Abstract The validation of many anuran species is based on a strictly descriptive, morphological analysis of a small number of specimens with a limited geographic distribution. The Scinax Wagler, 1830 genus is a controversial group with many doubtful taxa and taxonomic uncertainties, due a high number of cryptic species. One example is the pair of species Scinax constrictus and Scinax nebulosus, which share a similar morphology. Scinax constrictus is restricted to the Brazilian Cerrado savanna, while S. nebulosus is widely distributed throughout northern South America. Despite the validation of many anuran species, discriminations based only on morphological traits is quite difficult due to the high conservative morphology of some groups. In this context, the present study uses mitochondrial and nuclear genes to provide a more consistent diagnosis and test the validity of S. constrictus as a distinct species from S. nebulosus, as well as evaluate the position of these taxa within the Scinax genus. The topologies obtained herein uphold the monophyletic status of Scinax based on all molecular markers assessed in this study, in all analytical approaches, with high levels of statistical support.


Resumo A validação de muitas espécies de anuros é baseada em uma análise morfológica e descritiva de um pequeno número de espécimes com uma distribuição geográfica limitada. O gênero Scinax Wagler, 1830 é um grupo controverso com muitos táxons duvidosos e incertezas taxonômicas devido ao grande número de espécies crípticas. Um exemplo são as espécies, Scinax constrictus e Scinax nebulosus, que compartilham uma morfologia similar. Scinax constrictus é restrito à savana do Cerrado brasileiro, enquanto S. nebulosus é amplamente distribuído pelo norte da América do Sul. Apesar da validação de muitas espécies de anuros, a discriminação baseada apenas em características morfológicas é bastante difícil, devido à alta morfologia conservadora de alguns grupos. Neste contexto, o presente estudo utiliza genes mitocondriais e nucleares para fornecer um diagnóstico mais consistente e para testar a validade de S. constrictus como uma espécie distinta de S. nebulosus, bem como avaliar a posição destes táxons dentro do gênero Scinax. As topologias obtidas confirmaram o status monofilético de Scinax com base em todos os marcadores moleculares, em todas as abordagens analíticas, com altos níveis de suporte estatístico.

INTRODUCTION: Insulinomas are a rare type of pancreatic neuroendocrine tumours characterized by insulin hypersecretion. They are considered malignant when metastases are present. Traditional therapies often promote only temporarily symptomatic relief and may be associated with severe adverse effects. There is scarce experience in treating malignant insulinomas with peptide receptors radionuclide therapy (PRRNT). PATIENTS AND METHODS: We describe PRRNT results in four patients with inoperable malignant insulinomas with poorly controllable hypoglycaemia. All patients received therapy with 177Lu-DOTA-TATE after conventional therapies failed in controlling disease progression and symptoms. The activity administered per cycle was 4.8-7.4 GBq. The interval between cycles was 10-16 weeks. Haematology, liver and kidney function tests were performed before treatment initiation and 5 and 10 weeks after each cycle. RESULTS: Patient 1 presented significant clinical benefit for 13 months after PRRNT, with imaging improvement. Patient 2 obtained reduction of the number and severity of hypoglycaemic episodes during 15 months after therapy. Patient 3 is asymptomatic since PRRNT first cycle performed 23 months ago and revealed significant imaging improvement. Patient 4 had resolution of hypoglycaemia only 3 days after PRRNT first cycle and today, 16 months after therapy, the disease seems to be in remission and the patient maintains euglycaemic state. PRRNT was well tolerated, with only hematologic grade 2 toxicity in patient 1 and mild kidney toxicity in patient 3. CONCLUSIONS: After the start of 177Lu-DOTA-TATE all patients achieved hypoglycaemia symptomatic control and had evident improvement of their quality of life. Three patients showed imagiological improvement suggesting reduced tumour load.

Undernutrition represents a major public health challenge for middle- and low-income countries. This study aimed to evaluate whether a multideficient Northeast Brazil regional basic diet (RBD) induces acute morphological and functional changes in the ileum of mice. Swiss mice (∼25 g) were allocated into two groups: i) control mice were fed a standard diet and II) undernourished mice were fed the RBD. After 7 days, mice were killed and the ileum collected for evaluation of electrophysiological parameters (Ussing chambers), transcription (RT-qPCR) and protein expression (western blotting) of intestinal transporters and tight junctions. Body weight gain was significantly decreased in the undernourished group, which also showed decreased crypt depth but no alterations in villus height. Electrophysiology measurements showed a reduced basal short circuit current (Isc) in the undernourished group, with no differences in transepithelial resistance. Specific substrate-evoked Isc related to affinity and efficacy (glutamine and alanyl-glutamine) were not different between groups, except for the maximum Isc (efficacy) induced by glucose. Transcription of Sglt1 and Pept1 was significantly higher in the undernourished group, while SN-2 transcription was decreased. No changes were found in transcription of CAT-1 and CFTR, while claudin-2 and occludin transcriptions were significantly increased in the undernourished group. Despite mRNA changes, SGLT-1, PEPT-1, claudin-2 and occludin protein expression showed no difference between groups. These results demonstrate early effects of the RBD on mice, which include reduced body weight and crypt depth in the absence of significant alterations to villus morphology, intestinal transporters and tight junction expression.

