Bemisia tabaci Gennadius (Homoptera: Aleyrodidae) is the most devastating insect-pest in cotton crop. It is vector of the cotton leaf curl virus (CLCV) and is responsible for huge losses to cotton industry in Pakistan and worldwide. It is mainly controlled by insecticides but the injudicious use of insecticides has resulted in insecticide resistance and population resurgence in addition to various harmful effects on the humans, non-target organisms and the environment. Transgenerational studies are very helpful to choose a best insecticidal option. In the current study, age-stage two-sex life table analysis was used to identify transgenerational effects of sublethal doses of afidopyropen. The adults of B. tabaci were treated with three concentrations of afidopyropen i.e., LC10, LC30 and LC50. The results indicated significant changes in the progeny i.e. the fecundity decreased in treated population; and female and male longevity of their progeny were more in control as compared to treated populations. Similarly, population parameters like intrinsic rate of growth (r), net reproductive rate (R0) and limiting rate of growth (λ) were significantly decreased in the treated adult progeny with values of 0.08-0.11, 4.85-7.46 and 1.09-1.12 per day, respectively. Based on the reduced biotic potential, afidopyropen can be suggested as an effective alternative option for the management of B. tabaci.

Background: Mosquito Control Programs are articulated to control Mosquito Borne Diseases and success of such programs depends on the activities of field workers, and their adherence to the standard operating procedures (SOP's) is governed by their knowledge, attitudes, and practices (KAP). Present study was intended to assess the KAP of mosquito control workers of Pakistan to get an exact depiction of prevailing situation. Methods: A cross-sectional descriptive study was conducted in March-April 2020. Questionnaire containing 30 closed and open-ended questions were administrated to participants. Knowledge and practices were evaluated using a scoring system i.e., by giving 1 point to each correct answer while attitude questions were analyzed individually and expressed in percentage for each response. Results: Total 639 workers were interviewed, mean age was 29.8 (SD ±7.87) years, majority (65.1%) was in age group of 18-30 years. Mean knowledge score was 6.96±1.28 (range 0-9) with 77.36% correct answers (P= 0.073). Mean practices score was 7.00±1.62 (range 2-9) with 77.83% appropriate answers (P< 0.001). Both knowledge and practices scores were higher for permanent employees, practices score increased with increase in job experience. Very weak positive correlation (r= 0.127) was observed between knowledge and practice scores. Conclusion: Appropriate practice correlates with better knowledge and positive attitude towards control activities. Hence, training on protection and protective measures for having a positive attitude among healthcare workers is necessary against the fight with mosquitoes.

BACKGROUND: Under the family Tephritidae, Bactrocera zonata (Saunders) is a serious pest, attacking fruits and vegetables causing large quantitative and qualitative damages throughout the world. Fruit flies require proteinaceous food for sexual maturation and egg development. Therefore, food bait attractants are frequently utilized for fruit fly detection, monitoring, mass trapping, and control. Using a Y-shape olfactometer (behavioral tests), we selected the best synthetic proteinaceous food bait attractants to volatiles identified by fruit fly antennae. The responses of B. zonata adults, male and female, to some ammonium compounds (ammonium acetate (AA), trimethylamine (TMA), and putrescine) that were mixed with certain food attractants were evaluated under laboratory conditions. Using flies ranging in age from 5 to 30 days, possible mixtures were discovered that might be useful in developing fruit fly attractants for both males and females. So, four base baits were developed by mixing protein hydrolysate with jaggery, papaya powder, kachri powder, potassium hydroxide (KOH), and guava pulp. Finally, thirty-two (32) synthetic blends were developed when the above four base baits were mixed with synthetic attractants. RESULTS: The olfactometer bioassay indicated that protein hydrolysate and jaggery-based baits were effective in attracting both male and female flies throughout their adult lives when combined with AA + putrescine (Bait 6) and AA + TMA + putrescine (Bait 8). Similarly, protein hydrolysate + guava pulp-based baits combined with AA + putrescine (Bait 6) and AA + TMA + putrescine (Bait 8) was effective in attracting both male and female flies from 5 to 30 days of age. The pH of all 32 synthetic blends was measured and ranged from 4.77 to 11.35. CONCLUSIONS: According to our observation, the variation in pH may be due to differences in chemical composition between the attractants and food constituents. The pH of protein bait attractants may be an important factor in the attraction efficiency of B. zonata males and females.

