Background: This in vivo study evaluated the effect of graphene oxide and graphite coatings, coupled with the micro-arc oxidation (MAO) surface roughening technique, known for their mechanical strength, chemical stability, and antibacterial properties. The main objective was to assess the degree of improvement in osseointegration of titanium implants resulting from these interventions. Materials and methods: In this study, 32 female rats were utilized and randomly allocated into four groups (n = 8 each): machined surface titanium implants (control), those roughened by the MAO method, those coated with graphene oxide-doped MAO, and those with a graphite-doped MAO coating. Titanium implants were surgically placed in the right tibia of the rats. Rats undergoing no additional procedures during the 4-week experimental period were sacrificed at the end. Then, the implants and surrounding bone tissues were separated and embedded in acrylic blocks for reverse torque analysis. Using a digital torque device, the rotational force was applied to all samples using a hex driver and racquet until implant separation from the bone occurred, with the corresponding values recorded on the digital display. Then, statistical analysis was performed to analyze the data. Results: No statistically significant difference between the groups was observed in the biomechanical bone-implant connection levels (N/cm) (P = 0.268). Post-hoc tests were not required because no discernible differences were identified between the groups. Conclusion: Within the scope of this study, implants treated with the MAO method, along with those coated with graphene oxide- and graphite-doped MAO method, did not exhibit significant superiority in terms of osseointegration compared to machined surface titanium implants.

This study investigated the protective effects of p-coumaric acid (PCA) against bisphenol A (BPA)-induced testicular toxicity in male rats. The rats were divided into control, BPA, BPA+PCA50, BPA+PCA100, and PCA100 groups. Following a 14-day treatment period, various analyses were conducted on epididymal sperm quality and testicular tissues. PCA exhibited dose-dependent cytoprotective, antioxidant, and anti-inflammatory effects, ameliorating the decline in sperm quality induced by BPA. The treatment elevated antioxidant enzyme activities (SOD, GPx, CAT) and restored redox homeostasis by increasing cellular glutathione (GSH) and reducing malondialdehyde (MDA) levels. PCA also mitigated BPA-induced proinflammatory responses while reinstating anti-inflammatory IL-10 levels. Apoptotic parameters (p53 and p38-MAPK) were normalized by PCA in BPA-treated testicular tissue. Immunohistochemical and immunofluorescent analyses confirmed the cytoprotective and anti-inflammatory effects of PCA, evidenced by the upregulation of HO-1, Bcl-2, and Nrf-2 and the downregulation of the proapoptotic gene Bax in BPA-induced testicular intoxication. PCA corrected the disturbance in male reproductive hormone levels and reinstated testosterone biosynthetic capacity after BPA-induced testicular insult. In silico analyses suggested PCA's potential modulation of the oxidative stress KEAP1/NRF2/ARE pathway, affirming BPA's inhibitory impact on P450scc. This study elucidates BPA's molecular disruption of testosterone biosynthesis and highlights PCA's therapeutic potential in mitigating BPA's adverse effects on testicular function, showcasing its cytoprotective, anti-inflammatory, and hormone-regulating properties. The integrated in vivo and in silico approach offers a comprehensive understanding of complex mechanisms, paving the way for future research in reproductive health and toxicology, and underscores the importance of employing BPA-free plastic wares in semen handling.

