[Biotransformation and excretion of N,N'-bis(3-(2-ethoxyphenoxy)-2-hydroxypropyl)ethylenediamine dihydrochloride (Falirytmin) in rats]. / Biotransformation und Ausscheidung von N,N'-Bis[3-(2-ethoxy-phenoxy)-2- hydroxy-propyl]ethylendiamin-dihydrochlorid (Falirytmin) bei der Ratte.

Falirytmin (1) was metabolized in rats almost completely. Besides small amounts of 1 in urine and feces 18 metabolites could be separated. The proposed structures of 14 compounds demonstrate aromatic hydroxylation, N- and O-dealkylation, side-chain oxidation, alcohol dehydrogenation and N-acetylation. The main route of excretion of 1 and metabolites was with the feces. After p.o. or i.v. administration of 14C-O-ethyl-1 the average excretion of radioactivity in urine, feces and expired air was about 75% in 96 h. The residual activity in organs was about 2-2.5%. Whole-body autoradiography confirms these results. Only slight 14C-activity was seen in muscle, fat, liver, bone marrow and gut.

In connection with routine endoscopic examinations of 31 patients with familial adenomatosis coli and 28 health kindreds of patients with familial adenomatosis coli we investigated by autoradiography the in vitro incorporation of 3H-thymidine into bioptic mucosal particles from colon and adenomas. Within the 5-year follow-up period there was found a significantly increased labelling index of polyps and the surrounding mucosa in patients with polyposis independing on the time elapsed after operation. In 12 of 28 kindreds the histological normal colonic mucosa showed an increased labelling index too. The prognostic value of these results must be proofed by further controls.

Proliferation of cultivated cells from normal, preneoplastic and neoplastic human breast tissue--preliminary report.

The success of cultivation of epithelial cells from normal, neoplastic and preneoplastic tissues is reviewed. MCDB 170 medium with special supplements is useful for epithelial cell culture. The epithelial character was demonstrated by the reaction with 3 different monoclonal antibodies. Most cell populations from preneoplastic and neoplastic tissues were identified as fibroblasts. Cell proliferation as measured by cell doubling time, labeling index, flow cytometry did not differ between the tissue types examined. Only 3H-thymidine uptake showed an increased tendency in cells from preneoplastic and neoplastic tissues. The use of normal human breast epithelial cells for molecular biological studies is recommended.

[Possibilities of using monoclonal antibodies to CEA in scintigraphic diagnosis of tumors]. / Möglichkeiten des Einsatzes von monoklonalen Antikörpern gegen CEA in der szintigrafischen Tumordiagnostik.

Improved tumour localization by scintigraphy is the aim of application of radiolabelled antibodies in nuclear medical tumour diagnostics. Results from the literature until 1984 in the field of monoclonal antibodies (Mabs) against CEA are discussed in the following points: type and stability of the radiolabelling; accumulation and biologic activity of Mabs in the animal; distribution of Mabs in clinical application and possible injury for the patient; methods for increased accumulation in the tumor and improved tumour-imaging. Further work is needed in this field before introducing this method into the diagnostic routine.

Flow cytometry, cytomorphology, histology and autoradiography in human gastric hyperplastic polyps and the surrounding mucosa.

43 cases of gastric hyperplastic polyps, 33 tissues adjoining the polyps and 7 tissue samples of the corpus mucosa were investigated by flow cytometry, cytomorphology, autoradiography and histology, respectively. All cases revealed diploid DNA distribution patterns. The results attributed an increased proliferative tendency to special cases of hyperplastic polyps and the surrounding mucosa.

The effect of carbenoxolone on the chronic atrophic gastric mucosa determined by flow cytometry and histoautoradiography.

A trial with carbenoxolone at a dose of 150 mg/day for 6 months was performed in 24 patients with chronic atrophic gastritis. A control group of 21 patients received placebos. Gastric mucosa was examined by flow cytometry (FC) and histoautoradiography 2, 4, 12 and 24 weeks after starting therapy. Carbenoxolone had neither an effect on cell cycle stage distribution as determined by FC nor on the labelling indices obtained by histoautoradiography.
