INTRODUCTION AND AIMS: Altering the position and orientation of the root canal access cavity passway, or modifying the reduction of dentin volume, can influence the strength of dentition. This study aimed to compare the effects of different access cavities on the biomechanical performances of maxillary central incisors with a finite element analysis. METHODS: Based on the micro-computed tomography (CT) scan of a maxillary central incisor, the finite element models of the intact tooth and teeth with 4 access cavity designs: conservative incisal access cavity, incisal access cavity, conservative access cavity, and traditional access cavity were generated. Simulated occlusal forces were applied at the incisal edge of the incisor in the finite element analysis procedure. RESULTS: The maximum von Mises stress and maximum principal stress in the cervical area are highest in the traditional access cavity group, followed by the conservative access cavity group, incisal access cavity group, and conservative incisal access cavity group. CONCLUSION: The conservative access cavities minimise the extent of dentin removal from the cervical region, protecting the mechanical behaviour of the incisor. Moving the access cavity entry point to the incisal edge also improves the fracture resistance of the incisor. CLINICAL RELEVANCE: This study's findings would help clinicians select the most appropriate endodontics access cavity method when performing the root canal on maxillary central incisors.

Ochratoxin A (OTA) is a toxic fungal metabolite that is commonly found in cereals and animal feed. It is economically damaging and potentially hazardous to human health. Herein, we propose an electrochemical immunosensor for the rapid detection of OTA using anti-OTA antibodies and diazonium-functionalized, screen-printed electrodes. We attached 4-aminobenzoic acid to an electrode surface, activated the carboxyl groups on the surface with carbodiimide, and attached an antibody to the diazo layer. Subsequently, we used bovine serum protein as a blocker to prevent non-specific antigens from binding to the antibody. We evaluated the performance of the sensor by cyclic voltammetry, electrochemical impedance spectroscopy, and differential pulse voltammetry. The sensor is highly specific and sensitive, has good linear responses in the range 20-200 ng/mL, a limit of detection of 0.5 ng/mL, and good recoveries of 90.5%-100.9% in spiked samples. It can be stored at 4 °C for approximately 2 weeks, and is highly stable, with a current response variation of no more than 4.6%.

Peptide and protein postmodification have gained significant attention due to their extensive impact on biomolecule engineering and drug discovery, of which cysteine-specific modification strategies are prominent due to their inherent nucleophilicity and low abundance. Herein, the study introduces a novel approach utilizing multifunctional 5-substituted 1,2,3-triazine derivatives to achieve multifaceted bioconjugation targeting cysteine-containing peptides and proteins. On the one hand, this represents an inaugural instance of employing 1,2,3-triazine in biomolecular-specific modification within a physiological solution. On the other hand, as a powerful combination of precision modification and biorthogonality, this strategy allows for the one-pot dual-orthogonal functionalization of biomolecules utilizing the aldehyde group generated simultaneously. 1,2,3-Triazine derivatives with diverse functional groups allow conjugation to peptides or proteins, while bi-triazines enable peptide cyclization and dimerization. The examination of the stability of bi-triazines revealed their potential for reversible peptide modification. This work establishes a comprehensive platform for identifying cysteine-selective modifications, providing new avenues for peptide-based drug development, protein bioconjugation, and chemical biology research.

Silent information regulator 2 (Sir2) proteins typically catalyze NAD+-dependent protein deacetylation. The recently identified bacterial Sir2 domain-containing protein, defense-associated sirtuin 2 (DSR2), recognizes the phage tail tube and depletes NAD+ to abort phage propagation, which is counteracted by the phage-encoded DSR anti-defense 1 (DSAD1), but their molecular mechanisms remain unclear. Here, we determine cryo-EM structures of inactive DSR2 in its apo form, DSR2-DSAD1 and DSR2-DSAD1-NAD+, as well as active DSR2-tube and DSR2-tube-NAD+ complexes. DSR2 forms a tetramer with its C-terminal sensor domains (CTDs) in two distinct conformations: CTDclosed or CTDopen. Monomeric, rather than oligomeric, tail tube proteins preferentially bind to CTDclosed and activate Sir2 for NAD+ hydrolysis. DSAD1 binding to CTDopen allosterically inhibits tube binding and tube-mediated DSR2 activation. Our findings provide mechanistic insight into DSR2 assembly, tube-mediated DSR2 activation, and DSAD1-mediated inhibition and NAD+ substrate catalysis in bacterial DSR2 anti-phage defense systems.

