RESUMO

Coxiella burnetii is a zoonotic intracellular bacterium that is widely distributed and affects domestic animals, wildlife, humans and non-mammalian species. This systematic review was aimed at synthesizing research findings on C. burnetii in both domestic and wild animals of South Africa. The systematic review protocol was registered with Open Society Foundations of systematic reviews ( https://doi.org/10.17605/OSF.IO/8WS ). PRISMA guidelines were followed to collect and evaluate relevant scientific articles published on C. burnetii infecting domestic and wild animals in South Africa. Published articles were sourced from five electronic databases, namely, Google Scholar, PubMed and ScienceDirect, EBSCO and Scopus. Results showed 11 eligible studies involving four domestic animals, three wild animals and one ectoparasite species from seven provinces across South Africa. The occurrence of C. burnetii infection was high in Ceratotherium simum (white rhinoceros) (53.9%), medium in sheep (29.0%) and low in pigs (0.9%). Limpopo province (26%) had the most recorded infections followed by KwaZulu-Natal (19%) and Free State (3%) had the least reported occurrence of C. burnetii. The current study discovered that there is scarcity of published research on prevalence and distribution of C. burnetii infecting domestic and wild animals in South Africa, and this is of concern as this bacterium is an important zoonotic pathogen of "One Health" importance.

Background and Aim: Anthelmintic resistance (AR) and acaricide resistance (ACR) pose great economic threat to communal livestock raised by rural communities, limiting sustainable production. This study was conducted to assess the occurrence of AR and ACR against nematodes and ticks that infest small ruminants (sheep and goats) from small-scale farming communities in the North West Province of South Africa, as well as document the associated risk factors. Materials and Methods: The study was conducted on small-scale farming locations in two districts of the North West Province, namely, Dr. Ruth Segomotsi Mompati district and Dr. Kenneth Kaunda district, from November 2019 to March 2020. A questionnaire survey based specifically on antiparasitic treatment and related management practices was administered to 86 small-scale farmers. A fecal egg count reduction test (FECRT) was used to determine in vivo AR in small ruminants against benzimidazole (BZD), levamisole, and macrocyclic lactone on nine ruminant farms. Then, deoxyribonucleic acid was extracted from L3 larvae and resistant nematodes were identified using a polymerase chain reaction, targeting the internal transcribed spacer 2 gene. An egg hatch assay (EHA) and a larval mortality assay (LMA) were used to determine in vitro AR against thiabendazole (TBZ and BZD) in the same farms. Acaricide resistance against fluazuron-flumethrin (Drastic Deadline eXtreme) pour-on was assessed using an adult immersion test (AIT) on Rhipicephalus evertsi. Results: Questionnaire results indicated that most farmers (89%) relied solely on anthelmintics. Farmers used visual appraisal to estimate the dosage, which is the primary cause of resistance. The FECRT revealed AR in all the farms. Egg hatch assay results revealed AR development against TBZ in all districts, with >95% of the eggs hatching at variable doses. Larval mortality assay results revealed the development of resistance against BZD, with 50% of L3 larvae surviving at different doses in all farms. Adult immersion test results indicated that fluazuron-flumethrin (>99%) exhibited high acaricidal efficacy against R. evertsi by inhibiting tick oviposition. Conclusion: This investigation found that sheep and goats in the studied areas are developing AR to gastrointestinal parasites. The findings of in vivo tests showed resistance with fecal egg count reduction percentage of <95% or lower confidence limit of <90%. The results of EHA and LMA revealed no evidence of inhibition of egg development and larval mortality, indicating the development of resistance. Acaricide resistance was not detected against fluazuron-flumethrin, which is commonly used in the study areas. Thus, developing management methods for these economically significant livestock nematodes, including teaching small-scale farmers how to properly administer anthelmintics and acaricides to their livestock, is urgently needed.

The study of Toxoplasma gondii genotypes is beneficial for detecting strains linked to increased disease severity and uncovering the processes involved in the transmission and distribution of this zoonotic parasite. A systematic review of literature was conducted to investigate the present status of T. gondii genetic diversity in African countries and among host species on the continent. Data from the results in the included studies were sorted, reviewed and descriptively analysed using tables, graphs and maps. Results indicate that there is a relative amount of genetic diversity with a clear difference in the population structure between geographical regions and the propensity for unique and regional genotypes to be predominant in tropical rainforest biomes, near the equator. From a clinical perspective, connections between specific T. gondii genotypes and disease manifestations were found. Theories are outlined on the dissemination of African T. gondii genotypes to other continents. The overrepresentation of samples from one geographical area and dissimilar genotyping methodologies creates challenges when concluding on the genetic diversity of T. gondii in Africa. The need for uniform genotyping methods with a continent-wide sampling of an extensive host range involving humans, domestic animals and wildlife is emphasized.

