RESUMO

The high performance of biomass and metabolite biosynthesis by photosynthetic microorganisms is directly influenced by the cultivation system employed. Photobioreactors (PBRs) stand out as controlled and fundamental systems for increasing the production of biocompounds. However, the high costs associated with these systems hinder their viability. Thus, a more practical and economical approach is necessary. Accordingly, this study aimed to design and evaluate low-cost flat-panel photobioreactors on a laboratory scale for the cultivation of photosynthetic microorganisms, using economical materials and instruments. Additionally, internal optimization of the low-cost system was aimed to maximize growth and biomass production. The PBRs were designed and built with uniform dimensions, employing 4 mm translucent glass and agitation through compressors. The internally optimized system (PBR-OII) was equipped with perforated acrylic plates used as static mixers. To evaluate the performance of the low-cost PBR-OII, a comparison was made with the control photobioreactor (PBR-CI), of the same geometry but without internal optimization, using a culture of Synechocystis sp. CACIAM 05 culture. The results showed that the PBR-OII achieved maximum biomass yield and productivity of 6.82 mg/mL and 250 mg/L/day, respectively, values superior to the PBR-CI (1.87 mg/mL and 62 mg/L/day). Additionally, the chlorophyll concentration in the PBR-OII system was 28.89 ± 3.44 µg/mL, while in the control system, the maximum reached was 23.12 ± 1.85 µg/mL. Therefore, low-cost photobioreactors have demonstrated to be an essential tool for significantly increasing biomass production, supporting research, and reducing costs associated with the process, enabling their implementation on a laboratory scale.

Renewable energy has been recognized as an alternative to fossil fuels as a step to transform the energy produced and consumed worldwide. Cyanobacteria and microalgae are currently being considered as substitutes to the traditional feedstock used to produce biofuels due to their ability to achieve high amounts of lipids under cellular stress conditions. The aim of this study was to investigate the utilization of Tolypothrix sp. CACIAM 22 cyanobacterial biomass as a feedstock for biodiesel production, specifically by examining the effects of supplementing with hydrolysate of Brazil nutshell (HBNS) on biomass generation, lipid production, fatty acid composition, and quality of synthesized biodiesel. The supplementation of HBNS led to a significant increase of 12g.L-1 in wet biomass production. The lipid content reached 41 % of the biomass produced in HBNS supplemented cultures when nitrate source was deprived. The quality evaluation of cyanobacteria-derived biodiesel was performed using Biodiesel Analyzer ver 2.2 software, revealing superior quality compared to biodiesel produced from plant sources. The biodiesel exhibited values of 23 h for oxidative stability, 65 for cetane number, and an iodine index of 31 (g I2. 100 g-1 fat), indicating promising potential as a renewable source. This study is the first to utilize HBNS as an organic supplement for cyanobacteria culture medium and assess its impact on biomass and lipid production in Tolypothrix sp., supporting the hypothesis of utilizing this biomass as a renewable feedstock for biodiesel production as a viable alternative to plant sources based on biomass production, lipid productivity, and biodiesel quality.

RESUMO

Efforts are intensifying to identify new biofuel sources in response to the pressing need to mitigate environmental pollutants, such as greenhouse gases, which are key contributors to global warming and various worldwide calamities. Algae and microalgae present themselves as excellent alternatives for solid-gaseous fuel production, given their renewable nature and non-polluting characteristics. However, making biomass production from these organisms economically feasible remains a challenge. This article collates various studies on the use of lignocellulosic waste, transforming it from environmental waste to valuable organic supplements for algae and microalgae cultivation. The focus is on enhancing biomass production and the metabolites derived from these biomasses.