Objective: During the COVID-19 pandemic, people posted help-seeking messages on Weibo, a mainstream social media in China, to solve practical problems. As viruses, policies, and perceptions have all changed, help-seeking behavior on Weibo has been shown to evolve in this paper. Methods: We compare and analyze the help-seeking messages from three dimensions: content categories, time distribution, and retweeting influencing factors. First, we crawled the help-seeking messages from Weibo, and successively used CNN and xlm-roberta-large models for text classification to analyze the changes of help-seeking messages in different stages from the content categories dimension. Subsequently, we studied the time distribution of help-seeking messages and calculated the time lag using TLCC algorithm. Finally, we analyze the changes of the retweeting influencing factors of help-seeking messages in different stages by negative binomial regression. Results: (1) Help-seekers in different periods have different emphasis on content. (2) There is a significant correlation between new daily help-seeking messages and new confirmed cases in the middle stage (1/1/2022-5/20/2022), with a 16-day time lag, but there is no correlation in the latter stage (12/10/2022-2/25/2023). (3) In all the periods, pictures or videos, and the length of the text have a significant positive effect on the number of retweets of help-seeking messages, but other factors do not have exactly the same effect on the retweeting volume. Conclusion: This paper demonstrates the evolution of help-seeking messages during different stages of the COVID-19 pandemic in three dimensions: content categories, time distribution, and retweeting influencing factors, which are worthy of reference for decision-makers and help-seekers, as well as provide thinking for subsequent studies.

Pyrene-based derivatives have been widely deployed in organic luminescent materials because of their bright fluorescence, high charge carrier mobility, and facile modification. Nevertheless, the fluorescence output of conventional pyrenes is prone to quenching upon aggregation due to extensive intermolecular π-π stacking interactions. To address this issue, a set of new Y-shaped pyrene-containing luminogens are synthesized from a new bromopyrene chemical precursor, 2-hydroxyl-7-tert-butyl-1,3-bromopyrene, where the bromo and hydroxyl groups at the pyrene core can be readily modified to obtain the target products and provide great flexibility in tuning the photophysical performances. When the hydroxy group at the 2-position of pyrene was replaced by a benzyl group, the steric hindrance of the benzyl group not only efficiently inhibits the detrimental intermolecular π-π stacking interactions but also rigidifies the molecular conformation, resulting in a narrow-band blue emission. Moreover, the TPE-containing compounds 2c and 3c possessed characteristic aggregation-induced emission (AIE) properties with fluorescence quantum yields of up to 66% and 38% in the solid state, respectively. Thus, this article has methodically investigated the factors influencing the optical behavior, such as intermolecular interactions, and the steric effects of the substituent group, thereby opening up the potential to develop narrow-band pyrene-based blue emitters for OLED device applications.

Primary health care (PHC) is the most effective way to improve people's health and well-being, and primary care services should act as the cornerstone of a resilient health system and the foundation of universal health coverage. To promote high quality development of PHC, an International Symposium on Quality Primary Health Care Development was held on December 4-5, 2023 in Beijing, China, and the participants have proposed and advocated the Beijing Initiative on Quality Primary Health Care Development. The Beijing Initiative calls on all countries to carry out and strengthen 11 actions: fulfill political commitment and accountability; achieve "health in all policies" through multisectoral coordination; establish sustainable financing; empower communities and individuals; provide community-based integrated care; promote the connection and integration of health services and social services through good governance; enhance training, allocation and motivation of health workforce, and medical education; expand application of traditional and alternative medicine for disease prevention and illness healing; empower PHC with digital technology; ensure access to medicinal products and appropriate technologies; and last, strengthen global partnership and international health cooperation. The Initiative will enrich the content of quality development of PHC, build consensus, and put forward policies for quality development of PHC in China in the new era, which are expected to make contributions in accelerating global actions.

