RESUMO

Multilingualism has been demonstrated to lead to a more favorable trajectory of neurocognitive aging, yet our understanding of its effect on neurocognition across the lifespan remains limited. We collected resting state EEG recordings from a sample of multilingual individuals across a wide age range. Additionally, we obtained data on participant multilingual language use patterns alongside other known lifestyle enrichment factors. Language experience was operationalized via a modified multilingual diversity (MLD) score. Generalized additive modeling was employed to examine the effects and interactions of age and MLD on resting state oscillatory power and coherence. The data suggest an independent modulatory effect of individualized multilingual engagement on age-related differences in whole brain resting state power across alpha and theta bands, and an interaction between age and MLD on resting state coherence in alpha, theta, and low beta. These results provide evidence of multilingual engagement as an independent correlational factor related to differences in resting state EEG power, consistent with the claim that multilingualism can serve as a protective factor in neurocognitive aging.

INTRODUCTION: Emerging evidence links changes in the gut microbiome to late-onset Alzheimer's disease (LOAD), necessitating examination of AD mouse models with consideration of the microbiome. METHODS: We used shotgun metagenomics and untargeted metabolomics to study the human amyloid beta knock-in (hAß-KI) murine model for LOAD compared to both wild-type (WT) mice and a model for early-onset AD (3xTg-AD). RESULTS: Eighteen-month female (but not male) hAß-KI microbiomes were distinct from WT microbiomes, with AD genotype accounting for 18% of the variance by permutational multivariate analysis of variance (PERMANOVA). Metabolomic diversity differences were observed in females, however no individual metabolites were differentially abundant. hAß-KI mice microbiomes were distinguishable from 3xTg-AD animals (81% accuracy by random forest modeling), with separation primarily driven by Romboutsia ilealis and Turicibacter species. Microbiomes were highly cage specific, with cage assignment accounting for more than 40% of the PERMANOVA variance between the groups. DISCUSSION: These findings highlight a sex-dependent variation in the microbiomes of hAß-KI mice and underscore the importance of considering the microbiome when designing studies that use murine models for AD. HIGHLIGHTS: Microbial diversity and the abundance of several species differed in human amyloid beta knock-in (hAß-KI) females but not males. Correlations to Alzheimer's disease (AD) genotype were stronger for the microbiome than the metabolome. Microbiomes from hAß-KI mice were distinct from 3xTg-AD mice. Cage effects accounted for most of the variance in the microbiome and metabolome.

This study examines online processing and offline judgments of subject-verb person agreement with a focus on how this is impacted by markedness in heritage speakers (HSs) of Italian. To this end, 54 adult HSs living in Germany and 40 homeland Italian speakers completed a self-paced reading task (SPRT) and a grammaticality judgment task (GJT). Markedness was manipulated by probing agreement with both first-person (marked) and third-person (unmarked) subjects. Agreement was manipulated by crossing first-person marked subjects with third-person unmarked verbs and vice versa. Crucially, person violations with 1st person subjects (e.g., io *suona la chitarra "I plays-3rd-person the guitar") yielded significantly shorter RTs in the SPRT and higher accuracy in the GJT than the opposite error type (e.g., il giornalista *esco spesso "the journalist go-1st-person out often"). This effect is consistent with the claim that when the first element in the dependency is marked (first person), the parser generates stronger predictions regarding upcoming agreeing elements. These results nicely align with work from the same populations investigating the impact of morphological markedness on grammatical gender agreement, suggesting that markedness impacts agreement similarly in two distinct grammatical domains and that sensitivity to markedness is more prevalent for HSs.

This study examined the acquisition of numeral classifiers in 120 monolingual Japanese children. Previous research has argued that the complex semantic system underlying classifiers is late acquired. Thus, we set out to determine the age at which Japanese children are able to extend the semantic properties of classifiers to novel items/situations. Participants completed a comprehension task with a mouse-tracking extension and a production task with nonce and familiar items. While the comprehension results showed ceiling effects on familiar and nonce items, age significantly modulated a difference in accuracy between familiar and nonce items in the production task. The findings suggest that the acquisition of the underlying semantic system is acquired much earlier than previously argued. Previously attested issues with Japanese classifier production in young(er) children are more likely to reflect accessing difficulties than indexing the underlying grammatical competence of the classifier system.

