Oriental theileriosis caused by Theileria orientalis is a growing health concern of lactating cows in its endemic areas. Rapid and sensitive diagnostic tests are demand areas for appropriate and effective prophylactic and therapeutic measures. Quantitative polymerase chain reaction (qPCR) is the answer for both detection and quantification of parasites. Present study deals with qPCR for detection of parasitemia level of T. orientalis in apparently healthy and clinically affected cows. Major piroplasm surface protein (MPSP) gene present in T. orientalis was cloned in pUC57 vector and transformed into E. coli Top 10 cells. Single and mixed infections of hemoprotozoa other than T. orientalis, causing anemia were differentiated through blood smear examination and PCR tests. T. orientalis was detected in 108 (63.15%) ill and 48 (26.66%) healthy cows. Piroplasms detected per 1000 red blood cells (RBCs) was 0-1 in the healthy group as compared to 3-22 in those showing clinical signs. Parasitemia in ill cows ranged between 6.9 × 102 and 4.5 × 103 parasites / µl of blood which was significantly higher (p<0.05) than healthy group (2.6 × 102 - 5.7 × 102 parasites / µl of blood). Phylogenetic study of the isolates showed similarity with Buffeli type that unfolded its pathogenic form in apparently healthy and ill cows.

The present experiment was carried out to assess the comparative efficacy of ivermectin and fenbendazole individually for anthelmintic therapy for the hookworm infested dogs. Dogs presented to the Department of Veterinary Clinical Medicine or Veterinary Clinical Complex, Bhubaneswar were randomly screened for Ancylostoma caninum infection and the positive dogs were selected for the therapeutic trial Faecal samples were collected randomly from presented dogs immediately after defaecation or from the rectum directly using a faecal scoop. The collected sample was examined by floatation technique to detect the positive cases of Ancylostoma caninum infection. The dogs with normal clinical parameters and no eggs or ova in the faeces were included in in group 1 (n = 12). Dogs with faecal sample positive for Ancylostoma caninum ova were recruited for the comparative study (n = 24) which were grouped into two groups consisting of 12 dogs in each (group 2 and 3). Group 2 dogs (n = 12) were treated with ivermectin at 200 µg/kg body weight once orally repeated after 15 days with proper supportive therapy each time. Group 3 (n = 12) were treated with fenbendazole at 50 mg/kg body weight once orally repeated after 15 days with proper supportive therapy each time. Haematological examinations and serum biochemical tests were carried out in all groups each time on day 0, 15 and 30 of the experiment. The therapeutic efficacy of both the drugs was calculated on the basis of number of animals found free of Ancylostoma infection as determined by reduction in EPG count of the faeces of the group following the treatment. The reduction in eggs per gram (EPG) count on day 15 and day 30 was more significant in group 2 than group 3. The mean EPG count reduced significantly to 24.17 ± 11.44 on day 15 from day 0 level of 1650.00 ± 247.25 in fenbendazole-treated dogs. On day 30, the mean value further reduced to become nil.The 15th day after treatment, mean (± SE) value of protein, albumin and globulin was changed to 5.63 ± 0.12, 2.64 ± 0.12 and 2.99 ± 0.15 g/dl, respectively. The 30th day after treatment, the values were 6.23 ± 0.14, 3.20 ± 0.18 and 3.03 ± 0.21 g/dl, respectively. The total protein and albumin values were significantly changed from day 0 level in group 2 and 3 by 15th day and 30th day, respectively, at 1% level of significance. Following treatment with ivermectin, the 15th day haematological values increased significantly at 1% level (P < 0.01) of significance. There was significant increase in the values at 1% level on the 30th day compared to day 0 and the mean values were non-significantly comparable to the healthy control group except PCV and TEC.

