RESUMO

1. Aflatoxins (AFs) are metabolites which especially have toxic effects on proteins, and are detoxified by the aflatoxin-B1 aldehyde reductase (AFAR) pathway. In this pathway, the aldo-keto reductase family 7, member A2 (AKR7A2) enzyme, which is controlled by nucleic-related erythroid factor 2 (Nrf2), plays an active role. However, data on the efficacy of this critical pathway in broilers is limited.2. The aim of the following study was to investigate the changes in the expression levels of AKR7A2, Nrf2, and caspase-3, and the effects of Nigella sativa seeds (NS), thymoquinone (TMQ), and bentonite (BNT) in broilers exposed to AFs.3. One-hundred broilers were divided into ten groups (control (CNT); AF; NS; TMQ; BNT; AF+TMQ; AF+NS; AF+BNT; AF+BNT+NS; AF+BNT+TMQ) and fed for 28 d. AF, TMQ, NS and BNT were added to diets at levels of 2 mg/kg, 300 mg/kg, 50 g/kg and 10 g/kg respectively.4. The addition of AF to the diet decreased AKR7A2 and Nrf2 levels dramatically, but increased caspase-3 (P < 0.01). TMQ, NS and BNT additions to the diet eliminated all negative effects caused by AF (P < 0.01); and AKR7A2 and Nrf2 were further raised in TMQ and NS groups when compared to the control group. TMQ and NS showed a positive effect on detoxification parameters when given together with BNT.5. Supplementation with NS and TMQ enhanced AF detoxification via the AFAR pathway, by increasing AKR7A2 and Nrf2 levels, in addition to reducing hepatocyte apoptosis.

Sulfur mustard (SM) was responsible for more than 80% of all documented chemical casualties during the Great War. Recent literature on clinic picture of SM exposure remained so limited with the sporadic cases who were accidentally exposed to SM especially either in Western Europe or China. We reported a Syrian family of four who became victims of chemical terrorism due to SM exposure and we described the detailed clinical course of the family including the medical history, initial symptomatology, clinical examination, hematological data, and initial treatment in the first 48 hours after exposure at Kilis State Hospital, Turkey. The principles of our therapeutic approaches were designed according to the total affected body surface area, severity of cutaneous and respiratory lesions, and existing hematological disorders. SM is still considered as a critical vesicant agent and a current threat because of its ease of synthesis. Chemical terrorist attacks of non-state actors or terrorist organizations with "home-made" SM is likely such a threat which is targeting health systems of developed and developing countries. Except sarin attacks in Japan, the literature depends on real incidents of chemical terrorism is so rare and for this reason we have gaps and challanges in the prepardness of medical response system against chemical terrorism. Medical management could be performed adequetly only if the response system is well planned, well equipped, and well prepared for overburdened medical facilities filled with SM contaminated casualties after a chemical terrorist attack.

We evaluated the efficacy of yeast glucomannan (Mycosorb), incorporated into the diet at 0.5 and 1 g/kg, in reducing the detrimental effects of 2 mg aflatoxin/kg diet on growing broiler chicks from 1 to 21 d of age. A total of 240 male broiler chicks (Ross-308) was divided into 6 treatment groups [Control, Aflatoxin (AF), Yeast glucomannan (YG; 0.5 g/kg), AF plus YG (0.5 g/kg), YG (1 g/kg), and AF plus YG (1 g/kg)]. Ten chicks from each of the 6 groups were slaughtered and pathological examinations were performed on the liver, bursa of Fabricius, thymus, spleen and kidney. The aflatoxin treatment caused moderate to severe hydropic/fatty degeneration in the hepatocytes of the liver and the tubular epithelium of the kidneys, and follicular depletion in the bursa of Fabricius, thymus and spleen. Yeast glucomannan added to the aflatoxin-containing diet at 0.5 and 1 g/kg diminished the severity of pathological changes, slightly and moderately, respectively. The number of affected organs was also reduced in the group given 1 g/kg yeast glucomannan, compared to the aflatoxin group. These results show that yeast glucomannan effectively diminished the adverse effects of aflatoxin on the pathological changes and that the higher concentration of yeast glucomannan (1 g/kg) was more effective than the lower concentration (0.5 g/kg) and itself had no adverse effect.

