For the last two decades different scientific disciplines have focused on lacustrine dissolved organic matter (DOM) given its importance in the biogeochemistry of carbon and in ecosystem functioning. New satellites supply the appropriate resolutions to evaluate chromophoric dissolved organic matter (CDOM) in inland waters, opening the possibility to estimate DOM at appropriate spatiotemporal scales. This requires, however, a robust relationship between CDOM and dissolved organic carbon (DOC). In this work, we evaluated the use of CDOM as a proxy of DOC in 7 Andean Patagonian lakes. Considering the entire data set, CDOM absorption coefficients (a355 and a440) were linearly related with DOC. Shallow lakes, however, drove this relationship showing a moderate relationship, whereas, deep lakes with lower colour presented a weaker relationship. Therefore, we assessed the use of CDOM spectral shape information to improve DOC estimates regardless of observed DOM differences due to climatic seasonality and lakes' morphometry. The use of well-known CDOM spectral shape metrics (i.e., S275-295 and a250:a365 ratio) significantly improved DOC estimation. Particularly, using a Gaussian decomposition approach we found that much of the variation in the spectral shape, associated with the variability of CDOM:DOC ratio, was explained by differences in two dynamic regions centred at 270 and 320 nm. A strong nonlinear relationship was found between the a270:a320 ratio and the DOC-specific absorption coefficients a*355 and a*440. This was translated into a further improvement in DOC estimation yielding the higher R2 and lower mean absolute differences (MAPD < 16%), either considering the entire data set or shallow and deep lakes separately. Our results highlight that incorporating the CDOM spectral shape information improves the characterization of the DOC pool of inland waters, which is particularly relevant for remote and/or inaccessible sites and has significant implications for the environmental management, biogeochemical studies and future remote sensing applications.

OBJECTIVES: To investigate the survival and failure mode of fiber-post resin restorations over preparations with different ferrule designs when submitted to a fatigue load test. METHODS AND MATERIALS: Fifty bovine incisors were selected and divided into five groups (n=10) according to ferrule design: a no-ferrule group, a 2-mm circumferential ferrule group, a 2-mm buccal ferrule group, a 2-mm lingual ferrule group, and a 2-mm buccal and lingual ferrule group. The fiberglass post was cemented and the composite core was built up and prepared, followed by cementation of a full composite crown. The samples were subjected to a cyclic fatigue test with loading applicator at 135°; a staircase approach was used until fracture. Survival (cycles to fracture) and failure modes were recorded. Survival data were analyzed with the log-rank test, while Kruskal-Wallis and Fisher exact tests were used to analyze failure mode data (α=0.05). RESULTS: The median number of cycles to fracture ranged from 215,000 to 236,153. The log-rank test showed no statistically significant difference in survival rates among the groups (p=0.82). Regarding failure mode, three types were observed: I, post and/or core fracture; II, root fracture in the cervical third; and III, root fracture in the middle third. No statistical difference was observed among the groups (Kruskal-Wallis test, p=0.147). CONCLUSION: The ferrule design had no effect on fatigue resistance or failure mode of endodontically treated incisor teeth restored with a fiber post, composite core buildup, and composite crown.

A green and simple method has been proposed in this work for the simultaneous determination of V, Ni and Fe in fuel ash samples by solid sampling high resolution continuum source graphite furnace atomic absorption spectrometry (SS HR CS GFAAS). The application of fast programs in combination with direct solid sampling allows eliminating pretreatment steps, involving minimal manipulation of sample. Iridium treated platforms were applied throughout the present study, enabling the use of aqueous standards for calibration. Correlation coefficients for the calibration curves were typically better than 0.9931. The concentrations found in the fuel ash samples analysed ranged from 0.66% to 4.2% for V, 0.23-0.7% for Ni and 0.10-0.60% for Fe. Precision (%RSD) were 5.2%, 10.0% and 9.8% for V, Ni and Fe, respectively, obtained as the average of the %RSD of six replicates of each fuel ash sample. The optimum conditions established were applied to the determination of the target analytes in fuel ash samples. In order to test the accuracy and applicability of the proposed method in the analysis of samples, five ash samples from the combustion of fuel in power stations, were analysed. The method accuracy was evaluated by comparing the results obtained using the proposed method with the results obtained by ICP OES previous acid digestion. The results showed good agreement between them. The goal of this work has been to develop a fast and simple methodology that permits the use of aqueous standards for straightforward calibration and the simultaneous determination of V, Ni and Fe in fuel ash samples by direct SS HR CS GFAAS.