Hepatospecific delivery by ligand based receptor targeting is an established strategy to augment therapy associated with liver diseases and disorders. Previously, we have investigated the effect of ligand headgroup on cellular uptake mediated by the asialoglycoprotein receptor by in silico and in vitro approach. In this paper, we report the design of agarose based liposomes for delivery to liver cancer cells and provide a proof of concept of the targeting efficiency against galactose liposomes using an in vivo approach. Sorafenib Tosylate loaded targeting liposomes were developed and optimized using factorial design. Comparative evaluation including cell cytotoxicity, pharmacokinetics and biodistribution and hepatospecific uptake was performed for both the liposomal systems. The formulations possessed a particle size of 150 - 180 nm and a zeta potential of 30 - 60 mV depending on the amount of ligand and drug loading, with more than 90% entrapment efficiency. A two-fold increase in cytotoxicity was observed with agarose-based liposomes as compared to galactose based liposomes. In vivo PK evaluation indicated a reduction in half life of drug when loaded in agarose ligand loaded system, probably due to greater uptake in the liver as evidenced in biodistribution study. Intrahepatic disposition revealed a higher PC/NPC uptake ratio with the targeted systems as compared to conventional liposomes, although the agarose-based system resulted in highest uptake ratio. A biocompatible platform for specific delivery of drugs to hepatocytes was established validating a rational approach to design liver targeting systems.

Mahanadi is one of the major rivers of peninsular India. Like other Indian rivers, it is contaminated with sewages, industrial discharges, and agricultural runoff. Thus, necessity was felt to monitor its pollution status. Present work was part of that program and aimed to assess the sediment contamination due to the trace metals Cd, Cr, Cu, Mn, Pb, and Zn during 2012-2015. Sediment pollution status and ecological risks were evaluated calculating contamination factor (CF), geo-accumulation (Igeo), pollution load index (PLI), potential ecological risk (EiR), etc. The recorded metal concentrations were Cd BDL of flame mode of AAS; Cr BDL - 73.9; Cu BDL - 44.4; Mn 37.2 - 1887.0; Pb BDL - 29.5; and Zn BDL - 92.5 mg kg-1. As per US EPA guidelines, Cr concentrations at many locations were in the moderately polluted range. Igeo, CF, mCd, PLI, and EiR indicated low pollution levels and low ecological risks due to the trace metals assessed. The sediment quality guidelines (SQGs) indicated that Cr and Cu concentrations exceeded (16% sample) the threshold effect concentrations and may occasionally exhibit adverse biological effects. The association of sediment organic matter, conductivity and content of Cu, and their grouping in component 1 of PCA revealed that the anthropogenic input was dominant and so also the component 2 where Cr exhibited moderately good correlation with organic matter. Cluster analysis of the sampling sites based on pollution status yielded 3 groups: relatively uncontaminated (S3, S4), low to moderately contaminated (S2), and moderately contaminated (S1, S5, S6) stretches.

RESUMO

The postoperative period after craniopharyngioma surgery has a high likelihood of complications and its management can be challenging. We present the case of a 10-year-old boy who was operated for craniopharyngioma. In the postoperative period he developed lacunar infarct, endocrine disturbances, delayed vasospasm leading to cerebral infarction and status epilepticus. The likely cause of status epilepticus was likely delayed cerebral infarction compounded with dyselectrolytemia. We discuss the possible etiology of delayed cerebral infarction.

Coronary computed tomography angiography (CCTA) offers high-resolution anatomic characterization of the coronary vasculature but may be suboptimal for lesions dependent on real-time visualization of flow including chronic total occlusion (CTO). In CTOs, heavy calcification and distal vessel opacification from collateralization may confound luminal assessment. Several studies have examined the role of CCTA in characterizing known CTOs to guide percutaneous coronary intervention (PCI). However, the efficacy of CCTA in the de novo diagnosis of CTOs prior to coronary angiography (CAG) has not been demonstrated. A total of 233 consecutive patients who presented for CAG within a 3-month period of having CCTA were retrospectively reviewed. Those patients with prior diagnosis of CTO or prior bypass of the occluded vessels were excluded. Sensitivity and specificity analysis of CCTA in identifying CTOs using CAG as the gold standard was performed. The prevalence of CTO was 21.11% in the population that met criteria for analysis ( n = 199). The sensitivity of CCTA in predicting CTO was 57.1%, while the specificity was 96.8%. The positive predictive value and negative predictive value of CCTA in detection of CTO were 82.8 and 89.4%, respectively. Our study shows that CCTA has excellent specificity but poor sensitivity in the detection of CTO thus limiting its clinical use in de novo diagnosis. Further studies to determine the effect of de novo CTO diagnosis on clinically important procedural factors, such as radiation exposure, contrast use, and need for repeat procedures, are warranted and may implicate a role for CCTA in this setting.

