RESUMO

T-cell intracellular antigen 1 (TIA1) and TIA1-like/related protein (TIAL1/TIAR) are two members of the classical family of RNA binding proteins. Through their selective interactions with distinct RNAs and proteins, these multifunctional regulators are involved in chromatin remodeling, RNA splicing and processing and translation regulation, linking them to a wide range of diseases including neuronal disorders, cancer and other pathologies. From their discovery to the present day, many studies have focused on the behavior of these proteins in order to understand their impact on molecular and cellular processes and to understand their relationship to human pathologies. The volume of research on these proteins in various fields, including molecular biology, biochemistry, cell biology, immunology and cancer, has steadily increased, indicating a growing interest in these gene expression regulators among researchers. This information can be used to know the most productive institutions working in the field, understand the focus of research, identify key areas of involvement, delve deeper into their relationship and impact on different diseases, and to establish the level of study associated with them.

RESUMO

AIM: The aim of this study was to evaluate the predictors of burnout in female nurses during the first wave of the COVID-19 pandemic. It also sought to evaluate the relationship between each of the variables studied (anxiety, depression, sociodemographic and COVID-19 variables) and the dimensions of the burnout. BACKGROUND: One of the groups of health care workers worst affected by the COVID-19 crisis has been women working as nursing staff, due to the high percentage they account for at a global level and their direct contact with infected patients. DESIGN: This was a cross-sectional, quantitative study. METHODS: Four hundred forty-four Spanish female nurses from hospital and primary health care centres took part in the study. The data were obtained in 2020 by means of an online survey. RESULTS: Symptoms of depression are a common predictor variable to all the dimensions of burnout, whereas symptoms of anxiety predict emotional exhaustion and depersonalization. Age and years of experience in the job predict depersonalization, whereas the probability of contracting the infection is a predictor variable of emotional exhaustion and personal accomplishment. CONCLUSION: Predictor variables should be considered in the creation of prevention and intervention plans to reduce the levels of burnout in female nurses.

RESUMO

T-cell intracellular antigen 1 (TIA1)-related/like (TIAR/TIAL1) protein is a multifunctional RNA-binding protein (RBP) involved in regulating many aspects of gene expression, independently or in combination with its paralog TIA1. TIAR was first described in 1992 by Paul Anderson's lab in relation to the development of a cell death phenotype in immune system cells, as it possesses nucleolytic activity against cytotoxic lymphocyte target cells. Similar to TIA1, it is characterized by a subcellular nucleo-cytoplasmic localization and ubiquitous expression in the cells of different tissues of higher organisms. In this paper, we review the relevant structural and functional information available about TIAR from a triple perspective (molecular, cellular and pathophysiological), paying special attention to its expression and regulation in cellular events and processes linked to human pathophysiology.

RESUMO

T-cell intracellular antigen 1 (TIA1) is an RNA-binding protein that is primarily involved in the post-transcriptional regulation of cellular RNAs. Furthermore, it is a key component of stress granules (SGs), RNA, and protein aggregates that are formed in response to stressful stimuli to reduce cellular activity as a survival mechanism. TIA1 p.E384K mutation is the genetic cause of Welander distal myopathy (WDM), a late-onset muscular dystrophy whose pathogenesis has been related to modifying SG dynamics. In this study, we present the results obtained by analyzing two specific aspects: (i) SGs properties and dynamics depending on the amino acid at position 384 of TIA1; and (ii) the formation/disassembly time-course of TIA1WT/WDM-dependent SGs under oxidative stress. The generation of TIA1 variants-in which the amino acid mutated in WDM and the adjacent ones were replaced by lysines, glutamic acids, or alanines-allowed us to verify that the inclusion of a single lysine is necessary and sufficient to alter SGs dynamics. Moreover, time-lapse microscopy analysis allowed us to establish in vivo the dynamics of TIA1WT/WDM-dependent SG formation and disassembly, after the elimination of the oxidizing agent, for 1 and 3 h, respectively. Our observations show distinct dynamics between the formation and disassembly of TIA1WT/WDM-dependent SGs. Taken together, this study has allowed us to expand the existing knowledge on the role of TIA1 and the WDM mutation in SG formation.

RESUMO

OBJECTIVE: This study aims to examine whether there are differences in symptoms of posttraumatic stress, depression, anxiety, levels of burnout and resilience in Spanish healthcare staff between the first wave of the COVID-19 pandemic and after it, depending on several demographic and work-related variables. METHODS: A longitudinal study was conducted in April 2020 (T0), and July 2020 (T1). Symptoms of posttraumatic stress, depression, anxiety, burnout, levels of resilience, along with demographic and work-related variables in 443 workers were assessed. RESULTS: Symptoms and burnout were more pronounced at T0, whereas the levels of resilience were higher at T1. Being women, being young, holding a lower-level job, less years of experience, lower educational level, and/or working rotating shifts are associated with having more posttraumatic stress symptoms and burnout. CONCLUSION: These variables would be considered in similar situations.