Demographic data for Elkhorn coral, Acropora palmata, and in situ water temperature data from seven upper Florida Keys (USA) reefs revealed three warm thermal stress events between 2010 and 2016. During a mild bleaching event in 2011, up to 59% of colonies bleached, but no mortality resulted. In both 2014 and 2015, severe and unprecedented bleaching was observed with up to 100% of colonies bleached. A. palmata live tissue cover declined by one-third following the 2014-2015 events. Colony mortality of mildly- and non-bleached colonies did not differ but increased significantly with more severe bleaching. Increased bleaching prevalence corresponded to maximum daily average water temperatures above 31.3°C. However, the cumulative days with daily average exceeding 31.0°C provided a better predictor of bleaching response. The bleaching response of surviving colonies in 2015 was not consistent with acclimatization as most individual colonies bleached at least as badly as in 2014.

Initially discovered in Georgia in 2009, the exotic invasive plataspid, Megacopta cribraria (F.), has become a serious pest of soybean ( Glycine max (L.) Merrill). Managing M. cribraria in soybean typically involves the application of broad-spectrum insecticides. Soybean host plant resistance is an attractive alternative approach; however, no commercial soybean cultivars have been identified as resistant. During 2013 and 2014, we compared 40 and 44 soybean genotypes, respectively, for resistance to M. cribraria in a split-plot design under natural insect infestation in small-plot experiments. Soybean genotypes were selected to maximize diversity with respect to maturity group, pubescence type, leaf shape, seed size, nitrogen fixation, drought tolerance, seed protein content, and pest resistance. Megacopta cribraria egg masses, nymphs, and adults were counted during the growing season to identify potentially resistant soybean genotypes. Soybean seed yield was measured in insecticide-protected and unprotected conditions to determine tolerance to M. cribraria feeding. In both years, a range of host plant resistance was observed. The fewest M. cribraria adults and nymphs were found on narrow-leaf, small-seeded cultivars 'N7103' and 'Vance,' as well as the nonnodulating cultivar 'Nitrasoy.' Additionally, N7103 and Vance were among the least susceptible genotypes to M. cribraria oviposition in the field. Most 'Benning' cultivar insect-resistant near-isogenic breeding lines also displayed moderate levels of resistance to M. cribraria . Seed yields of Vance and N7103 were less affected by M. cribraria in 2013 than most other soybean genotypes. These results may be useful to soybean breeders to develop cultivars with resistance to M. cribraria.

BACKGROUND: A randomised study was conducted on the number of attempts made during the conventional endotracheal intubation of a mannequin using two polyvinyl (PVC) tracheal tubes, apparently similar but from different manufactures: INTERSURGICAL (IS; Intersurgical S.L., Madrid, Spain) and Mallinckrodt (ML; Mallinkrodt Medical S.A., Madrid, Spain). METHODS: A total of 26 anaesthesiologists, in randomly established order (generated by Epidat 3.1) intubated a mannequin twice using a different tube each time. The tubes were masked by painting them to prevent recognition. The main outcome of the study was to compare the number of attempts needed to complete the manoeuvre for each tube. Data on intubation time and failed intubations were also collected. RESULTS: The number of attempts with the ML tube was significantly lower than with the IS tube. Intubation was completed on the first attempt with the ML tube in 93.3% of cases, while using the IS tube the percentage fell to 30.8% (Fisher exact test, P<.001). The time required to complete the manoeuvre was greater with the IS tube (median 10.8seconds, interquartile range 6-22) than with the ML tube (median 4.4seconds, interquartile range 3.5 to 6.3). CONCLUSIONS: The PVC tube from the ML manufacturer was superior when compared with the IS, the latter was also associated with a larger number of attempts to complete intubation using a conventional Macintosh blade.