The indiscriminate application of synthetic pesticides on horticultural plants for the management of Bactrocera zonata has both economic and environmental implications due to the biomagnification of harmful residues in humans via the food chain. This necessitates the use of some eco-friendly control measures such as insect growth regulators (IGRs) as an alternative. A laboratory experiment was laid out to determine the potential chemosterilant effect of five IGRs including pyriproxyfen, novaluron, lufenuron, buprofezin, and flubendiamide at six different concentrations on B. zonata after treating adult diet. Using the oral bioassay, B. zonata were fed on IGRs impregnated diet (50-300 ppm/5 mL diet) which was replaced with the normal diet after 24 h of feeding. Ten pairs of B. zonata were placed in a separate plastic cage accommodating an ovipositor attractant guava for eggs collection and calculation. An analysis of the result revealed that fecundity and hatchability were higher at a low dose and vice versa. Lufenuron at 300 ppm/5 mL of diet significantly decreased the fecundity rate (31.1%) as compared to pyriproxyfen, novaluron, buprofezin and flubendiamide, which had 39.3%, 39.3%, 43.8%, and 47.5%, respectively. The lowest hatchability (19.9%) was noted in lufenuron treated diet followed by pyriproxyfen, novaluron, buprofezin, and flubendiamide, which had 22.1%, 25.0%, 30.9%, and 31.6%, respectively. Furthermore, in a population of crosses between the lufenuron treated male and female, a significant decrease in fecundity (45.5%) and hatchability (51.7%) was noted as compared to other IGRs. Overall, this study identified the chemosterilant potential of lufenuron on the population of B. zonata which can be integrated for its management strategy.

Selenium (Se) is an essential nutrient for both plants and animals and is usually provided as a supplement to livestock. Se bioaccumulation promotes plant growth by enhancing the accumulation of organic solutes and the activation of antioxidant system. In animals, the Se supplements reduce the chances of mastitis and white muscle disease, and improve the immunity, health, and reproduction, particularly in lactating dairy cows. Therefore, the enrichment or biofortification of fodder crops with Se may improve the nutritional quality of forages and thereby reduce malnutrition in cattle. However, Se hyper-accumulation in plants or plant parts can cause direct toxic effects on insects especially bees. Berseem is a highly cross-pollinated fodder crop that attracts a large number of pollinators. However, little or no reports are available regarding the effects of Se biofortification on the foraging behavior of pollinators in berseem. Therefore, the current study was planned to evaluate the effect of exogenous application of Se on the foraging behavior of native pollinators visiting the berseem crop. Five different doses of Se were applied to evaluate its effect on abundance, foraging behavior (visit duration and visitation rate), and single-visit efficacy of native pollinators that may affect berseem seed yield. Our results showed maximum abundance of pollinators in plants supplemented with low Se level, i.e., 5 g ha-1 while the minimum abundance was observed at high Se doses (15 and 20 g ha-1). Also, the seed yield attributes, i.e., the number of seeds per head, seed weight per head, 1000 seed weight, were the highest in plants treated with a low dose of Se, whereas the seed yield of berseem decreased with an increase in Se concentrations. Hence, our study provides evidence that high doses of Se negatively affect the foraging behavior of pollinators (visitation rate and visit duration) in plants. We conclude that the application of moderate Se dose positively influences the pollination ecology of berseem, consequently improving seed yield. HIGHLIGHTS: Selenium (Se) is essential for animals and beneficial for plants and may become toxic at high level. Se is delivered to the environment due to agriculture. Se toxicity affected berseem growth and considerably reduced the seed yield. High dose of Se reduced the abundance of pollinators and negatively affected their foraging behavior.