RESUMO

T-cell intracellular antigen 1 (TIA1) is a multifunctional RNA-binding protein involved in regulating gene expression and splicing during development and in response to environmental stress, to maintain cell homeostasis and promote survival. Herein, we used TIA1-deficient murine embryonic fibroblasts (MEFs) to study their role in mitochondria homeostasis. We found that the loss of TIA1 was associated with changes in mitochondrial morphology, promoting the appearance of elongated mitochondria with heterogeneous cristae density and size. The proteomic patterns of TIA1-deficient MEFs were consistent with expression changes in molecular components related to mitochondrial dynamics/organization and respiration. Bioenergetics analysis illustrated that TIA1 deficiency enhances mitochondrial respiration. Overall, our findings shed light on the role of TIA1 in mitochondrial dynamics and highlight a point of crosstalk between potential pro-survival and pro-senescence pathways.

RESUMO

BACKGROUND: During the COVID-19 pandemic, the police force was one of the institutions that was most exposed to possible infections, in addition to suffering stressful situations that affect health. This study aims to independently assess the relationship between demographic, work, and COVID-19 variables, and resilience, engagement, and the perception of health, as well as to evaluate which variables are more important when predicting health perceptions. METHOD: 640 Spanish police officers were evaluated through a cross-sectional design and online survey. RESULTS: The perception of the lack of personal protective equipment, believing that it is easy to become infected with COVID-19, or working as a police officer were associated with low levels of engagement and resilience and a worse perception of health. More years of experience in the police force was associated with worse health and lower levels of engagement and resilience. Engagement and resilience would be protective variables of health. CONCLUSIONS: It is necessary to promote practices related to engagement and resilience in the police force. The discussion emphasizes that more experienced police officers demonstrate less engagement and resilience.

RESUMO

The Effort-Reward Imbalance Questionnaire (hereinafter, ERIQ) has been largely used worldwide to assess job stress, but it has not yet been applied in Spanish police. The objective of this study was to examine the construct validity and the internal consistency of the ERIQ in police officers. A cross-sectional study was carried out, using a nonprobability sampling (quota). A total of 217 Spanish police officers participated, 192 men (88.47%) and 25 women (11.53%). The mean age was 41 years (SD = 7.51). These police officers completed the ERIQ together with some other questionnaires (DECORE-21, MBI, GHQ and STAI) in order to provide evidence for validity based on the relationships to other constructs. A confirmatory factor analysis was performed and a matrix of correlations with the rest of constructs was created. The results showed an appropriate fit to the original model consisting of three scales. In addition, the scales of the ERIQ presented the expected relationship with the other constructs. The ERIQ is a valid instrument for assessing occupational stress in Spanish police officers and can improve the interventions in this professional group.

RESUMO

OBJECTIVES: The IMPACT trial has compared the benefit in the reduction of moderate/severe exacerbations of single inhaler triple therapy (SITT) with fluticasone furoate (FF)/umeclidinium (UMEC)/vilanterol (VI) versus dual therapy with FF/VI (ICS/LABA) and UMEC/VI (LAMA/LABA) in the treatment of patients with chronic obstructive disease (COPD). This study performs a subgroup analysis of the cohort from Spain in the IMPACT study. MATERIALS AND METHODS: In IMPACT, a 52-week randomized, double-blind, parallel-group, multicenter study (N = 10,355), patients ⩾40 years of age with COPD and ⩾1 moderate/severe exacerbations in the previous year were randomized 2:2:1 to once-daily FF/UMEC/VI 100/62.5/25 µg, FF/VI 100/25 µg or UMEC/VI 62.5/25 µg administered via the Ellipta inhaler. Here, we present a subgroup analysis of the 499 patients from Spain, included in the intent-to-treat (ITT) population in the study. Endpoint assessed included exposure-adjusted rate of moderate and severe exacerbations. RESULTS: In the Spain cohort, the exposure-adjusted rate of on-treatment moderate/severe COPD exacerbations per year for FF/UMEC/VI was 1.31 versus 1.43 and 1.57 for FF/VI and UMEC/VI, respectively. No new adverse events were identified. The results are consistent with those observed in the overall ITT study population. CONCLUSION: In the Spain cohort of the IMPACT study, patients receiving triple therapy with FF/UMEC/VI had a lower exposure-adjusted rate of exacerbations compared with FF/VI and UMEC/VI, similar to the overall population.Study Title: A Phase III, 52 Week, Randomized, Double-blind, 3-arm Parallel Group Study, Comparing the Efficacy, Safety and Tolerability of the Fixed Dose Triple Combination FF/UMEC/VI With the Fixed Dose Dual Combinations of FF/VI and UMEC/VI, All Administered Once-daily in the Morning Via a Dry Powder Inhaler in Subjects With Chronic Obstructive Pulmonary DiseaseURL: https://www.clinicaltrialsregister.eu/ctr-search/search?query=CTT116855/ https://clinicaltrials.gov/ct2/show/NCT02164513Registration number: GSK (CTT116855/NCT02164513).The reviews of this paper are available via the supplemental material section.