Cadmium (Cd) is a toxic heavy metal with significant environmental health hazards. It enters the body through various routes with tissue accumulation. The relatively longer half-life with slow body clearance significantly results in hepatotoxicity during its liver detoxification. Therefore, researchers are exploring the potential use of herbal-derived phytocomponents to mitigate their toxicity. Here, we investigated, for the first time, the possible ameliorative effect of the phytochemical Morin (3,5,7,29,49-pentahydroxyflavone) against acute Cd-induced hepatotoxicity while resolving its underlying cellular mechanisms in a rat animal model. The study involved 50 adult male Sprague-Dawley rats weighing 200-250 g. The animals were divided into five equal groups: control, Cd, Morin100 + Cd, Morin200 + Cd, and Morin200. The 2nd, 3rd, and 4th groups were intraperitoneally treated with Cd (6.5 mg/kg), while the 3rd, 4th, and 5th groups were orally treated with Morin (100 and 200 mg/kg) for 5 consecutive days. On the 6th day, hepatic function (serum ALT, AST, ALP, LDH enzyme activities, and total bilirubin level) testing, transcriptome analysis, and immunohistochemistry were performed to elucidate the ameliorative effect of Morin on hepatotoxicity. In addition to restoring liver function and tissue injury, Morin alleviated Cd-induced hepatic oxidative/endoplasmic reticulum stress in a dose-dependent manner, as revealed by upregulating the expression of antioxidants (SOD, GSH, Gpx, CAT, and Nrf2) and decreasing the expression of ER stress markers. The expression of the proinflammatory mediators (TNF-α, IL-1-ß, and IL-6) was also downregulated while improving the anti-inflammatory (IL-10 and IL-4) expression levels. Morin further slowed the apoptotic cascades by deregulating the expression of pro-apoptotic Bax and Caspase 12 markers concomitant with an increase in anti-apoptotic Blc2 mRNA expression. Furthermore, Morin restored Cd-induced tissue damage and markedly suppressed the cytoplasmic expression of JNK and p-PERK immunostained proteins. This study demonstrated the dose-dependent antioxidant hepatoprotective effect of Morin against acute hepatic Cd intoxication. This effect is likely linked with the modulation of upstream p-GRP78/PERK/ATF6 pro-apoptotic oxidative/ER stress and the downstream JNK/BAX/caspase 12 apoptotic signaling pathways.

This study investigated the effects of Selenium (Se) on testis toxicity induced by Acrylamide (ACR) in rats. In our study, 50 male adult rats were used, and the rats were divided into five groups; control, ACR, Se0.5 + ACR, Se1 + ACR, and Se1. Se and ACR treatments were applied for 10 days. On the 11th day of the experimental study, intracardiac blood samples from the rats were taken under anesthesia and euthanized. Sperm motility and morphology were evaluated. Dihydrotestosterone, FSH, and LH levels in sera were analyzed with commercial ELISA kits. MDA, GSH, TNF-α, IL-6, and IL-1ß levels and SOD, GPx, and CAT, activities were measured to detect the level of oxidative stress and inflammation in rat testis tissues. Expression analysis of HSD17B1, StAR, CYP17A1, MAPk14, and P-53 as target mRNA levels were performed with Real Time-PCR System technology for each cDNA sample synthesized from rat testis RNA. Testicular tissues were evaluated by histopathological, immunohistochemical, and immunofluorescent examinations. Serum dihydrotestosterone and FSH levels decreased significantly in the ACR group compared to the control group, while LH levels increased and a high dose of Se prevented these changes caused by ACR. A high dose of Se prevented these changes caused by ACR. ACR-induced testicular oxidative stress, inflammation, apoptosis, changes in the expression of reproductive enzymes, some changes in sperm motility and morphology, DNA, and tissue damage, and Se administration prevented these pathologies caused by ACR. As a result of this study, it was determined that Se prevents oxidative stress, inflammation, apoptosis, autophagy, and DNA damage in testicular toxicity induced by ACR in rats.

BACKGROUND We conducted a finite element analysis to evaluate stress levels in incisor teeth restored with custom polyetheretherketone (PEEK) dental post-cores compared to conventional post-cores. MATERIAL AND METHODS Using micro-computed tomography (µCT) imaging data, a 3D model of a maxillary incisor was created. For each material type, 3D mesh models were developed via specialized software. Two post diameters, 2.5 mm and 3.5 mm, were considered. Five different post materials were examined: Unfilled polyetheretherketone (Group UP); Glass fiber-reinforced polyetheretherketone (Group GP); Carbon fiber-reinforced polyetheretherketone (Group CP); Metal (Group M); and Zirconia ceramic (Group Z). Each model underwent finite element analysis, after which the von Mises equivalent stress values were determined. RESULTS For models involving both wide and narrow diameter posts across the crown, crown cement, post cement, and dentin, PEEK posts (Group UP, GP, and CP) exhibited higher von Mises stress values than Groups Z and M. However, the reverse trend was noticed in the post model itself. In the post cement model, stress values appeared similar only for the narrow-diameter post groups. Notably, results for Groups Z and M were largely consistent with each other. CONCLUSIONS PEEK posts, which have a lower modulus of elasticity, demonstrated different stress values when contrasted with zirconia and metal posts. As the post diameter expanded, the residual dentin decreased, influencing the stress values among various materials. Further in vitro and clinical examinations are essential to comprehensively understand PEEK posts.

