RESUMO

Cataract disease results from non-amyloid aggregation of eye lens proteins and is the leading cause of blindness in the world. A variety of studies have implicated both essential and xenobiotic metals as potential etiological agents in cataract disease. Essential metal ions, such as copper and zinc, are known to induce the aggregation in vitro of human γD crystallin, one of the more abundant γ-crystallins in the core of the lens. In this study, we expand the investigation of metal-crystallin interactions to heavy metal ions, such as divalent lead, cadmium and mercury. The impact of these metal ions in the non-amyloid aggregation, protein folding and thermal stability of three homologous human lens γ-crystallins has been evaluated using turbidity assays, electron microscopy, electronic absorption and circular dichroism spectroscopies. Our results show that Hg(II) ions can induce the non-amyloid aggregation of human γC and γS crystallins, but not γD crystallin. The mechanism of Hg-induced aggregation involves direct metal-protein interactions, loss of thermal stability, partial unfolding of the N-terminal domain of these proteins, and formation of disulfide-bridged dimers. Putative Hg(II) binding sites in γ-crystallins involved in metal-induced aggregation are discussed. This study reveals that mercury ions can induce the aggregation of human lens proteins, uncovering a potential role of this heavy metal ion in the bioinorganic chemistry of cataract disease.

Built-up on top of ancient lake deposits, Mexico City experiences some of the largest seismic site effects worldwide. Besides the extreme amplification of seismic waves, duration of intense ground motion from large subduction earthquakes exceeds three minutes in the lake-bed zone of the basin, where hundreds of buildings collapsed or were seriously damaged during the magnitude 8.0 Michoacán earthquake in 1985. Different mechanisms contribute to the long lasting motions, such as the regional dispersion and multiple-scattering of the incoming wavefield from the coast, more than 300 km away the city. By means of high performance computational modeling we show that, despite the highly dissipative basin deposits, seismic energy can propagate long distances in the deep structure of the valley, promoting also a large elongation of motion. Our simulations reveal that the seismic response of the basin is dominated by surface-waves overtones, and that this mechanism increases the duration of ground motion by more than 170% and 290% of the incoming wavefield duration at 0.5 and 0.3 Hz, respectively, which are two frequencies with the largest observed amplification. This conclusion contradicts what has been previously stated from observational and modeling investigations, where the basin itself has been discarded as a preponderant factor promoting long and devastating shaking in Mexico City.

In most clinical trials, human mesenchymal stem cells (hMSCs) are expanded in vitro before implantation. The genetic stability of human stem cells is critical for their clinical use. However, the relationship between stem-cell expansion and genetic stability is poorly understood. Here, we demonstrate that within the normal expansion period, hMSC cultures show a high percentage of aneuploid cells that progressively increases until senescence. Despite this accumulation, we show that in a heterogeneous culture the senescence-prone hMSC subpopulation has a lower proliferation potential and a higher incidence of aneuploidy than the non-senescent subpopulation. We further show that senescence is linked to a novel transcriptional signature that includes a set of genes implicated in ploidy control. Overexpression of the telomerase catalytic subunit (human telomerase reverse transcriptase, hTERT) inhibited senescence, markedly reducing the levels of aneuploidy and preventing the dysregulation of ploidy-controlling genes. hMSC-replicative senescence was accompanied by an increase in oxygen consumption rate (OCR) and oxidative stress, but in long-term cultures that overexpress hTERT, these parameters were maintained at basal levels, comparable to unmodified hMSCs at initial passages. We therefore propose that hTERT contributes to genetic stability through its classical telomere maintenance function and also by reducing the levels of oxidative stress, possibly, by controlling mitochondrial physiology. Finally, we propose that aneuploidy is a relevant factor in the induction of senescence and should be assessed in hMSCs before their clinical use.