Lactose intolerance affects approximately 65% of the global adult population, leading to the demand for lactose-free products. The enzyme ß-galactosidase (ßG) is commonly used in the industry to produce such products, but its recovery after lactose hydrolysis is challenging. In this scenario, the study aims to encapsulate ßG within capsules, varying in dimensions and wall materials, to ensure their suitability for efficient industrial recovery. The enzyme ßG was encapsulated through ionic gelation using alginate and its blends with pectin, maltodextrin, starch, or whey protein as wall materials. The capsules produced underwent evaluation for encapsulation efficiency, release profiles, activity of the ßG enzyme, and the decline in enzyme activity when reused over multiple cycles. Alginate at 5% wt/vol concentrations, alone or combined with polymers such as maltodextrin, starch, or whey protein, achieved encapsulation efficiencies of approximately 98%, 98%, 80%, and 88%, respectively. The corresponding enzyme recovery rates were 34%, 19%, 31%, and 48%. Capsules made with an alginate-pectin blend exhibited no significant hydrolysis and maintained an encapsulation efficiency of 79%. Encapsulation with alginate alone demonstrated on poor retention of enzyme activity, showing a loss of 74% after just 4 cycles of reuse. Conversely, when alginate was mixed with starch or whey protein concentrate, the loss of enzyme activity was less than 40% after 4 reuses. These results highlight the benefits of combining encapsulation materials to improve enzyme recovery and reuse, offering potential economic advantages for the dairy industry.

For the first time, a method for the simultaneous analysis of fifteen polycyclic aromatic hydrocarbons (PAHs), including light and heavy PAHs, in açaí-based food products (AFPs) was developed using vacuum-assisted sorbent extraction (VASE) combined with gas chromatography-mass spectrometry (GC-MS). The method requires no organic solvents and is amenable to full automation. To achieve optimal analytical extraction conditions, VASE parameters including stirring rate, extraction time, desorption temperature, desorption time, preheat time, and preheat temperature were optimized using sequential multivariate optimization. The method was validated and yielded limits of quantification below 1 µg kg-1 for all analytes, with recoveries ranging from 65 % to 112 % and good precision (≤11 % relative standard deviation). Additionally, the greenness and practical aspects of the method were investigated using the Green Analytical Procedure Index (GAPI), eco-scale, and the Blue Applicability Grade Index (BAGI), respectively. The VASE-GC-MS approach is suitable for routine analysis and exhibits characteristics of a green analytical method. No PAHs were detected above the limits of detection in thirty samples of AFPs.

The vulnerable status of the Amazon manatee, Trichechus inunguis, indicates the need to seek measures to guarantee its conservation. In this context, the cultivation of cells in vitro is a strategy that should at least guarantee the preservation of their genetic material. Thus, we established for the first time a primary culture of Amazonian manatee fibroblasts (TINsf) from a skin biopsy of a young male. Karyotypic analysis of the 3rd, 7th, and 12th passages confirmed the taxonomic identity of the species T. inunguis (2n = 56/NF = 92) and indicated that this culture presents genomic stability. Gene and protein expression of vimentin at the 13th passage show the predominant presence of fibroblasts in TINsf. To test the cell line's responsiveness to materials and demonstrate a possible application of this culture, it was exposed to andiroba seed oil (ASO), and its viability and proliferative capacity were evaluated. ASO demonstrated toxic effects at the highest concentrations and longest exposure times tested, reproducing results observed in human cultures, indicating the applicability of TINsf in toxicological and biotechnological studies. After cryopreservation, the TINsf line maintained its proliferative potential, indicating the establishment of a new culture available for future studies.