BACKGROUND: Theileria annulata is a tick-borne apicomplexan parasite that affects bovine and causes severe economic losses. Aims: Our study aimed to determine the molecular prevalence of T. annulata infection in asymptomatic carrier cattle in Odisha, India, to study the association of potential risk factors with theileriosis, and to investigate the effect of the parasite infection on hematological parameters in naturally affected animals. METHODS: A total of 226 cattle blood samples were collected from seven districts of Odisha, India. Molecular diagnoses of tropical theileriosis by polymerase chain reaction (PCR), cloning, sequencing, and phylogenetic analysis of isolated parasites were performed. Potential risk factors were investigated by univariable and multivariable logistic regression statistical analysis. Hematological parameters were compared between positive and negative animals. RESULTS: All animals included in our study were clinically normal, however, 54.86% (124/226) of examined animals were positive by PCR for T. annulata. The multivariable logistic regression showed that contact with other cattle from different herds during grazing (P<0.0001; OR: 12.75; 95% CI: 5.21-31.21), previous history of clinical signs (P=0.002; OR: 3.31; 95% CI: 1.53-6.31), and frequency of a ectoparasiticides application pre year (P<0.0001; OR: 9.22; 95% CI: 3.03-28.09) were the potential risk factors for the occurrence of tropical theileriosis. Nucleotide sequence identity data demonstrated that T. annulata strain (MN818858) Odisha shared homology of 99.6%, 99.49%, and 99.36% with Uttar Pradesh, India (MF346035), Bahrain (AF214797), and Hyderabad, India (MK034702), respectively. CONCLUSION: This is the first study to gain insight into the molecular epidemiology, risk factors, phylogeny, and hematological analysis of asymptomatic T. annulata infected cattle from India.

The incidence of symptomatic radiation induced lung pneumonitis (RILP), a major dose limiting side effect of thoracic radiotherapy, is in the range of 15-40%. Therapeutic options for the prevention and treatment of RILP are limited. Hence there is a need for developing novel radioprotectors to prevent RILP which can be patient compliant. This study sought to evaluate the efficacy of oral 3,3'-diselenodipropionic acid (DSePA), a novel selenocystine derivative to prevent RILP. C3H/HeJ (pneumonitis responding) mice received a single dose of 18 Gy, whole thorax irradiation and a subset were treated with DSePA orally (2.5 mg/kg), three times per week beginning 2 h post irradiation and continued till 6 months. DSePA delayed onset of grade ≥ 2 RILP by 45 days compared to radiation control (~105 versus ~60 days). It also reversed the severity of pneumonitis in 3/10 radiation treated mice leading to significant improvement in asymptomatic survival compared to radiation control (~180 versus ~102 days). DSePA significantly (p < 0.05) reduced the radiation-mediated infiltration of polymorphonuclear neutrophils (PMN) and elevation in levels of cytokines such as IL1-ß, ICAM-1, E-selectin, IL-17 and TGF-ß in the bronchoalveolar lavage fluid. Moreover DSePA lowered PMN-induced oxidants, maintained glutathione peroxidase activity and suppressed NF-kB/IL-17/G-CSF/neutrophil axis in the lung of irradiated mice. Additionally, this compound did not protect A549 (lung cancer) derived xenograft tumor from radiation exposure in SCID mice. DSePA offers protection to normal lung against RILP without affecting radiation sensitivity of tumors. It has the potential to be developed as an oral agent for preventing RILP.

Peste des petits ruminants (PPR) is an economically important livestock disease which affects a vast section of the small ruminant population in India. However, data on the incidence of PPR are limited and scant literature is available on the economic losses caused by the disease. In the present study, a structured sampling design was adopted, which covered the major agro-climatic regions of the country, to ascertain the morbidity and mortality rates of PPR. Available estimates of the economic losses in India due to various livestock diseases are based on single values of various epidemiological and economic parameters. Stochastic modelling was used to estimate the economic impact of PPR. Overall annual morbidity and mortality rates of PPR for small ruminants in India have been estimated from the sample as being 8%and 3.45%, respectively. The authors have analysed variations in these rates across species, age group, sex, season and region. The expected annual economic loss due to PPR in India ranges from as little as US $2 million to $18 million and may go up to US $1.5 billion; the most likely range of expected economic losses is between US $653 million and $669 million. This study thus reveals significant losses due to the incidence of PPR in small ruminants in India.