The red piranha, Pygocentrus nattereri, is an important resource for artisanal and commercial fisheries. The present study determines the genetic differentiation among P. nattereri populations from the northeastern Brazilian state of Maranhão. The DNA was isolated using a standard phenol-chloroform protocol and the Control Region was amplified by PCR. The PCR products were sequenced using the didesoxyterminal method. A sequence of 1039 bps was obtained from the Control Region of 60 specimens, which presented 33 polymorphic sites, 41 haplotypes, Ò» =0.978 and π =0.009. The neutrality tests (D and Fs) were significant (P < 0.05) for most of the populations analyzed. The AMOVA indicated that most of the molecular variation (72%) arises between groups. The fixation index was highly significant (FST = 0.707, P < 0.00001). The phylogenetic analyses indicated that the specimens represented a monophyletic group. Genetic distances between populations varied from 0.8% to 1.9%, and were <0.5% within populations. The degree of genetic differentiation found among the stocks of P. nattereri indicates the need for the development of independent management plans for the different river basins in order to preserve the genetic variability of their populations.

Eucalyptus plantations are frequently used for the establishment of bee yards. This study was carried on at Fazenda Brejão, northwestern region of the State of Minas Gerais, Brazil. This farm is covered both with native Cerrado vegetation (Brazilian savanna) and eucalyptus plantations. This paper reports on the botanic origin of pollen pellets and honey collected from honeybee (Apis mellifera) hives along a thirteen-month period (January 2004 to January 2005). The most frequent pollen types found in the pollen pellets during the rainy season were Trema micrantha (Ulmaceae), Copaifera langsdorffii (Fabaceae), an unidentified Poaceae, unidentified Asteraceae-2, Cecropia sp. 1 (Cecropiaceae) and Eucalyptus spp. (Myrtaceae); during the dry season the most frequent pollen types were Acosmium dasycarpum (Fabaceae), Cecropia sp. 1 (Cecropiaceae) and Eucalyptus spp. (Myrtaceae). Pollen grains of Baccharis sp. (Asteraceae), Cecropia sp. 1 (Cecropiaceae), Copaifera langsdorffii (Fabaceae), Mimosa nuda (Fabaceae), Eucalyptus spp. (Myrtaceae) and Trema micrantha (Ulmaceae) were present in the honey samples throughout the study period.

Resumo A piranha vermelha, Pygocentrus nattereri, é um recurso importante para pesca artesanal e comercial. O presente estudo determinou a diferenciação genética entre populações de P. nattereri no nordeste do estado brasileiro do Maranhão. O DNA foi isolado utilizando o protocolo de Fenol-clorofórmio e a Região Controle foi amplificada por PCR. Os produtos da PCR foram sequenciados usando o método didesoxiterminal. Uma sequência de 1039 pbs foi obtida da Região Controle de 60 espécimes, que apresentaram 33 sítios polimórficos, 41 haplótipos, һ= 0.978 e π= 0.009. Os testes de neutralidade (D and Fs) foram significativos (P < 0.05) para a maioria das populações analisadas. A AMOVA indicou que a maior parte da variação molecular (72%) surge entre os grupos. O índice de fixação foi altamente significativo (FST = 0.707, P = < 0.00001). As análises filogenéticas indicaram que os espécimes representam um grupo monofilético. Distâncias genéticas entre as populações variaram de 0.8% a 1.9%, e de <0.5% dentro das populações. O grau de diferenciação genética encontrada entre os estoques de P. nattereri indicam a necessidade para o desenvolvimento de planos de manejo independentes para as diferentes bacias hidrográficas, a fim de preservar a variabilidade genética dessas populações.