RESUMO

A visible-light mediated deoxygenative radical addition of carboxylic acids to dehydroalanines has been disclosed. The method can be used in ß-acyl alanine derivative synthesis, including those chiral and deuterated variants, and late-stage peptide modification with various functional groups, both in the homogeneous phase and on the resin in SPPS. It provides a new tool kit for rapid construction of bioactive peptide analogues, which has been demonstrated by modification of the antimicrobial peptide Feleucin-K3.

Charge-transfer (CT) cocrystals consisting of an electron donor and acceptor have gained attention for designing photothermal (PT) conversion materials with potential for biomedical and therapeutic use. However, the applicability of CT cocrystals is limited by their low stability and aqueous dispersity in biological settings. In this study, we present the self-assembly of CT cocrystals within hydrogen-bonded organic frameworks (HOFs), which not only allows for the dispersion and stabilization of cocrystals in aqueous solution but also promotes the CT interaction within the confined space of HOFs for photothermal conversion. We demonstrate that the CT interaction-driven self-assembly of tetrathiafulvalene (TTF) and tetracyanoquinodimethane (TCNQ) with PFC-1 HOFs results in the formation of cocrystal-encapsulated TQC@PFC-1 while retaining the crystalline structure of the cocrystal and PFC-1. TQC@PFC-1, in particular, exhibits significant absorption in the second near-infrared region (NIR-II) and excellent photothermal conversion efficiency, as high as 32%. Cellular delivery studies show that TQC@PFC-1 can be internalized in different types of cancer cells, leading to an effective NIR-II photothermal therapy effect both in cultured cells and in vivo. We anticipate that the strategy of self-assembly and stabilization of CT cocrystals in nanoscale HOFs opens the path for tuning their photophysical properties and interfacing cocrystals with biological settings for photothermal therapeutic applications.

The concept of triboelectric nanogenerator (TENG)-based fibrous air filters, in which the electroactive fibers are ready to enhance the electrostatic adsorption by sustainable energy harvesting, is appealing for long-term respiratory protection and in vivo real-time monitoring. This effort discloses a self-reinforcing electroactivity strategy to confer extreme alignment and refinement of the electrospun poly(lactic acid) (PLA) nanofibers, significantly facilitating formation of electroactive phases (i.e., ß-phase and highly aligned chains and dipoles) and promotion of polarization and electret properties. It endowed the PLA nanofibrous membranes (NFMs) with largely increased surface potential and filtration performance, as exemplified by efficient removal of PM0.3 and PM2.5 (90.68 and 99.82%, respectively) even at the highest airflow capacity of 85 L/min. With high electroactivity and a well-controlled morphology, the PLA NFMs exhibited superior TENG properties triggered by regular respiratory vibrations, enabling 9.21-fold increase of surface potential (-1.43 kV) and nearly 68% increase of PM0.3 capturing (94.3%) compared to those of conventional PLA membranes. The remarkable TENG mechanisms were examined to elaborately monitor the personal respiration characteristics, particularly those triggered large and rapid variations of output voltages like coughing and tachypnea. Featuring desirable biocompatibility and degradability, the self-powered PLA NFMs permit promising applications in the fabrication of ecofriendly air filters toward high-performance purification and intelligent monitoring.