Background: Clavicle fractures are among the most common upper limb fractures in adults, with the midshaft region being the most frequently affected site. Minimally invasive plate osteosynthesis (MIPO) has emerged as an alternative to the traditional open reduction and internal fixation (ORIF) technique, offering potential advantages. The purpose of this study was to conduct a systematic review to explore the results of this technique in the existing literature, with emphasis on the occurrence of surgical complications and functional outcomes and also to provide a comprehensive comparison of MIPO and ORIF in the management of midshaft clavicle fractures. Methods: We conducted a systematic review to evaluate the complication incidence and clinical outcomes of MIPO for midshaft clavicle fractures. We searched PubMed/Medical Literature Analysis and Retrieval System Online (MEDLINE), Scopus, the Cochrane Database of Controlled Trials, and the Cochrane Database of Systematic Reviews databases without language or date restrictions. Studies focusing on midshaft clavicle fractures treated with MIPO were included, while other clavicle fractures and nonclinical studies were excluded. The risk of bias was assessed using the Methodological Index for Nonrandomized Studies criteria and the Risk of Bias Tool 2 Cochrane tool. Data synthesis included qualitative analysis, and if applicable, quantitative analysis and meta-analysis. Adherence to Preferred Reporting Items for Systematic Reviews and Meta-analyses guidelines ensured reporting quality. Results: A total of 107 studies were initially identified, after applying inclusion and exclusion criteria, 22 studies were included for data extraction. These studies involved the evaluation of 714 clavicles treated with the MIPO technique. Of the 714 MIPO cases, 11 cases of implant failure, 5 nonunions, 2 infections, and 28 cases with neurological impairment were observed. Quantitative analysis comparing MIPO with ORIF revealed that MIPO had significantly shorter surgery time (mean difference -12.95, 95% confidence interval [-25.27 to -0.63], P = .04) and lower occurrence of numbness (odds ratio 0.29, 95% CI [0.15-0.56], P = .0002) compared to ORIF. Time to bone union, functional outcomes, and other complications were similar between MIPO and ORIF at the final follow-up. An overall moderate risk of bias was found across the studies. Conclusion: The MIPO technique yields good and comparable results to ORIF for midshaft clavicle fractures. Additionally, the MIPO technique may offer advantages such as reduced surgical time and lower chances of neurological impairment.

In this study, polymeric ionic liquids featuring different functional moieties were applied as sorbent coatings in direct-immersion solid-phase microextraction (DI-SPME) for the extraction of 2-methylimidazole (2-MI) and 4-methylimidazole (4-MI) from açaí-based food products followed by gas chromatography-mass spectrometry (GC-MS) analysis. The analytical method was optimized using a sequential experimental design. Variables used in GC-MS such as desorption time, as well as for SPME-DI, including extraction time, extraction temperature, incubation time of extraction, amount of NaCl in the extract, and stirring rate, were optimized. The fitness-for-purpose of the method was verified by the linearity of matrix-matched calibration curves (R2 ≥ 0.9921), adequate recoveries (81.7-89.7 %), and precision (relative standard deviations ≤11.2 %). The method was applied to twenty-five samples of açaí-based food products. 4-MI was found in four samples whereas 2-MI was not detected above the limit of detection. The method was found to be suitable for quality control analysis.

For the first time, benzophenone and related compounds were investigated in açaí-based food products. An extraction method based on the dilute-and-shoot approach, combined with the use of in-situ formed metal-containing ionic liquids (MCILs) followed by high-performance liquid chromatography, was developed and validated. A nickel and cobalt-based MCIL, in addition to the ratio of MCIL to lithium bis[(trifluoromethyl)sulfonyl]imide salt ([Li+][NTf2-]) for the ensuing metathesis reaction, were optimized. Parameters of the in-situ formed MCIL step, namely, the amount of MCIL, centrifugation time, and dilution step, were analyzed using a multivariate optimization approach, including central composite rotatable design and Derringer and Suich's tool. Optimum extraction performance was achieved using 50.98 mg of nickel-based MCIL and a MCIL to ([Li+][NTf2-]) ratio of 1:3 (m/m), a centrifuge time of 22 min, and 10.53 mL of water for the dilution step. This condition was used to perform analytical validation, which yielded satisfactory results with R2 ≥ 0.995, limits of detection (LOD) ranging from 0.0025 to 0.5 mg kg-1, and limits of quantification (LOQ) between 0.008 and 1.5 mg kg-1. The recovery rate ranged from 87 % to 107 % and precision values (as percent relative standard deviation) were equal or lower than 13 %. The validated method was applied to 25 samples of açaí-based food products purchased from Brazil and the United States. None of the samples showed analyte concentration levels above the LOD. The method's suitability was demonstrated for future monitoring of complex samples, such as foodstuffs.