La peste des petits ruminants (PPR) est une maladie du bétail à fort impact économique. En Inde, une grande partie de la population des petits ruminants est affectée. Cependant, les données disponibles sur l'incidence de la PPR sont rares et très peu d'articles ont été consacrés aux pertes économiques causées par la maladie. Les auteurs présentent une étude basée sur un échantillonnage structuré couvrant les principales régions agro-climatiques du pays, visant à déterminer avec certitude les taux de morbidité et de mortalité de la PPR. Les estimations disponibles des pertes économiques induites par diverses maladies des animaux d'élevage sont basées sur des valeurs uniques correspondant à divers paramètres épidémiologiques et économiques. Les auteurs ont évalué l'impact économique de la PPR en utilisant un modèle stochastique. En se basant sur l'échantillon, les taux annuels de morbidité et de mortalité de la PPR chez les petits ruminants en Inde ont été respectivement estimés à 8 % et à 3,45 %. Les auteurs ont également analysé les variations de ces taux en fonction de l'espèce, du groupe d'âge, du sexe, de la saison et de la région. Les pertes annuelles attendues imputables à la PPR en Inde fluctuent d'un minimum de 2 millions de dollars US (USD) à 18 millions d'USD, mais elles peuvent atteindre 1,5 milliard d'USD ; la fourchette la plus probable des pertes économiques attendues se situe entre 653 millions et 669 millions d'USD. La peste de pequeños rumiantes (PPR) es una enfermedad del ganado que reviste importancia económica y afecta a un vasto segmento de la población de pequeños rumiantes de la India. Sin embargo, existen pocos datos sobre su incidencia y muy escasas referencias bibliográficas sobre las pérdidas económicas que ocasiona. Los autores describen un estudio encaminado a determinar las tasas de morbilidad y mortalidad por PPR a partir de un muestreo estructurado que abarcaba las principales regiones agroclimáticas del país. Las estimaciones existentes de las pérdidas económicas causadas en la India por diversas enfermedades del ganado están basadas en valores únicos de diversos parámetros epidemiológicos y económicos. Para estimar el impacto económico de la PPR se utilizó una modelización estocástica. A partir de la muestra se calculó que, en los pequeños rumiantes del país, los índices anuales de morbilidad y mortalidad totales por PPR se cifran en un 8% y un 3,45%, respectivamente. Los autores analizaron después las variaciones que exhiben esos índices por especie, grupo de edad, sexo, estación y región. La cuantía prevista de las pérdidas económicas anuales causadas por la PPR en el país oscila: de apenas 2 a 18 millones de dólares estadounidenses puede llegar hasta los 1 500 millones. El intervalo más probable de pérdidas económicas se sitúa entre 653 y 669 millones. El estudio demuestra pues que la incidencia de la PPR entre los pequeños rumiantes de la India provoca pérdidas de importante magnitud.

Rock crushing is a common process in mining and related operations. Although a number of particle crushing tests have been proposed in the literature, most of them are concerned with single-particle crushing, i.e., a single rock sample is crushed in each test. Considering the realistic scenario in crushers where many fragments are involved, a laboratory crushing apparatus is developed in this study. This device consists of a Hopkinson pressure bar system and a piston-holder system. The Hopkinson pressure bar system is used to apply calibrated dynamic loads to the piston-holder system, and the piston-holder system is used to hold rock samples and to recover fragments for subsequent particle size analysis. The rock samples are subjected to three to seven impacts under three impact velocities (2.2, 3.8, and 5.0 m/s), with the feed size of the rock particle samples limited between 9.5 and 12.7 mm. Several key parameters are determined from this test, including particle size distribution parameters, impact velocity, loading pressure, and total work. The results show that the total work correlates well with resulting fragmentation size distribution, and the apparatus provides a useful tool for studying the mechanism of crushing, which further provides guidelines for the design of commercial crushers.