Most sheet facial masks for skincare are made of nonwovens and loaded with liquid active ingredients, which are usually opaque and require additives for long-term preservation. Herein, a Transparent Additive-Free Fibrous (TAFF) facial mask is reported for skin moisturizing. The TAFF facial mask consists of a bilayer fibrous membrane. The inner layer is fabricated by electrospinning functional components of gelatin (GE) and hyaluronic acid (HA) into a solid fibrous membrane to get rid of additives, the outer layer is an ultrathin PA6 fibrous membrane that is highly transparent, especially after absorbing water. The results indicate that the GE-HA membrane can quickly absorb water and become a transparent hydrogel film. By employing the hydrophobic PA6 membrane as the outer layer, directional water transport is achieved, which enables TAFF facial mask with excellent skin moisturizing effect. The skin moisture content is up to 84% ± 7% after placing the TAFF facial mask on the skin for 10 min. In addition, the relative transparency of the TAFF facial mask on the skin reaches 97.0% ± 1.9% when ultrathin PA6 membrane is used as the outer layer. The design of the transparent additive-free facial mask may serve as a guideline for developing new functional facial masks.

High-performance air filtration materials are important for addressing the airborne pollutants. Herein, we propose an unprecedented access to biodegradable poly(lactic acid) (PLA)-based MOFilters with excellent filtering performance and antibacterial activity. The fabrication involved a stepwise in situ growth of zeolitic imidazolate framework-8 (ZIF-8) crystals at the surface of microfibrous PLA membranes, followed by mechanical polarization under high pressure and low temperature (5 MPa, 40 °C) to trigger the ordered alignment of dipoles in PLA chains and ZIF-8. The unique structural features allowed these PLA-based MOFilters to achieve an exceptional combination of excellent tensile properties, high dielectric constant (up to 2.4 F/m), and enhanced surface potential as high as 4 kV. Arising from the remarkable surface activity and electrostatic adsorption effect, a significant increase (from over 12% to nearly 20%) in PM0.3 filtration efficiency was observed for the PLA-based MOFilters compared to that of pure PLA counterparts, with weak relation to the airflow velocities (10-85 L/min). Moreover, the air resistance was controlled at a considerably low level for all the MOFilters, that is, below 183 Pa even at 85 L/min. It is worth noting that distinct antibacterial properties were achieved for the MOFilters, as illustrated by the inhibitive rates of 87 and 100% against Escherichia coli and Staphylococcus aureus, respectively. The proposed concept of PLA-based MOFilters offers unprecedented multifunction integration, which may fuel the development of biodegradable versatile filters with high capturing and antibacterial performances yet desirable manufacturing feasibility.

High-performance flexible barium titanate (BaTiO3)-based piezoelectric devices have gained much attention. However, it is still a challenge to prepare flexible polymer/BaTiO3-based composite materials with uniform distribution and high performance due to the high viscosity of polymers. In this study, novel hybrid BaTiO3 particles were synthesized with assistance of TEMPO-oxidized cellulose nanofibrils (CNFs) via a low-temperature hydrothermal method and explored for their application in piezoelectric composites. Specifically, Ba2+ was adsorbed on uniformly dispersed CNFs with a large amount of negative charge on their surface, which nucleated, resulting in the synthesis of evenly dispersed CNF-BaTiO3. The obtained CNF-BaTiO3 possessed a uniform particle size, few impurities, high crystallinity and dispersity, high compatibility with the polymer substrate and surface activity due to the existence of CNFs. Subsequently, both polyvinylidene fluoride (PVDF) and TEMPO-oxidized CNFs were employed as piezoelectric substrates for the fabrication of a CNF/PVDF/CNF-BaTiO3 composite membrane with a compact structure, displaying the tensile strength of 18.61 ± 3.75 MPa and elongation at break of 3.06 ± 1.33%. Finally, a thin piezoelectric generator (PEG) was assembled, which output a considerable open-circuit voltage (4.4 V) and short-circuit current (200 nA), and could also power a light-emitting diode and charge a 1 µF capacitor to 3.66 V in 500 s. Its longitudinal piezoelectric constant (d 33) was 5.25 ± 1.04 pC N-1 even with a small thickness. It also exhibited high sensitivity to human movement, outputting a voltage of about 9 V and current of 739 nA for only a footstep. Thus, it exhibited good sensing property and energy harvesting property, presenting practical application prospects. This work provides a new idea for the preparation of hybrid BaTiO3 and cellulose-based piezoelectric composite materials.