This study was conducted to evaluate the pathological changes in broilers fed a diet containing low-levels of aflatoxin (AF) and clinoptilolite (CLI) until 42 days of age. A total of 576 one-day-old Ross-308 type broiler chicks were treated with varying levels of AF and CLI (15 g kg(-1)). The gross and histopathological changes in the liver, kidneys, spleen, thymus and bursa of Fabricius were investigated and relative organ weights were calculated. Compared to controls, significant changes (P<0.05), such as slight to moderate hydropic degeneration and/or fatty change (8 cases of 10), bile-duct hyperplasia (7 of 10) and periportal fibrosis (5 of 10), were found in chicks fed 100 ppb AF-containing diet. No gross-pathological changes were observed in any treatments. The addition of CLI to the 100 ppb AF-containing diet significantly decreased the number of affected broilers and/or the severity of lesions (hydropic degeneration and bile-duct hyperplasia) in the livers (P<0.05). The addition of CLI to the AF-free diet did not produce any significant lesions compared with the controls.

The effects of different dietary levels of zinc and vitamin A on tissue mineral accumulation and histopathological alterations were examined in laying hens. For this purpose, 130 Hisex brown laying hens aged 56 weeks were divided into two main groups. Each group of hens were fed on diets supplemented with two different levels of vitamin A (0-10,000 IU kg(-1)). Then, laying hens in both of the main groups were subdivided into five treatment groups given different levels of zinc (0, 25, 50, 100, and 200 mg kg(-1)) for 12 weeks. Dietary additions of zinc and/or vitamin A did not significantly affect the zinc concentrations of the pancreas, liver, gizzard, thyroid, and copper concentrations of the pancreas, gizzard, and thyroid. The group fed the highest zinc (200 mg kg(-1)) in the diet had twofold iron concentration in their thyroid tissue compared to the concentration of the control group receiving no extra zinc. Histopathologically, there was slight oedema, degranulation, and vacuolation in exocrine cells and some fibrotic areas in the pancreas and reduced follicular size. Follicular cell hypertrophy and hyperplasia occurred in the thyroids, and gizzard erosions were seen in some cases.

This study was conducted to determine the pathological changes in testes and epididymides and plasma testosterone levels of adult roosters during experimentally induced aflatoxicosis. In the study, 24 months of age, 32 Babcock breeder males were used, and they were divided into four groups each containing 8 animals. The groups were designed as follows; group 1: Control, no aflatoxin (AF), group 2: 5 ppm (parts per million) total aflatoxin (AF; B1, B2, G1, G2), group 3: 10 ppm AF and group 4: 20 ppm AF in the diet, and the birds were fed for 8 weeks. Grossly, it was seen that the testes of all AF-treatment groups birds were significantly (P < 0.001) atrophied when compared with those of control birds. Histopathologically, there was no spermatogenesis in the testes of 4, 5 and 6 cocks fed on a diet containing AF 5, 10 and 20 ppm, respectively. Furthermore, abnormal spermatozoa were observed in some of AF-treatment groups (in 2 cases in each of 5 and 10 ppm AF-treated groups, and in one case in 20 ppm AF-treated group). There were also mononuclear cell infiltration and/or focal lymphoid cell accumulation in the intertubular areas of the testes and epididymides in all AF-treatment groups. In conclusion, it has been shown that AF might totally or partially (dose related) suppress spermatogenesis, cause abnormality in spermatozoa and atrophy in testes. Furthermore, there was degeneration and desquamation in the epithelium and decrease in the size and thickness of the germinative layer of the seminiferous tubules, and lowered plasma testosterone levels in adult roosters.

The amelioration of aflatoxicosis in broiler chickens was examined by feeding two concentrations of natural zeolite (clinoptilolite). Clinoptilolite (ClI), incorporated into the diet at 1.5 and 2.5 per cent, was evaluated for the ability to reduce the deleterious effects of 2.5 mg total aflatoxin (AF) kg(-1)diet on growing broiler chicks from 1 to 21 days of age. A total of 360 broiler chicks were divided into six treatment groups [Control, AF, CLI (1.5 per cent), AF plus CLI (1.5 per cent), CLI (2.5 per cent), and AF plus CLI (2.5 per cent)] each consisting of 60 chicks. Compared to controls, the AF consuming chicks showed increases in the relative weights of liver and kidney; and gross-histopathologic hepatic lesions such as paleness, friability, diffuse hydropic degeneration and/or fatty change, bile-duct hyperplasia and periportal fibrosis. Glumerular hypertrophy, increases in the number of mesengial cells and hydropic degeneration of tubuler epithelium in kidneys of chicks fed diet AF alone were also observed. Atrophy and lymphoid depletion were seen in the thymuses and bursa of Fabricius from the chicks fed AF alone. The additions of CLI (1.5 and 2.5 per cent) to the AF -containing diet moderately (significantly in some cases) decreased the number of affected broilers and/or the severity of lesions. The addition of CLI to the AF-free diet did not produce any significant changes compared with the controls. These results suggest that CLI was effective for the protection of AF-toxication in broilers and it could contribute to a solution of the AF problem in poultry production.