Background: Lung cancer is the leading cause of cancer-related deaths across the world. In this study, we present therapeutically relevant genetic alterations in lung adenocarcinoma of Indian origin. Materials and methods: Forty-five primary lung adenocarcinoma tumors were sequenced for 676 amplicons using RainDance cancer panel at an average coverage of 1500 × (reads per million mapped reads). To validate the findings, 49 mutations across 23 genes were genotyped in an additional set of 363 primary lung adenocarcinoma tumors using mass spectrometry. NIH/3T3 cells over expressing mutant and wild-type FGFR3 constructs were characterized for anchorage independent growth, constitutive activation, tumor formation and sensitivity to FGFR inhibitors using in vitro and xenograft mouse models. Results: We present the first spectrum of actionable alterations in lung adenocarcinoma tumors of Indian origin, and shows that mutations of FGFR3 are present in 20 of 363 (5.5%) patients. These FGFR3 mutations are constitutively active and oncogenic when ectopically expressed in NIH/3T3 cells and using a xenograft model in NOD/SCID mice. Inhibition of FGFR3 kinase activity inhibits transformation of NIH/3T3 overexpressing FGFR3 constructs and growth of tumors driven by FGFR3 in the xenograft models. The reduction in tumor size in the mouse is paralleled by a reduction in the amounts of phospho-ERK, validating the in vitro findings. Interestingly, the FGFR3 mutations are significantly higher in a proportion of younger patients and show a trend toward better overall survival, compared with patients lacking actionable alterations or those harboring KRAS mutations. Conclusion: We present the first actionable mutation spectrum in Indian lung cancer genome. These findings implicate FGFR3 as a novel therapeutic in lung adenocarcinoma.

Thin carbon-backed isotopically enriched 208Pb targets were required for our experiment aimed to study the reaction dynamics for 48Ti + 208Pb system, populating the near super-heavy nucleus 256Rf, through mass-energy correlation of the fission fragments. Purity and thickness of the targets are of utmost importance in such studies as these factors have strong influence on the measurement accuracy of mass and energy distribution of fission fragments. 208Pb targets with thickness ranging from 60 µg/cm2 to 250 µg/cm2 have been fabricated in high vacuum environment using physical vapor deposition method. Important points in the method are as follows: •208Pb was deposited using resistive heating method, whereas carbon (backing foil) deposition was performed by using the electron beam bombardment technique.•Different characterization techniques such as Particle Induced X-ray Emission (PIXE), Energy Dispersive X-Ray Fluorescence (EDXRF) and Rutherford Backscattering Spectrometry (RBS) were used to assert the purity and thickness of the targets.•These targets have successfully been used to accomplish our experimental objectives.

Sarcoptes scabies var. cuniculi and Psoroptes cuniculi are most common mites prevailed in rabbits, but Notoedres cati, is the rarest mite ever been reported in rabbit. Two New Zealand white rabbits were presented with clinical signs of pruritus, alopecia, scab and crust formation and lichenification on the upper lip, ear pinnae, eyelids, lower jaw and limbs. Deep skin scraping was taken separately from 4 to 5 different skin lesions from each rabbit, revealed mixed infestations of N. cati, S. cuniculi and P. cuniculi. Subcutaneous injection of ivermectin at weekly intervals for four weeks resulted in remission of clinical signs and improvement of health condition in rabbits. This is the first report of N. cati infestation of rabbit in Odisha.

RESUMO

Perioperative management of deaf and dumb patients can be a challenging task. For smooth postoperative recovery, proper care should begin in the preoperative period. Understanding the patients' needs and training him to follow the instructions requires to involve a communication specialist. Judicious use of sedatives and analgesics is essential to keep the patient pain-free and comfortable. Postoperatively, the patient should be kept awake, enough to understand the internal need of the body and to make a meaningful response to external stimuli. Adequate preoperative planning and coordinated team efforts with involvement of specialists can help in delivering better postoperative care.

