Wastewater-based epidemiology has been widely used as a cost-effective method for tracking the COVID-19 pandemic at the community level. Here we describe COVIDBENS, a wastewater surveillance program running from June 2020 to March 2022 in the wastewater treatment plant of Bens in A Coruña (Spain). The main goal of this work was to provide an effective early warning tool based in wastewater epidemiology to help in decision-making at both the social and public health levels. RT-qPCR procedures and Illumina sequencing were used to weekly monitor the viral load and to detect SARS-CoV-2 mutations in wastewater, respectively. In addition, own statistical models were applied to estimate the real number of infected people and the frequency of each emerging variant circulating in the community, which considerable improved the surveillance strategy. Our analysis detected 6 viral load waves in A Coruña with concentrations between 103 and 106 SARS-CoV-2 RNA copies/L. Our system was able to anticipate community outbreaks during the pandemic with 8-36 days in advance with respect to clinical reports and, to detect the emergence of new SARS-CoV-2 variants in A Coruña such as Alpha (B.1.1.7), Delta (B.1.617.2), and Omicron (B.1.1.529 and BA.2) in wastewater with 42, 30, and 27 days, respectively, before the health system did. Data generated here helped local authorities and health managers to give a faster and more efficient response to the pandemic situation, and also allowed important industrial companies to adapt their production to each situation. The wastewater-based epidemiology program developed in our metropolitan area of A Coruña (Spain) during the SARS-CoV-2 pandemic served as a powerful early warning system combining statistical models with mutations and viral load monitoring in wastewater over time.

The quantification of the SARS-CoV-2 RNA load in wastewater has emerged as a useful tool to monitor COVID-19 outbreaks in the community. This approach was implemented in the metropolitan area of A Coruña (NW Spain), where wastewater from a treatment plant was analyzed to track the epidemic dynamics in a population of 369,098 inhabitants. Viral load detected in the wastewater and the epidemiological data from A Coruña health system served as main sources for statistical models developing. Regression models described here allowed us to estimate the number of infected people (R2 = 0.9), including symptomatic and asymptomatic individuals. These models have helped to understand the real magnitude of the epidemic in a population at any given time and have been used as an effective early warning tool for predicting outbreaks in A Coruña municipality. The methodology of the present work could be used to develop a similar wastewater-based epidemiological model to track the evolution of the COVID-19 epidemic anywhere in the world where centralized water-based sanitation systems exist.

In regions with a mild climate, pesticides are often used around homes for pest control. Recent monitoring studies have linked pesticide use in residential areas to aquatic toxicity in urban surface water ecosystems, and suggested dust particles on paved surfaces as an important source of pesticides. To test the hypothesis that dust on hard surfaces is a significant source of pesticides, we evaluated spatial and temporal patterns of current-use insecticides in Southern California, and further explored their distribution as a function of particle sizes. Pyrethroid insecticides were detected in dust from the driveway, curb gutter and street at 53.5-94.8%, with median concentrations of 1-46 ng g(-1). Pyrethroid residues were uniformly distributed in areas adjacent to a house, suggesting significant redistribution. The total levels of pyrethroids in dust significantly (p < 0.01) decreased from October to February, suggesting rainfalls as a major mechanism to move pesticide residues offsite. Fipronil as well as its degradation products, were detected at 50.6-75.5%, and spatial and temporal patterns of fipronil residues suggested rapid transformations of fipronil to its biologically active intermediates. Moreover, pyrethroids were found to be enriched in fine particles that have a higher mobility in runoff than coarse particles. Results from this study highlight the widespread occurrence of pesticides in outdoor dust around homes and the potential contribution to downstream surface water contamination via rain-induced runoff.