Resumo Plantações de Eucalyptus são, frequentemente, utilizadas como locais de instalação para colmeias. Este estudo foi realizado na Fazenda Brejão, localizada no noroeste de Minas Gerais, Brasil. Esta fazenda é coberta por Cerrado nativo (savana brasileira) e por plantações de eucaliptos. Este trabalho indica a origem botânica de bolotas e mel coletados em colmeias de Apis mellifera por um período de 13 meses (Janeiro/2004 a janeiro/2005). Os tipos polínicos mais frequentes nas amostras de pólen durante a estação chuvosa foram Trema micrantha (Ulmaceae), Copaifera langsdorffii (Fabaceae), Poaceae, Asteraceae não identificada 2, Cecropia sp. 1 (Cecropiaceae) e Eucalyptus spp. (Myrtaceae); na estação seca, os tipos polínicos mais frequentes foram Acosmium dasycarpum (Fabaceae), Cecropia sp. 1 (Cecropiaceae) e Eucalyptus spp. (Myrtaceae). As espécies Baccharis sp. (Asteraceae), Cecropia sp. 1 (Cecropiaceae), Copaifera langsdorffii (Fabaceae), Mimosa nuda (Fabaceae), Eucalyptus spp. (Myrtaceae) e Trema micrantha (Ulmaceae) estiveram presentes durante todo o período amostrado.

RESUMO

The aim of this study was to estimate the genetic diversity and structure of the only known population of minnow Astyanax xavante, which inhabits a stretch of river including several waterfalls. The FST values among the samples were not significant, except between two populations separated by a 30 m waterfall. Nevertheless, haplotype and nucleotide diversity increased in the downstream direction, indicating that gene flow is unidirectional, which indicates this genetic pattern as downstairs gene flow, as it has the effect of increasing genetic diversity in the downstream direction.

INTRODUCTION: Tobacco smoke is a risk factor for Chronic Obstructive Pulmonary Disease and a major public health problem. Prenatal maternal smoking and post-natal environmental tobacco smoke (ETS) lead to dose-dependent decrease in lung function and respiratory morbidity. Influence of different socioeconomic indicators and ETS in the home has also been suggested. METHODS: Data on 313 children (52 % male) from 4 public schools in Lisbon was analyzed [1st (46 %) and 4th graders]. ETS assessment and respiratory symptoms were based on a self-answered questionnaire. All children performed standard spirometry in the school setting and 54 % were acceptable according to ATS/ERS criteria. Descriptive and bivariate analysis of the most relevant variables was done, followed by multiple logistic regression analysis adjusted to the variables with clinical/statistical relevance. RESULTS: ETS in the home was found in 41 % (maternal smoking during pregnancy 18 %, smoking mother 32 %, smoking father 38 %). Smoking fathers had lower education and less qualified occupation. Cough was more frequent in children with a smoking mother (adjusted OR = 2.1 95CI 1.1-4.0) and wheezing in children with maternal smoking during pregnancy and smoking parents. All differences were significant (p < 0.05). No association was found between parental education and cough/wheeze or ETS and respiratory infections/asthma/decreased spirometric values. CONCLUSIONS: Children in Lisbon are frequently exposed to ETS which results in significant respiratory morbidity. Targeted interventions must have social conditions in consideration. In this study, field spirometry was not helpful in early detection of lung function disability in children associated with ETS.

A set of primers to amplify 10 microsatellite DNA loci was developed for the Neotropical fish Cichla piquiti, one of the largest sized cichlids in the Amazon Basin. These loci were used to genotype individuals from two populations, one native population from the Tocantins River, the other an introduced population in southeast Brazil, Upper Paraná River. Cross-amplification was also successful for another species of peacock bass, C. kelberi. An average of 4.4 alleles per locus (2-9 alleles) was detected. These markers will be useful for the characterization of genetic structure of native populations, and also for invasive biology studies since Cichla species have been introduced in many river basins outside their native ranges.

RESUMO

Dipteryx odorata is an intensely exploited Amazonian tree legume. Microsatellite markers were developed to study the genetic structure, gene flow and reproductive biology of D. odorata. Eight highly polymorphic microsatellite markers were isolated from enriched repeat libraries screened for microsatellite repeats. An average of 16 alleles and 0.964 phenotype diversity per locus were found in 76 individuals from the Tapajos National Forest, in the state of Pará in the Brazilian Amazon.

RESUMO

The Dendrogene Project (Genetic Conservation within Managed Forests in Amazonia) aims to understand the genetic and ecological processes that underpin tree species survival and in particular their response to forest management regimes. As part of the project, we developed eight microsatellite markers for Jacaranda copaia to be used for genetic structure, gene flow and reproductive biology studies. Polymorphism was evaluated using 96 adult trees from the Tapajos National Forest in the Eastern Brazilian Amazon. An average of 22 alleles per locus were detected, with expected heterozygosity ranging from 0.731 to 0.94.

