RESUMO

Marchalina hellenica (Hemiptera: Marchalinidae), an endemic species in Greece and Turkey, is a major contributor to the annual honey production in its native range. However, in the areas that it invades, lacking natural enemies, it has detrimental effects on pine trees and potentially contributes to tree mortality. Although it was originally reported as thelytokous, males were later reported in Turkey and on several of the islands of Greece. To further disambiguate the exact parthenogenetic reproduction strategy of M. hellenica, we studied the emergence pattern of male individuals in Greece for two consecutive years (2021 and 2022). Furthermore, we examined the genetic variation among 15 geographically distant populations of M. hellenica in Greece using a mitochondrial DNA marker and compared the results with data from Turkey. The findings of this study document the existence of an additional M. hellenica population in its native range that repeatedly produces males, apart from the areas of Greece and Turkey in which they were initially reported, suggesting that males play a major, so far unknown role in the reproduction of this species. The populations in Greece and Turkey exhibited a strong genetic affinity, while human-aided dispersal seems to have obscured the genetic pattern acquired.

The American Academy of Ophthalmology and the National Heart, Lung and Blood Institute recommend patients with sickle cell disease (SCD) undergo dilated funduscopic exams (DFE) every 1-2 years to screen for sickle retinopathy. There is a paucity of data on the adherence rate to these guidelines; a retrospective study was performed to evaluate our institution's adherence. A chart review of 842 adults with SCD, seen 3/2017-3/2021 in the Montefiore healthcare system (All Patients), was done. Only about half of All Patients (n = 842) had >1 DFE during the study period (Total Examined Patients, n = 415). The Total Examined Patients were categorized as screening, those without retinopathy (Retinopathy-, n = 199), or follow-up, including individuals previously diagnosed with retinopathy (Retinopathy+, n = 216). Only 40.3% of screening patients (n = 87) had DFE at least biennially. As expected, there was a significant decrease in the average DFE rate of the Total Examined Patients after the COVID-19 pandemic started (13.6%) compared to pre-COVID (29.8%, p < 0.001). Similarly, there was a significant decrease in the screening rate of Retinopathy- patients from 18.6% on average pre-COVID to 6.7% during COVID (p < 0.001). This data shows the sickle retinopathy screening rate is low and innovative approaches may need to be employed to remedy this issue.

Background: A telemedicine screening initiative was implemented by the Montefiore Health System to improve access to eyecare for a multi-ethnic, at-risk population of diabetic patients in a largely underserved urban community in the Bronx, New York. This retrospective, cross-sectional analysis evaluates the societal benefit and financial sustainability of this program by analyzing both cost and revenue generation based on current standard Medicare reimbursement rates. Methods: Non-mydriatic fundus cameras were placed in collaboration with a vendor in eight outpatient primary care sites throughout the Montefiore Health Care System, and data was collected between June 2014 and July 2016. Fundus photos were electronically transmitted to a central reading center to be systematically reviewed and coded by faculty ophthalmologists, and patients were subsequently scheduled for ophthalmic evaluation based upon a predetermined treatment algorithm. A retrospective chart review of 2251 patients was performed utilizing our electronic medical record system (Epic Systems, Verona WI). Revenue was projected utilizing standard Medicare rates for our region while societal benefit was calculated using quality adjusted life years (QALY). Results: Of the 2251 patient charts reviewed, 791 patients (35.1%) were seen by Montefiore ophthalmologists within a year of the original screening date. Estimated revenue generated by these visits was $276,800, with the majority from the treatment of retinal disease ($208,535), and the remainder from other ophthalmic conditions detected in the fundus photos ($68,265). There was a societal benefit of 14.66 quality adjusted life years (QALYs) with an estimated value of $35,471/QALY. Conclusion: This telemedicine initiative was successful in identifying many patients with diabetic retinopathy and other ophthalmic conditions who may otherwise not have been formally evaluated. Our analysis demonstrates the program to generate a downstream revenue of nearly $280K with a cost benefit below <50% of the threshold of $100,000/QALY, and therefore cost-effective in marginalized communities.

