RESUMO

Antibody-drug conjugates (ADCs) have greatly improved the outcomes of advanced breast tumors. However, the treatment of breast tumors with existing ADCs is still hindered by many issues, such as tumor antigen heterogeneity and drug resistance. Therefore, ADCs against new targets would provide options for the treatment of these challenges. Sortilin-1 (SORT1) may be a promising target for ADC as it is upregulated in breast cancer. To evaluate the possibility of SORT1 as an ADC target, a humanized antibody_8D302 with high affinity against SORT1 was generated. Additionally, 8D302 was conjugated with MMAE and DXd to generate two ADCs_8D302-MMAE and 8D302-DXd, respectively. Both 8D302-MMAE and 8D302-DXd showed effective cytotoxicity against SORT1 positive breast tumor cell lines and induced bystander killing. Consequently, 8D302-MMAE showed relatively better anti-tumor activity than 8D302-DXd both in vitro and in vivo, but 8D302-DXd had superior safety profile and pharmacokinetics profile over 8D302-MMAE. Furthermore, SORT1 induced faster internalization and lysosomal trafficking of antibodies and had a higher turnover compared with HER2. Also, 8D302-DXd exhibited superior cell cytotoxicity and tumor suppression over trastuzumab-DXd, a HER2-targeted ADC. We hypothesize that the high turnover of SORT1 enables SORT1-targeted ADC to be a powerful agent for the treatment of SORT1-positive breast tumor.

This study proposes a multidimensional uncalibrated technique for tracking and grasping dynamic targets by a robotic arm in the eye-in-hand mode. This method avoids complex and cumbersome calibration processes, enabling machine vision tasks to be adaptively applied in a variety of complex environments, which solved the problem of traditional calibration methods being unstable in complex environments. The specific method used in this study is first, in the eye-in-hand mode, the robotic arm moves along the x, y, and z axes in sequence, and images are taken before and after each movement. Thereafter, the image Jacobian matrix is calculated from the three (or more) sets of images collected. Finally, the robotic arm converts the target coordinates in the real-time captured images by the camera into coordinates in the robotic arm coordinate system through the image Jacobian matrix and performs real-time tracking. This study tests the dynamic quasi-Newton method for estimating the Jacobian matrix and optimizes the initialization coupling problem using the orthogonal moving method. This optimization scheme significantly shortens the iteration process, making the uncalibrated technology more fully applied in the field of dynamic object tracking. In addition, this study proposes a servo control algorithm with predictive compensation to mitigate or even eliminate the systematic error caused by time delay in dynamic target tracking in robot visual servo systems.

RESUMO

Head and neck primary soft tissue sarcoma is a rare adult connective tissue malignant tumor derived from mesenchymal tissue, which can occur in the paranasal sinuses, throat or neck space.The clinical manifestations are local spread masses in the head and neck or difficulty breathing, swallowing, etc al. MRI and enhanced CT examination are the most commonly used to diagnose such diseases. Pathological diagnosis requires immunohistochemistry combined with FISH to detect MDM2 and CDK4. In this report,two cases of primary soft tissue sarcoma were reported,one is parotid high-differentiated liposarcoma and the other is laryngeal dedifferentiated leiomyosarcoma, introducing the characteristics diagnosis and treatment, and reviewing the relevant literature.

In this paper, we introduce a novel three-dimension chaotic system with strange characteristic by applying construction of a 3D chaotic circuit method. Multiple equilibria and abundant coexisting attractors exist in this system. A mathematical model is developed and detailed stability analyses for equilibrium points are executed with obtaining significant results of the period-doubling bifurcation patterns confirmed by phase plane plots and Lyapunov exponent spectra. By varying the initial value and unique controlled parameter, the double-scroll chaotic attractor is broken up into a pair of symmetric singular attractors. Then, the local basins of attraction are investigated concerning the initial condition. Next, the circuit synthesis results generated by Multisim simulation tool validate the self-excitation characteristics of this system. Finally, the feedback control technique is used to study difference synchronization of this system. Main conclusions prove the validity and reliability of difference synchronization.

RESUMO

Laryngeal schwannoma is a rare clinical benign neurogenic tumor that originated in the larynx. Large laryngeal schwannoma can cause foreign body sensation in the pharynx and swallowing obstruction. Clinical symptoms includehoarseness and dyspnea. The enhanced CT scan illustrates that the localized mass is isolated, with clear boundary with the surrounding tissues in the larynx. A 68-year-old woman presented with a swallowing discorder,endoscopy demonstrated a smooth mass of the left aryepiglottic fold.The patient then underwent endoscopy surgey supported by the low temperature plasma instead of an open incision.

