystem. In this regard, a hybrid polyvinylidene fluoride (PVDF)-polyvinyl pyrrolidone (PVP) hollow fiber was synthesized with hydrophilic nano-titanium oxide (TiO2) at varied loadings of 0, 0.5, 1.0, 1.5, and 2.0 wt% using the phase inversion technique. The resultant membranes were characterized in terms of Scanning Electron Microscopy (SEM), Energy Dispersive X-ray Spectroscopy (EDX), contact angle, porosity, and zeta potential. The permeability flux was assessed using both pure water and leachate; also, rejection performance was evaluated based on boron removal from the leachate. The results revealed that the membrane with 1.0 wt% loading had the highest flux alongside an upturn in boron rejection percentage of 223 L/m2·h and 94.39%, respectively. In addition, the lowest contact angle of 50.01° was recorded with 1.0 wt% TiO2 loading, and this implies that it is the most hydrophilic. Throughout the experiment cycles, the fiber with 1.0 wt% TiO2 loading demonstrated a high flux recovery varying between 92.82% and 76.26% after 9 h filtration time. The physicochemical analysis of the permeate revealed that the boron concentration was significantly reduced to 0.43 mg/L, which is far lower than the discharge limit of 1.0 mg/L.
RESUMO
Advancements in availability and specificity of light-emitting diodes (LEDs) have facilitated trait modification of high-value edible herbs and vegetables through the fine manipulation of spectra. Coriander (Coriandrum sativum L.) is a culinary herb, known for its fresh, citrusy aroma, and high economic value. Studies into the impact of light intensity and spectrum on C. sativum physiology, morphology, and aroma are limited. Using a nasal impact frequency panel, a selection of key compounds associated with the characteristic aroma of coriander was identified. Significant differences (P < 0.05) were observed in the concentration of these aromatics between plants grown in a controlled environment chamber under the same photosynthetic photon flux density (PPFD) but custom spectra: red (100%), blue (100%), red + blue (RB, 50% equal contribution), or red + green + blue (RGB, 35.8% red: 26.4% green: 37.8% blue) wavelengths. In general, the concentration of aromatics increased with increasing numbers of wavelengths emitted alongside selective changes, e.g., the greatest increase in coriander-defining E-(2)-decenal occurred under the RGB spectrum. This change in aroma profile was accompanied by significant differences (P < 0.05) in light saturated photosynthetic CO2 assimilation, water-use efficiency (Wi), and morphology. While plants grown under red wavelengths achieved the greatest leaf area, RB spectrum plants were shortest and had the highest leaf:shoot ratio. Therefore, this work evidences a trade-off between sellable commercial morphologies with a weaker, less desirable aroma or a less desirable morphology with more intense coriander-like aromas. When supplemental trichromatic LEDs were used in a commercial glasshouse, the majority of compounds, with the exception of linalool, also increased showing that even as a supplement additional wavelength can modify the aromatic profile increasing its complexity. Lower levels of linalool suggest these plants may be more susceptible to biotic stress such as herbivory. Finally, the concentration of coriander-defining aromatics E-(2)-decenal and E-(2)-hexenal was significantly higher in supermarket pre-packaged coriander leaves implying that concentrations of aromatics increase after excision. In summary, spectra can be used to co-manipulate aroma profile and plant form with increasing spectral complexity leading to greater aromatic complexity and intensity. We suggest that increasing spectral complexity progressively stimulates signaling pathways giving rise to valuable economic traits.
RESUMO
Much of humanity relies on rice (Oryza sativa) as a food source, but cultivation is water intensive and the crop is vulnerable to drought and high temperatures. Under climate change, periods of reduced water availability and high temperature are expected to become more frequent, leading to detrimental effects on rice yields. We engineered the high-yielding rice cultivar 'IR64' to produce fewer stomata by manipulating the level of a developmental signal. We overexpressed the rice epidermal patterning factor OsEPF1, creating plants with substantially reduced stomatal density and correspondingly low stomatal conductance. Low stomatal density rice lines were more able to conserve water, using c. 60% of the normal amount between weeks 4 and 5 post germination. When grown at elevated atmospheric CO2 , rice plants with low stomatal density were able to maintain their stomatal conductance and survive drought and high temperature (40°C) for longer than control plants. Low stomatal density rice gave equivalent or even improved yields, despite a reduced rate of photosynthesis in some conditions. Rice plants with fewer stomata are drought tolerant and more conservative in their water use, and they should perform better in the future when climate change is expected to threaten food security.