Throughout the year 2019, Nigeria had sporadic outbreaks of yellow fever (YF), which began in the northern region of the country. Indeed, controlling the bites and population of Aedes mosquitoes and vaccination are the only effective means of preventing YF. Vectorial migration, sylvan-to-urban spillover, immunization failure and, perhaps, genetic modification of YFV could be reasons for the re-emergence of YF at the community, state and national levels. This article offers a critical review of the vector biology, YF vaccine immunodynamics and environmental drivers of YFV infections, with the aim of understanding the interplay of these factors in the re-emergence of YF and risk assessment of living in or travelling to areas where YF is endemic.

Summary: T helper 17 (Th17) are a CD4+ T subpopulation cells which are involved in the host protection against microbes such as extracellular and intracellular bacteria, parasites, fungi, and viruses. Monogenic defects including those mutations in some genes such as the signal transducer and activator of transcription (STAT)1 and 3, dedicator of cytokinesis 8 (DOCK8), autoimmune regulator (AIRE), and interleukin 17 receptor A (IL-17RA) can lead to impairment in Th17 cell development and function along with the concomitant increased risk for chronic mucocutaneous candidiasis (CMC). The immunologic abnormalities in these patients include low frequency of Th17 cells; defective cutaneous or in vitro T cell response to Candida species, and/or autoantibodies against relevant cytokines. This review outlines the biological characteristics and functionality of Th17 cells, as well as the clinical features of individuals with genetic defects associated with Th17 deficiency.

Cancer is a debilitating disease that is on the increase in both developed and developing countries. Anticancer drugs are often expensive, have narrow spectrum of activities, and are associated with toxicities and side effects such as myelosuppression, immunosuppression, gastrointestinal disturbance, alopecia, skin toxicity, and hepatotoxicity. Plants have been the major source of anticancer drugs both in orthodox and traditional medicine. Many of the plants claimed by the traditional medicine practitioners (TMPs) to be effective in the treatment of cancer are yet to be evaluated scientifically. In this work, five medicinal plants used by TMPs in Borno State, Nigeria, were tested against two brain tumor cell lines. Ethanol extracts of Securidaca longepedunculata, Andira inermis subsp. rooseveltii, Annona senegalensis, Carissa edulis, and Parinari polyandra were used. U87 and U231 brain tumor cell lines were used for proliferation assay, U251 cell line was used for the invasion assay in collagen V coated inserts, and U87 cell line was used for the western blot detection of cleaved Poly-ADP-Ribose-Polymerase (PARP). The result revealed that all tested extracts significantly (p < 0.05) inhibited the proliferation of U87 and U231 cell lines with the respective IC50 values ranging between 8 and 20 µg/ml for S. longepedunculata and 100 and 90 µg/ml for P. polyandra. The five extracts significantly (p < 0.05) inhibited the invasion of U251 cell line at the concentration of 10 µg/ml (S. longepedunculata), 20 µg/ml (A. inermis), 50 µg/ml (A. senegalensis), 50 µg/ml (C. edulis), and 50 µg/ml (P. polyandra). Securidaca longepedunculata extract induced the cleavage of PARP. It was concluded that these medicinal plants have antiproliferative and anti-invasive activities and possess good prospects as source of anticancer agents especially S. longepedunculata which induced apoptosis in U87 cell line.

