In this study, two nanoemulsions were formulated with essential oil (EO) of Ocimum gratissimum with (EON) or without (EOE) cashew gum (CG). Subsequently, inhibition of melanosis and preservation of the quality of shrimp stored for 16 days at 4 ± 0.5 °C were evaluated. A computational approach was performed to predict the system interactions. Dynamic light scattering (DLS) and atomic force microscopy (AFM) were used for nanoparticle analysis. Gas chromatography and flame ionization detector (GC-FID) determined the chemical composition of the EO constituents. Shrimps were evaluated according to melanosis's appearance, psychrotrophic bacteria's count, pH, total volatile basic nitrogen, and thiobarbituric acid reactive substances. EON exhibited a particle size three times smaller than EOE. The shrimp treated with EON showed a more pronounced sensory inhibition of melanosis, which was considered mild by the 16th day. Meanwhile, in the other groups, melanosis was moderate (EOE) or severe (untreated group). Both EON and EOE treatments exhibited inhibition of psychrotrophic bacteria and demonstrated the potential to prevent lipid oxidation, thus extending the shelf life compared to untreated fresh shrimp. EON with cashew gum, seems more promising due to its physicochemical characteristics and superior sensory performance in inhibiting melanosis during shrimp preservation.

As described in the literature, Solieria filiformis lectin (SfL) from the marine red alga S. filiformis was found to have antinociceptive and anti-inflammatory effects. In this study, we characterized two SfL variants, SfL-1 and SfL-2, with molecular mass of 27,552Da and 27,985Da, respectively. The primary structures of SfL-1 and SfL-2 consist of four tandem-repeat protein domains with 67 amino acids each. SfL-1 and -2 showed high similarity to OAAH-family lectins. 3D structure prediction revealed that SfL-1 and -2 are composed of two ß-barrel-like domains formed by five antiparallel ß-strands, which are connected by a short peptide linker. Furthermore, the mixture of isoforms (SfLs) showed anticancer effect against MCF-7 cells. Specifically, SfLs inhibited 50% of viability in MCF-7 cells after treatment at 125µg.mL-1, while the inhibition of Human Dermal Fibroblasts (HDF) was 34% with the same treatment. Finally, 24h after treatment, 25% of MCF-7 cells were in early apoptosis and 35% in late apoptosis. Evaluation of pro- and anti-apoptotic gene expression of MCF-7 cells revealed that SfLs induced caspase-dependent apoptosis within 24h.

Seedling establishment in saline conditions is crucial for plant survival and productivity. This study was performed to elucidate the biochemical and physiological mechanisms involved with the recovery and establishment of cashew seedlings subjected to salinity. The changes in the Na+ levels and K/Na ratios, associated with relative water content, indicated that osmotic effects were more important than salt toxicity in the inhibition of seedling growth and cotyledonary protein mobilization. Salinity (50mM NaCl) induced a strong delay in protein breakdown and amino acid accumulation in cotyledons, and this effect was closely related to azocaseinolytic and protease activities. In parallel, proline and free amino acids accumulated in the leaves whereas the protein content decreased. Assays with specific inhibitors indicated that the most important proteases in cotyledons were of serine, cysteine and aspartic types. Proteomic analysis revealed that most of the cashew reserve proteins are 11S globulin-type and that these proteins were similarly degraded under salinity. In the late establishment phase, the salt-treated seedlings displayed an unexpected recovery in terms of leaf growth and N mobilization from cotyledon to leaves. This recovery coordinately involved a great leaf expansion, decreased amino acid content and increased protein synthesis in leaves. This response occurred in parallel with a prominent induction in the cotyledon proteolytic activity. Altogether, these data suggest that a source-sink mechanism involving leaf growth and protein synthesis may have acted as an important sink for reserve mobilization contributing to the seedling establishment under salinity. The amino acids that accumulated in the leaves may have exerted negative feedback to act as a signal for the induction of protease activity in the cotyledon. Overall, these mechanisms employed by cashew seedlings may be part of an adaptive process for the efficient rescue of cotyledonary proteins, as the cashew species originates from an environment with N-poor soil and high salinity.