Online research methods have the potential to facilitate equitable accessibility to otherwise-expensive research resources, as well as to more diverse populations and language combinations than currently populate our studies. In psycholinguistics specifically, webcam-based eye tracking is emerging as a powerful online tool capable of capturing sentence processing effects in real time. The present paper asks whether webcam-based eye tracking provides the necessary granularity to replicate effects-crucially both large and small-that tracker-based eye tracking has shown. Using the Gorilla Experiment Builder platform, this study set out to replicate two psycholinguistic effects: a robust one, the verb semantic constraint effect, first reported in Altmann and Kamide,  Cognition 73(3), 247-264 (1999), and a smaller one, the lexical interference effect, first examined by Kukona et al. Journal of Experimental Psychology: Learning, Memory, and Cognition, 40(2), 326 (2014). Webcam-based eye tracking was able to replicate both effects, thus showing that its functionality is not limited to large effects. Moreover, the paper also reports two approaches to computing statistical power and discusses the differences in their outputs. Beyond discussing several important methodological, theoretical, and practical implications, we offer some further technical details and advice on how to implement webcam-based eye-tracking studies. We believe that the advent of webcam-based eye tracking, at least in respect of the visual world paradigm, will kickstart a new wave of more diverse studies with more diverse populations.

The present study investigated the qualitative nature of grammatical gender knowledge and processing in heritage speakers (HSs) of Spanish living in the United States. Forty-four adult Spanish HS bilinguals participated, completing a behavioral grammatical gender assignment task and a grammaticality judgment task (GJT) while their brain activity was recorded using electroencephalography (EEG). The EEG GJT task included grammatical and ungrammatical sentences with grammatical gender violations on inanimate nouns, where transparency of the morpho(phono)logical cue and markedness were manipulated. The results of this study revealed that grammatical gender violations elicited the typical P600 effect across all relevant conditions, indicating that the grammatical representations and processing of grammatical gender in HSs are qualitatively similar to those in Spanish-dominant native speakers. Given the experimental manipulation in this study, these findings also suggest that both morphological transparency and markedness play significant roles in how grammatical gender is processed. However, the results of this study differ from those reported in previous studies with Spanish-dominant native speakers, as the P600 effect found was accompanied by a biphasic N400 effect. This pattern of results is interpreted as further evidence that the bilingual experience of HSs modulates certain aspects of morphosyntactic processing, particularly conferring a greater reliance on morphology. Additionally, the results of this study highlight the importance of incorporating neurolinguistic online processing methods to better understand what underlies HS bilingual competence and processing outcomes.

Human oral microbial communities are diverse, with implications for oral and systemic health. Oral microbial communities change over time; thus, it is important to understand how healthy versus dysbiotic oral microbiomes differ, especially within and between families. There is also a need to understand how the oral microbiome composition is changed within an individual including by factors such as environmental tobacco smoke (ETS) exposure, metabolic regulation, inflammation, and antioxidant potential. Using archived saliva samples collected from caregivers and children during a 90-month follow-up assessment in a longitudinal study of child development in the context of rural poverty, we used 16S rRNA gene sequencing to determine the salivary microbiome. A total of 724 saliva samples were available, 448 of which were from caregiver/child dyads, an additional 70 from children and 206 from adults. We compared children's and caregivers' oral microbiomes, performed "stomatotype" analyses, and examined microbial relations with concentrations of salivary markers associated with ETS exposure, metabolic regulation, inflammation, and antioxidant potential (i.e., salivary cotinine, adiponectin, C-reactive protein, and uric acid) assayed from the same biospecimens. Our results indicate that children and caregivers share much of their oral microbiome diversity, but there are distinct differences. Microbiomes from intrafamily individuals are more similar than microbiomes from nonfamily individuals, with child/caregiver dyad explaining 52% of overall microbial variation. Notably, children harbor fewer potential pathogens than caregivers, and participants' microbiomes clustered into two groups, with major differences being driven by Streptococcus spp. Differences in salivary microbiome composition associated with ETS exposure, and taxa associated with salivary analytes representing potential associations between antioxidant potential, metabolic regulation, and the oral microbiome. IMPORTANCE The human oral cavity is a multi-environment habitat that harbors a diversity of microorganisms. This oral microbiome is often transmitted between cohabitating individuals, which may associate oral and systemic health within family members. Furthermore, family social ecology plays a significant role in childhood development, which may be associated with lifelong health outcomes. In this study, we collected saliva from children and their caregivers and used 16S rRNA gene sequencing to characterize their oral microbiomes. We also analyzed salivary biomeasures of environmental tobacco smoke exposure, metabolic regulation, inflammation, and antioxidant potential. We show there are differences in individuals' oral microbiomes mainly due to Streptococcus spp. that family members share much of their microbes, and several bacterial taxa associate with the selected salivary biomeasures. Our results suggest there are large-scale oral microbiome patterns, and there are likely relationships between oral microbiomes and the social ecology of families.