The acrosome reaction occurs in vivo following sperm capacitation and is essential for the acquisition of sperm fertilization ability. However, little is known about the molecular identity of the physiological acrosome reaction regulators. In addition to progesterone, which is produced by cumulus oophorus cells and known to regulate acrosome reaction by activating the specific calcium channel CatSper, endogenous opioid peptides such as beta-endorphin and met-enkephalin are present at high concentrations in the follicular fluid suggesting that the opioid system may be involved in the mechanisms regulating the acrosome reaction in humans. By using Reverse Transcription-PCR, western blot and immunofluorescence approaches, we described the presence and localization of the beta-endorphin precursor, pro-opiomelanocortinin the middle section and in flagellum of human spermatozoa, and inside the seminiferous tubules of human testis. Flow cytometry and intracellular calcium analyses showed that beta-endorphin causes an inversely dose-dependent increase in the percentage of acrosome-reacted sperm cells by a calcium-independent protein kinase C pathway. These findings are important for future studies of sperm physiology and provide new insight into the function of the opioid system as a target of fertility management.

The influence of the charge on the permeability of microgel particles is studied in the presence of electric fields. Electrophoresis experiments performed on pH-ionizable pNIPAM-AA microgels show that particles behave as permeable spheres when the network is ionized. However, they keep non-permeable in the absence of charge. The ionic nature of the network thus controls the permeability of the soft particles. A salt-dependent local viscosity explains these permeability changes. This is confirmed by NMR as alternative independent technique. Strongly hydrated counterions located around fixed charges on the network are considered responsible for the local viscosity variations.

OBJECTIVES: This study tested the hypothesis that photoactivation timing and resin cement affect mechanical properties and bond strength of fiberglass posts to root dentin at different depths. METHODS: Fiberglass posts (Exacto, Angelus) were luted with RelyX Unicem (3M ESPE), Panavia F 2.0 (Kuraray), or RelyX ARC (3M ESPE) using three photoactivation timings: light curing immediately, after three minutes, or after five minutes. Push-out bonding strength, PBS (n=10) was measured on each root region (coronal, middle, apical). The elastic modulus (E) and Vickers hardness (VHN) of the cement layer along the root canal were determined using dynamic indentation (n=5). A strain-gauge test was used to measure post-gel shrinkage of each cement (n=10). Residual shrinkage stress was assessed with finite element analysis. Data were analyzed with two-way analysis of variance in a split-plot arrangement and a Tukey test (α=0.05). Multiple linear regression analysis was used to determine the influence of study factors. RESULTS: The five-minute delay photoactivation timing significantly increased the PBS for all resin cements evaluated. The PBS decreased significantly from coronal to apical root canal regions. The mean values for E and VHN increased significantly with the delayed photoactivation for RelyX Unicem and decreased from coronal to apical root regions for all resin cements with the immediate-curing timing. CONCLUSIONS: The PBS of fiber posts to root dentin, E, and VHN values were affected by the root canal region, photoactivation timing, and resin cement type. Shrinkage stress values decreased gradually with delayed photoactivation for all the cements.

We used allometric models to identify the optimal body size/shape characteristics associated with physical and motor performance tests in Peruvian schoolchildren. The sample consisted of 3624 subjects (1669 boys and 1955 girls) aged 11-17 years from 31 public schools belonging to four cities located in the three natural regions in central Peru. Motor performance included 12-min run, standing long jump, grip strength, curl-ups, shuttle run, and sit and reach. The reciprocal Ponderal index (RPI), a characteristic sometimes referred to as the somatotype "ectomorphy," was found to be the most suitable body shape indicator associated with 12-min run, standing long jump, curl-up, and shuttle run performance. A positive maturation offset parameter was also associated with greater standing long jump, grip strength, shuttle run, and sit-and-reach performances. With the exception of the sit-and-reach flexibility, sex differences are pervasive in all tests favoring boys. Rainforest schoolchildren are best performers in the power and flexibility tests, whereas those from high altitude were superior in the 12-min endurance test even after taking their much lighter body size characteristics into account. This latter finding suggests that living at high altitude in Peru benefits children's endurance performance both before and even after controlling for differences in the confounding variable of body size/shape.