In order to increase the number and contact probability of electric dipole on cellulose, acid and alkali treatment was employed to extract hemicellulose and lignin from original wood to gain a highly oriented cellulose frame. The combined means with 2,2,6,6-tetramethylpiperidine-1-oxyl-NaBr-NaClO oxidation and impregnation of PDMS with compression was subsequently used to enhance its mechanical performance and electromechanical conversion. The assembled wooden electromechanical device (10 mm × 10 mm × 1 mm) exhibits the maximum open-circuit voltage (V OC) of 11.75 V and short-circuit current (I SC) of 211.01 nA as stepped by foot. It can be sliced to fabricate a flexible sensor with high sensitivity displaying V OC of 2.88 V and I SC of 210.09 nA under the tapped state. Its highly oriented wood fiber makes it display significant anisotropy in terms of mechanical and electromechanical performance for multidirectional sense. This strategy will exactly provide reference for developing other high-performance piezoelectric devices.

Problem-based learning (PBL) is increasingly being used in medical education globally, but its effectiveness in teaching remains controversial. A randomized controlled trial (RCT) is the method of choice for evaluating its effectiveness. The quality of an RCT has a significant effect on this evaluation, but to date we have not seen an assessment of the quality of RCTs for PBL. Two researchers searched MEDLINE and EMBASE for RCTs addressing PBL in medical education. The overall quality of each report was measured on a 28-point overall quality score (OQS) based on the 2010 revised Comprehensive Standards for Reporting and Testing (CONSORT) Statement. Furthermore, to study the key factors affecting OQS more effectively, a linear regression model of those factors was established using SPSS. After literature screening, 30 RCTs were eventually included and analyzed. The median OQS was 15 (range, 7-20), which meant that half of the items in the revised 2010 CONSORT statement were poorly reported in at least 40% of the RCTs analyzed. The regression model showed that the year of publication of RCTs and the impact factors of the journals in which they were published were the main factors affecting OQS. The overall quality of reporting of RCTs on PBL teaching in medical education was not satisfactory. Some RCTs were subjectively selective in reporting certain items, leading to heterogeneity in quality. It is expected that statisticians will develop new standards more suitable for evaluating RCTs related to teaching research and that editors and peer reviewers will be required to review the relevant RCTs more strictly.