Free-range chickens are an integral part of poultry production in many developing countries. In the Mountain Kingdom of Lesotho, the majority of the population own free-range chickens, which serve a variety of purposes including being a source of meat, eggs and use for cultural rituals amongst others. However, there is lack of scientific studies on occurrence of parasitic infections on free-range chickens in Lesotho. The aim of this study was to document common gastrointestinal parasites infecting free-range chickens in four villages of Mafeteng District in Lesotho. A total number of 462 pooled faecal samples were collected from various households in HaKubutu (n = 114), HaMatjeka (n = 120), HaMpalipali (n = 120) and Thabang Villages (n = 108) which were subjected to microscopic examination using McMaster technique. The prevalence of gastrointestinal parasite infection was as follows: Eimeria tenella (12.8%), Ascaridia galli (10.4%) and Heterakis gallinarum (5%). The prevalence for H. gallinarum and Ascaridia galli were comparatively higher during the hot-wet season (7.1% and 2.8% respectively) than the cold-dry season (3.2% and 1.9% respectively) and varied significantly (P < 0.05). For E. tenella, the oocysts per gram were slightly higher in the cold-dry season than the hot-wet season. Polymerase chain reaction only amplified DNA from six (29%) adult A. galli worms of which two amplicons were successfully sequenced. The obtained cytochrome C oxidase subunit 1 partial gene sequences displayed 98-100% identity with South African A. galli isolates. This is the first scientific study on prevalence and molecular characterization of nematodes and coccidia species infecting free-range village chickens in Lesotho. The findings can be used to review management of gastrointestinal nematodes and protozoal parasites of free-range chickens in Lesotho.

BACKGROUND AND AIM: Previous studies recorded the prevalence of gastrointestinal nematodes (GIN) in Limpopo Province. However, the studies did not address the seasonal patterns of infection and did not cover all districts of Limpopo Province, namely; Capricorn, Sekhukhune, Waterberg, Mopani, and Vhembe. It is, therefore, important to provide up to date information on the prevalence and seasonal occurrence data of GIN in all districts of Limpopo province. The present study was conducted to determine the occurrence of anthelmintic resistance (AR) and document the prevalence of GIN infecting sheep in five districts of Limpopo Province, South Africa. MATERIALS AND METHODS: Forty animals in each district were used for fecal egg count reduction test (FECRT) to determine AR against ivermectin (0.2 mg/kg), levamisole (LEV) (5 mg/kg), and albendazole (7.5 mg/kg). Egg hatch test (EHT) was used to determine AR against thiabendazole (TBZ) and micro-agar larval development test (MALDT) was used for both TBZ and LEV. Naturally, infected sheep (n=780) were sampled for prevalence across five districts of Limpopo. FAMACHA© eye-color score estimations were also performed for each study animal. RESULTS: FECRT showed occurrence of AR in most of the districts and a few with suspected resistance. EHT results showed AR development against TBZ for all districts, while the MALDT showed no AR against LEV in all districts, but detected AR against TBZ in Sekhukhune, Capricorn, and Waterberg. Haemonchus contortus was the most resistant species. A high nematode prevalence (88-100%) and 1210-1861 eggs per gram (EPG) was observed in all districts during the hot wet season, decreasing to 75-80% (453-1202 EPG) during the cold dry season. The sheep revealed a FAMACHA© mean score of 3, indicating mild anemia during the hot wet season except for Vhembe district that revealed a FAMACHA© mean score of 4 during the hot wet season, indicating anemia. CONCLUSION: AR recorded in Limpopo Province may be due to under-dosing caused by lack of weighing equipment and high treatment frequencies due to lack of proper training on anthelmintic use. The detection of AR in Limpopo is an important finding because it will help in outlining effective management systems against GIN.

