RESUMO

A summer study and a winter study were conducted using an incomplete factorial structure in a complete randomized design. Within season, the factors studied were 1) type of diet, which included 2 levels of ME, classified as either concentrate (3.04 Mcal of ME/kg) or roughage (2.63 Mcal of ME/kg) diets, and 2) daily ME intakes (MEI) of 11, 18, and 25 Mcal of ME/d for the roughage diets and 18, 25, and 32 Mcal of ME/d for the concentrate diets. In Exp. 1 (summer study), 30 steers (5 steers/treatment combination) were used to collect tympanic temperatures (TT). In Exp. 2 (winter study), 24 steers (4 steers/treatment combination) were used to collect TT. Mean TT was 0.3°C greater for summer than winter (38.9 vs. 38.6°C, respectively; P < 0.05). Steers fed diets based on concentrate tended to display greater TT than steers fed diets based on roughage. Season × diet × hour interactions were found for TT (P = 0.01). In the winter, greater TT (P < 0.05) were found from 0900 to 1400 h when an equal amount of MEI was derived from a concentrate-based vs. roughage-based diet. In cattle fed roughage-based diets during the summer, TT = 38.63 + 0.0114•MEI, whereas for cattle fed concentrate-based diets, TT = 38.69 + 0.0114•MEI. During the winter, for cattle fed a roughage-based diet, TT = 37.65 + 0.0856•MEI - 0.0018•MEI(2), whereas for cattle fed a concentrate-based diet, TT = 35.37 + 0.2635•MEI - 0.0051•MEI(2). In summary, results demonstrate that increases in the energy of the diet resulted in increases in TT. However, the response was dependent on season of the year, with a linear response in TT for summer and a quadratic response during winter.

Malolactic fermentation (MLF), the conversion of malate to lactate, is an important process leading to the deacidification of hard apple cider. MLF is dependent on the levels of inhibitory factors such as sulfur dioxide and ethanol. To assess the effect of these 2 factors on MLF, hard apple cider was produced from pasteurized, unfiltered apple cider (Malus domestica cvs Red Delicious, Golden Delicious, Braeburn, and Fuji). Apple cider was treated with 2 levels of sulfur dioxide (50 and 80 ppm) and then fermented using Saccharomyces cerevisiae montrachet. After the primary fermentation, 1 set of the samples remained unadjusted and 100% ethyl alcohol was used to adjust other sets of samples to 7%, 9%, or 11% (v/v) ethanol. Following the ethanol adjustment, Oenococcus oeni MCW was used to initiate the MLF in half of the samples. Cider parameters monitored throughout the fermentations included organic acid content, titratable acidity, pH, ethanol production, and sugar content. Since samples containing either sulfur dioxide level had similar sugar utilization rates and ethanol production it was concluded that sulfur dioxide had no effect on the primary fermentation. Sulfur dioxide content was shown to have an impact on MLF. There was no difference in the rate of malic acid consumption, but lactic acid production was faster in the 50-ppm sulfur dioxide samples. MLF was not inhibited by ethanol content.

Thermal hysteresis may be used to quantify and characterize the amount of heat stress an animal experiences. Heat stress studies of farm animals suggest that above a certain threshold body temperature (Tb) is driven by ambient temperature (Ta). Patterns in the Tb-Ta phase diagram indicate the presence of hysteresis. When hysteresis is present, there are two values for Tb depending on whether Ta is increasing or decreasing. A theoretical delay-relay model is proposed to illustrate the hysteretic dynamics of the Tb-Ta relationship when Ta is cyclic. Two types of hysteresis, transitional and thermal, are identified. Thermal hysteresis becomes prominent when Ta forces Tb into an elliptical limit cycle. The area of the ellipse is an indication of the animal's heat load. Also, the resulting Tb-Ta pattern depends on the delay (or lag) between Tb and Ta. The delay suggests possible levels of heat stress. A greater delay between Tb and Ta indicates more time is needed to dissipate the heat load, implying the animal is suffering more heat stress. For a given Ta, the Tb on the decreasing (recovery) path is always higher than the Tb on the increasing (challenge) path. Essentially, the animal requires more energy to dissipate heat than to absorb it.