Nonylphenol (NP), one of the priority hazardous substances, is in fact a mixture of numerous isomers. It is inconclusive whether or not biodegradation during wastewater treatment process is isomer-specific, leading to the environmental release of NP in different isomer profiles. In this study, we evaluated the isomer selectivity of 19 NP isomers in a laboratory-scale continuous flow conventional activated sludge bioreactor under various operational conditions. The removal efficiency of NP isomers ranged from 90 to 99%, depending on the operational conditions and isomer structures. Isomer selective biodegradation resulted in the increase of composition of recalcitrant isomers, such as, NP193a/b, NP110a and NP194 in the effluent. Moreover, biodegradability was related to the bulkiness of α-substituents and followed α-dimethyl > α-ethyl-α-methyl > α-methyl-α-n-propyl > α-iso-propyl-α-methyl. Steric effect index, a quantitative descriptor of steric hindrance, was linearly correlated with residues of NP isomers in the effluent (R² = 0.76). Decrease of temperature to 10 °C decreased the overall biodegradability and also enhanced the relative enrichment of recalcitrant isomers. These findings suggest that isomer compositions of NP entering the environment may be different from those in technical mixtures and that isomeric selectivity should be taken into account to better understand the occurrence, fate, and ecological risks of NP.

In light of rapid global change, the demand for wastewater treatment is increasing rapidly and will continue to do so in the near future. Wastewater management is a complex puzzle for which the proper pieces must be combined to achieve the desired solution, requiring the simultaneous consideration of technical, economic, social and environmental issues. In this context, a knowledge-based methodology (KBM) for the conceptual design of wastewater treatment plant (WWTP) process flow diagrams (PFDs) and its application for two scenarios is presented in this paper. The core of the KBM is composed of two knowledge bases (KBs). The first, a specification knowledge base (S-KB), summarizes the main features of the different treatment technologies: pollutants removal efficiency, operational costs and technical reliability. The second, a compatibility knowledge base (C-KB), contains information about the different interactions amongst the treatment technologies and determines their degree of compatibility. The proposed methodology is based on a decision hierarchy that uses the information contained in both KBs to generate all possible WWTP configurations, screening and selecting appropriate configurations based on user-specified requirements and scenario characteristics. The design of the most adequate treatment train for small and medium sized wastewater treatment plants (2000 and 50,000 p.e. respectively) according to different restrictions (space constraints, operation simplicity and cost optimization) was the example in order to show the usefulness of the KBM.

The influence of operation condition on the fate of 16 pharmaceutical and personal care products (PPCPs) in a single-sludge nitrifying/denitrifying pilot plant was assessed. Volatilisation, sorption and degradation were included in the mass balances to determine the most relevant removal mechanisms during PPCP treatment. Sludge retention time (SRT) was an important factor for the removal of compounds that significantly sorb onto sludge, as ethinylestradiol, whose removal increased 11% when working at SRT above 20 d. The internal recirculation ratio was significant for the removal of moderately biodegradable compounds, as citalopram. The positive effect of operating at warmer temperatures was particularly significant for two antibiotics, implying a 30% increase in their transformations. In the case of naproxen, an influence of sludge acclimation and concentration was observed, leading to removal efficiencies from 27% to 99%. Concerning removal mechanisms, most compounds were removed due to biotransformation, although for fragrances sorption and volatilisation played a role.

The occurrence and fate of eight pharmaceutical and personal care products (PPCPs) during sewage treatment has been studied in a pilot-scale treatment plant, comprising a primary settler (2.85 m(3)), an aeration tank (1.845 m(3)) and a secondary clarifier (0.5 m(3)), placed on site at a wastewater treatment works in the north west of the UK. It was fed both with raw sewage and the return liquor produced after sludge centrifugation, thus representing the most common configuration for a municipal sewage treatment plant based on the activated sludge process. Samples were taken at six different locations, including the return liquor stream, and analysed for musk fragrances and pharmaceutically active compounds belonging to various therapeutic groups such as anti-inflammatory drugs, tranquillisers and antiepileptics. Mass balances were conducted for those PPCPs that were quantifiable. The fate of the PPCPs was found to differ according to their physical-chemical characteristics. Anti-inflammatories underwent a degradation process and were almost completely removed from sewage during the biological treatment step. Musk fragrances were only partially removed, through adsorption onto the primary suspended solids and the biomass in the aerobic process, due to their strong lipophilic characteristics. The results of this study provide increasing evidence that the partial removal of these substances through the sewage treatment process contribute to the environmental occurrence of PPCPs. Consequently, existing STPs should be upgraded in order to attenuate the release of these substances into the aquatic environment.