The study re-visited malaria burden and pre-hospital medication among malarious subjects in Maiduguri, Northeast Nigeria. A total of 1,657 febrile subjects were screened for malaria by microscopy at two health institutions. Giemsa-stained blood smears were examined for parasitaemia and gametocytaemia; and parasite density (PD), gametocyte density (GD) and gametocyte sex ratio (GSR) were determined. The mean age of the 1,657 subjects was 27.5 ± 12.2 years and 7.8% (130/1,657) of the subjects aged <5 years. Sex distribution showed 47.0% (778/1,657) males and 53.0% (879/1657) females. Parasitaemia was recorded in 22.6% (375/1,657) with geometric mean PD of 8,925 (320-275,000) parasites/µl blood. The prevalence of parasitaemia was highest among subjects <5 years (χ2 = 401.1; df = 5; p < 0.0001) and in August and September (χ2 = 406.9; df = 11; p < 0.0001). Prevalence of gametocytaemia was 12.8% (48/375) with geometric mean GD of 109 (8-464) gametocytes/µl blood. The prevalence was higher in dry (16.5%, 29/176) than wet (9.5%, 19/199) months (χ2 = 4.0; df = 1; p = 0.045). The weighted mean GSR was 0.4 ± 0.1 with highest value in March (0.7 ± 0.2). Pre-hospital medication was recorded in 74.1% (278/375) of the subjects with parasitaemia. Analgesics (51.7%; 194/375) accounted for the highest proportion of drug consumed while 9.3% (35/375) of the subjects took antimalarial drugs. Malaria persisted in Maiduguri especially among subjects <5 years during wet months and pre-hospital medication is a common practice. These findings could serve as guide for policy decision that could contribute to effective treatment and control of malaria in the region.

Incidence of cancer is estimated to be on the increase and current anticancer drugs are characterized by narrow margin of safety and side effects. There is the need to explore new drugs especially from plants since plants serve as major source of drugs. Securidaca longipedunculata Fresen plant is called the mother of all medicines in northern Nigeria and is used traditionally in the treatment of cancers by most traditional medicine practitioners in the region. This study is aimed at evaluating the anticancer activity of the plant extract using U87 brain tumor cell line. Ethanol extract of its root bark was prepared and fractionated by silica gel column chromatography. In vitro activity of the extract and fractions were assessed on the viability of U87 malignant brain tumor cell line by using hemacytometer, annexin V-PE and 7AAD flow cytometry and western blot detection of Poly-ADP-Ribose-Polymerase (PARP) cleavage. The results showed that the extract significantly (p<0.01) inhibited proliferation of U87 cell line with IC50 of 20.535 µg/ml. Apoptosis was induced by the extract (41.53 ± 10.33%) and the polar fraction (47.3 ± 2.7%) via cleavage of PARP. It was concluded that the ethanol extract of S. longipedunculata root bark inhibited proliferation of U87 cell line and induced apoptosis by cleavage of PARP, thus supporting folkloric use of the plant in the management of cancers.

Among the many vascular complications of sickle cell disease (SCD), retinopathy is the most prevalent and represents a leading cause of blindness. Hydroxycarbamide therapy ameliorates many symptoms of SCD, and high fetal haemoglobin (HbF) levels have been shown to protect against the development of retinopathy in children with HbSS. Its effect on adults with SCD, who are at a much higher risk of developing retinopathy, has not been studied. We aimed to investigate the effect of hydroxycarbamide use and HbF level on sickle cell retinopathy development in adults. We performed a retrospective cross-sectional study and collected demographics, comorbidities, and ocular and haematological data from 300 adult sickle cell subjects examined at the Henkind Eye Institute at Montefiore Medical Center during a 5-year period, from October 2012 to November 2017. The cohort was comprised mainly of Black and Hispanic subjects with all SCD genotypes, aged 18-71 years. Results show that in HbSS patients treated with hydroxycarbamide, those with retinopathy had significantly lower HbF levels compared to patients without retinopathy (P = 0·018). Our study identified the optimal HbF cut-off point for retinopathy protection to be 14·87%. Thus, a HbF level of 15% appears to be the threshold above which the odds for developing retinopathy in SS patients are reduced by 50%.