Scomberomorus cavalla is a pelagic fish species widely distributed on the Atlantic west coast, and a noticeable decrease in its capture level in the USA and Gulf of Mexico is occurring, compared to the levels reached by the species in the past. Likewise, in some areas of Brazil, there has been indication of over-harvesting. However, there are no molecular studies focusing on the management of such an important item. Thus, in the present study, 380 nucleotide base pairs of the mitochondrial DNA D-Loop region of samples from Macapá, Bragança, and Fortaleza were sequenced. Phylogenetic and population analyses revealed that there is only one panmitic population, and low levels of genetic variability were verified. These results, as well as the noticed over-harvesting of S. cavalla, represent very important data to determine the management of such stock in order to prevent a collapse or the risk of future extinction.


Scomberomorus cavalla é uma espécie de peixe pelágico amplamente distribuído na costa oeste do Atlântico, e uma diminuição no seu nível de captura tem sido verificada nos E.U.A e Golfo do México, comparada com os níveis alcançados pela espécie no passado. Da mesma forma, em algumas áreas do Brasil, há indícios de sobre-exploração. Entretanto, não existem estudos moleculares que visam o manejo deste importante item. Desta forma, no presente estudo, foram seqüenciados 380 pares de bases nucleotídicas da região da Alça-D do DNA mitocondrial de amostras provenientes de desembarque em Macapá, Bragança e Fortaleza. As análises filogenéticas e populacionais revelaram que há apenas uma população panmítica e baixos níveis de variabilidade genética foram observados. Estes resultados, assim como a observada sobre-exploração de S. cavala, representam dados muito importantes para o estabelecimento do manejo deste estoque a fim de prevenir um colapso ou risco de extinção no futuro.

AIM: To investigate in individuals with symptomatic dental abscesses the occurrence of functional polymorphisms within five genes involved with the immune response. The functional gene polymorphisms analysed were CD14 (-260 C/T), IL1B (+3954 C/T), IL6 (-174 G/C,), IL10 (-1082 G/A) and TNFA (-308 G/A). METHODOLOGY: Genomic DNA obtained from oral swabs from individuals with symptomatic dental abscesses and asymptomatic inflammatory periapical lesions, without previous exacerbation, was submitted to restriction fragment length polymorphism (RFLP) analyses to determine each individual genotype. The chi-square and principal components analysis tests were used for statistical analysis. RESULTS: A significant association was observed between the occurrence of the GG genotype or the G allele expression of the polymorphic locus-174 (G/C) of the IL6 gene, and the presence of the symptomatic dental abscesses in women and in individuals < or =35 years old. The principal components analysis suggested predominance of the symptomatic dental abscesses in individuals displaying: high-producer IL6 genotype; intermediate and high-producer IL1B genotypes and low-producer TNFA genotype. CONCLUSIONS: The present study suggests that genetic factors are associated with susceptibility to develop symptomatic dental abscesses.

Foram analisadas as concentrações séricas de retinol de 16 éguas gestantes em haras de duas diferentes regiões de Minas Gerais (Vale do Rio Doce e Sul de Minas). Os animais foram mantidos em pastagens com sal mineral à vontade, sem qualquer suplementação concentrada. Amostras de sangue foram coletadas a cada 30 dias, nos meses de maio a novembro de 2002, objetivando verificar a necessidade de suplementação das éguas com vitamina A nos meses de seca. A concentração de betacaroteno das pastagens também foi correlacionada com as concentrações sorológicas de vitamina A, a fim de verificar a utilização da reserva hepática dessa vitamina pelos eqüinos. As análises do retinol sorológico e do betacaroteno das pastagens foram realizadas por cromatografia líquida de alta eficiência (HPLC). O experimento foi em parcelas subdivididas, no delineamento inteiramente ao acaso, e os resultados foram comparados pelo teste Duncan. A correlação entre as concentrações séricas de retinol e betacaroteno das pastagens foi estimada pelo método de Pearson. Os resultados demonstraram que, no ano de 2002, não houve necessidade de suplementação das éguas dos haras das regiões estudadas nos meses de seca e que não houve correlação entre os teores de retinol plasmático das éguas com os de betacaroteno das pastagens.


Retinol serum concentrations were analyzed from 16 pregnant mares raised in farms of two different regions of Minas Gerais state, Brazil (Vale do Rio Doce and Sul de Minas). The animals were maintained on pasture with free access to mineral supplement without grain concentrate supplementation. Blood samples were collected monthly from May to November 2002, aiming to verify the need of vitamin A supplementation during the dry season. The concentration of beta-carotene from pasture samples was also correlated with blood plasma concentration to verify the utilization of hepatic stock of vitamin A. High performance liquid chromatography (HPLC) was used to determine vitamin A and beta-carotene concentrations. The used statistics was the split plot design and the results were compared by Duncan test. The Pearson correlation between the serum concentration of retinol and beta-carotene from pasture was estimated. The results demonstrated that the supplementation of pregnant mares in farms of the studied regions was not necessary during the dry season and that no significant correlation between the serum concentration of retinol and beta-carotene from pasture was observed.