The aim of the study was to evaluate daily rhythms of rectal temperature (RT) and body surface temperature (BST) of adult, yearling and foal donkeys during the cold-dry (harmattan) and hot-dry seasons in a tropical savanna. The RTs and BSTs of 30 clinically healthy, free-ranging donkeys were recorded using digital and infrared thermometers. The RT and BST of each donkey and thermal environmental parameters inside the pen were measured bi-hourly from 0600 to 0600 hours (GM + 1) on each experimental day. Application of single cosinor procedure showed that RT and BST exhibited varying levels of daily rhythmicity in both seasons, with values usually higher during the afternoon periods of the photophase of the light/dark cycle. The oscillatory pattern differed with season, with greater amplitude recorded during the cold-dry season than the hot-dry season. The acrophases of the RT and BST were mostly restricted to the photophase of the light/dark cycle in both seasons. The mean values of RT in the adult (37.10 ± 0.10 °C), yearling (37.19 ± 0.10 °C) and foal (37.45 ± 0.11 °C) donkeys were higher (F = 93.41, P < 0.0001) during the hot-dry season than the cold-dry season (35.40 ± 0.05 °C, 35.50 ± 0.04 °C and 36.20 ± 0.05 °C, respectively). In conclusion, seasonal changes influenced significantly the daily rhythmicity of RT and BST in donkeys, kept under natural light/dark cycle.

OBJECTIVES: To determine the prevalence of macrovascular and microvascular complications of type 2 diabetes mellitus and its associated determinants. Methods: We evaluated the online medical records of patients with type 2 diabetes mellitus who have been seen in chronic disease unit between April and June 2014. Seven hundred and forty-eight participants satisfied the inclusion criteria. Of these, 317 (42.4%) were males with a mean age of 57.9 years, 681 (64.3%) were obese with body mass index (BMI) of greater than 30, while only 9.6% had normal BMI (less than 25). Results: Only 158 (21.1%) had controlled hemoglobin A1c level. Retinopathy prevalence was 14.8% and neuropathy was 5.6%. Macrovascular complications accounted for 12.1%. Patients' age and duration of diabetes were main predictors of developing complications. Patients with more than 20 years history of diabetes have 30% more odds of developing the complications when compared to patients with less than 10 years of diabetes history. Each increment in age by one year increases the odds of developing microvascular complications by 4% and macro vascular complication by 2%. Conclusion: Our results showed diabetic patients with poor glycemic control, and longer duration of diabetic history had higher prevalence of both macrovascular and microvascular.

INTRODUCTION: For more than seven decades orthodontist used cephalometric analysis as one of the main diagnostic tools which can be performed manually or by software. The use of computers in treatment planning is expected to avoid errors and make it less time consuming with effective evaluation and high reproducibility. AIM: This study was done to evaluate and compare the accuracy and reliability of cephalometric measurements between computerized method of direct digital radiographs and conventional tracing. MATERIALS AND METHODS: Digital and conventional hand tracing cephalometric analysis of 50 patients were done. Thirty anatomical landmarks were defined on each radiograph by a single investi-gator, 5 skeletal analysis (Steiner, Wits, Tweeds, McNamara, Rakosi Jarabaks) and 28 variables were calculated. RESULTS: The variables showed consistency between the two methods except for 1-NA, Y-axis and interincisal angle measurements which were higher in manual tracing and higher facial axis angle in digital tracing. CONCLUSION: Most of the commonly used measurements were accurate except some measurements between the digital tracing with FACAD® and manual methods. The advantages of digital imaging such as enhancement, transmission, archiving and low radiation dosages makes it to be preferred over conventional method in daily use.
RESUMO
Smoking is accountable for the fatality of a substantial number of persons and increases the likelihood of cancer and cardiovascular diseases. Although data have shown high prevalence rates of cigarette smoking in Saudi Arabia, relatively little is known about the broader scope. The objectives of this study were to investigate socio-demographic factors, patterns of use and cessation behavior associated with smoking in Saudi Arabia (KSA). The study utilized a cross-sectional, multi-step design of sampling. Residents (N=1,497; aged 15 years and older) were recruited from seven administrative areas in Southwest Saudi Arabia. A pretested questionnaire was utilized to obtain data on participant cigarette smoking, including their daily use, age, education, income, marital status and employment status. The current study is the first of its kind to gather data cessation behavior of Saudi subjects. With the exception of 1.5% females, all the respondents were male. The majority of the respondents were married, had a university level of education, were employed, and were younger than 34 years old. The same trends were also observed among smokers' samples. The current prevalence of cigarette smoking was 49.2% and 65.7% of smokers had smoking at less than 18 years of age. The mean daily use amongst smokers was 7.98 cigarettes (SD=4.587). More than 50% of the study sample had tried at least once to quit smoking. However, 42% of the smokers participating had never. On the other hand, about 25% of the respondents were willing to consider quitting smoking in the future. Modeling of cigarette smoking suggested that the most significant independent predictors of smoking behavior were geographic area, gender, marital status, education, job and age. Considerable variation in smoking prevalence was noted related with participant sociodemographics. Findings recommend the necessity for control and intervention programs in Saudi community.