Sexual behavior in animals is important in ensuring the continuity of the generation. These behaviors differ in animal species. Sexual behaviors are shaped under the control of the reproductive system. Physiological stimuli produced by the reproductive system find their counterparts in the organism as reproductive activity. Reproductive activity display a critical role by transferring on the genetic heritage of organisms to the next generations. This activity, which is built on delicate balances, is associated with many systems in the organism. Nervous system, hormonal system, and circulatory system are the main ones. The regular formation of the reproductive activity in species is due to the effect of various factors. In domestic mammals, the reproductive activity is regulated by hormones secreted from brain and endocrine glands. Many hormones have duties in terms of the sustainability of reproductive activity. GnRH is the main hormone responsible for initiating this reproductive activity. Gonadotropin-releasing hormone (GnRH), which is a small molecule peptide from certain nerve cells in the nucleus infundibularis region of the hypothalamus and consists of different amino acids, is secreted under the influence of smell, temperature, light, and physical stimulation. Besides, GnRH release is controlled by various neurotransmitters (adrenaline, noradrenaline, dopamine, acetylcholine, serotonin). On the other hand, various genetic factors in secretory glands, gonadal cells, reproductive tissues can lead to significant changes on reproductive activity through specific molecular pathways and mechanisms.

Bisphenol A (BPA) is one of the chemicals that cause dysfunction and infertility in testicles. Therefore, it is crucial to develop effective treatments against this damage. In this study, the effects of Hesperidin (HESP), a flavonoid in testicular toxicity induced by BPA in rats, on oxidative stress, inflammation, apoptosis, histological damage, spermatogenesis, steroidogenic enzymes and reproductive hormones were investigated. Our study used 52 Sprague Dawley male rats weighing 250-300 g, and four experimental groups were formed. From the experimental groups, 1 ml of olive oil was administered to the control group, HESP at a dose of 50 mg/kg to the HESP group, BPA at a dose of 100 mg/kg to the BPA group, HESP at a dose of 50 mg/kg to the BPA + HESP group and 100 mg/kg BPA was administered intragastrically (ig) for 14 days. We determined that BPA administration causes apoptosis, histological damage, inflammation, oxidative stress and toxic effects on spermatogenesis and steroidogenic enzymes in testicles. We observed that the administration of HESP with BPA attenuated oxidative stress, inflammation and apoptosis resulting in therapeutic effects on both steroidogenic enzymes and spermatogenesis and reproductive hormones (FSH, LH and testosterone). Our findings from this study clearly showed that while HESP treatment alleviates oxidative damage, inflammation and apoptosis in testicles of rats treated with BPA, it has regulatory effects on steroidogenic enzymes, spermatogenesis and serum reproductive hormones.

PURPOSE: To evaluate the fracture strength of polyetheretherketone (PEEK) single crowns veneered with different materials produced by computer-aided design (CAD)-computer-aided manufacture (CAM) after aging. METHODS: 60 stainless-steel master dies were prepared with a 1 mm-wide deep chamfer. Sixty PEEK frameworks were produced with a CAD-CAM system for the right maxillary first molar tooth on the dies. PEEK frameworks were divided into six groups (n= 10) according to veneering materials (five CAD-CAM materials and a resin composite). Group ZR: monolithic zirconia (Upcera ST-Color); Group EC: lithium disilicate glass-ceramic (IPS e.max CAD); Group LU: resin nano-ceramic (Lava Ultimate); Group VM: feldspathic ceramic (Vitablocs Mark II); Group VS: zirconia-reinforced lithium silicate glass-ceramic (VITA Suprinity); and Group CR: indirect resin composite (Gradia). All samples were subjected to a fracture strength test in a universal test device after thermo-mechanical aging and then the results were analyzed statistically using one-way ANOVA and Tukey's post hoc test. In addition, post-fracture analyses of all samples were performed with a stereomicroscope. RESULTS: The differences in fracture strength values of different veneer materials in single crowns with a PEEK framework were statistically significant (P< 0.05). Fracture strength was highest in Group ZR (1665 N), followed by Groups LU (1440 N), EC (1249 N), CR (918 N), VS (754 N), and VM (655 N). CLINICAL SIGNIFICANCE: PEEK frameworks may have the potential to be used with many veneer materials of different content and properties in fixed partial prostheses.

