RESUMO

The soil bacterium Pseudomonas putida KT2440 has been shown to produce selenium nanoparticles aerobically from selenite; however, the molecular actors involved in this process are unknown. Here, through a combination of genetic and analytical techniques, we report the first insights into selenite metabolism in this bacterium. Our results suggest that the reduction of selenite occurs through an interconnected metabolic network involving central metabolic reactions, sulphur metabolism, and the response to oxidative stress. Genes such as sucA, D2HGDH and PP_3148 revealed that the 2-ketoglutarate and glutamate metabolism is important to convert selenite into selenium. On the other hand, mutations affecting the activity of the sulphite reductase decreased the bacteria's ability to transform selenite. Other genes related to sulphur metabolism (ssuEF, sfnCE, sqrR, sqr and pdo2) and stress response (gqr, lsfA, ahpCF and sadI) were also identified as involved in selenite transformation. Interestingly, suppression of genes sqrR, sqr and pdo2 resulted in the production of selenium nanoparticles at a higher rate than the wild-type strain, which is of biotechnological interest. The data provided in this study brings us closer to understanding the metabolism of selenium in bacteria and offers new targets for the development of biotechnological tools for the production of selenium nanoparticles.

RESUMO

BACKGROUND: Transcranial direct current stimulation (tDCS) seems to be a potential tool to optimize the long-term effects of multimodal physical training (MPT) on fitness components in post-stroke patients. OBJECTIVE: We investigated the effects of cortical tDCS combined with MPT on motor function reflected by strength, motor performance, and cardiorespiratory capacity in chronic stroke patients. METHODS: This double-blind randomized controlled trial included 18 volunteers (55 ± 10 y, 72 ± 13 kg), who underwent MPT preceded by either sham stimulation (SHAM) or 2 mA bi-hemispheric tDCS. MPT consisted of 24 sessions of 60-70 min performed 2 d/wk within 12-16 weeks, with individualized intensity. Outcomes were Fugl-Meyer scores for lower limbs (FM-LL), and total (FM-Total); speed in the 10-m walk test (10MWT); oxygen uptake and work output at maximal effort (VO2max and Wmax), and gas exchange threshold (VO2-GET and W-GET); peak torque of isokinetic knee extension (PT-EXT) and flexion (PT-FLEX) of paretic and non-paretic limbs; bilateral strength deficit during knee extension (DS-EXT) and flexion (DS-FLEX). RESULTS: Pre- vs. post-intervention improvements were detected in tDCS vs. SHAM (p < 0.05) for FM-total (29.6% vs. 15.9%; effect size [ES] = 0.78), FM-LL (35.9% vs. 9.0%; ES = 1.23), 10MWT (10.6% vs. 3.8%; ES = 0.67), Wmax (75.0% vs. 4.3%; ES = 1.68), W-GET (91.6% vs. 12.4%; ES = 1.62), PT-EXT (25.6% vs. -6.5%; ES = 1.94) and PT-FLEX (26.3% vs. 9.8%; ES = 0.65) of the paretic limb, and DS-EXT (-13.7% vs. 2.5; ES = 1.43). CONCLUSION: Bi-hemispheric cortical tDCS optimized the effects of MPT performed with moderate volume and intensity upon muscle strength, motor function, and cardiorespiratory performance in stroke hemiparetic survivors. RESUMO

Tellurium oxyanions are chemical species of great toxicity and their presence in the environment has increased because of mining industries and photovoltaic and electronic waste. Recovery strategies for this metalloid that are based on micro-organisms are of interest, but further studies of the transport systems and enzymes responsible for implementing tellurium transformations are required because many mechanisms remain unknown. Here, we investigated the involvement in tellurite uptake of the putative phosphate transporter PitB (PP1373) in soil bacterium Pseudomonas putida KT2440. For this purpose, through a method based on the CRISPR/Cas9 system, we generated a strain deficient in the pitB gene and characterized its phenotype on exposing it to varied concentrations of tellurite. Growth curves and transmission electronic microscopy experiments for the wild-type and ΔpitB strains showed that both were able to internalize tellurite into the cytoplasm and reduce the oxyanion to black nano-sized and rod-shaped tellurium particles, although the ΔpitB strain showed an increased resistance to the tellurite toxic effects. At a concentration of 100 µM tellurite, where the biomass formation of the wild-type strain decreased by half, we observed a greater ability of ΔpitB to reduce this oxyanion with respect to the wild-type strain (~38 vs ~16 %), which is related to the greater biomass production of ΔpitB and not to a greater consumption of tellurite per cell. The phenotype of the mutant was restored on over-expressing pitB in trans. In summary, our results indicate that PitB is one of several transporters responsible for tellurite uptake in P. putida KT2440.