Ivermectin is one of the most effective and widely used antiparasitic agents ever discovered because of its broad spectrum activity against nematodes and arthropods. Therefore, the study was conducted to determine the comparative anthelmintic efficacy of ivermectin by giving through oral and subcutaneous routes in clinically infected dogs and also to check the prevalence of gastrointestinal nematode in dogs in Bhubaneswar, Odisha. Total 154 dogs with clinical signs of gastroenteritis were examined for the presence of nematodal ova in their faecal sample and 64 were found positive (41.56 %). The screened dogs were divided in to two groups (n = 10 each), first group of animals were treated with ivermectin @ 0.5 mg/kg body weight by subcutaneous route and the second group with same dose of ivermectin through oral route. Efficacy of ivermectin was evaluated based on egg per gram (EPG), haematological and biochemical parameters. It is concluded that, Prevalence of gastrointestinal nematodes are high in dogs in the climatic condition of Bhubaneswar, Odisha. Ivermectin was found effective when administered through either of the routes. Haemato-biochemical parameters were restored after administration of ivermectin and also significant reduction in EPG was found in dogs treated with ivermectin by both the routes, but no significant difference was observed in EPG between oral and subcutaneous treated dogs in various intervals of study.

Structure, vibrational and nuclear magnetic resonance spectra, and antioxidant action of ascorbic acid towards hydroxyl radicals have been studied computationally and in vitro by ultraviolet-visible, nuclear magnetic resonance and vibrational spectroscopic techniques. Time dependant density functional theory calculations have been employed to specify various electronic transitions in ultraviolet-visible spectra. Observed chemical shifts and vibrational bands in nuclear magnetic resonance and vibrational spectra, respectively have been assigned with the help of calculations. Changes in the structure of ascorbic acid in aqueous phase have been examined computationally and experimentally by recording Raman spectra in aqueous medium. Theoretical calculations of the interaction between ascorbic acid molecule and hydroxyl radical predicted the formation of dehydroascorbic acid as first product, which has been confirmed by comparing its simulated spectra with the corresponding spectra of ascorbic acid in presence of hydrogen peroxide.

Nicorandil is a commonly used antianginal agent, which has both nitrate-like and ATP-sensitive potassium (K ATP ) channel activator properties. Activation of potassium channels by nicorandil causes expulsion of potassium ions into the extracellular space leading to membrane hyperpolarization, closure of voltage-gated calcium channels and finally vasodilatation. However, on the other hand, being an activator of K ATP channel, it can expel K + ions out of the cells and can cause hyperkalemia. Here, we report a case of nicorandil induced hyperkalemia unresponsive to medical treatment in a patient with diabetic nephropathy.

AIM: Studies on the prevalence of gastrointestinal helminths infection in Banaraja fowls of three blocks (Chandua, Shamakhunta and Bangriposi) of Mayurbhanj district in Odisha with respect to semi-intensive system of rearing. MATERIALS AND METHODS: A total of 160 Banaraja birds (30 males and 130 females) belonging to two age groups (below 1 month age and above 1 month) were examined for the presence of different species of gastrointestinal helminth infection over a period of 1-year. The method of investigation included collection of fecal sample and gastrointestinal tracts, examination of fecal sample of birds, collection of parasites from different part of gastrointestinal tract, counting of parasites, and examination of the collected parasites by standard parasitological techniques followed by morphological identification as far as possible up to the species level. RESULTS: Overall, 58.75% birds were found infected with various gastrointestinal helminths. Total five species of parasites were detected that included Ascaridia galli (25.63%), Heterakis gallinarum (33.75%), Raillietina tetragona (46.25%), Raillietina echinobothrida (11.87%), and Echinostoma revolutum (1.87%). Both single (19.15%) as well as mixed (80.85%) infection were observed. Highest incidence of infection was observed during rainy season (68.88%) followed by winter (66.66%) and least in summer season (41.81%). Sex-wise incidence revealed slightly higher occurrence among females (59.23%) than males (56.67%). Age-wise prevalence revealed that chicks were more susceptible (77.77%) than adults (51.30%) to gastrointestinal helminths infection. CONCLUSIONS: Present study revealed that mixed infection with gastrointestinal helminths of different species was more common than infection with single species and season-wise prevalence was higher in rainy season followed by winter and summer. Chicks were found to be more prone to this parasitic infection and a slight higher prevalence among female birds was observed.

RESUMO

Immune thrombocytopenic purpura (ITP) patients are at high-risk for bleeding complications during and after cardiac surgeries involving cardiopulmonary bypass. We report a patient with ITP with severe coronary artery disease and mitral valve regurgitation who underwent uncomplicated coronary artery bypass grafting and mitral valve replacement. Three weeks later, the patient was readmitted in a very low general condition with signs of pericardial tamponade. We describe our experience of managing the case.