Mating is preceded by a series of interdependent events that can be broadly categorized into searching and courtship. Long-range signals convey species- and sex-specific information during searching, while short-range signals provide information specific to individuals during courtship. Studies have shown that cuticular hydrocarbons (CHCs) can be used for mate recognition in addition to protecting insects from desiccation. In Psylloidea, four species rely on semiochemicals for long-range mate attraction. Psyllid mating research has focused on long-range mate attraction and has largely ignored the potential use of cuticular hydrocarbons (CHCs) as mate recognition cues. This study investigated whether CHCs of Aacanthocnema dobsoni have semiochemical activity for long- and short-range communication prior to mating. Using a solid sampler for solvent-less injection of whole psyllids into coupled gas chromatography/mass spectrometry, we found quantitative, sex- and age-related differences in CHC profiles. Males had higher proportions of 2-MeC28, 11,15-diMeC29, and n-C33 alkanes, while females had higher proportions of 5-MeC27, 3-MeC27, 5,15-diMeC27, n-C29 and n-C30 alkanes. In males and females, 84 and 68 % of CHCs varied with age, respectively. Y-tube olfactometer bioassays provided no evidence that males or females responded to odors emanating from groups of conspecifics of the opposite sex. Tests of male and female psyllids for attraction to branchlets previously occupied by conspecifics showed no evidence of attraction to possible semiochemical residues. Our short-range chemoreception bioassay showed that males were as indifferent to freshly killed individuals of either sex with intact CHC profiles as to those treated with hexane (to remove CHCs). Aacanthocnema dobsoni utilizes substrate-borne vibrations (SBVs) for communication. Therefore, our results indicate that SBVs are probably more important than semiochemicals for long-range mate attraction. Furthermore, CHCs are unlikely to mediate short-range mate recognition or provide mate assessment cues.

Background: Widespread dr ug-resistant Plasmodium falciparum strains have challenged the pivotal role played by 4-aminoquinolines, including chloroquine (CQ), which has been delisted for the treatment of malaria in most parts of the world. This study assessed the in vitro sensitivity of P. falciparum clinical isolates (PfCIs) to amodiaquine (AQ) and CQ in Northeast Nigeria. Materials and methods: PfCIs were collected from subjects with uncomplicated P. falciparum malaria in Azare, Bauchi State and Maiduguri, Borno State following an informed consent. The in vitro sensitivity was assessed by micro-test (MarkIII) method and the IC50 of AQ and CQ was determined using HN-NonLin Version VI.1 software. The reference standard cut-off values for in vitro AQ and CQ resistance of 80 and 160 nmol/l, respectively, were used. Isolates that were inhibited by lower AQ and CQ concentrations were referred to as sensitive. Results: Valid in vitro assay r esults were obtained for 88.9% (80/90) of the PfCIs; Azare had 93.3% (28/30) and Maiduguri had 86.7% (52/60) [χ2 = 0.35; df = 1; p = 0.486]. The geometric mean (GM) IC50 of AQ and CQ were 24.2 nmol/l (95% CI, 10.5 - 49.6 nmol/l) and 39.5 nmol/l (95% CI, 34.5 - 49.6 nmol/l), respectively. The AQ (p = 0.922) and CQ (p = 0.085) GM IC50 were similar between Azare and Maiduguri PfCIs. Only one isolate showed in vitro resistance to AQ giving a sensitivity of 98.8% (79/80) while 17 PfCIs showed in vitro resistance to CQ giving a sensitivity of 78.8% (63/80). The CQ sensitivity was similar between Azare (67.9%; 19/28) and Maiduguri (84.6%; 44/52) PfCIs (χ2 = 3.05; df = 1; p = 0.081). Conclusions: The findings may suggest that the AQ in vitro sensitivity remains high and the isolates in Northeast Nigeria may appear more sensitive to CQ than isolates from other parts. These findings may affect malaria treatment and control policy in Nigeria.

Neoplasms of the brain are often overlooked in resource-limited countries. Our literature search via AJOL and PubMed demonstrated that brain tumor research is still a rarity in these regions. We highlight the current status, importance, challenges, and methods of improving brain tumor research in West Africa. We suggest that more attention be given to basic, clinical, and epidemiological brain tumor research by national governments, private organizations, international organizations, non-governmental organizations (NGOs), and individuals in this region.