Stereological methods for estimating the 3D particle size and density from 2D projections are essential to many research fields. These methods are, however, prone to errors arising from undetected particle profiles due to sectioning and limited resolution, known as 'lost caps'. A potential solution developed by Keiding, Jensen, and Ranek in 1972, which we refer to as the Keiding model, accounts for lost caps by quantifying the smallest detectable profile in terms of its limiting 'cap angle' (ϕ), a size-independent measure of a particle's distance from the section surface. However, this simple solution has not been widely adopted nor tested. Rather, model-independent design-based stereological methods, which do not explicitly account for lost caps, have come to the fore. Here, we provide the first experimental validation of the Keiding model by comparing the size and density of particles estimated from 2D projections with direct measurement from 3D EM reconstructions of the same tissue. We applied the Keiding model to estimate the size and density of somata, nuclei and vesicles in the cerebellum of mice and rats, where high packing density can be problematic for design-based methods. Our analysis reveals a Gaussian distribution for ϕ rather than a single value. Nevertheless, curve fits of the Keiding model to the 2D diameter distribution accurately estimate the mean ϕ and 3D diameter distribution. While systematic testing using simulations revealed an upper limit to determining ϕ, our analysis shows that estimated ϕ can be used to determine the 3D particle density from the 2D density under a wide range of conditions, and this method is potentially more accurate than minimum-size-based lost-cap corrections and disector methods. Our results show the Keiding model provides an efficient means of accurately estimating the size and density of particles from 2D projections even under conditions of a high density.

RESUMO

The sequence revolution revealed that bacteria-infecting viruses, known as phages, are Earth's most abundant biological entities. Phages have far-reaching impacts on the form and function of microbial communities and play a fundamental role in ecological processes. However, even well into the sequencing revolution, we have only just begun to explore the murky waters around the phage biology iceberg. Many viral reads cannot be assigned to a culturable isolate, and reference databases are biased toward more easily collectible samples, which likely distorts our conclusions. This minireview points out alternatives to mapping reads to reference databases and highlights innovative bioinformatic and experimental approaches that can help us overcome some of the challenges in phage research and better decipher the impact of phages on microbial communities. Moving beyond the identification of novel phages, we highlight phage metabolomics as an important influencer of bacterial host cell physiology and hope to inspire the reader to consider the effects of phages on host metabolism and ecosystems at large. We encourage researchers to report unassigned/unknown sequencing reads and contigs and to continue developing alternative methods to investigate phages within sequence data.