PURPOSE: To evaluate the effects of ultrasonic scaling (US) and air polishing (AP) on four polyetheretherketone (PEEK) composites. METHODS: One hundred-twenty 15 × 3 mm discs of PEEK specimens were divided into four groups (n=30): Unfilled PEEK(U-PEEK), carbon fiber-reinforced PEEK(CFR-PEEK), glass fiber-reinforced PEEK(GFR-PEEK), and ceramic-filled PEEK(CF-PEEK). Each group was further divided into three subgroups (n = 10): control, US, and AP. Profilometry and scanning electron microscopy were used to analyze and evaluate surface roughness (SR). Statistical analyses of the data obtained were conducted using Shapiro-Wilk, Welch, and Games-Howell tests. RESULTS: When the SR values of the specimens with US cleaning were evaluated, a statistically significant difference was found between the groups (P< 0.05). When the SR values of the specimens with AP cleaning were analyzed, there was a statistically significant difference in the CF-PEEK group (P< 0.05), whereas the other groups were not significantly different (P> 0.05). More studies are needed on CFR-PEEK and GFR-PEEK materials offered as alternatives to CF-PEEK in dentistry. CLINICAL SIGNIFICANCE: Dental instruments affect the different PEEK materials, as well as causing surface roughness in many restorative materials used in dentistry. Surface roughness that occurs in dental restorations can cause bacterial adhesion. It is clinically important to choose the dental instrument according to the type of PEEK used for dental implant or prosthetic restoration in the clinic.

Abstract The aim of this in vitro study was to compare the long-term effect of overnight use of denture cleansers with different chemical compositions on the color stability of denture base polymers (DBPs). The four DBPs evaluated were PEEK (PK group), thermoinjection-molded polyamide (PA group), auto-polymerized polymethylmethacrylate (PMMA) (AP group), and heat-polymerized resin PMMA (HP group). The cleaning agents used were Corega tablet (CT), Protefix tablet (PT), and 0.5% sodium hypochlorite (NaOCl) solution (SH). Distilled water (DW) served as a control. Forty-eight disc-shaped specimens (10mm × 2mm) were prepared from each DBP and randomly immersed in the different storage media (n=12 in each group). Color measurements of the specimens before and after immersion in the denture cleansers were made using a spectrophotometer (VITA Easyshade V). The color changes were measured and recorded in L* a* b*. After 120 d, the CIEDE2000 formula was used to calculate color changes (ΔE00). For analysis of the data, a multivariate analysis of variance was used (p<0.05). The results of the statistical analysis revealed significant color change differences in the DBPs immersed in the various denture cleansers (p<0.05). Groups PA and PK showed the highest values for all solutions. AP-SH showed the highest ΔE00 value in group AP, no significant difference was found between other solutions. In group HP, there was no statistically significant difference between the average ΔE00 values of all solutions. Long-term daily use of the denture cleansers affected the color stability of all the DBPs, although the level of color change was acceptable. Laboratory and clinical studies on the color stability of PEEK are needed to confirm the results of this study.


Resumen El objetivo de este estudio in vitro fue comparar el efecto a largo plazo del uso nocturno de limpiadores de prótesis con diferentes composiciones químicas sobre la estabilidad del color de los polímeros base de prótesis (DBP). Los cuatro DBP evaluados fueron PEEK (grupo PK), poliamida moldeada por termoinyección (grupo PA), polimetilmetacrilato (PMMA) autopolimerizado (grupo AP) y resina PMMA termopolimerizada (grupo HP). Los agentes de limpieza utilizados fueron Corega (CT), Protefix (PT) y la solución de hipoclorito de sodio (NaOCl) al 0,5% (SH). El agua destilada (DW) sirvió de control. Se prepararon 48 especímenes en forma de disco (10mm × 2mm) de cada DBP y se sumergieron al azar en los diferentes medios de almacenamiento (n=12 en cada grupo). Las mediciones del color de las muestras antes y después de la inmersión en los limpiadores de dentaduras se realizaron con un espectrofotómetro (VITA Easyshade V). Los cambios de color se midieron y registraron en L* a* b*. Después de 120 d, se utilizó la fórmula CIEDE2000 para calcular los cambios de color (ΔE00). Para el análisis de los datos, se utilizó un análisis multivariante de la varianza (p<0,05). Los resultados del análisis estadístico revelaron diferencias significativas en el cambio de color de los DBP sumergidos en los distintos limpiadores de dentaduras (p<0,05). Los grupos PA y PK mostraron los valores más altos para todas las soluciones. AP-SH mostró el valor ΔE00 más alto en el grupo AP, no se encontraron diferencias significativas entre las demás soluciones. En el grupo HP, no hubo diferencias estadísticamente significativas entre los valores medios de ΔE00 de todas las soluciones. El uso diario a largo plazo de los limpiadores de dentaduras afectó a la estabilidad del color de todos los DBP, aunque el nivel de cambio de color fue aceptable. Se necesitan estudios de laboratorio y clínicos sobre la estabilidad del color del PEEK para confirmar los resultados de este estudio.