The aim of this work was to evaluate the effect of buffer addition and process temperature (ambient and 35°C) on H2 production in batch fermentation of cheese whey (CW). When the H2 production reached a plateau, the headspace of the reactors were flushed with N2 and reactors were re-incubated. Afterwards, only the reactors with phosphate buffer showed a second cycle of H2 production and 48% more H2 was obtained. The absence of a second cycle in non-buffered reactors could be related to a lower final pH than in the buffered reactors; the low pH could drive the fermentation to solvents production. Indeed a high solvent production was observed in non-buffered bioreactors as given by low ρ ratios (defined as the ratio between sum of organic acid production and sum of solvents production). Regarding the process temperatures, no significant difference between the H2 production of reactors incubated at ambient temperature and at 35°C was described. After flushing the headspace of bioreactors with N2 at the end of the second cycle, the H2 production did not resume (in all reactors).

The stromal compartment surrounding epithelial-derived pancreatic tumors is thought to have a key role in the aggressive phenotype of this malignancy. Emerging evidence suggests that cancer-associated fibroblasts (CAFs), the most abundant cells in the stroma of pancreatic tumors, contribute to the tumor's invasion, metastasis and resistance to therapy, but the precise molecular mechanisms that regulate CAFs behavior are poorly understood. In this study, we utilized immortalized human pancreatic CAFs to investigate molecular pathways that control the matrix-remodeling and invasion-promoting activity of CAFs. We showed previously that palladin, an actin-associated protein, is expressed at high levels in CAFs of pancreatic tumors and other solid tumors, and also in an immortalized line of human CAFs. In this study, we found that short-term exposure of CAFs to phorbol esters reduced the number of stress fibers and triggered the appearance of individual invadopodia and invadopodial rosettes in CAFs. Molecular analysis of invadopodia revealed that their composition resembled that of similar structures (that is, invadopodia and podosomes) described in other cell types. Pharmacological inhibition and small interfering RNA knockdown experiments demonstrated that protein kinase C, the small GTPase Cdc42 and palladin were necessary for the efficient assembly of invadopodia by CAFs. In addition, GTPase activity assays showed that palladin contributes to the activation of Cdc42. In mouse xenograft experiments using a mixture of CAFs and tumor cells, palladin expression in CAFs promoted the rapid growth and metastasis of human pancreatic tumor cells. Overall, these results indicate that high levels of palladin expression in CAFs enhance their ability to remodel the extracellular matrix by regulating the activity of Cdc42, which in turn promotes the assembly of matrix-degrading invadopodia in CAFs and tumor cell invasion. Together, these results identify a novel molecular signaling pathway that may provide new molecular targets for the inhibition of pancreatic cancer metastasis.

OBJECTIVES: Finite element analysis (FEA) was used to study polymerization shrinkage stress in molars restored with composites and to correlate those stresses with experimentally measured tooth deformation. METHODS: Three composites (Filtek LS, Aelite LS Posterior, Filtek Supreme) and three filling techniques (bulk, 2.0-mm increments, and 1.0-mm increments) for restoring a molar were simulated in a two-dimensional FEA. Polymerization shrinkage was modeled using post-gel shrinkage, which was measured using the strain gauge technique (n=10). Cuspal tooth deformation, measured at the buccal and lingual surfaces with strain gauges in a laboratory study, was used to validate the analysis. Residual shrinkage stresses were expressed in modified von Mises equivalent stresses. Linear Pearson correlations were determined between the laboratory and FEA results. RESULTS: Post-gel shrinkage values (in volume %) were: Filtek LS (0.11 ± 0.03) < Aelite LS Posterior (0.51 ± 0.02) < Filtek Supreme (0.62 ± 0.09). The 1.0-mm increment filling caused substantially higher stresses and strains in the cervical enamel region. Significant correlations were found between: elastic modulus and FEA strain, elastic modulus and FEA stress, post-gel shrinkage and FEA strain, post-gel shrinkage and FEA stress, FEA strain and cuspal deformation by strain gauge, and FEA stress and cuspal deformation by strain gauge (p<0.05). CONCLUSIONS: Increasing the number of increments and high post-gel shrinkage and/or elastic modulus values caused higher stresses in the remaining tooth structure and tooth/restoration interface. Cuspal deformation measured with the strain gauge method validated the finite element analyses.