RESUMO

Abstract The aim of the present study was to investigate the level of physical activity and associated factors of the academic community of a Brazilian federal higher education institution. Overall, 483 students, 49 teachers and 153 administrative technicians of both genders participated in this stdy. Sociodemographic questionnaire, BMI evaluation and classification of the level of physical activity (IPAQ) were used. Statistical analysis was performed by the Pearson Chi-square test to observe possible associations between independent variables and LPA and Poisson regression analysis with 95% confidence intervals through the SPSS software version 22.0. Among the 61.8% of individuals classified as physically active, 48.90% belonged to the health area and 52.40% had Graduation / Specialization level. Of these, 45.7% showed normal BMI and 62.2% of them did not practice exercises before being admitted to the extension project. Teachers and administrative technicians were, respectively, 3.62 (95% CI = 1.11-11.82) and 2.15 (95% CI = 1.03-4.66) times more likely of having sedentary behavior when compared to the group of students. LPA was higher in undergraduate students, while teachers and technicians were more likely of being sedentary. The majority of participants had normal BMI and the largest portion of physically active individuals was composed of students of the health area. Thus, it is necessary to encourage interventions for the practice of physical activity within the academic community, as well as the implementation of internal policies to mobilize individuals to improve the worrying situation of sedentary behavior.

Bernardes, WL, Montenegro, RA, Monteiro, WD, de Almeida Freire, R, Massaferri, R, and Farinatti, P. Optimizing a treadmill ramp protocol to evaluate aerobic capacity of hemiparetic poststroke patients. J Strength Cond Res 32(3): 876-884, 2018-A correct assessment of cardiopulmonary capacity is important for aerobic training within motor rehabilitation of poststroke hemiparetic patients (PSHPs). However, specific cardiopulmonary exercise testing (CPET) for these patients are scarce. We proposed adaptations in a protocol originally developed for PSHPs by Ovando et al. (CPET1). We hypothesized that our adapted protocol (CPET2) would improve the original test, by preventing early fatigue and increasing patients' peak performance. Eleven PSHPs (52 ± 14 years, 10 men) performed both protocols. CPET2 integrated changes in final speed (100-120% vs. 140% maximal speed in 10-m walking test), treadmill inclination (final inclination of 5 vs. 10%), and estimated test duration (10 vs. 8 minutes) to smooth the rate of workload increment of CPET1. Peak oxygen uptake (V[Combining Dot Above]O2peak) (20.3 ± 6.1 vs. 18.6 ± 5.0 ml·kg·min; p = 0.04), V[Combining Dot Above]O2 at gas exchange transition (V[Combining Dot Above]O2-GET) (11.5 ± 2.9 vs. 9.8 ± 2.0 ml·kg·min; p = 0.04), and time to exhaustion (10 ± 3 vs. 6 ± 2 minutes; p < 0.001) were higher in CPET2 than in CPET1. Slopes and intercepts of regressions describing relationships between V[Combining Dot Above]O2 vs. workload, heart rate vs. workload, and V[Combining Dot Above]O2 vs. heart rate were similar between CPETs. However, standard errors of estimates obtained for regressions between heart rate vs. workload (3.0 ± 1.3 vs. 3.8 ± 1.0 b·min; p = 0.004) and V[Combining Dot Above]O2 vs. heart rate (6.0 ± 2.1 vs. 4.8 ± 2.4 ml·kg·min; p = 0.05) were lower in CPET2 than in CPET1. In conclusion, the present adaptations in Ovando's CPET protocol increased exercise tolerance of PSHPs, eliciting higher V[Combining Dot Above]O2peak and V[Combining Dot Above]O2-GET, preventing earlier fatigue, and providing better physiological relationships along submaximal workloads.