Aerobic training is related to an increase in blood oxidation markers. The purpose of the present study was to investigate the antioxidant capacity of Lippia citriodora extracts (PLX(®) ) on plasma and blood cell oxidative status of university students beginning a 21 days aerobic training routine (3 days/week). Using a double-blind design, 15 male athletes (21 ± 2.1 years) were assigned to a group consuming 1.8 g/day of the plant extract (PLX(®) -group) or a placebo (PLB-group). Two blood extractions were performed at day 0 and 21, from which lymphocytes, erythrocytes and plasma were isolated. Several circulating parameters, antioxidant enzyme activities and oxidative stress markers were measured. The PLX(®) -group displayed an increased HDL-cholesterol, a modest decrease in erythrocyte number and an increased circulating urea. Activation of glutathione (GSH)-reductase was observed in erythrocytes and lymphocytes of PLX(®) -group, accompanied by lower levels of oxidative stress markers, such as malondialdehyde and protein carbonyls in plasma. The antioxidant action exerted by PLX(®) on GSH-reductase seems to be post-translational and mainly due to verbascoside, a phenylpropanoid that represents 10% (w/w) of extract content. In conclusion, PLX(®) shows antioxidant properties that could play an important role in modulating GSH-reductase activity in lymphocytes and erythrocytes and protecting plasma from exercise oxidative damage.

Expansion of human stem cells before cell therapy is typically performed at 20% O(2). Growth in these pro-oxidative conditions can lead to oxidative stress and genetic instability. Here, we demonstrate that culture of human mesenchymal stem cells at lower, physiological O(2) concentrations significantly increases lifespan, limiting oxidative stress, DNA damage, telomere shortening and chromosomal aberrations. Our gene expression and bioenergetic data strongly suggest that growth at reduced oxygen tensions favors a natural metabolic state of increased glycolysis and reduced oxidative phosphorylation. We propose that this balance is disturbed at 20% O(2), resulting in abnormally increased levels of oxidative stress. These observations indicate that bioenergetic pathways are intertwined with the control of lifespan and decisively influence the genetic stability of human primary stem cells. We conclude that stem cells for human therapy should be grown under low oxygen conditions to increase biosafety.

Fet3p is a multicopper oxidase recently isolated from the yeast, Saccharomyces cerevisiae. Fet3p is functionally homologous to ceruloplasmin (Cp) in that both are ferroxidases. However, by sequence homology Fet3p is more similar to fungal laccase, and both contain a type 1 Cu site that lacks the axial methionine ligand present in the functional type 1 sites of Cp. To determine the contribution of the electronic structure of the type 1 Cu site of Fet3p to the ferroxidase mechanism, we have examined the absorption, circular dichroism, magnetic circular dichroism, electron paramagnetic resonance, and resonance Raman spectra of wild-type Fet3p and type 1 and type 2 Cu-depleted mutants. The spectroscopic features of the type 1 Cu site of Fet3p are nearly identical to those of fungal laccase, indicating a very similar three-coordinate geometry. We have also examined the reactivity of the type 1 Cu site by means of redox titrations and stopped-flow kinetics. From poised potential redox titrations, the E degrees of the type 1 Cu site is 427 mV, which is low for a three-coordinate type 1 Cu site. The kinetics of reduction of the type 1 Cu sites of four different multicopper oxidases with two different substrates were compared. The type 1 site of a plant laccase (Rhus vernicifera) is reduced moderately slowly by both Fe(II) and a bulky organic substrate, 1,4-hydroquinone (with 6 equiv of substrate, k(obs) = 0.029 and 0.013 s(-)(1), respectively). On the other hand, the type 1 site of a fungal laccase (Coprinus cinereus) is reduced very rapidly by both substrates (k(obs) > 23 s(-)(1)). In contrast, both Fet3p and Cp are rapidly reduced by Fe(II) (k(obs) > 23 s(-)(1)), but only very slowly by 1,4-hydroquinone (10- and 100-fold more slowly than plant laccase, respectively). Semiclassical theory is used to analyze the origin of these differences in reactivity in terms of type 1 Cu site accessibility to specific substrates.