INTRODUCTION: The species Lantana camara is used in folk medicine. The biological activities of this medicinal plant are attributable to the presence of various derivatives of triterpenoids and phenolic compounds present in its preparations, indicating excellent economic potential. OBJECTIVE: In this study, the operational conditions of ultrasound-assisted extraction (UAE) and microwave-assisted extraction (MAE) were optimized using Box-Behnken design to improve the total phenolic content (TPC) recovered in hydroethanolic extracts of L. camara leaves. MATERIAL AND METHODS: The TPC, total flavonoid content (TFC), and antioxidant activities of the hydroalcoholic extracts of L. camara, prepared by UAE and MAE under the optimized extraction conditions, were compared with those of the extracts obtained by conventional extraction methods. RESULTS: Under the optimal conditions, the extracts obtained by UAE (35% ethanol, 25 min, and a solvent-to-solid ratio of 60:1 mL/g) and by MAE (53% ethanol, 15 min, and 300 W) provided high yields of 32.50% and 38.61% and TPC values of 102.89 and 109.83 mg GAE/g DW, respectively. The MAE extract showed the best results with respect to TPC, TFC, and antioxidant activities, followed by extracts obtained by UAE, Soxhlet extraction, decoction, maceration, and infusion, in that order. CONCLUSION: The results obtained indicate that L. camara may be used as an important source of antioxidant phenolic compounds to obtain products with high biological and economic potential, especially when the extraction process is performed under appropriate conditions using MAE and/or UAE, employing environmentally friendly solvents such as water and ethanol.

Pentaclethra macroloba (Willd.), whose common name is "pracaxi," is naturally found in the Amazon region. The present study is aimed at analyzing the anatomy, seed histochemistry, and chemical composition in fatty acid profile of P. macroloba seed oils. Seeds were collected in the cities of Belém, Marituba, and São Domingos do Capim-PA. For the study in light microscopy, scanning electron microscopy, and histochemistry, seeds were sectioned in cross and longitudinal sections of the embryonic axis and fixed in formaldehyde, acetic acid, and 50% ethyl alcohol; neutral-buffered formalin; and formaldehyde and ferrous sulfate and stored in 70% ethyl alcohol. For the anatomical study, the seeds were subjected to the usual techniques of plant anatomy. Histochemical tests were performed on plant material, freehand sectioned, and embedded in histological paraffin with DMSO. The fatty acid profile was determined for gas chromatography (GC-FID). Integument is divided into three strata, monoseriate exotesta, mesotesta formed by several layers of parenchyma cells, and monoseriate endotesta, formed by compressed cells. Cotyledons are composed of thin-walled parenchyma cells with several secretory cavities and secretory idioblasts. The main metabolic classes are lipids, phenolic compounds, carbohydrates, proteins, and alkaloids. The main fatty acids found in P. macroloba oil are oleic, behenic, lignoceric, and linoleic. P. macroloba seeds have important anatomical characteristics for their circumscription in Leguminosae and also in Caesalpinioideae, and their oil is rich in fatty acids essential to the human diet, providing many benefits to the human health, such as fatty acids belonging to the omega family (linoleic, oleic).

In the Amazon, some species of Loricariidae are at risk of extinction due to habitat loss and overexploitation by the ornamental fish market. Cytogenetic data related to the karyotype and meiotic cycle can contribute to understanding the reproductive biology and help management and conservation programs of these fish. Additionally, chromosomal mapping of repetitive DNA in Loricariidae may aid comparative genomic studies in this family. However, cytogenetics analysis is limited in Amazonian locariids. In this study, chromosomal mapping of multigenic families was performed in Scobinancistrus aureatus, Scobinancistrus pariolispos and Spatuloricaria sp. Meiotic analyzes were performed in Hypancistrus zebra and Hypancistrus sp. "pão". Results showed new karyotype for Spatuloricaria sp. (2n=66, NF=82, 50m-10sm-6m). Distinct patterns of chromosomal organization of histone H1, histone H3 and snDNA U2 genes were registered in the karyotypes of the studied species, proving to be an excellent cytotaxonomic tool. Hypotheses to explain the evolutionary dynamics of these sequences in studied Loricariidae were proposed. Regarding H. zebra and H. sp. "pão", we describe the events related to synapse and transcriptional activity during the meiotic cycle, which in both species showed 26 fully synapsed bivalents, with high gene expression only during zygotene and pachytene. Both Hypancistrus species could be used may be models for evaluating changes in spermatogenesis of Loricariidae.