Municipal wastewater provides a representative sample of human fecal waste across a catchment area and contains a wide diversity of microbes. Sequencing wastewater samples provides information about human-associated and medically important microbial populations, and may be useful to assay disease prevalence and antimicrobial resistance (AMR). Here, we present a study in which we used untargeted metatranscriptomic sequencing on RNA extracted from 275 sewage influent samples obtained from eight wastewater treatment plants (WTPs) representing approximately 16 million people in Southern California between August 2020 - August 2021. We characterized bacterial and viral transcripts, assessed metabolic pathway activity, and identified over 2,000 AMR genes/variants across all samples. Because we did not deplete ribosomal RNA, we have a unique window into AMR carried as ribosomal mutants. We show that AMR diversity varied between WTPs (as measured through PERMANOVA, P < 0.001) and that the relative abundance of many individual AMR genes/variants increased over time (as measured with MaAsLin2, Padj < 0.05). Similarly, we detected transcripts mapping to human pathogenic bacteria and viruses suggesting RNA sequencing is a powerful tool for wastewater-based epidemiology and that there are geographical signatures to microbial transcription. We captured the transcription of gene pathways common to bacterial cell processes, including central carbon metabolism, nucleotide synthesis/salvage, and amino acid biosynthesis. We also posit that due to the ubiquity of many viruses and bacteria in wastewater, new biological targets for microbial water quality assessment can be developed. To the best of our knowledge, our study provides the most complete longitudinal metatranscriptomic analysis of a large population's wastewater to date and demonstrates our ability to monitor the presence and activity of microbes in complex samples. By sequencing RNA, we can track the relative abundance of expressed AMR genes/variants and metabolic pathways, increasing our understanding of AMR activity across large human populations and sewer sheds.

Tobamoviruses are agriculturally relevant viruses that cause crop losses and have infected plants in many regions of the world. These viruses are frequently found in municipal wastewater, likely coming from human diet and industrial waste across wastewater catchment areas. As part of a large wastewater-based epidemiology study across Southern California, we analyzed RNA sequence data from 275 influent wastewater samples obtained from eight wastewater treatment plants with a catchment area of approximately 16 million people from July 2020 to August 2021. We assembled 1,083 high-quality genomes, enumerated viral sequencing reads, and detected thousands of single nucleotide variants from eight common tobamoviruses: bell pepper mottle virus, cucumber green mottle mosaic virus, pepper mild mottle virus, tobacco mild green mosaic virus, tomato brown rugose fruit virus, tomato mosaic virus, tomato mottle mosaic virus, and tropical soda apple mosaic virus. We show that single nucleotide variants had amino acid-altering consequences along with synonymous mutations, which represents potential evolution with functional consequences in genomes of these viruses. Our study shows the importance of wastewater sequencing to monitor the genomic diversity of these plant-infecting viruses, and we suggest that our data could be used to continue tracking the genomic variability of such pathogens. IMPORTANCE Diseases caused by viruses in the genus Tobamovirus cause crop losses around the world. As with other viruses, mutation occurring in the virus's genomes can have functional consequences and may alter viral infectivity. Many of these plant-infecting viruses have been found in wastewater, likely coming from human consumption of infected plants and produce. By sequencing RNA extracted from influent wastewater obtained from eight wastewater treatment plants in Southern California, we assembled high-quality viral genomes and detected thousands of single nucleotide variants from eight tobamoviruses. Our study shows that Tobamovirus genomes vary at many positions, which may have important consequences when designing assays for the detection of these viruses by agricultural or environmental scientists.

This study examined potential sources of grammatical gender variability in heritage speakers (HSs) of Italian with a focus on morphological markedness. Fifty-four adult Italian HSs living in Germany and 40 homeland Italian speakers completed an online Self-Paced Reading Task and an offline Grammaticality Judgment Task. Both tasks involved sentences with grammatical and ungrammatical noun-adjective agreement, manipulating markedness. In grammatical sentences, both groups showed a markedness effect: shorter reading times (RTs) and higher accuracy for sentences containing masculine nouns as compared to sentences with feminine nouns. In ungrammatical sentences, although both groups were sensitive to ungrammaticality, only HSs showed a markedness effect, that is, they had significantly longer RTs and higher accuracy when violations were realized on feminine adjectives. Proficiency in the HL was a significant predictor of accuracy and RTs at the individual level. Taken together, results indicate that HSs acquire and process gender in a qualitatively similar way to homeland native speakers. However, RT evidence seems to suggest that at least under particular experimental methods, markedness considerations are more prevalent for HSs resulting in a speed-accuracy tradeoff.

