Sechium edule (Jacq.) Sw. (Cucurbitaceae) is a species native to Mexico and Central America. The collection, characterization, and evaluation of accessions maintained in genebanks is essential for the conservation of this species. However, there are no specific varietal descriptors that differ from those used in a phenetic approach and are adapted to international registration guidelines to help distinguish, improve, cluster, and protect intraspecific variants of common use and those obtained by breeding. Therefore, 65 morphological descriptors (qualitative and quantitative) were evaluated in 133 accessions obtained from Mexico, Guatemala, and Costa Rica located in the National Germplasm Bank of S. edule in Mexico. These characteristics were observed to be phenetically stable for five generations under the same agroclimatic conditions. In addition, an analysis of amplified fragment length polymorphism (AFLP) was applied to 133 samples from a set of 245 accessions. According to the multivariate analysis, 26 of the 65 descriptors evaluated (qualitative and quantitative) enabled differentiation of varieties of S. edule. The AFLP analysis showed a high level of polymorphism and genetic distance between cultivated accessions and their corresponding wild ancestor. The variations in S. edule suggest that the morphological characteristics have differentiated from an essentially derived initial edible variety (ancestral original variety), but unlike other cucurbits, there is no evidence of the ancestral edible for Sechium since the seed is unorthodox and there are no relicts.

Cucurbitacins are a class of secondary metabolites initially isolated from the Cucurbitaceae family. They are important for their analgesic, anti-inflammatory, antimicrobial, antiviral, and anticancer biological actions. This review addresses pharmacokinetic parameters recently reported, including absorption, metabolism, distribution, and elimination phases of cucurbitacins. It includes recent studies of the molecular mechanisms of the biological activity of the most studied cucurbitacins and some derivatives, especially their anticancer capacity, to propose the integration of the pharmacokinetic profiles of cucurbitacins and the possibilities of their use. The main botanical genera and species of American origin that have been studied, and others whose chemo taxonomy makes them essential sources for the extraction of these metabolites, are summarized.

Agricultural biodiversity includes many species that have biological variants (natives, ecotypes, races, morphotypes). Their use is restricted to local areas because they do not fulfill the commercial requirements; however, it is well documented that these species are a source of metabolites, proteins, enzymes, and genes. Rescuing and harnessing them through traditional genetic breeding is time-consuming and expensive. Inducing mutagenesis may be a short-time option for its genetic improvement. A review of outstanding research was carried out, in order to become familiar with gene breeding using gamma radiation and its relevance to obtain outstanding agronomic characteristics for underutilized species. An approach was made to the global panorama of the application of gamma radiation in different conventional crop species and in vitro cultivated species, in order to obtain secondary metabolites, as well as molecular tools used for mutation screening. The varied effects of gamma radiation are essentially the result of the individual responses and phenotypic plasticity of each organism. However, even implicit chance can be reduced with specific genetic breeding, environmental adaptation, or conservation objectives.

Exogenous silicon (Si) can enhance plant resistance to various abiotic factors causing osmotic stress. The objective of this research was to evaluate the application of 1 and 2 mM Si to plants under normal conditions and under osmotic stress. Morelos A-98 rice seedlings, were treated with 1 and 2 mM SiO2 for 28 d. Subsequently, half of the plants were subjected to osmotic stress with the addition of 10% polyethylene glycol (PEG) 8000; and continued with the addition of Si (0, 1 and 2 mM SiO2) for both conditions. The application of Si under both conditions increased chlorophyll b in leaves, root volume, as well as fresh and dry biomass of roots. Interestingly, the number of tillers, shoot fresh and dry biomass, shoot water content, concentration of total chlorophyll, chlorophyll a/b ratio, and the concentration of total sugars and proline in shoot increased with the addition of Si under osmotic stress conditions. The addition of Si under normal conditions decreased the concentration of sugars in the roots, K and Mn in roots, and increased the concentration of Fe and Zn in shoots. Therefore, Si can be used as a potent inorganic biostimulant in rice Morelos A-98 since it stimulates plant growth and modulates the concentration of vital biomolecules and essential nutrients.

Hibiscus sabdariffa L. (Hs) calyxes, rich in organic acids, are included in diets in different countries. In recent years, some phytochemicals have been shown to reduce bacterial virulence at sublethal concentrations by interfering with quorum sensing (QS) systems. Therefore, in this study the antivirulence properties of Hs calyxes and two γ-lactones (hibiscus acid [HA] and its methyl ester) in Pseudomonas aeruginosa were analyzed. Acetone and methanol extracts of Hs showed anti-QS activity by inhibiting violacein production (60% to 80% with 250 µg/mL). In molecular docking analysis, the γ-lactones registered a good binding score, which suggests strong interaction with the active site of LasR protein. To verify their effect in vitro, they were isolated from Hs and evaluated in six QS-regulated phenotypes, as well as in ExoU toxin that is released by the type III secretion system (T3SS). At 500 µg/mL they reduced alkaline protease (29-52%) and elastase (15-37%) activity, biofilm formation (∼75%), and swarming (50%), but there was no effect on pyocyanin production, hemolytic activity, or type III secretion. In a mouse abscess/necrosis model, HA at sublethal concentrations (15 and 31.2 µg/mL) affected infection establishment and prevented damage and systemic spread. In conclusion, HA is the first molecule identified with antivirulence properties in Hs with the potential to prevent infections caused by P. aeruginosa.