RESUMO

Post-stroke patients usually exhibit reduced peak muscular torque (PT) and/or force steadiness during submaximal exercise. Brain stimulation techniques have been proposed to improve neural plasticity and help to restore motor performance in post-stroke patients. The present study compared the effects of bihemispheric motor cortex transcranial direct current stimulation (tDCS) on PT and force steadiness during maximal and submaximal resistance exercise performed by post-stroke patients vs. healthy controls. A double-blind randomized crossover controlled trial (identification number: TCTR20151112001; URL: http://www.clinicaltrials.in.th/) was conducted involving nine healthy and 10 post-stroke hemiparetic individuals who received either tDCS (2 mA) or sham stimulus upon the motor cortex for 20 min. PT and force steadiness (reflected by the coefficient of variation (CV) of muscular torque) were assessed during unilateral knee extension and flexion at maximal and submaximal workloads (1 set of 3 repetitions at 100% PT and 2 sets of 10 repetitions at 50% PT, respectively). No significant change in PT was observed in post-stroke and healthy subjects. Force steadiness during knee extension (~25-35%, P < 0.001) and flexion (~22-33%, P < 0.001) improved after tDCS compared to the sham condition in post-stroke patients, but improved only during knee extension (~13-27%, P < 0.001) in healthy controls. These results suggest that tDCS may improve force steadiness, but not PT in post-stroke hemiparetic patients, which might be relevant in the context of motor rehabilitation programs.

RESUMO

The effects of a recreational soccer program (RSP) upon body composition, heart rate variability (HRV), biochemical markers, cardio-respiratory fitness, and endothelial function in obese adolescents were investigated. A randomised controlled clinical trial was conducted with 30 adolescents aged 12-17 years and body mass index (BMI) >2 standard deviations of WHO reference values, which were assigned to RSP (n = 10, 2 girls) and obese control (n = 10, 4 girls) groups. The 12-week RSP included 60-min sessions performed 3 times/week. BMI, waist circumference, blood pressure, blood glucose, lipid profile, insulin, C-reactive protein, HRV, and maximal oxygen consumption (VO2peak) were evaluated following standardised procedures. Body composition was determined by dual-energy X-ray absorptiometry and endothelial function by venous occlusion plethysmography. After intervention, RSP exhibited significant reductions in BMI (-0.7 ± 0.2 kg · m(-2)), waist circumference (-8.2 ± 1.4 cm), %body fat (-2.2 ± 0.4%), systolic blood pressure (-5.0 ± 2.3 mmHg), total cholesterol (-16.2 ± 5.8 mg · dL(-1)), triglycerides (-20.5 ± 12.9 mg · dL(-1)), C-reactive protein (-0.06 ± 0.01 mg · dL(-1)), insulin resistance (HOMA-IR, -1.4 ± 0.6), and sympathetic activity (LF, -13.9 ± 6.6 un) vs. controls (P < 0.05). Significant increase was observed in parasympathetic activity (HF, 13.9 ± 6.6 un), VO2peak (7.9 ± 2.8 ml · kg(-1) · min(-1)), and high-density lipoprotein cholesterol (11.0 ± 6.3 mg · dL(-1)) (P < 0.05). Vascular conductance (19.5 ± 8.1 ml · min(-1) · 100 ml, P = 0.005) increased and vascular resistance (-5.9 ± 2.4 ml · min(-1) · 100 ml, P = 0.041) decreased in RSP, but not in controls. A 12-week recreational soccer intervention was effective to improve biochemical, cardiovascular, and fitness health markers in obese adolescents.

RESUMO

AIM: This study investigated the accuracy and reliability of short-term resting heart rate variability (HRV) assessment by fingertip photoplethysmography (PPG) and using a cardiofrequencimeter (Polar RS800cx) in obese adolescents. Obesity in adolescents may be related to autonomic dysfunction due to insulin resistance and/or increased central fat. This fact may affect the accuracy and reliability of the evaluation of autonomic activity at rest. METHODS: Fourteen adolescents (15±2 years) classified as obese and exhibiting insulin resistance underwent the following procedures: (a) anthropometric measurements, and (b) two 25-min HRV recordings in a supine position conducted simultaneously using ECG, PPG, and Polar RS800cx. RESULTS: The results showed significant correlations between HRV markers obtained using Polar RS800cx and PPG and those obtained from ECG, with coefficients of determination (R) and intraclass correlation coefficients ranging from 0.60 to 0.98 (P-value ranging from 0.03 to 0.05) and 0.70 to 0.99 (P-value ranging from 0.01 to 0.05), respectively. The technical error of measurement and intraclass correlation coefficients for test-retest reliability ranged from 8.9 to 45.8% and 0.38 to 0.68 for ECG, 8.0 to 30.5% and 0.29 to 0.61 for Polar RS800cx, and 7.2 to 54.6% and 0.45 to 0.64 for PPG, respectively. CONCLUSION: HRV indices calculated by Polar RS800cx and PPG appear to be as accurate and reproducible as data from ECG when evaluating the autonomic control of heart rate at rest in obese adolescents.