Objective: Investigations of the effects of proton pump inhibitors (PPIs) on bone healing have revealed that they affect bone regeneration negatively. The exact mechanism by which this adverse effect on bone tissue is not known. The aim of this study is to biomechanic and biochemical investigation of the effects of the PPIs on guided periimplant bone regeneration. Material & methods: Spraque dawley rats were divided controls (n = 8): there is no treatment during 8 week experimental period, PPI- Dosage 1 (n = 8) and Dosage 2 (n = 8): 5 mg/kg and 10 mg/kg omeprazol applied 3 times in a week with oral gavage during 8 weeks respectfully. Bone defects created half of the implant length circumferencial after implant insertion and defects filled with bone grafts. After experimental period the rats sacrified and implants with surrounding bone tissues were removed to reverse torque analysis (Newton), blood samples collected to biochemical analysis (glucose, AST, ALT, ALP, urea, creatinin, calcium, P). Results: Biomechanic reverse torque values did not revealed any statistical differences between the groups (P > 0,05). Conclusion: According the biomechanical and biochemical parameters PPIs does not effect the periimplant guided bone regeneration.

OBJECTIVES: The aim of this experimental animal study was to evaluate the effects of systemic propranolol on new bone formation in peri-implant bone defects. MATERIAL AND METHODS: Implant slots were created 4mm long and 2.5 mm wide. After the titanium implants were placed in the sockets, 2 mm defects were created in the neck of the implants. Bone grafts were placed in these defects. Then the rats were randomly divided into three equal groups: control (n = 8), propranolol dose-1 (PRP-1) (n = 8), and propranolol dose-2 (PRP-2) (n = 8) groups. In the control group, the rats received no further treatment during the eight-week experimental period after the surgery. The rats in the PRP-1 and PRP-2 groups were given 5 mg/kg and 10 mg/kg propranolol, respectively, every three days for the eight-week experimental period after the surgery. At the end of the experimental period, the rats were euthanized. Blood serum was collected for biochemical analysis, and the implants and surrounding bone tissues were used for the histological analysis. RESULTS: There were no significant differences in the histological analysis results and the biochemical parameters (alkaline phosphatase, calcium, creatinine and phosphorus) of the groups (P > 0.05). Also, in the test groups, there was numerically but not statistically more new bone formation detected compared with the controls. CONCLUSIONS: Within the limitations of this study, propranolol did not affect the new bone formation in peri-implant defects.

BACKGROUND: This study aimed to investigate the effects of systemic omeprazole treatment on the osseointegration of titanium implants. MATERIAL AND METHODS: After surgical insertion of titanium implants into the metaphyseal part of rats' both right and left tibial bones, the animals were randomly divided into three equal groups: control (n = 8), omeprazole dosage-1 (n = 8) (OME-1), and omeprazole dosage-2 (n = 8) (OME-2) and totally 48 implants were surgically integrated. The rats in the control group received no treatment during the four-week postoperative experimental period. In the OME-1 and OME-2 groups, the rats received omeprazole in doses of 5 and 10 mg/kg, respectively, every 3 days for 4 weeks. After the experimental period, the rats were euthanized. One rat died in each group and the study was completed with seven rats in each group. Blood serum was collected for biochemical analysis, and the implants and surrounding bone tissue were used for biomechanical reverse-torque analysis. In the biomechanical analysis, implants that were not properly placed and were not osseointegrated were excluded from the evaluation. RESULTS: One-way analysis of variance and Tukey's honestly significant difference test and Student's t test were used for statistical analysis. The reverse-torque test (control (n = 9), OME-1 (N = 7), and OME-2 (n = 7)) analysis of biochemical parameters (alkaline phosphatase, calcium, phosphorus, aspartate aminotransferase, alanine amino transferase, urea, and creatinine) revealed no significant differences between the groups (control (n = 7), OME-1 (N = 7), and OME-2 (n = 7)) (P > 0.05). CONCLUSIONS: Omeprazole had no biomechanical or biochemical effects on the osseointegration process of titanium implants.