We developed a neuropsychological battery for assessment of cognitive processes that was standardized in 150 neurologically intact subjects from different socioeducational levels in Mexico City (Ostrosky et al., 1985, 1986). The present study was designed to explore the capacity of this neuropsychological battery to discriminate a brain-injured population from a normal one. Thirty-four patients attending the neurological service of two hospitals institutions in Mexico City were studied. The reasons for going to the hospital included both neurological and neuropsychological symptoms. The group was divided into two subgroups: twenty-four patients who showed brain damage confirmed by brain scans, and ten patients with a normal brain scan. A control group of 19 normal subjects was also studied and paired with the other groups by sex, age and sociocultural level. The results show that the neuropsychological battery was able to recognize 83.3% of the patients with scanographically confirmed brain damage: the total percentage of successful diagnosis was 88.2% and there were no false positives. These results indicate that neuropsychological assessment is a powerful diagnostic procedure that also evaluates the patient's cognitive-behavioral activity and can help to predict the possibilities for rehabilitation and return to work.

We describe the pre- and postoperative neuropsychological profiles of seven patients who received an autograft of adrenal medullary tissue to the caudate nucleus for the treatment of Parkinson's Disease (PD). The preoperative neuropsychological evaluations revealed specific cognitive deficits of varying degree. The patients showed frontal lobe-type deficits with alterations in behavioral programming leading to difficulties in the organization of motor sequences and alternating programs. They also showed memory disorders and visuospatial and visuoperceptual deficiences such as a loss of figure-ground perspective and fragmentation. Postoperative evaluations, carried out 3 months after neurosurgery, revealed a significant amelioration of the frontal lobe-type symptoms and visuospatial deficits, as well as an improvement in memory tasks that require an active organization of the response. Immediate and delayed memory difficulties remained unchanged. These observations were compared to neuropsychological data obtained from neurologically intact subjects and from unoperated PD patients. The improvements of the operated PD patients resulted in performance levels close to normal values and clearly distinguishable from those of unoperated PD patients, and were unrelated to improved mood, increased alertness, or sustained attention. Autotransplantation of adrenal medullary tissue to the caudate nucleus of PD patients showing a decreased effective response to L-dopa therapy can partially restore motor functions and frontal-type cognitive symptoms.

The group selected for this evaluation consisted of 109 normal people with an average age of 25, coming from two different sociocultural levels (high and low) of Mexico City. All were completely evaluated by Luria's battery for neuropsychological assessment, adapted by Ardila, Ostrosky, and Canseco, 1981. This group of tests measures nine different areas: Motor Functions, Somatosensory Knowledge, Auditory Knowledge, Visuospatial Knowledge, Cognitive Processes, Language, Reading, Writing and Basic Calculations. For all of these, the higher performance standards were achieved by the subjects from the high sociocultural level. A significant interaction between sociocultural level and sex was observed. The differences between sexes appear only in subjects from the low sociocultural level. Factor analysis of the battery revealed that the most sensitive items to sociocultural level were those related on one hand, to the handling of complex structural and conceptual aspects of language and on the other hand, to the organization of motor sequences and in general motor programming. Research related to the differences found is reviewed and implications for clinical assessment are discussed.

Three experiments are reported investigating individual differences in interhemispheric electroencephalogram (EEG) activity and cognitive responses to persuasion. Experiment 1 indicated that subjects who were characterized by relative left hemispheric EEG activity over the parietal areas also produced a less affectively polarized profile of thought listings about the persuasive communication. Moreover, this individual difference emerged only when subjects were confronted by the forewarning and message; the basal patterns of interhemispheric EEG activity, which were obtained prior to the announcement of the attitudinal recommendation, did not portend distinguishable profiles of cognitive responding. Experiment 2 replicated the major findings of Experiment 1 using different topics and a within-subjects rather than a between-subjects design. Further analyses suggested that thinking about an attitude issue rather than responding to a persuasive communication was sufficient to obtain the above relationship between interhemispheric EEG alpha abundance and cognitive response. Experiment 3 used Tesser's time-to-think procedure to assess interhemispheric EEG patterning as a function of the affective polarization of topic-relevant thinking. The results supported the expectation that as subjects thought longer about attitude issues they exhibited a shifting of relative hemispheric EEG activity from the left toward the right parietal areas of the cerebral hemispheres. The significance and limitations of these findings for research on attitude change and the utility of including psychophysiological approaches to elusive research problems in personality and social psychology are discussed.