RESUMO

The influence of transcranial direct current stimulation (tDCS) upon maximal strength performance in exercises recruiting large muscle mass has not been established in healthy populations. The purpose of this study was to investigate whether anodal tDCS was able to increase the performance during maximal strength exercise (MSEX) in healthy subjects. Fourteen volunteers (age: 26 ± 4 yrs) performed two MSEX after anodal or sham tDCS (2mA; 20min prior MSEX), involving knee extensors and flexors in concentric isokinetic muscle actions of the dominant limb (3 sets of 10 repetitions). The electrical muscle activity (sEMG) of four recruited muscles was recorded during MSEX. Anodal tDCS was not able to improve force production (i.e., total work and peak torque), fatigue resistance, or electromyographic activity during MSEX when compared to sham condition. In conclusion, anodal tDCS applied upon the contralateral motor cortex was not capable of increasing the strength performance of knee extensors and flexors in young healthy subjects.

BACKGROUND: The temporal and insular cortex (TC, IC) have been associated with autonomic nervous system (ANS) control and the awareness of emotional feelings from the body. Evidence shows that the ANS and rating of perceived exertion (RPE) regulate exercise performance. Non-invasive brain stimulation can modulate the cortical area directly beneath the electrode related to ANS and RPE, but it could also affect subcortical areas by connection within the cortico-cortical neural networks. This study evaluated the effects of transcranial direct current stimulation (tDCS) over the TC on the ANS, RPE and performance during a maximal dynamic exercise. METHODS: Ten trained cyclists participated in this study (33±9 years; 171.5±5.8 cm; 72.8±9.5 kg; 10-11 training years). After 20-min of receiving either anodal tDCS applied over the left TC (T3) or sham stimulation, subjects completed a maximal incremental cycling exercise test. RPE, heart rate (HR) and R-R intervals (as a measure of ANS function) were recorded continuously throughout the tests. Peak power output (PPO) was recorded at the end of the tests. RESULTS: With anodal tDCS, PPO improved by ~4% (anodal tDCS: 313.2±29.9 vs 301.0±19.8 watts: sham tDCS; p=0.043), parasympathetic vagal withdrawal was delayed (anodal tDCS: 147.5±53.3 vs 125.0±35.4 watts: sham tDCS; p=0.041) and HR was reduced at submaximal workloads. RPE also increased more slowly during exercise following anodal tDCS application, but maximal RPE and HR values were not affected by cortical stimulation. CONCLUSIONS: The findings suggest that non-invasive brain stimulation over the TC modulates the ANS activity and the sensory perception of effort and exercise performance, indicating that the brain plays a crucial role in the exercise performance regulation.

RESUMO

This study compared resting oxygen uptake (V̇O2) with the standard metabolic equivalent (MET) value of 3.5 mL·kg(-1)·min(-1), tested the accuracy of a previously published prediction model for resting V̇O2, and proposed a new prediction model for a more homogeneous population. One hundred and twenty-five apparently healthy men, aged 17-38 years, visited the laboratory for the assessment of resting V̇O2. The mean resting V̇O2 of 3.21 mL·kg(-1)·min(-1) (95% confidence interval (CI), 3.13 to 3.30) was significantly lower than the standard MET value of 3.5 mL·kg(-1)·min(-1) (mean difference, 0.29; 95% CI, 0.20 to 0.37; t = 6.7; p < 0.001). The prediction model proposed by a previous study, derived from a heterogeneous sample, exhibited no predictive ability in our more homogeneous sample. However, our population-specific regression model, which included body surface area and percent body fat as predictors, demonstrated relatively poor predictive ability, with a low R(2) (0.22) and high standard error of the estimate (0.42 mL·kg(-1)·min(-1)). Pearson's correlation coefficients for body surface area and resting V̇O2, and for percent body fat and resting V̇O2, were 0.20 (p = 0.022) and -0.36 (p < 0.001), respectively. In conclusion, the standard MET value of 3.5 mL·kg(-1)·min(-1) considerably overestimates mean resting V̇O2 in a relatively large group of apparently healthy men. Our population-specific prediction model for resting V̇O2 demonstrated relatively poor accuracy, although it was considerably more accurate than the previously published model. Further research needs to be conducted to establish accurate population-specific prediction models.