ABSTRACT Objectives: The effects of weightlessness on the liver were studied using a tail suspension (TS) male mouse model. Methods: The effects of 0-, 2- and 4-week TS (CON, TS2 and TS4 groups) on glycogen and lipid content, as well as on the molecular processes of the synthesis and degradation pathways, were examined. Results: (1) The number of glycogenosomes under ultrastructure and the glycogen content were considerably larger in the TS4 group than in the other two groups. (2) In the TS4 group, glycogen synthase activity remained constant while glycogen phosphorylase activity dropped, indicating that glycogen breakdown was reduced. (3) The livers of the TS2 group had the highest lipid and triglyceride content, indicating lipid buildup in the liver at this time. (4) In the TS2 group, the activities of the fatty acid synthesis-related factors acetyl-CoA carboxylase and fatty acid synthase increased, while hepatic lipase decreased, indicating that lipid synthesis increased, while decomposition decreased. (5) In the TS2 group, the protein expression of glucose transporters 1 and 2 increased. Conclusions: From TS2 weeks to TS4 weeks, the main energy consumption mode in the livers of mice transitioned from glucose metabolism to lipid metabolism as glucose use decreased. RESUMEN Objetivos: Se estudiaron los efectos de la antigravedad en el hígado utilizando un modelo de ratón macho en prueba de suspensión de la cola (TS, tail suspension). Métodos: Se examinaron los efectos de la TS a las 0, 2 y 4 semanas (grupos CON, TS2 y TS4) sobre el contenido de glucógeno y lípidos, así como sobre los procesos moleculares de las vías de síntesis y degradación. Resultados: (1) El número de glucogenosomas ultraestructurales y el contenido de glucógeno fueron expresivamente más altos en el grupo TS4 que en los otros dos grupos. (2) En el grupo TS4, la actividad de la glucógeno sintasa se mantuvo constante, mientras que la actividad de la glucógeno fosforilasa disminuyó, lo que indica que la degradación del glucógeno se redujo. (3) Los hígados del grupo TS2 presentaron el mayor contenido de lípidos y triglicéridos, lo que indica la acumulación de lípidos en el hígado en ese momento. (4) En el grupo TS2, la actividad de los factores relacionados con la síntesis de ácidos grasos acetil-CoA carboxilasa y ácido graso sintasa aumentó, mientras que la lipasa hepática disminuyó, indicando que la síntesis de lípidos aumentó mientras que la descomposición disminuyó. (5) En el grupo TS2, la expresión proteica de los transportadores de glucosa 1 y 2 aumentó. RESUMO Objetivos: Os efeitos da antigravidade no fígado foram estudados usando um modelo de camundongo macho com a suspensão pela cauda (TS, tail suspension). Métodos: Foram examinados os efeitos da TS em 0, 2 e 4 semanas (grupos CON, TS2 e TS4) sobre o conteúdo de glicogênio e lipídios, bem como nos processos moleculares das vias de síntese e degradação. Resultados: (1) O número de glicogenossomos ultraestruturais e o teor de glicogênio foram expressivamente maiores no grupo TS4 do que nos outros dois grupos. (2) No grupo TS4, a atividade de glicogênio sintase permaneceu constante, enquanto a atividade de glicogênio fosforilase caiu, indicando que a degradação do glicogênio foi reduzida. (3) Os fígados do grupo TS2 tiveram o maior teor lipídico e de triglicérides, indicando acúmulo de lipídios no fígado no momento. (4) No grupo TS2, a atividade dos fatores relacionados com a síntese de ácidos graxos acetil-CoA carboxilase e ácido graxo sintase aumentaram, enquanto a lipase hepática diminuiu, indicando que a síntese de lipídios aumentou, enquanto a decomposição diminuiu. (5) No grupo TS2, a expressão proteica dos transportadores de glicose 1 e 2 aumentou. Conclusões: De TS2 semanas para TS4 semanas, o principal modo de consumo de energia no fígado de camundongos passou do metabolismo da glicose para o metabolismo lipídico, à medida que o uso de glicose diminuiu. As a representative polyolefin, high-density polyethylene (HDPE) has become one of the most commonly used commercial plastics with a wide range of applications in the world. However, its applications are limited due to poor mechanical properties. Hence, it is indispensable to develop composites with improved mechanical properties to overcome this disadvantage. In our work, basalt fiber (BF) and polyamide 6 (PA6)-reinforced HDPE composites were prepared. The effects of adding fiber, organic filler, and polar component maleic anhydride (MAH) on the microstructural characteristics of composites were investigated. Microstructural characterization evidenced that the binary-dispersed phase (PA6/BF) possesses a core-shell structure in which the component PA6 encapsulates the component BF, and the extent of encapsulation declines with the increase of MAH addition. It has been confirmed by scanning electron microscopy (SEM) observation that the microstructure is related to the interfacial tension of components. The effects of multicomponents on the crystallization behavior of composites were studied. The differential scanning calorimeter (DSC) analysis exhibited a significant change in the HDPE microstructure. Results showed that, as nucleating agents, PA6 and BF improve the crystallization rate in the cooling process. Furthermore, the rheological behavior of multicomponent composites was studied. With the increase of MAH, a clear improvement of complex viscosity and storage modulus was observed, of which the mechanism has been discussed in detail. The relationship between microstructure and heat resistance of composites was studied by a thermal deformation test under static fore. It is confirmed that the thermally conductive fiber BF and other components can form a thermally conductive network and channels, thus improving the heat resistance. It can become a composite material, which is suitable for special environments.