Thousands of tons of residual lignocellulosic biomass are produced and discarded by agroindustries in the Amazon. These biomasses could be harnessed and used in the preparation of activated carbon, in view of the growing demand for this product with high added value, however, little is known about their characteristics, in addition to their potential as precursors of activated carbon. Therefore, the aim of this work was to evaluate the potential of four different biomasses in the preparation and quality of activated carbon. Residues from the processing of the fruits of acai, babassu, Brazil nut, and oil palm were collected, characterized, carbonized, physically activated with CO2, and characterized. The contents of the total extractives, insoluble lignin, minerals, holocellulose, and elemental (CHNS-O) were analyzed. The surface area and surface morphology were determined from the AC produced, and adsorption tests for methylene blue and phenol were performed. The four biomasses showed potential for use in the preparation of CA; the residues presented high contents of lignin (21.83-55.76%) and carbon (46.49-53.79%). AC were predominantly microporous, although small mesopores could be observed. The AC had a surface area of 569.65-1101.26 m2 g-1, a high methylene blue (93-390 mg g-1), and phenol (159-595 mg g-1) adsorption capacities. Babassu-AC stood out compared to the AC of the other analyzed biomasses, reaching the best results.

Neural tissue-related illnesses have a high incidence and prevalence in society. Despite intensive research efforts to enhance the regeneration of neural cells into functional tissue, effective treatments are still unavailable. Here, a novel therapeutic approach based on vertically aligned carbon nanotube forests (VA-CNT forests) and periodic VA-CNT micropillars produced by thermal chemical vapor deposition is explored. In addition, honeycomb-like and flower-like morphologies are created. Initial viability testing reveals that NE-4C neural stem cells seeded on all morphologies survive and proliferate. In addition, free-standing VA-CNT forests and capillary-driven VA-CNT forests are created, with the latter demonstrating enhanced capacity to stimulate neuritogenesis and network formation under minimal differentiation medium conditions. This is attributed to the interaction between surface roughness and 3D-like morphology that mimics the native extracellular matrix, thus enhancing cellular attachment and communication. These findings provide a new avenue for the construction of electroresponsive scaffolds based on CNTs for neural tissue engineering.

Alternanthera littoralis P. Beauv is a plant native to Brazil that exhibits various beneficial activities including antioxidant, antibacterial, antifungal, antiprotozoal, anti-hyperalgesic, and anti-inflammatory properties. The aim of this study was to assess the impact of the ethanol extract of Alternanthera littoralis (EEAl) on reproductive outcomes, embryofetal development, and DNA integrity of pregnant female mice. Pregnant Swiss female mice were randomly assigned to three experimental groups (n = 10): controls were administered either 1% Tween 80 (vehicle), EEAl 100 mg/kg or EEAl 1000 mg/kg. Treatment was administered through gavage during the gestational period until day 18. On gestational days 16, 17, and 18, a peripheral blood sample from the tail vein was obtained for DNA integrity analysis (micronucleus test). After the last collection, animals were euthanized by cervical dislocation. Maternal organs and fetuses were collected, weighed, and subsequently analyzed. Reproductive outcome parameters were assessed by measurement of number of implants, live fetuses, and resorptions. Embryonic development was determined by adequacy of weight for gestational age as well as determination of external, visceral, and skeletal malformations. Data demonstrated that EEAl did not produce maternal toxicity at either dose associated with no marked alterations in any of the reproductive outcome parameters including implantation sites, live/dead fetuses ratio, fetal viability, post-implantation losses, resorptions, and resorption rate. However, EEAl 1000 group reduced embryofetal development by lowering placental weight. In addition, there was an increase in the frequency of external and skeletal malformations in the EEAl 1000 group, which could not be attributed to extract exposure as these values were within control levels. Based upon our findings, evidence indicates that the EEAl at the concentrations employed in our study may be considered safe for use during pregnancy and extracts of this plant show potential for development of phytomedicines to be used in pregnancy.