The present study uses EEG time-frequency representations (TFRs) with a Flanker task to investigate if and how individual differences in bilingual language experience modulate neurocognitive outcomes (oscillatory dynamics) in two bilingual group types: late bilinguals (L2 learners) and early bilinguals (heritage speakers-HSs). TFRs were computed for both incongruent and congruent trials. The difference between the two (Flanker effect vis-à-vis cognitive interference) was then (1) compared between the HSs and the L2 learners, (2) modeled as a function of individual differences with bilingual experience within each group separately and (3) probed for its potential (a)symmetry between brain and behavioral data. We found no differences at the behavioral and neural levels for the between-groups comparisons. However, oscillatory dynamics (mainly theta increase and alpha suppression) of inhibition and cognitive control were found to be modulated by individual differences in bilingual language experience, albeit distinctly within each bilingual group. While the results indicate adaptations toward differential brain recruitment in line with bilingual language experience variation overall, this does not manifest uniformly. Rather, earlier versus later onset to bilingualism-the bilingual type-seems to constitute an independent qualifier to how individual differences play out.

The growing interdisciplinary research field of psycholinguistics is in constant need of new and up-to-date tools which will allow researchers to answer complex questions, but also expand on languages other than English, which dominates the field. One type of such tools are picture datasets which provide naming norms for everyday objects. However, existing databases tend to be small in terms of the number of items they include, and have also been normed in a limited number of languages, despite the recent boom in multilingualism research. In this paper we present the Multilingual Picture (Multipic) database, containing naming norms and familiarity scores for 500 coloured pictures, in thirty-two languages or language varieties from around the world. The data was validated with standard methods that have been used for existing picture datasets. This is the first dataset to provide naming norms, and translation equivalents, for such a variety of languages; as such, it will be of particular value to psycholinguists and other interested researchers. The dataset has been made freely available.

Models on L3/Ln acquisition differ with respect to how they envisage degree (holistic vs. selective transfer of the L1, L2 or both) and/or timing (initial stages vs. development) of how the influence of source languages unfolds. This study uses EEG/ERPs to examine these models, bringing together two types of bilinguals: heritage speakers (HSs) (Italian-German, n = 15) compared to adult L2 learners (L1 German, L2 English, n = 28) learning L3/Ln Latin. Participants were trained on a selected Latin lexicon over two sessions and, afterward, on two grammatical properties: case (similar between German and Latin) and adjective-noun order (similar between Italian and Latin). Neurophysiological findings show an N200/N400 deflection for the HSs in case morphology and a P600 effect for the German L2 group in adjectival position. None of the current L3/Ln models predict the observed results, which questions the appropriateness of this methodology. Nevertheless, the results are illustrative of differences in how HSs and L2 learners approach the very initial stages of additional language learning, the implications of which are discussed.

The present study employed a self-paced reading task in conjunction with concurrent acceptability judgements to examine how similar or different English natives and Chinese learners of English are when processing non-local agreement. We also tested how determiner-number specification modulates number agreement computation in both native and non-native processing by manipulating number marking with demonstrative determiners (the versus that/these). Results suggest both groups were sensitive to non-local agreement violations, indexed by longer reading times for sentences containing number violations. Furthermore, we found determiner-number specification facilitated processing of number violations in both native and non-native groups in an acceptability judgement task only, with stronger sensitivity to violations with demonstrative determiners than those with bare determiners. Contrary to some theories that predict qualitative differences between native and non-native processing, we did not find any significant differences between native and non-native speakers, despite the fact that the Chinese speakers of English had to process a novel linguistic feature absent in their native language.

RESUMO

As a result of advances in healthcare, the worldwide average life expectancy is steadily increasing. However, this positive trend has societal and individual costs, not least because greater life expectancy is linked to higher incidence of age-related diseases, such as dementia. Over the past few decades, research has isolated various protective "healthy lifestyle" factors argued to contribute positively to cognitive aging, e.g., healthy diet, physical exercise and occupational attainment. The present article critically reviews neuroscientific evidence for another such factor, i.e., speaking multiple languages. Moreover, with multiple societal stakeholders in mind, we contextualize and stress the importance of the research program that seeks to uncover and understand potential connections between bilingual language experience and cognitive aging trajectories, inclusive of the socio-economic impact it can have. If on the right track, this is an important line of research because bilingualism has the potential to cross-over socio-economic divides to a degree other healthy lifestyle factors currently do not and likely cannot.