Serotonin (5-HT), a neurotransmitter synthesized from tryptophan, has been proposed as a feedback inhibitor of lactation. We determined that the gene coding for tryptophan hydroxylase 1, the rate-limiting enzyme for 5-HT synthesis, is expressed in bovine mammary epithelial cells in vitro and is upregulated by prolactin. In addition, 5-HT reduced the expression of alpha-lactalbu-min and casein genes in vitro. Furthermore, inhibiting 5-HT synthesis with p-chlorophenylalanine or blocking the 5-HT receptor with methysergide (METH) increased milk protein gene expression. We then evaluated effects of intramammary 5-HT or METH infusion on production and milk composition in 6 multiparous Holstein cows. Cows were assigned to a repeated measures design of contralateral intramammary infusions of METH (20 mg/quarter per d) or saline for 3 d followed by a 7-d washout period before administering 5-HT (50 mg/quarter/d) or SAL for 3 d. For each udder half, milk yield was recorded twice and composition was determined once per day. Blood samples were harvested each day for plasma to determine glucose and nonesterified fatty acid concentrations. Evaporative heat loss, respiration rate, left and right udder temperatures, and rectal temperatures were obtained after each milking to evaluate possible systemic effects of infusions. During METH and saline infusions milk yield increased 10.9%. During 5-HT and saline infusion milk yield decreased 11.1%. Milk yield and physiological responses suggested intramammary 5-HT and METH doses were high enough to cause systemic effects. Infusing saline, METH, and 5-HT increased milk SCC. Infusing 5-HT tended to reduce mean lactose concentration (4.3 vs. 4.6%) relative to saline. Milk protein content was decreased by METH and SAL (2.0%) and was increased (5.8%) by 5-HT followed by a 33% decrease postinfusion. Infusion of METH increased evaporative heat loss 11%, which decreased 11% postinfusion. Infusions of 5-HT or METH did not affect plasma nonesterified fatty acid or glucose concentrations, respiration rate, or milk fat content. We conclude 5-HT infusion reduced milk synthesis, whereas blocking the 5-HT receptor with METH increased milk synthesis. Doses of 5-HT and METH used in this study likely resulted in systemic effects. These data support the concept that 5-HT is a feedback inhibitor of lactation in the bovine.

Heat stress in feedlot cattle causes reduced performance, and in the most severe cases, death of the animals, thus causing the loss of millions of dollars in revenue to the cattle industry. A study was designed to evaluate dynamics of thermoregulation and feeding activities when feeder cattle were exposed to simulated heat waves, in comparison with repeated sinusoidal hot and thermoneutral environments. Nine beef steers were randomly assigned to an individual pen in one of three environmental chambers. Each chamber was subjected to each of three temperature regimes (heatwave simulation from Rockport, Mo., 1995, heatwave simulation from Columbia, Mo., 1999, and Controlled heat stress treatment of 32+/-7 degrees C) for a period of 18 days, according to a Latin square treatment design, with a 10-day thermoneutral period (18+/-7 degrees C) separating treatment periods. Respiration rate, core body temperature, heat production, feed intake, and feeding behavior were measured on each animal for the duration of the experiment. Differences were found in all treatments for all parameters except feeding behavior. It was shown that the two simulated heat waves elicited very different thermoregulatory responses. Based on these results the heat wave centered at Rockport, Mo. in 1995 was devastating because the animals were not acclimated to hot conditions, thus causing an acute response to heat stress. The responses of cattle to conditions at Columbia, Mo. showed some acclimation to heat prior to the peak stress days, and therefore a dampened response was seen. It appears the extreme conditions at Columbia, Mo., 1999 were made severe by environmental conditions not simulated during this study (low wind speed and intensive solar radiation). Overall, it was determined while a cyclic heat stress treatment is a representative model to test heat stress in cattle, further heat stress experiments should be conducted in an actual feedlot.

Net portal absorption of AA during the 6-h postprandial period was measured in eight gilts (48.5 +/- 1.6 kg BW) in a crossover design. The pigs had chronic catheters placed in the portal vein, carotid artery, and ileal vein, and were trained to consume 1.2 kg of a standard grower diet once daily. Blood samples were taken every 30 min for 4 h and then hourly until 6 h after feeding. The first set of blood samples was taken after pigs were fed a meal of the test 16% CP corn-soybean meal diet (16% CP) or the test 12% CP corn-soybean meal diet supplemented with crystalline lysine, threonine, and tryptophan (12% CP + AA) to equal the three AA levels in the 16% CP diet. Pigs were then fed the standard diet for 2 d. Following that, blood samples were again taken after the pigs were fed a meal of the test diet that was not given to them at the first sampling period. Net portal AA absorption was calculated by multiplying porto-arterial plasma AA concentration difference by portal vein plasma flow rate (PVPF), estimated by an indicator-dilution technique employing p-aminohippuric acid as the indicator infused into the ileal vein. Plasma concentrations of lysine and threonine of pigs were affected by the diet x time interaction (P < 0.01). Portal and arterial plasma lysine and threonine concentrations in pigs attained the maximal level by 1 h postprandial when the 12% CP + AA diet was fed, but reached the peak level at 2.5 h postprandial when the 16% CP diet was given. The PVPF of pigs over the 6 h postprandial was less (P < 0.01) when the 12% CP + AA diet was given than when the 16% CP diet was fed. Net portal absorptions of lysine and threonine also were affected (P < 0.05) by time x diet interaction. The peak portal absorption of both lysine and threonine in pigs appeared at 0.5 h postprandial when the 12% CP + AA diet was given, but at 2.5 h postprandial with the feeding of the 16% CP diet. The early appearance of peak portal absorption of lysine and threonine from feeding the 12% CP + AA compared with the 16% CP diet indicates that crystalline lysine and threonine are absorbed more rapidly than protein-bound lysine and threonine in pigs fed once daily.