Lip prints in a narrow sense are normal lines and fissures in the form of wrinkles and grooves present in the zone of transition of lips. The aims of this study were to determine the types and association of lip prints as well as the role of the lip print as genetic marker in ethnic differentiation in multi-ethnic population of Nigeria. A total of 820 individuals (414 males and 406 females) participated. The study sample was drawn from different ethnic groups of Nigeria. The three major ethnic groups were identified and the minor groups were lumped together. This was to provide clear evidence on the ethnic differentiation based on lip print types among Nigerians. The lip prints were collected on microscopic glass slides and developed using carbon black powder. Each print was then divided into 10 compartments and analyzed using a magnifying lens. Chi-squared test was used for association between ethnicity and lip print types; p<0.05 was set as a level of significance. The result of the study showed the following lip print pattern in Nigerian population where Type V (31.39%) was predominant, followed by Type III (24.18%), Type IV (18.70%), Type I (14.87%), Type II (10.29%) and least frequent was Type I' (0.57%). Statistically significant association (p<0.05) of lip print types with ethnicity was found in upper left lateral compartments (ULL). It was concluded that lip prints show ethnic differences. Hence, they may hold potential promise as a supplementary tool in ethnic differentiation and in personal identification.

The antineuraminidase activity of different organic soluble fractions of Ganoderma lucidum extract was investigated using inhibition of hemagglutination and elution of chicken erythrocytes by Newcastle disease virus (NDV). Fractions of methanol, ethylacetate, and normal butanol (n-butanol) of the G. lucidum were tested against neuraminidase producing NDV as antigen. Different dilutions of the organic soluble fractions inhibited elution of 1% red blood cells by neuraminidase of NDV While the methanolic and n-butanol extracts inhibited neuraminidase activity even at a dilution of 1:16 and that of ethylacetate fraction inhibited even at 1:32 respectively. This finding indicates that G. lucidum has some antineuraminidase activity against NDV and may be exploited in the management of NDV infection.

Mate finding and courtship involve complex interactions that require close coordination between individuals of the opposite gender. Well-organized signalling systems, sometimes involving a combination of signal modalities, are required to convey species-specific and individual information to members of the opposite gender. Previous studies of psyllids have focused on single-signal modalities and have largely ignored the potentially interdependent nature of different types of signals. Several studies have shown that semiochemicals play a role in psyllid mate finding. However, long-range semiochemical sex attractants, such as the highly volatile sex pheromones used by many Lepidoptera (molecular weights <300), are yet to be identified. The compounds identified thus far, namely 13-methylheptacosane (from Cacopsylla pyricola) and dodecanoic acid (from Diaphorina citri), seem to have short range activity or no activity under field conditions. The possible role played by cuticular hydrocarbons in psyllid courtship remains largely ignored. Conversely, many psyllid species rely on vibrational signals for mate finding and mate assessment during courtship. This apparent disproportional reliance on vibrational rather than semiochemical signals suggests that vibrational signals have been more influential in sexual selection in psyllids. However, male fitness, female choice and benefits accrued from selecting fitter males remain poorly understood.

The proximate, minerals and amino acids contents of wild melon (Citrullus ecirrhosus) seeds were determined. The results for proximate analysis (% DW) showed a composition of 3.73 +/- 0.25 moisture, 2.12 +/- 0.08 ash 26.36 +/- 0.10 crude protein, 50.67 +/- 10.76 crude lipid, 2.17 +/- 0.29 crude fibre, 18.69 +/- 0.82 available carbohydrate and energy value of 601.7 +/- 8.75 kcal/100 g. The seeds amino acids profile revealed that for adults but leucine, lysine and threonine are below the requirement value for children. The overall result implies that seed of the wild melon possessed the potential to be used as a source of nutrition.