Nephrotoxicity is a very important complication of 5-fluorouracil (5-FU)-treated cancer patients. Increased oxidative stress, kidney damage, and apoptosis play an important role in the pathogenesis of nephrotoxicity caused by 5-FU. In this study, protective effects of two natural compounds, hesperidin and curcumin, on experimentally induced kidney damage in mice with 5-FU were determined. Application of 5-FU resulted in severe histopathological changes and severe renal failure with increased serum urea and creatinine levels. Also, 5-FU-induced kidney damage, increased levels of malondialdehyde (MDA), decreased superoxide dismutase (SOD), catalase (CAT), glutathione reductase (GR) activity, and glutathione (GSH) level have been demonstrated. Also, where 5-FU is in the concentration of caspase-3 and 8-OHdG immune-positive cells and therefore causes apoptosis and DNA damage in kidney tissue cells. However, especially high doses of hesperidin and curcumin treatment significantly improved 5-FU-induced oxidative stress/lipid peroxidation, apoptosis/DNA damage, and renal dysfunction. Based on these data, our results suggest that hesperidin and curcumin may be used as new and promising agents against 5-FU-induced nephrotoxicity.

ABSTRACT: This study aimed to investigate the effects of systemic propranol on the osseointegration of titanium implants. After the surgical insertion of titanium implants into the metaphyseal part of the tibial bone, the rats were randomly divided into 2 equal groups: the control (CNT) (n = 10) and propranol group (P) (n = 10); CNT: Rats received no further treatment during the 4 week experimental period after surgery. Rats received 10 mg/kg propranol in every day during the 4 week experimental period in PRP group after the surgical insertion of the implants. After the experimental period, the rats were euthanized, blood serum were collected to biochemical analysis and the implants and surrounding bone tissues were used for the histopathologic analysis. To analysis of the data between tests and controls student T test was used. There were no significant differences in the biochemical parameters (alcaline phosphatase, calcium, phosphor) of the groups (P > 0.05). Bone implant connection (BIC) ratios was detected higher in test animals compared with the controls (P < 0.05). Systemic propranolol may increases titanium implant osseointegration.

We sought to determine the effects of selenium (Se) on acrylamide (ACR)-induced nephrotoxicity in rats. In our study, 50 adult male Sprague-Dawley rats weighing 200-250 g were randomly divided into five groups. The control group was given intra-gastric (i.g.) saline (1 mL) for 10 days. The ACR group was given i.g. ACR in saline (38.27 mg/kg titrated to 1 mL) for 10 days. The Se0.5 + ACR and Se1 + ACR groups were administered Se in saline (0.5 and 1 mg/kg, respectively) for 10 days and given i.g. ACR (38.27 mg/kg) one hour after the Se injections. The Se1 group was administered i.g. Se (1 mg/kg) for 10 days. On day 11, intracardiac blood samples were obtained from the rats while they were under anesthesia, after which they were euthanized by decapitation. Urea and creatinine concentrations of blood serum samples were analyzed with an autoanalyzer. Enzyme-linked immunosorbence immunosorbent assay (ELISA) was used to quantify malondialdehyde (MDA), superoxide dismutase (SOD), glutathione (GSH), glutathione peroxidase (GPx), catalase (CAT), tumor necrosis factor-α (TNF-α), nuclear factor-κB (NF-κB), interleukin (IL)-33, IL-6, IL-1ß, cyclooxygenase-2 (COX-2), kidney injury molecule-1 (KIM-1), mitogen-activated protein kinase-1 (MAPK-1), and caspase-3 in kidney tissues. Renal tissues were evaluated by histopathological and immunohistochemical examinations for 8-hydroxylo-2'-deoxyguanosin 8-hydroxy-2'-deoxyguanosine (8-OhDG) and Bax. Serum urea and creatinine levels were higher in the ACR group than in the control, and these ACR-induced increases were prevented by high doses of Se. Additionally, ACR induced the renal oxidative stress, inflammation, apoptosis, and damage to DNA and tissue; likewise, these were prevented by high doses of Se. Taken with ACR, Se confers protection against ACR-induced nephrotoxicity in rats by reducing oxidative stress, inflammation, apoptosis, and DNA damage.