Accumulated evidence has demonstrated that miRNAs are closely implicated in lung carcinogenesis. Herein, we explored the expression pattern of miR-30b-5p in lung cancer, and aimed to uncover miR-30b-5p roles in lung cancer progression and drug resistance. miR-30b-5p expression profiles in lung cancer tissues and the matched non-tumor tissues were determined by using qPCR. Cell viability, migration, invasion and in vivo tumorigenesis were determined by using the CCK-8, colony formation, wound healing, transwell chambers experiments and tumor xenograft models. RNA immunoprecipitation (RIP) and dual luciferase reporter experiments were applied to evaluate the relationship between miR-30b-5p and LRP8. The results demonstrated that miR-30b-5p showed a low expression profile in lung cancer tissues and cells, and closely linked to poor prognosis and malignant clinical process. Cell viability, migration, invasiveness and tumorigenesis were significantly weakened following miR-30b-5p overexpression in A549 and NCI-H1299 cells, while cell apoptosis rates were increased. In addition, miR-30b-5p was lowly expressed in A549/DDP (a cisplatin drug resistant cell line) as compared with A549 cells, and miR-30b-5p increased A549/DDP cell sensitivity to DDP. However, these above roles of miR-30b-5p were all significantly impaired following the overexpression of LRP8 which was overexpressed in lung cancer tissues. Collectively, this study demonstrated that miR-30b-5p functions as a tumor suppressor in lung cancer, and re-sensitizes lung cancer cells to DDP by targeting LRP8.

Although modern radiotherapy offers excellent local control in the treatment of nasopharyngeal carcinoma (NPC), current therapeutic decisions remain burdensome due to the frequency of local recurrence and treatment failure at distant sites. One potential and promising strategy for the prevention or treatment of cancers is the use of bioactive components of plant origin, including dietary plant products. Herein, we studied one class of these bioactive compounds, grape seed proanthocyanidins (GSPs), and explored their effect on NPC CNE-2 cells, as well as the primary mechanism underlying this effect. Our results revealed that treatment of human NPC CNE-2 cells with GSPs reduced cell viability in a dose- and time-dependent manner, and moreover, markedly induced cell cycle arrest at the G2/M phase, leading to induction of apoptosis. In addition, we found that the underlying mechanism was associated with increased expression of the pro-apoptotic protein Bax, decreased expression of the anti-apoptotic proteins Bcl-2 and Bcl-xL, upregulation of cleaved caspase-3 and cleaved poly(ADP-ribose) polymerase (PRAP) protein, and the loss of mitochondrial membrane potential (MMP) (Δψm). Furthermore, GSPs upregulated the Bcl-2 homology 3 (BH3)-only proteins, Bim and Bad, in a concentration-dependent manner. Taken together, these data supported our hypothesis that, in human NPC CNE-2 cells, GSPs could induce apoptosis through the mitochondrial pathway and ultimately reduce cell viability. Collectively, the results discussed above provide substantive evidence for the potential of GSPs as an effective bioactive phytochemical for the treatment of NPC.

OBJECTIVE: To study the relationship between the radiotherapy resistance and autophagy. To provide a theoretiacal basis for drugs that regulate autophagy to improve radiotherapy sensitivity. METHOD: Flow cytometry (FCM) was performed to analyze the distribution of the cell cycle of CNE2 and CNE2/DDP cells under the action of X radiation. The expression of autopagy-specific gene Beclin1 and microtubule-associated protein light chain 3ß (MAPLC3ß) in CNE2 and CNE2/DDP cells was determined by real time PCR and Immumofluorescence staining. RESULT: CNE2/DDP and their parental CNE2 cells produced the G2-M phase arrest under the action of X radiation. With the radiation dose increasing,The cells which in the G2-M phase were more and more (P<0. 05). The G2-M phase arrest in CNE2/DDP cells was more obvious than in CNE2 cells (P<0. 05). The expression of Beclin1 and MAPLC3ß in CNE2 and CNE2/DDP cells increased under the action of X radiation. What's more, the raise was more and more obvious with the increase of the irradiation dose(P<0. 05). The expression levels of Beclin1 and MAPLC3ß in CNE2/DDP was lower than that in CNE2 cells (P<0. 05). CONCLUSION: Autophagic cell death may be the one manner of death in nasopharyngeal carcinoma CNE2 and CNE2/DDP cells under the action of X radiation. The radiation resistance of CNE2/DDP cells may be related to the low expression of autophagy-related genes.