Porcine and human cysticercosis, caused by the larval stage of tapeworm Taenia solium, is a zoonosis in southern Africa and known to be endemic in South Africa, mainly in Eastern Cape Province. No efforts to control or eradicate this parasite have been made, despite the increasing occurrence in most Eastern Cape districts, except for routine meat inspection at local abattoirs. The parasite poses a potentially serious agricultural problem, public health risk and economic loss amongst Eastern Cape smallholder pig production communities. The objective of this study was to determine the effectiveness of routine meat inspection for the detection of porcine cysticercosis in pigs from rural smallholder/subsistence production systems in Eastern Cape Province villages. The effectiveness of meat inspection, by registered meat inspectors, in the detection of pigs infected with T. solium cysts was assessed and compared with whole carcass dissection as the "gold standard" method. The commercial antigen enzyme linked immunosorbent assay (B158/B160 Ag-ELISA) kit screened all the slaughtered animals. The proportion of pigs found infected with T. solium cysts, as measured by meat inspection, was lower (5%, 9/180) than with carcass dissection (18.9%, 34/180) and B158/B60 Ag-ELISA test (21.6%, 38/176). Four out of 180 carcasses were heavily infested with T. solium cysts, evenly distributed throughout the carcasses, to a level impossible to enumerate. Of the remaining 176 carcasses, approximately 526 cysticerci, distributed at various anatomical regions of the pig, were counted during carcass dissection. Sites with higher cyst counts, such as the back and hind leg, do not form part of the normal meat inspection regime. The level of agreement (Kappa statistic) between dissection (gold standard) and meat inspection of the two districts was negative (-0.1955). There was a slight agreement in the Kappa statistic (0.0328) between dissection and B158/B60 Ag-ELISA. This study confirms that current meat inspection procedures alone are not sufficiently sensitive to detect all cases of porcine cysticercosis at the abattoirs and require modifications, or should be supplemented by other methods. A risk-based meat safety assurance system, such as HACCP, that considers specific food safety aspects before and after the abattoir (point of slaughter) should be followed. Before slaughter, aspects such as origin, husbandry practices and on-farm animal health control should be considered; after slaughter, the abattoir should inform the next entity in the supply chain of the limitations of meat inspections and the real meaning of an "Approval" stamp. New validated testing methods that can be routinely used should be developed, and government should develop policies and legislation that promotes a risk-based meat safety assurance system throughout the food supply chain.

Anthelmintic treatment is the most common way of controlling nematode infections in ruminants even though several countries have reported anthelmintic resistance (AR), resulting in limitation for sustainable small ruminant production. The aim of the present study was to evaluate the knowledge of resource-poor sheep farmers in Limpopo province of South Africa on the use of anthelmintics. A questionnaire regarding helminthosis control practices was administered to small ruminant farmers in five districts of Limpopo province namely Capricorn, Sekhukhune, Waterberg, Vhembe, and Mopani. A total of 77 resource-poor farmers were interviewed between June and August of 2017 using a structured questionnaire with a combination of qualitative and quantitative open-ended questions. The interviewed farmers were divided into three groups based on their farming experience (< 5; 6-10, and ˃ 10 years of farming experience). Limited farming experience was shown as one of the risks, as farmers that owned sheep for less than 10 years could not identify the symptoms of gastrointestinal parasites infection and did not know how nematodes are transmitted to animals. However, no significant difference (p < 0.05) was found to exist between the three groups of farmers in terms of clinical signs identification and correct application of anthelmintics. About 43% of the respondents were unaware of gastrointestinal nematodes (GI) that infect sheep, could not identify the clinical symptoms of gastrointestinal nematodes infection, and only 34% knew how animals become infected. Although 67.5% of farmers mentioned that they never dose their sheep, 32.5% use anthelmintics at varying times in a year. None of the farmers weighed their sheep before dosing them instead visual appraisal of individual weight was the most common means of estimating the anthelmintic dose. The above information is an indication of risks associated with possible occurrence of anthelmintic resistance in the study areas. There is therefore, a need to give training to resource-poor farmers of small stock on proper application of anthelmintic treatment and to educate them on how to prevent development of AR. Future studies on AR should also be conducted in the province in flocks with high-treatment frequencies to establish the occurrence of AR using both in vivo and in vitro methods. The most common risk factor associated with the occurrence of AR in all the five districts of Limpopo province was found to be the use of anthelmintics without weighing the animals to determine the correct dosage.