The hormetic effect may cause disease control measures to fail due to inadequate treatment of human disease vectors such as houseflies. Age-stage, two-sex life table is used for accurate estimation of the hermetic impacts on insects as it allows to study sub-lethal or transgenerational effects. Pyrethroids insecticides are primarily used for the management of houseflies. This study used lambda-cyhalothrin (a pyrethroid insecticide) to quantify its transgenerational impacts on houseflies. Life table parameters of a progeny of adult houseflies exposed to LC10, LC30, and LC50 of lambda-cyhalothrin were computed. Statistically higher fecundity (71.31 per female) was observed in control treatment, while it was the adults exposed to LC50 recorded the lowest progeny. Significantly higher values for intrinsic rate of growth (r), limiting rate of growth (λ), and net reproductive rate (Ro) (0.16, 1.16, and 31.38 per day, respectively) were recorded for the control treatment of the study. Contrarily, lower values for λ, Ro, and r were (0.10, 1.10, and 9.24 per day, respectively) were noted in the LC50 treatment. Decreased population parameters suggest that lambda-cyhalothrin can be successfully used in indoor environments to control houseflies.

Housefly, Musca domestica L. (Diptera: Muscidae), is an important insect-pest owing to its disease transmitting capability in both humans and animals e.g. diarrhea, gastroenteritis diseases, lumpy skin disease, cholera. Residual insecticides are widely used to control houseflies that pose serious threat to human and animal health as well as to the environment. The population resurgence after insecticidal spray is also a common problem in many cases. Transgenerational studies can be very helpful to see the fate of insecticidal sprays. In the current study, we determined transgenerational effects of sublethal doses of diafenthiuron using Age-stage two-sex life table analysis, while focusing on potential use of diafenthiuron in baits. After treating adult flies with three different doses of diafenthiuron viz. LC10, LC20 and LC50, the progeny depicted significant changes regarding pre-adult duration (shorter in control), male and female longevity of their progeny were observed (more adult longevity in control). Additionally, population growth parameters viz. intrinsic rate of growth (r) (-0.03-0.12 per day), net reproductive rate (R0) (0.50-9.98), limiting rate of growth (λ) (0.96-1.13 per day) were also significantly reduced in the progeny of treated adults. Based on the decreased biotic potential i.e. intrinsic rate of growth and other population parameters, diafenthiuron can be recommended as an effective insecticide even at lower doses.

Although Pakistan has rich biodiversity, many groups are poorly known, particularly insects. To address this gap, we employed DNA barcoding to survey its insect diversity. Specimens obtained through diverse collecting methods at 1,858 sites across Pakistan from 2010-2019 were examined for sequence variation in the 658 bp barcode region of the cytochrome c oxidase 1 (COI) gene. Sequences from nearly 49,000 specimens were assigned to 6,590 Barcode Index Numbers (BINs), a proxy for species, and most (88%) also possessed a representative image on the Barcode of Life Data System (BOLD). By coupling morphological inspections with barcode matches on BOLD, every BIN was assigned to an order (19) and most (99.8%) were placed to a family (362). However, just 40% of the BINs were assigned to a genus (1,375) and 21% to a species (1,364). Five orders (Coleoptera, Diptera, Hemiptera, Hymenoptera, Lepidoptera) accounted for 92% of the specimens and BINs. More than half of the BINs (59%) are so far only known from Pakistan, but others have also been reported from Bangladesh (13%), India (12%), and China (8%). Representing the first DNA barcode survey of the insect fauna in any South Asian country, this study provides the foundation for a complete inventory of the insect fauna in Pakistan while also contributing to the global DNA barcode reference library.

Resistance management is very important for devising control strategies of polyphagous insect-pests like Helicoverpa armigera Hübner (Lepidoptera: Noctuidae). Considering the importance of resistance management, demographic features of selected and unselected populations of H. armigera were studied in 6 different treatments viz. emamectin benzoate, Helicoverpa armigera Nucleopolyhedrosis Virus (HaNPV), emamectin benzoate+HaNPV, spinetoram, spinetoram+HaNPV and control. Higher values for fecundity, intrinsic rate, the finite rate of increase (λ) were recorded in the control of selected as compared to the rest of treatment. Similarly, higher values for these population parameters viz. oviposition days, fecundity, intrinsic rate, the finite rate of increase were calculated in the unselected control. Similarly, net reproductive rate (R0) for selected and unselected control was higher as compared to the rest of the treatments. It may happen because these kinds of selection pressures can result in decreased fitness of the test insect thus decreased fitness of H. armigera in different treatments was observed as compared to the control. Additionally, quicker development of susceptible insects was observed because susceptible insects were growing without any stressor (xenobiotics) as compared to the rest which contributed to their faster development.