The aim of this study was to investigate the ameliorative effect of Lawsonia inermis Linn used traditionally against trypanosomosis. Twenty-five adult Wistar rats of both sex were individually infected intraperitoneally (IP) with 106Trypanosoma congolense per ml of blood. Following establishment of infection, the rats were randomly divided into five groups of 5 rats each. Rats in groups I, II, and III were treated with 125, 250 and 500 mg/kg of the extract, respectively, while rats in groups IV and V were treated with 3.5 mg/kg and 2 ml/kg of diminazene aceturate (DM) once and physiological buffered saline, respectively. All treatments except DM were given orally for 7 days IP. The antitrypanosomal effect of the plant was assessed by observing the level of parasitaemia daily, packed cell volume (PCV) weekly, erythrocyte osmotic fragility (EOF) and malondialdehyde (MDA) concentration on day 21. Phytochemical screening of the extract revealed the presence of alkaloids, carbohydrates, triterpenes, steroids, cardiac glycosides, saponins, tannins and flavonoids. The extract significantly (P < 0.05) reduced levels of parasitaemia at 250 mg/kg. PCV was higher (P > 0.05) in extract treated groups but significantly higher (P < 0.05) in group II at week 2 when compared to group V. Rats in group II had significantly lower values of EOF and MDA when compared with groups IV and V. Thus, the leaf of L. inermis has in addition to an antitrypanosomal effect against T. congolense in rats, an attenuating effect on the trypanosomosis pathology probably mediated via protection of the erythrocyte membrane against trypanosome-induced oxidative damage to the erythrocytes.
RESUMO
The aim of the study was to investigate the effects of age and season on haematological parameters of donkeys at rest during the rainy and cold-dry seasons. Thirty healthy donkeys divided into three groups based on their age served as the subjects. During each season, blood sample was collected from each donkey thrice, 2 weeks apart, for haematological analysis, and the dry-bulb temperature (DBT), relative humidity (RH) and temperature-humidity index (THI) were obtained thrice each day during the experimental period using standard procedures. During the rainy season, the mean DBT (33.05 ± 0.49 °C), RH (73.63 ± 1.09 %) and THI (84.39 ± 0.71) were higher (P < 0.0001) than the corresponding values of 24.00 ± 0.44 °C, 36.80 ± 0.92 % and 64.80 ± 0.62, during the cold-dry season. Packed cell volume (PCV), erythrocyte count [red blood cell (RBC)], haemoglobin concentration (Hb), mean corpuscular volume (MCV), mean corpuscular haemoglobin (MCH), platelet count (PLT), leucocyte count [white blood cell (WBC)], lymphocyte count (LYM) and neutrophil/lymphocyte ratio (N/L) were higher (P < 0.05) in adults than foals during the rainy season. The MCV, MCH, WBC, NEU, LYM and PLT of adult and yearling donkeys were higher (P < 0.05) during the rainy than the cold-dry season. The PCV, RBC, Hb, MCV, MCH, and NEU of foals were higher in the rainy than the cold-dry season. The N/L of adult and foal donkeys were higher (P < 0.05) in the rainy than in the cold-dry season. In conclusion, PCV, RBC, Hb and LYM were considerably higher in foals than yearlings or adults during the rainy season, while erythrocytic indices and platelet counts were higher in adults or yearlings than in foals in both seasons. Erythrocytic indices, PLT and N/L were higher in the rainy than the cold-dry season in adults, yearlings and foals.

The present experiment was performed with the aim of investigating the effect of season on behavioral activities of donkeys during the rainy and harmattan seasons in the Northern Guinea zone of Nigeria. Sixteen apparently healthy donkeys were used as subjects and divided into four groups based on age. During each season, behavioral activities of each donkey were evaluated for three weeks using the focal animal sampling technique. The dry-bulb temperature (DBT), relative humidity (RH), and temperature-humidity index (THI) were obtained three times each day during the experimental period using standard procedures. In the rainy season, the mean DBT (31.65 ± 0.49°C), RH (73.63 ± 1.09%), and THI (84.39 ± 0.71) were significantly (P<0.0001) higher than the corresponding values of 24.00 ± 0.44°C, 36.80 ± 0.92%, and 64.80 ± 0.62 in the harmattan season. During the rainy season, the donkeys spent 60.00 ± 0.77%, 25.40 ± 0.69%, and 2.94 ± 0.21% on grazing, resting, and grooming, respectively. During the harmattan season, the donkeys spent the most time on grazing (76.76 ± 0.43%), less time on resting (11.97 ± 0.38%), and the least time on grooming (0.89 ± 0.05%). In conclusion, season and seasonal variations affect the daytime behavioral activities of donkeys in the zone, and this should be considered in husbandry practices for donkeys.