BACKGROUND: To describe the pattern of patients admitted due to rare diseases corresponding to congenital anomalies in a regional hospital. METHODS: Retrospective transversal study. We considered hospital discharges for the years 2009-2012 with principal diagnosis between codes CIE 9R MC 740-759. The source of information was the Basic Minimum Data Set. Socio-demographic and clinical variables were analyzed. RESULTS: One point six percent (1.6%) of the population was admitted to hospital due to rare congenital diseases. Fifty-eight point five percent (58.5%) were male, with average age 21.4 ± 21.5 years. The major diagnostic categories were: diseases of the nervous system (86.9%), circulatory systems diseases (51.7%) and musculoskeletal system diseases (50.3%). Eighteen percent (18%) of hospital admissions corresponded to patient readmissions. The service with the greatest number of episodes was Pediatric Surgery, 29%, followed by Neurosurgery, 20%. CONCLUSIONS: The pattern of rare congenital disease in the "Virgen de Nieves" University Hospital corresponds to a young patient, with a disease belonging to the diseases of the nervous system group of the major diagnostic categories, treated surgically, and with a low percentage of readmissions.

Megacopta cribraria (F.) (Hemiptera: Plataspidae) was first reported in 2009 near Atlanta, GA. The insect undergoes two generations per year. The first-generation is reported mainly in kudzu during May and June, with the second establishing on both kudzu and soybean during July and August. A greenhouse study was conducted to determine the suitability of two legumes as hosts for first generation M. cribraria. First generation M. cribraria successfully developed on caged potted soybean plants. Conversely, snap beans were not a suitable host under the conditions of this study. A range of 45-50 d was needed to transition from the egg to adult on soybean plants. Although this study was limited to the greenhouse, kudzu may not be an obligate host for the development of first-generation M. cribraria. An important implication of this finding is the establishment for this pest on spring-planted soybean and for the possible expanded geographic range for this pest beyond that of kudzu.

AIM: To evaluate the effect of luting agent and fibreglass post design on bond strength to root dentine at different depths within the canal. METHODOLOGY: Ninety single-rooted teeth were root filled and prepared to receive either a parallel-sided and serrated fibreglass post (Reforpost no. 2) or a tapered and smooth fibreglass post (Exato Cônico). The posts were cemented with the following resin cements: dual-cured resin cement (Rely X ARC), two self-adhesive resin cements (Rely X Unicem and MaxCem) and a self-cured resin cement (Cement-Post). The roots were cross-sectioned to obtain two 1-mm-thick discs for each cervical, middle and apical third of the prepared root portion. The posts were submitted to a micropush-out test at a speed of 0.5 mm min(-1), and the bond strength values (MPa) were submitted to anova in a split-plot arrangement and Tukey's test (P < 0.05). RESULTS: The RelyX Unicem demonstrated significantly higher bond strength values (P < 0.001) along the root dentine. The RelyX ARC and Cement-Post had similar bond strength values in the cervical third; however, the bond strength decreased significantly (P < 0.001) in an apical direction for the RelyX ARC. Significantly lower bond strength values (P < 0.001), irrespective of canal region, were found for MaxCem cement. The bond strength was similar for both post configurations irrespective of the resin cement and canal region. CONCLUSIONS: The retention of glass fibre posts remained unaffected by surface roughness but was influenced by resin cement type. The self-adhesive cement RelyX Unicem yielded a significantly greater (P < 0.001) bond strength value when cementing the fibreglass posts.