Lucerne (Medicago sativa L.) is a cross-pollinated crop and requires entomophilous pollination for tripping of flowers and subsequent pod and seed set. To discover the best pollinators for lucerne seed production, a two-year field trial was carried out at the research farm of MNS University of Agriculture, Multan, Pakistan. Abundance and diversity of insect pollinators along with the foraging behavior were recorded in terms of tripping efficiency, stay time, visitation rate and pollen harvest. The single-visit efficiency of abundant insect pollinators was also evaluated in terms of number of seeds and seed weight per raceme along with germination percentage. Ten most abundant floral visitors (five solitary bee species, three honeybee species and two syrphid fly species) were tested for their pollination efficiency. Honeybees were most abundant in both the years followed by the solitary bees and syrphid flies. Single-visit efficacy in terms of number of pods per raceme, number of seeds per raceme, 1000 seed weight and germination percentage revealed Megachile cephalotes as the most efficient insect pollinator followed by Megachile hera and Amegilla sp. Future studies should investigate the biology and ecology of these bee species with special emphasis on their nesting behavior and seasonality.

Termites are a significant pest of buildings, agriculture, and trees, and are mainly controlled by baiting. However, baiting systems are available for only lower termites (Rhinotermitidae) not for higher termites (Termitidae). Termite foraging behavior associated with baiting systems varies among species and families, and plays a significant role in baiting success. Here, foraging behavior of Odontotermes obesus (Blattodea: Termitidae: Macrotermitinae), a fungus-growing higher termite, was investigated relative to three bait-station sizes (small, medium, and large) containing different quantities of food. Significantly more workers recruited to large stations (470/station) compared to medium (246/station) and small (124/station) stations. Abundance of O. obesus in large and medium stations significantly positively correlated with relative humidity whereas negative but non-significant correlations were observed with temperature in large and medium stations. Total and continuous contacts with the stations increased with time and were greater in large stations. Station abandonment due to disturbance was significantly less in large stations (3%) followed by medium (9%) and small stations (20%). Our results suggest that large stations (≈8 litres volume) work best for population management of O. obesus and other related fungus-growing higher termites.

Green peach aphid [Myzus persicae (Sulzer) (Hemiptera: Aphididae)] is a significant pest with a known history of insecticide resistance. Neonicotinoids could manage this pest; however, their frequent use led to the evolution of resistance in field populations of M. persicae. Toxicity data for neonicotinoid insecticides synergized with pipernyl butoxide (PBO) in a field population (FP) were collected and compared to a laboratory susceptible clone (SC) of aphids. The enhanced expression of metabolic resistance-related cytochrome P450 gene CYP6CY3 and an arginine-threonine substitution were detected in FP, causing a single point mutation (R81T) at ß1 subunit of nicotinic acetylcholine receptor (nAChR) within D loop. High level of resistance to imidacloprid was developed in FP with 101-fold resistance ratio and moderate resistance level (10.9-fold) to acetamiprid. The results of PBO synergized bioassay suggested that cytochrome P450 enzymes were involved in the resistance to neonicotinoids. The mRNA transcriptional level of CYP6CY3 gene was significantly higher (3.74 fold) in FP compared to SC. The R81T mutation associated with neonicotinoid resistance had 26% resistant allele frequency in FP. Both P450 enzymes and R81T mutation of nAChR were found in field-evolved neonicotinoid resistance. It is concluded that field-evolved resistance in green peach aphid could be managed by using appropriate synergists such as PBO.

American bolloworm, Helicoverpa armigera Hubner (Noctuidae: Lepidoptera) is considered as a major pest of various crops all over the world. It is mainly controlled by indiscriminate use of synthetic insecticides in the world due to which this pest developed resistance to most of the available insecticides. Therefore, in the current study, the efficacy of virulent strain of HaNPV (0.5 × 109 PIB/ml) alone and in combination with recommended doses of spintoram (20 g/100 L of water) and emamectin benzoate (200 ml/100 L of water) was tested in field. The combination of HaNPV with spintoram and emamectin benzoate 100% reduced the larval population as compared to emamectin benzoate and HaNPV alone. This suggested that the combination of spintoram and emamectin benzoate with HaNPV could be used in field to manage the infestation of H. armigera.