Age-associated postural changes directly affect daily-task performance. Exercise has been used in order prevent impairments caused by aging process. This study has aimed to compare the postural profile at the sagittal plane of elderly women participants of the Multidimensional Study of Elderly in Porto Alegre (EMIPOA) and elderly women engaged in an exercise program (GEF) through posture grad assessment. Posterior image analysis and posture classification was done as described by Kendall, Mccreary e Provance (1995). We used descriptive statistics and the Kolmogorov-Smirnov test (α<0.05) to compare the results of the groups. Both groups presented the kyphosis-lordosis as predominant posture profile. There was no significant difference in prevalence of postural profile between EMIPOA and GEF. Results suggest that the exercise program was not effective to produce postural changes at the sagittal plane for elderly women.

RESUMO

This study investigated whether transcranial direct current stimulation (tDCS) on dorsolateral prefrontal cortex (DLPFC) isolated or combined with aerobic exercise influenced the desire to eat, hunger, and satiety in overweight subjects. Nine volunteers underwent anodal or sham tDCS (2 mA; 20 min) over DLPFC and isocaloric exercise bouts (70%VO(2)R; ~200 kcal). The appetite sensations were evaluated by visual analogue scales at four moments: I - Baseline; II - After tDCS; III - Post-Exercise and IV - 30-min Post-Exercise. The tDCS on left DLPFC decreased the desire to eat at baseline (tDCS -26% vs. -14% Sham). The tDCS associated with exercise had greater suppressing effect in desire to eat compared to either tDCS or exercise alone (tDCS -39% vs. -27% Sham). Moreover, the tDCS associated with exercise decreased hunger (tDCS -48% vs. 36% Sham) and increased satiety (tDCS 28% vs. 7% Sham) immediately after exercise. The post-exercise 30-min recovery elicited an overall increase in appetite. However the increase in desire to eat and hunger after recovery was lower after tDCS (29% and 13%, respectively) compared to sham stimulation (77% and 113%, respectively). These findings in overweight subjects indicate that the combination of tDCS over DLPFC and aerobic exercise induced greater decrease in appetite sensations compared to anodal tDCS or exercise alone.

The last two decades has followed an increase in interest from a large number of researchers interested in investigating the ergogenic potential of different resources that might assist in improving the physical performance of athletes in different sports. Most studies have focused on the mechanisms underlying the development of fatigue as this complex phenomenon is a limiting factor in sports performance. Thus, ergogenic resources that are effective in delaying the development of fatigue may improve athletic performance. Despite many discussions about fatigue, its exact cause is still a controversial subject. However, it has been suggested that cortical mechanisms may be involved and studies that have tried to modulate specific brain regions can provide valuable answers to better understand this phenomenon. Currently, (tDCS) has been used as a noninvasive tool that can modulate the excitability of specific regions of the cerebral cortex. Preliminary data have shown that tDCS anode is able to increase the excitability of motor cortex for up to one hour after its application and also increase the time until exhaustion in isometric exercise. These data suggest that tDCS can probably be used as a tool in research on mechanisms involved in the development of fatigue and perhaps as a potential ergogenic aid for athletes..

RESUMO

To investigate whether the manipulation of brain excitability by transcranial direct current stimulation (tDCS) modulates the heart rate variability (HRV), the effect of tDCS applied at rest on the left temporal lobe in athletes (AG) and non-athletes (NAG) was evaluated. The HRV parameters (natural logarithms of LF, HF, and LF/HF) was assessed in 20 healthy men before, and immediately after tDCS and sham stimulation. After anodal tDCS in AG the parasympathetic activity (HF(log)) increased (P<0.01) and the sympathetic activity (LF(log)) and sympatho-vagal balance (LF/HF(log)) decreased (P<0.01), whereas no significant effects were detected in NAG (P>0.05). No significant changes in HRV indexes were provoked by sham stimulation in both AG and NAG (P>0.05). In conclusion, tDCS applied on the left temporal lobe significantly increased the overall HRV in AG, enhancing the parasympathetic and decreasing the sympathetic modulation of heart rate. Consequently the sympatho-vagal balance decreased at rest in AG but not in NAG. Releasing a weak electric current to stimulate selected brain areas may induce favorable effects on the autonomic control to the heart in highly fit subjects.

RESUMO

The concentrations of Legionella pneumophila in cooling towers may vary considerably over short periods of time, producing significant fluctuations throughout the year. Despite genetic variability, in small geographical areas the same indistinguishable pulsed-field gel electrophoresis patterns may be shared among different cooling towers and persist over time.