Benefiting from the multi-directional load-bearing capability, the three-dimensional braided composites (3DBC) have found a wide application in primary structures. It is therefore of great importance to fully understand their mechanical behavior and failure modes. In the present paper, the tensile and compressive tests were carried out, according to standardized testing methods, for eight types of 3DBC, which were manufactured by resin transfer molding (RTM). It was found that the mechanical properties of the 3DBCs decreased with an increasing braiding angle. When the braiding angle was 20°, 3D 5-directional braided composite (3D5dBC) exhibited the best mechanical properties, while for the braiding angle of 40°, the mechanical properties of 3D6dBC were the most prominent. Moreover, the tensile strength of the 3DBCs is approximately two times as much as the compressive strength; however, the compressive modulus is always 10% higher than the tensile modulus. The failure modes of the 3DBCs with a braiding angle of 20°greatly depended on the braiding structures. However, they tend to be consistent when the braiding angle increases to 40°.

The model electroactive bacterium Geobacter sulfurreducens can acquire electrons directly from solid donors including metals and other species. Reports on this physiology concluding that solid donors are the only electron sources were conducted with fumarate believed to serve exclusively as the terminal electron acceptor (TEA). Here, G. sulfurreducens was repeatedly transferred for adaptation within a growth medium containing only fumarate and no other solid or soluble substrate. The resulting evolved strain grew efficiently with either the C4-dicarboxylate fumarate or malate acting simultaneously as electron donor, carbon source, and electron acceptor via disproportionation. Whole-genome sequencing identified 38 mutations including one in the regulator PilR known to repress the expression of the C4-dicarboxylate antiporter DcuB essential to G. sulfurreducens when growing with fumarate. Futhermore, the PilR mutation was identical to the sole mutation previously reported in an evolved G. sulfurreducens grown in a co-culture assumed to derive energy solely from direct interspecies electron transfer, but cultivated with fumarate as the TEA. When cultivating the fumarate-adapted strain in the presence of stainless steel and fumarate, biocorrosion was observed and bacterial growth was accelerated 2.3 times. These results suggest that G. sulfurreducens can conserve energy concomitantly from C4-dicarboxylate disproportionation and the oxidation of a solid electron donor. This co-metabolic capacity confers an advantage to Geobacter for survival and colonization and explains in part why these microbes are omnipresent in different anaerobic ecosystems.

Catalpol, a major bioactive component from Rehmannia glutinosa, which has been used to treat diabetes. The present study was designed to elucidate the anti-diabetic effect and mechanism of action for catalpol in db/db mice. The db/db mice were randomly divided into six groups (10/group) according to their blood glucose levels: db/db control, metformin (positive control), and four dose levels of catalpol treatment (25, 50, 100, and 200 mg·kg-1), and 10 db/m mice were used as the normal control. All the groups were administered orally for 8 weeks. The levels of fasting blood glucose (FBG), random blood glucose (RBG), glucose tolerance, insulin tolerance, and glycated serum protein (GSP) and the globe gene expression in liver tissues were analyzed. Our results showed that catalpol treatment obviously reduced water intake and food intake in a dose-dependent manner. Catalpol treatment also remarkably reduce fasting blood glucose (FBG) and random blood glucose (RBG) in a dose-dependent manner. The RBG-lowering effect of catalpol was better than that of metformin. Furthermore, catalpol significantly improved glucose tolerance and insulin tolerance via increasing insulin sensitivity. Catalpol treatment significantly decreased GSP level. The comparisons of gene expression in liver tissues among normal control mice, db/db mice and catalpol treated mice (200 and 100 mg·kg-1) indicated that there were significant increases in the expressions of 287 genes, whichwere mainly involved in lipid metabolism, response to stress, energy metabolism, and cellular processes, and significant decreases in the expressions of 520 genes, which were mainly involved in cell growth, death, immune system, and response to stress. Four genes expressed differentially were linked to glucose metabolism or insulin signaling pathways, including Irs1 (insulin receptor substrate 1), Idh2 (isocitrate dehydrogenase 2 (NADP+), mitochondrial), G6pd2 (glucose-6-phosphate dehydrogenase 2), and SOCS3 (suppressor of cytokine signaling 3). In conclusion, catalpol ecerted significant hypoglycemic effect and remarkable therapeutic effect in db/db mice via modulating various gene expressions.