Nucleopolyhedrosis viruses can be utilized for effective management of agriculture pests. Their efficacy can be increased if they are mixed with certain insecticides. In the current study, HaNPV was mixed with two insecticides: spinetoram and emamectin benzoate in various combinations and applied to larvae of H. armigera in laboratory conditions. There were a total of 15 combinations of HaNPV with each of the two insecticides in addition to five doses of HaNPV and three doses of insecticides alone. The synergistic and antagonistic effects of combinations were explored. The results revealed that there was synergistic effect of HaNPV @ 0.5 × 109 PIB/ml × Spinetoram @ 40, 20, 10 ml/100 L of water. In case of emamectin benzoate, synergistic effects were recorded at 1 × 109 PIB/ml HaNPV × emamectin benzoate @ 100 ml/100 L of water. However, 0.5 × 109 PIB/ml HaNPV has synergistic effects with all three doses of emamectin benzoate. The results suggested that HaNPV can be used in combination with spinetoram and emamectin benzoate for the management of resistant population of H. armigera.

Red palm weevil (Rhynchophorus ferrugineus) is a voracious pest of date palm worldwide. Pakistan ranks sixth in date palm production globally. Losses to date palm plantations in Pakistan sometimes surpass 10%-20%. Most of the traditional management strategies used by farmers have been found insignificant to combat this voracious pest. The entomopathogenic fungi, Beauveria bassiana [QA-3(L) and QA-3(H)] and insecticides, Nitenpyram (Active 10% SL) [NIT (L) and NIT (H)] were applied to larval (2nd, 4th, and 6th), pupal and adult stages of R. ferrugienus. Integration or alone application of fungi with insecticides at different concentration under laboratory conditions. Combined application was depicted additive and synergistic interactions. Contrarily, highest cumulative mortality (100%) was recorded in 2nd instar larvae as compared to later instar larvae at combined application. The maximum pupal and adult mortality remained 89% and 66% respectively after treatment with [QA-3 (H) + NIT (L)]. The combination of B. bassiana at higher concentration whereas Nitenpyram at lower dose was found more lethal to larvae, pupae and adults of R. ferrugineus. This signifies the need of combining B. bassiana and bio-rational insecticides that can reduce the cost of management with least harm to environment and natural enemies.

Helicoverpa armigera Hübner (Lepidoptera: Noctuidae) is serious pests of cotton and several other crops. Helicoverpa armigera Nucleopolyhedrovirus (HaNPV) can be important alternative to synthetic insecticides for the management of H. armigera. However, the efficacy of HaNPV can vary in horizontal and vertical transmission. In the current study, we evaluated the efficacy of HaNPV of a virulent strain (vertically transmitted up to six generations) and wild strains (used after isolation from the field infected larvae). Both strains were applied to the 2nd instar larvae of H. armigera @ 1 × 109 polyhedral inclusion bodies (PIB)/ml. There were six replications of each strain (strains). The results indicated higher mortalities in larvae exposed to virulent strains (68.33 ±â€¯6.07%) as compared to wild strain (45 ±â€¯2.24%). Virulent strains killed the larvae quite faster than wild strain. The lethal time (LT50) to kill 50% of the larvae by virulent strain was 7.15 days and for wild strain it was 19.47 days. The results showed that multiple passage of HaNPV through several generations enhances its efficacy to kill H. armigera larvae faster. The results of this study will be helpful to manage H. armigera and other related lepidopoterous pests.