RESUMO

In the context of a diet transition from animal protein to plant protein, both for sustainable and healthy scopes, innovative plant-based foods are being developing. A combination with milk proteins has been proposed as a strategy to overcome the scarce functional and sensorial properties of plant proteins. Based on this mixture were designed several colloidal systems such as suspensions, gels, emulsions, and foams which can be found in many food products. This review aims to give profound scientific insights on the challenges and opportunities of developing such binary systems which could soon open a new market category in the food industry. The recent trends in the formulation of each colloidal system, as well as their limits and advantages are here considered. Lastly, new approaches to improve the coexistence of both milk and plant proteins and how they affect the sensorial profile of food products are discussed.

Cattle trypanosomiasis negatively impacts animal husbandry due to high morbidity, productivity losses, and mortality rates. Knowledge regarding Trypanosoma evansi infections in locally adapted breeds remains limited. Some cattle breeds exhibit trypanotolerance, requiring the determination of prevalence, as well as related tolerance and resistance characteristics, for disease control programs. This study aimed to determine T. evansi prevalence in Crioula Lageana cattle and associate clinical, hematological, and biochemical aspects with the infection to further research on tolerance in this population. Blood samples from 310 Crioula Lageana cattle were tested using Polymerase Chain Reaction (PCR) and Indirect Immunofluorescence Reaction (IIFR). T. evansi prevalence was 8% (24/310) using PCR and 4% (11/310) using IIFR. Positive animals showed increased ruminal movements, elevated eosinophil counts, and reduced monocyte numbers, but both latter within the reference range for the species. Albumin concentrations were low in positive cases and remained below the reference range limit for both groups. However, triglycerides exceeded the physiological range for the species in both positive and negative groups. Increased gamma-glutamyltransferase (GGT) activity was observed in positive animals. In conclusion, Crioula Lageana cattle exhibited enzootic instability with a low T. evansi infection prevalence when assessed using PCR and IIFR techniques. Furthermore, the animals did not display clinical, hematological, or biochemical alterations attributable to the presence of hemoparasites.

This study aimed to investigate the suitability of the application of high-intensity ultrasounds (HIUS) to improve the acid induced gelation of mixed protein systems formed by casein micelles (CMs) and pea. The protein suspensions were prepared in different protein ratios CMs: pea (100:0, 80:20, 50:50, 20:80, 0:100) at 8% (w/w) total protein concentration. In the suspensions, the ultrasound treatment produced an increase in solubility, surface hydrophobicity, and a decrease in the samples' viscosity, with more remarkable differences in protein blends in which pea protein was the major component. However, the replacement of 20% of CMs for pea proteins highly affected the gel elasticity. Hence, the creation of smaller and more hydrophobic building blocks before acidification due to the HIUS treatment increased the elasticity of the gels up to 10 times. Therefore, high-intensity ultrasounds are a suitable green technique to increase the gelling properties of CMs: pea systems.

The occurrence of persistent organic pollutants in aquatic bodies, namely polycyclic aromatic hydrocarbons (PAHs), has been increasingly detected. The presence of such contaminants represents a serious threat to human health due to their toxicity. Therefore, aiming to provide a novel and efficient alternative for PAHs' removal from water, the present study assesses the effect of oligo(ß-pinene) blended with chitosan for the adsorption of these pollutants. Oligo(ß-pinene) with phenyl end-groups was synthesized by organocatalyzed atom transfer radical polymerization (O-ATRP) and incorporated in different concentrations (6, 12, and 18 %) to chitosan films. The oligo(ß-pinene) loading in the chitosan matrix impressively improved this polysaccharide adsorption capacity. The formulation containing 12 % of oligomer demonstrated a contaminant removal performance three times higher (298.82 %) than pure chitosan during only 1 h of the decontamination process. Adsorption isotherms showed an improved uptake of PAHs with the increase of the contaminants' concentration in the aqueous media due to the formation of a higher concentration gradient. Additionally, a comprehensive characterization of oligo(ß-pinene)/chitosan formulation was performed to provide a better understanding of the interactions between the components of the blends. Overall, it was concluded that oligo(ß-pinene)/chitosan blends can be used as a high-performance and sustainable alternative for PAHs removal.
