The olfactory system uses hundreds of odorant receptors (ORs), the largest group of the G-protein-coupled receptor (GPCR) superfamily, to detect a vast array of odorants. Each OR is activated by specific odorous ligands, and like other GPCRs, antagonism can block activation of ORs. Recent studies suggest that odorant antagonisms in mixtures influence olfactory neuron activities, but it is unclear how this affects perception of odor mixtures. In this study, we identified a set of human ORs activated by methanethiol and hydrogen sulfide, two potent volatile sulfur malodors, through large-scale heterologous expression. Screening odorants that block OR activation in heterologous cells identified a set of antagonists, including ß-ionone. Sensory evaluation in humans revealed that ß-ionone reduced the odor intensity and unpleasantness of methanethiol. Additionally, suppression was not observed when methanethiol and ß-ionone were introduced simultaneously to different nostrils. Our study supports the hypothesis that odor sensation is altered through antagonistic interactions at the OR level.

In the modern information society, people are constantly exposed to stress due to complex work environments and various interpersonal relationships. Aromatherapy is attracting attention as one of the methods for relieving stress using aroma. A method to quantitatively evaluate such an effect is necessary to clarify the effect of aroma on the human psychological state. In this study, we propose a method of using two biological indexes, electroencephalogram (EEG) and heart rate variability (HRV), to evaluate human psychological states during the inhalation of aroma. The purpose is to investigate the relationship between biological indexes and the psychological effect of aromas. First, we conducted an aroma presentation experiment using seven different olfactory stimuli while collecting data from EEG and pulse sensors. Next, we extracted the EEG and HRV indexes from the experimental data and analyzed them with respect to the olfactory stimuli. Our study found that olfactory stimuli have a strong effect on psychological states during aroma stimuli and that the human response to olfactory stimuli is immediate but gradually adapts to a more neutral state. The EEG and HRV indexes showed significant differences between aromas and unpleasant odors especially for male participants in their 20-30s, while the delta wave and RMSSD indexes showed potential for generalizing the method to evaluate psychological states influenced by olfactory stimuli across genders and generations. The results suggest the possibility of using EEG and HRV indexes to evaluate psychological states toward olfactory stimuli such as aroma. In addition, we visualized the psychological states affected by the olfactory stimuli on an emotion map, suggesting an appropriate range of EEG frequency bands for evaluating psychological states applied to the olfactory stimuli. The novelty of this research lies in our proposed method to provide a more detailed picture of the psychological responses to olfactory stimuli using the integration of biological indexes and emotion map, which contributes to the areas such as marketing and product design by providing insights into the emotional responses of consumers to different olfactory products.

To synthesize a biodegradable alternative to spherical polyacrylic acid absorbent resin, spherical hydrogel particles were prepared from carboxymethyl cellulose (CMC) dissolved in an aqueous solution, using ethylene glycol diglycidyl ether (EGDE) as a crosslinking agent. The effect of varying the initial CMC concentration and feed amount of EGDE on the shape, water absorbency, water-holding capacity, and enzyme degradability of the resultant CMC hydrogels was determined. The reaction solution was poured into fluid paraffin, and spherical hydrogel particles were obtained via the shear force from stirring. The shape and diameter of the spherical hydrogel particles in the swollen state depended on the CMC concentration. The spherical hydrogel particles obtained by increasing the amount of EGDE resulted in a decrease in absorbency. Additionally, all the spherical hydrogel particles were degraded by cellulase. Thus, spherical biodegradable hydrogel particles were prepared from CMC, and the particle size and water absorption of the hydrogel could be controlled in the range of 5-18 mm and 30-90 g·g-1 in the swollen state, respectively. As an alternative to conventional superabsorbent polymers, the spherical CMC hydrogels are likely to be useful in industrial and agricultural applications.

Intracranial arachnoid cysts(ACs)are thought to develop during infancy and remain clinically asymptomatic in most cases. AC is often diagnosed incidentally during imaging studies. The prognosis for the recovery of newly developed symptoms is usually favorable. However, the prognosis following surgery for chronic symptoms is still unclear. Herein, we report the case of an adult patient with right convexity AC whose chronic symptoms improved after surgery. The patient was a 63-year-old woman who had an AC in her right temporal convexity since the age of 59, without any symptoms, which was incidentally diagnosed on MRI. At 61 years of age, she visited a hospital complaining of slight weakness in her left upper limb. No changes were found on the MRI scan, but her visual field indicated the presence of a left homonymous hemianopia, and her cognitive function had decreased slightly. Two years later, she developed modest left hemiparesis;therefore, she underwent cystoventriculostomy to improve the symptoms of hemiparesis. After the surgery, the left hemiparesis recovered fully, and a remarkable improvement was noted in the left homonymous hemianopia and the degenerated cognitive functions. There are no clear guidelines regarding the surgical indications for AC in adults. Many reports suggest that new symptoms related to AC can be improved with surgery. However, amelioration of chronic symptoms has rarely been reported. The present case indicates that it is possible to improve the chronic symptoms of AC in adults with surgical procedures.

BACKGROUND AND PURPOSE: Transient global amnesia (TGA) is a rare clinical entity with a sudden onset of anterograde amnesia that recovers within 24 hours. The underlying pathophysiology is uncertain. Imaging studies are controversial, but diffusion-weighted images often show small diffusion-restricted lesions in the hippocampus, which may suggest ischemic damage. Thus, we conducted the first clinical study using neurite orientation dispersion and density imaging (NODDI) and arterial spin labeling (ASL) to examine whether the microstructure and perfusion status of the hippocampus are influenced by the presence of diffusion-restricted lesions. METHODS: Ten patients with a clinical diagnosis of TGA were evaluated by conventional MRI, NODDI, and ASL. The intracellular volume fraction (ICVF) and orientation dispersion index (ODI) on NODDI and cerebral blood flow (CBF) determined by ASL in the hippocampus were calculated and compared by diffusion-weighted imaging (DWI) positivity. Correlations among ICVF, ODI, and CBF were also analyzed. RESULTS: Three patients had typical unilateral DWI-positive lesions. No significant differences in any of the three parameters were detected between DWI-positive and DWI-negative hippocampi. A statistically significant correlation was detected between the ODI and CBF (R = .51, P = .021). CONCLUSIONS: The first NODDI and ASL study in patients after TGA demonstrated no obvious microstructural or perfusion abnormalities in the hippocampus with typical DWI-positive lesions, which may indicate that TGA does not cause destructive damage or involve baseline microstructure or perfusion abnormalities in the hippocampus in relation to diffusion-restricted lesions.

A bundle of polymer-coated filaments was successfully introduced as an extraction medium for the preconcentration of an aqueous solution of aromatic compounds. The extraction was simply carried out with pumping the aqueous sample solution to the extraction capillary at ambient temperature. The extracted analytes were sequentially eluted with a flow of pure water using temperature-programmed heating of the extraction capillary in an oven. The results clearly suggest that the polymer-coated fiber-packed capillary could be employed in the sample preparation process for the analysis of various aqueous samples. Introducing the fractions eluted from the fiber-packed capillary to a conventional microcolumn liquid chromatography (micro-LC) system via a home-made valve-based modulator, an on-line coupled extraction/separation system was developed and a possibility to a pseudo-two-dimensional (pseudo-2D) LC separation of aromatic compounds in aqueous matrices has also been demonstrated.