The objective was to describe variability in prevalence, incidence, and duration of fecal shedding of naturally occurring E. coli O157:H7 by a group of feedlot cattle over time. One hundred steers, randomly assigned to 10 pens, were fed a high-concentrate finishing diet for 136 days (19 weeks). Rectal feces from each animal were tested for E. coli O157:H7 every week for 19 weeks. E. coli O157:H7 was recovered from each animal that completed the study and was detected from at least one animal every week. Average pen prevalence of cattle shedding E. coli O157:H7 varied significantly over time (P < 0.0001) and across pens (P < 0.0001), ranging from 1 to 80%. Pairwise comparisons of mean pen prevalence of E. coli O157:H7 between weeks and estimation of the predicted probability of an incident case of E. coli O157:H7 over time allowed the definition of three distinct phases--namely, the preepidemic, epidemic, and postepidemic periods. Average pen prevalence varied significantly over time (P < 0.01) and across pens (P < 0.001) for all time periods. The odds of an incident case were significantly greater during epidemic and postepidemic periods relative to the preepidemic period (P = 0.0002 and P = 0.03, respectively). Duration of infection was significantly longer for first or second infections that began during epidemic or postepidemic periods relative to the preepidemic period (P < 0.001). Both incidence and duration of shedding peaked during the epidemic period. Pen-level prevalence of cattle shedding E. coli O157:H7 was affected by both incidence and duration of shedding and could be explained by time- or pen-dependent risk factors, or both.

Three experiments were conducted to evaluate the effect of different management strategies on body temperature of feedlot steers finished in the summer months. In Exp. 1, 24 crossbred steers were chosen to assess the effect of altered feed intake and feeding time on tympanic temperature (TT) response. Managed feeding (MF) treatments were applied for 22 d only and provided 1) ad libitum access to feed at 0800 (ADLIB), 2) feed at 1600 with amount adjusted so that no feed was available at 0800 (BKMGT), 3) feed at 1600 at 85% of predicted ad libitum levels (LIMFD). During heat stress conditions on d 20 to 22 of MF, LIMFD and BKMGT had lower (P < 0.05) TT than ADLIB from 2100 through 2400. A carryover effect of limit-feeding was evident during a severe heat episode (d 36 to 38) with LIMFD steers having lower (P < 0.05) TT than ADLIB. In Exp. 2, TT were obtained from 24 crossbred steers assigned to three treatments, consisting of no water application (CON), water applied to feedlot mound surfaces from 1000 to 1200 (AM) or 1400 to 1600 (PM). From 2200 to 0900 and 1200 to 1400, steers assigned to morning sprinkling treatment had lower (P < 0.05) TT than steers assigned to afternoon sprinkling treatment. In Exp. 3, 24 steers were utilized in a 2 x 2 factorial arrangement of treatments with factors of feeding time [0800 (AMF) and 1400 (PMF)] and sprinkling (WET and DRY). Tympanic temperatures were monitored under hot environmental conditions on d 30 to 32 and 61 to 62. A feeding time x sprinkling interaction (P < 0.001) was evident on d 30 to 32, although AMF/DRY steers had the highest (P < 0.05) TT. On d 61 to 62, TT of PMF steers was higher (P < 0.05) than AMF between 1500 to 1800. Use of sprinklers can effectively reduce TT of feedlot cattle, whereas shifting to an afternoon vs morning feeding time was most beneficial when bunks were empty several hours prior to feeding.