Soils contain one of the largest carbon (C) pools in the biosphere with the greatest potential of C sequestration to mitigate climate change impacts. The present study aimed at comparing C sequestration potential of various land use systems including forestlands, croplands, agroforests, and orchards in the arid region of Pakistan. Soil samples from the layers of 0-20, 20-40, 40-60, and 60-80 cm depths were collected and analysed for soil physico-chemical properties namely texture, pH, EC, NPK-soil, organic matter (SOM), and soil organic C (SOC). Additionally, the above and below ground plant biomass and C contents were estimated. Results revealed that the highest C sequestration potential (64.54 Mg ha-1) was in the above ground biomass of forest land and the lowest (33.50 Mg ha-1) in cropland. The below ground plant biomass at 0-20 cm soil depth was 14.09, 12.38, 11.78, 11.76, and 10.92 Mg ha-1 for forest land, mango orchards, agroforests, citrus orchards and cropland, respectively. The respective values in case of total C content were, 6.84, 6.79, 6.10, 5.69 Mg ha-1. Irrespective to the soil depth, below ground biomass and total C followed the order: forest land > mango orchard > citrus orchard > agroforests > crop lands. It is concluded that the forest land have greater potential for C sequestration than the other land use systems studied in the arid region of Pakistan. Therefore, in order to cope up with climate change disasters in Pakistan the massive reforestation project - named the Billion Tree Tsunami of the Government of Pakistan will prove beneficial.

Storing grains remain vulnerable to insect pest attack. The present study developed a biopesticide using biomolecules and their encapsulation in nanoparticles. A 25 kDa cysteine protease extracted from seeds of Albizia procera (ApCP) was encapsulated in graphene quantum dots (GQDs). The insecticidal activity of ApCP, with or without GQDs, against two stored grain insect pests, Tribolium castaneum (Herbst) and Rhyzopertha dominica (Fabricius) was explored. Insects were exposed to three concentrations 7.0, 3.5 and 1.7 mg of ApCP per a gram of wheat flour and grains. The insecticidal activity of ApCP encapsulated with GQDs was improved compared to that of ApCP without GQDs for both insect pests. The number of eggs and larvae of T. castaneum was reduced by 49% and 86%, respectively. Larval mortality was increased to 72%, and adult eclosion of T. castaneum was reduced by 98% at a 7.0 mg/g concentration of ApCP with GQDs compared to that of ApCP without GQDs. Exposure to 7.0 mg/g ApCP with GQDs, the number of R. dominica eggs and larvae was reduced by 72% and 92% respectively, larval mortality was increased by 90%, and eclosion was reduced by 97%. The extraction, purification, characterization, quantification and encapsulation of ApCP with GQDs were also studied. Cysteine protease nanocarriers have the potential to control stored grain insect pests.

Plutella xylostella L. (Lepidoptera: Plutellidae) is an important pest causing significant losses to vegetables worldwide. Insecticides resistance in P. xylostella is a serious issue for scientists since last 30 years. However, deltamethrin and Bt Cry1Ac are commonly used insecticides against P. xylostella but studies involving development of resistance in P. xylostella against these two insecticides at different temperatures are lacking. The current study was aimed to find out the toxicity of deltamethrin and Bt Cry1Ac, and resistance development in P. xylostella. Results showed that the positive correlation between the temperature and toxicities of deltamethrin and Bt Cry1Ac. The results indicated -0.051, -0.049, -0.047, and -0.046 folds of deltamethrin resistance at 15 °C, 20 °C, 25 °C, and 30 °C temperatures, respectively from 1st to 12th generations. The toxicity of Bt Cry1Ac after 24 h was 2.2 and 4.8 folds on 1st generation at 20 °C and 25 °C temperatures, respectively compared to the toxicity recorded at 15 °C (non-overlapping of 95% confidence limits). Based on the results of this study, it is concluded that the temperature has a positive correlation with the toxicity of deltamethrin and Bt Cry1Ac against the larvae of P. xylostella. This study suggests that deltamethrin and Bt Cry1Ac can be included in the management program of P. xylostella on many vegetable crops. The baseline susceptibility data might be helpful to understand the resistance mechanisms in P. xylostella.

RESUMO

In developed and underdeveloped countries, ticks are important vectors transmitting various pathogens that cause diseases of veterinary and public health importance, like babesiosis, theileriosis, Crimean­Congo haemorrhagic fever (CCHF) and many more. Many species of ticks have been reported in scientific literature from Pakistan, which need to be listed for ready reference. For this purpose, a checklist of tick species recorded in Pakistan is presented here after comprehensive review of the available literature on the subject. Overall, nine genera and 53 species of ticks infesting animals in Pakistan are presented in this checklist.