The Sechium edule Perla Negra cultivar is a recently-obtained biological material whose progenitors are S. edule var. nigrum minor and S. edule var. amarus silvestrys, the latter of which has been reported to have antiproliferative activity against the HeLa P-388 and L-929 cancer cell lines. The present study aimed to determine if the methanolic extract of the fruit of the Perla Negra cultivar had the same biological activity. The methanolic extract was phytochemically characterized by thin layer chromatography (TLC) and column chromatography (CC), identifying the terpenes and flavonoids. The compounds identified via high performance liquid chromatography (HPLC) were Cucurbitacins B, D, E, and I for the terpene fractions, and Rutin, Phlorizidin, Myricetin, Quercetin, Naringenin, Phloretin, Apigenin, and Galangin for the flavonoid fractions). Biological activity was evaluated with different concentrations of the methanolic extract in the HeLa cell line and normal lymphocytes. The methanolic extract inhibited the proliferation of HeLa cells (IC50 1.85 µg·mL-1), but the lymphocytes were affected by the extract (IC50 30.04 µg·mL-1). Some fractions, and the pool of all of them, showed inhibition higher than 80% at a concentration of 2.11 µg·mL-1. Therefore, the biological effect shown by the methanolic extract of the Perla Negra has some specificity in inhibiting tumor cells and not normal cells; an unusual feature among molecules investigated as potential biomedical agents.

The antiproliferative potential of a crude extract from the chayote hybrid H-837-07-GISeM® and its potential for apoptosis induction were assessed in leukaemic cell lines and normal mouse bone marrow mononuclear cells (BM-MNCs). The extract strongly inhibited the proliferation of the P388, J774, and WEHI-3 cell lines (with an IC50 below 1.3 µg·mL(-1)), reduced cell viability, and induced apoptotic body production, phosphatidylserine translocation, and DNA fragmentation. However, the extract had no effect on BM-MNCs. We postulate that these properties make the extract a good candidate for an anti-tumour agent for clinical use.

Se estudió el efecto de la citocinina 6-bencilaminopurina (BAP) en el intercambio de gases y relaciones hídricas de Triticum aestivum L. durante el retraso de la senescencia foliar en invernadero. Plántulas de 21 días después de la siembra (DDS) fueron asperjadas con BAP 0,1mM o agua (testigo) cada 3 días por 20 días. El retraso de senescencia foliar en las plantas tratadas con BAP fue monitoreado mediante cuantificación de clorofila y proteína soluble total. El intercambio de gases se estimó mediante conductancia estomática (gs) y tasa de fijación de CO2. Las relaciones hídricas se evaluaron midiendo el potencial de agua total (YA) y sus componentes, osmótico (Ys) y de turgencia (Yt), cada 5 días por 20 días. Las hojas con BAP mostraron mayor concentración de clorofila y proteína total que el control tras 26 DDS, diferencias que aumentaron con el tiempo y a los 41 DDS fueron 7 veces mayores que el control. La gs registró valores no significativamente diferentes al estado verde pre-senescente. Estas respuestas se asociaron con tasas mayores de fijación de CO2, con concentraciones cercanas a las hojas verdes pre-senescentes (6,2μmol·CO2·m-²·s-¹). El YA se mantuvo constante bajo BAP y Ys fue significativamente menor que en los testigos, favoreciendo la retención de solutos en el citoplasma de hojas con retraso de senescencia. El Yt de las hojas tratadas fue mayor que su respectivo testigo de 31 a 41 DDS. Por tanto, la citocinina BAP promovió el mantenimiento de la tasa de fijación de CO2 y las relaciones hídricas durante el retraso de la senescencia foliar.

El potencial osmótico, el balance iónico y las relaciones nutrientes del medio de cultivo son determinantes en el medio de crecimiento y distribución de nutrientes de plantas cultivadas in vitro; sin embargo, en la mayoría de las investigaciones los primeros no se consideran. Se evaluó el efecto de diferentes potenciales osmóticos de las sales (-0,106; -0,113; -0,120; -0,120; -0,128; -0,143 y -0,157Pa) y del contenido de Ca2 en el medio de cultivo sobre la distribución de Ca2+ y K+, y el peso de biomasa seca de explantes de vid. La concentración iónca de los explantes se midió con un microscopio electrónico de barrido de bajo vació (MEB-LV). El mayor potencial osmótico del medio de cultivo (-0,106MPa) favoreció la translocación de Ca2+ y K+ de los tallos hacia las hojas, mientras que, el potencial más negativo (-0157MPa) permitió que los mayores contenidos se acumularan el tallos. El amyor peso de biomasa seca (57mg) y los menores porcentajes de explantes con síntomas de deficiencia de Ca+ (20 por ciento) se obtuvieron con el potencial osmótico de -0,106MPa y con concentración de Ca+ en el medio de cultivo de 1,5mM