After the surgical insertion of titanium implants into the metaphyseal part of the tibiae of rats, the rats were randomly divided into three equal groups: the control (n = 8), propranolol dosage-1 (PRP-1) (n = 8), and propranolol dosage-2 (PRP-2) (n = 8) groups. After the surgical insertion of the implants, the rats in the PRP-1 and PRP-2 groups were given 5 mg/kg and 10 mg/kg propranolol, respectively, every 3 days for the 4-week experimental period. After the experimental period, the rats were euthanized. Blood sera were collected for biochemical analysis, and the implants and surrounding bone tissues were used for the biomechanical reverse torque analysis. One-way ANOVA and Tukey's honest significant difference test were used for statistical analysis. The student t-test was used to analyze the data obtained from the tests and the controls. There were no significant differences in the reverse torque analysis results and the biochemical parameters (alkaline phosphatase, calcium, and phosphorus) of the groups (P > 0.05). Alkaline phosphatase was, however, found to be higher in test animals compared to the controls (P < 0.05). Also, propranolol did not biomechanically affect the osseointegration of titanium implants, while alkaline phosphatase activity was higher in the test animals.

ABSTRACT: The aim of this study was to compare the biomechanical osseointegration of titanium implants after guided bone regeneration (GBR) with a hydroxyapatite graft, deproteinized bovine bone graft, human-derived allograft, and calcium sulfate bone graft. Thirty-two female Sprague Dawley rats were divided into four groups, each containing eight (n = 8) rats: hydroxyapatide (HA), deproteinized bovine bone graft (DPBB), allograft (ALG), and calcium sulfate. Bone defects were created in the tibia of the rats, which were grafted with HA, DPBB, ALG, or CP bone grafts for the purpose of GBR. Ninety days after surgery, machine-surfaced titanium implants were inserted into the area where GBR had been undertaken. After 90 days of the surgical insertion of the implants, the rats were sacrificed, the implants with surrounding bone tissue were removed, and biomechanical osseointegration (N/cm) analysis was performed. No statistically significant differences were found among the groups in osseointegration (N/cm) three months after the GBR procedures (P > 0.05). According to the biomechanical results, none of the grafts used in this study was distinctly superior to any of the others.

PURPOSE: To compare the stresses generated after the use of low elastic modulus polyetheretherketone (PEEK) as dental post and crown material with glass fiber post and metal supported ceramic crowns using finite element analysis. METHODS: First an image of a maxillary central incisor taken on micro-computed tomography (micro-CT) was uploaded to a computer program. Modeling was made of all the materials in the computer environment and 3-dimensional (3D) virtual models were obtained. Using two different post materials (glass fiber and PEEK post) and two different crown materials (metal supported ceramic and PEEK crown), the modeling of the maxillary central tooth was completed and a total of four groups were obtained. Finally, 135° and 100 N forces were applied from the midpoint of the palatal surface. RESULTS: The FEA analysis showed the use of PEEK as post material decreased the stress values in the post, post cement and composite core models and did not have any significant change in the dentin and crown cement models. The use of composite veneer with PEEK substructure as crown material reduced stress values in dentin, the post and composite core and increased stress values in both the post cement and the crown cement. CLINICAL SIGNIFICANCE: Since the elastic modulus of the PEEK material is very close to dentin tissue, finite element stress analysis, less stress formation in the tooth and post core restoration compared to other post systems was observed. In this context, PEEK post may reduce irreversible complications.

INTRODUCTION: The aim of this study was to make a quantitative comparison of the cytotoxic potentials of four different polymerised and unpolymerised self-etching adhesives which were newly used clinically. MATERIALS AND METHODS: Cytotoxic effects of both polymerised and unpolymerised forms of all test adhesives were evaluated against L929 cell line using the MTT test. The activity for unpolymerised adhesives was assessed in different doses and incubation times manner. On the other hand, cytotoxicity of the polymerised adhesives prepared at different extraction times were evaluated as dependent on incubation times. Two-Way Variance Analysis and Bonferroni post-test was used for statistical evaluation. RESULTS: There were statistically significant differences between the groups (p < 0.05). In general, it was shown that unpolymerised and polymerised forms of each of the test compounds exhibited a time-dependent cytotoxic effect. However, the effect on polymerised forms was found to be independent of the duration of the extraction, while the effect on the unpolymerised forms increased dose-dependently. It was also determined that the most cytotoxic material in the unpolymerised form was Dentsply and in the polymerised form was Tokuyama. CONCLUSIONS: Dentsply should be preferred over Tokuyama to be able to provide clinically long-lasting restorations.