The mobility of pooled turkey sperm following various storage regimens was assessed by objectively measuring the ability of sperm to penetrate a 2% Accudenz [5-(N-2,3-dihydroxypropylacetamido)-2,4,6-tri-iodo-N,N'-bis(2,3-dihydroxypropyl)isophthalamide] solution at 41 C. When semen was diluted with Beltsville poultry semen extender and stored at 5 C with agitation at 150 rpm, sperm mobility declined as the storage interval increased (P < or = 0.05), with mobility scores (mean +/- SEM) of 0.440+/-0.029, 0.374+/-0.031, 0.282+/-0.011, 0.202+/-0.019, and 0.130+/-0.019 for 0-, 3-, 6-, 24-, and 48-h storage, respectively. For a 10-wk fertility trial using the same storage method, sperm mobility and fertilizing capacity of semen were significantly reduced following 24-h storage compared with values for unstored semen. The sperm mobility scores were 0.404+/-0.051 and 0.101+/-0.046 for unstored and 24-h stored semen, respectively, whereas the percentage of fertilized eggs was 95.9+/-5.1 for unstored semen and 48.0+/-5.1 for 24-h stored semen. When caffeine or pentoxifylline was added to semen at 2.5, 5, or 10 mM, no significant effect on sperm mobility was seen, regardless of whether these compounds were added to unstored semen, were present during 6-h storage, or were added following the 6-h storage interval. These studies demonstrate that sperm mobility and fertilizing capacity of pooled turkey semen declines with storage, and that addition of caffeine or pentoxifylline either during or after storage does not affect sperm mobility.

The objectives of this project were to see whether heart rate, respiration rate, blood pressure, and vocalization could be used to evaluate stress of castration in pigs. Six groups of pigs 1, 2, 4, 8, 16, and 24 d of age were used in the study, a total of 172 pigs. Half of each group of pigs received lidocaine before castration, injected subcutaneously over the testicle and infiltrated around the cord; the other half were left as controls. Pigs castrated without lidocaine had a higher heart rate (P < .02) and higher frequency of highest energy (HEF) measurements of vocalization (P < .05). Incising the scrotum and severing the spermatic cord elicited the greater heart rate response (P < .05) to castration without anesthetic, whereas HEF was lower during cutting of the cord. Both the heart rate and HEF data suggest that castration without anesthetic is of greater stress for pigs 8 d of age or older. Respiration rate was not a viable measure of stress associated with castration.

To study the use of electromagnetic scanning in prediction of lean content in beef carcasses and cuts, 100 beef cattle (60 steers and 40 heifers), representing a broad range in external fat thickness (.1 to 2.9 cm) and live weight (414 to 742 kg), were selected. Chilled right sides were divided into streamlined (foreshank, brisket, and ventral plate removed) forequarters (FQ) and full hindquarters (HQ) and scanned. Primal rounds, loins, ribs, and chucks were fabricated from the right side, scanned, and physically separated into lean, fat, and bone. Prediction equations for dissected lean content and percentage of lean included the peak of the electromagnetic scan response curve (obtained from scanning the HQ or FQ), length, temperature and weight of the scanned cut, and fat thickness at the 12th rib. Using the coefficient of determination, root mean square error, and Mallows' Cp statistic, the best model for each dependent variable (weight and percentage of lean) that included up to five independent variables was selected. Prediction equations for the HQ or FQ of steers accounted for 84 to 93% of the variation in lean weight of beef sides and quarters and 71 to 93% of primals. Sixty-one to 75% of the variation in percentage of lean in sides and quarters and 48 to 65% of primals was also explained. Similar results were obtained for heifer carcasses. Predicting percentage of lean in any scanned cut, rather than weight of lean, accounted for less of the variation. Weight and fat thickness contributed significantly when predicting percentage of lean.(ABSTRACT TRUNCATED AT 250 WORDS)

Albendazole (methyl 5-propylthio-1 H-benzimidazol-2-yl carbamate) was used as an anthelmintic in a 3-yr study involving 578 beef cows and 438 nursing calves. Infection levels for nematodes, coccidia, and tapeworm were relatively low throughout the 3-yr period. Eggs per gram of feces in cows and calves were lower (P less than .01) 2 wk posttreatment but were not different 5 mo later, when calves were weaned. Cow weight gain, rate and time of conception, and adjusted calf weaning weights were not affected significantly by deworming of either cows or calves. Level of nematode infection measured as eggs per gram of feces was higher (P less than .01) in younger cows than in mature cows. Although deworming with Albendazole lowered (P less than .01) nematode infection levels, no responses were observed in cow or calf performance.