RESUMO

The number of health workers infected with COVID-19 in Spain is one of the highest in the world. The aim of this study is to analyse posttraumatic stress, anxiety and depression during the COVID-19 pandemic. Associations between burnout, resilience, demographic, work and COVID-19 variables are analysed. Cross-sectional data on 1422 health workers were analysed. A total of 56.6% of health workers present symptoms of posttraumatic stress disorder, 58.6% anxiety disorder, 46% depressive disorder and 41.1% feel emotionally drained. The profile of a health worker with greater posttraumatic stress symptoms would be a person who works in the Autonomous Community of Madrid, in a hospital, is a woman, is concerned that a person he/she lives with may be infected, and thinks that he/she is very likely to be infected. The risk variables for anxiety and depression would be a person that is a woman, working 12- or 24-h shifts, and being worried that a family member could be infected. High scores on emotional exhaustion and depersonalization are risk factors for mental health, with resilience and personal fulfilment being protective variables. Data are provided to improve preventive measures for occupational health workers.

RESUMO

Conventionally, engagement has been portrayed as an opposing construct to burnout and academic stress, and has been positively related to performance in the educational environment; nevertheless, few studies include at the same time both of these in addition to the time perspective of individuals. The purpose of this investigation is to analyse the construct of engagement and time perspective factors associated with academic performance. To this effect, a cross-sectional study was performed in which 298 postgraduate university students participated and multiple regression analyses were performed. From the selected independent variables, three were identified as statistically significant in the final model. Results show that gender, time perspective oriented towards the future and engagement are associated to the average score of the student's academic record. Consequently, it becomes necessary to consider the future time dimension as well as engagement to promote improvement of the university system and access to teaching professions.

RESUMO

Among the variables associated with occupational stress, two of the most studied are the adverse perceptions of psychosocial risk factors in the workplace and burnout. With the rise of positive psychology, other variables of the individual type, such as hardy personality have also been the subject of study. No studies have been found that jointly examine these variables related to mental health in police officers. The aim of this research was to analyze which variables were associated with mental health in police officers. A total of 223 policemen (202 men and 21 women) participated in a cross-sectional study. Of all the variables, emotional exhaustion and perception of problems as challenges were the only factors introduced in the regression model. These factors must be considered to improve both human resource interventions and occupational health practices in this professional group.

RESUMO

There is scarce research on stress in Spanish police officers and on the use of instruments to assess this construct in this professional group. In Spain, the DECORE questionnaire has been one of the most used. Nevertheless, it had not been previously applied to police officers. The aim of this study was to analyze both the construct validity and the reliability of the original model of 44 items. A cross-sectional design and a non-probabilistic quota sampling were used. A total of 223 Spanish police officers participated. 202 police personnel were men (90.6%) and 21 were women (9.4%). The average age was 41 years old (SD = 7.52). These police officers answered the DECORE questionnaire. Confirmatory Factor Analysis was carried out. The results showed an unsatisfactory adjustment using the original model of 44 items. A final solution of 21 items and four factors was formulated with both suitable validity and reliability indexes. In this model, 23 items that did not weigh highly in any of the four factors were removed. The DECORE-21 questionnaire is recommended to assess work-related stress in police officers.

RESUMO

OBJECTIVES: To assess the linguistic equivalence of the COPD Assessment Test versions in Catalan (CAT-C), Galician (CAT-G) and Basque (CAT-V) with the validated Spanish version (CAT) in COPD patients able to use both official languages. METHODS: Study performed in primary care centers in Catalonia, Galicia and the Basque Country. Half of the patients completed the questionnaire in their local language followed by the CAT in Spanish, while the other half did so in reverse order. RESULTS: 151 COPD patients were included in the study, with a mean age of 69.0 (SD: 9.7) years. Most (79.5%) were men, 11.3% were educated up touniversity level, and 31.8% were current smokers. Mean FEV1 was 61.4% (SD: 16.8) predicted and 83.9% of patients were GOLD grade i or ii. Concordance between CAT-C, CAT-G and CAT-V and CAT was high, with differences between scores from 0.4 to-0.5. Reliability (Cronbach's alpha) ranged from0.72 to0.86. Convergent validity, when correlated with the Medical Research Council Dyspnea scale (P=.003) and Saint George's Respiratory Questionnaire (ICC, r=0.74) scores, was significant. CONCLUSIONS: CAT-C, CAT-G and CAT-V scores were similar to CAT scores, with high correlation and concordance. These results show the equivalence between the validated Spanish CAT and the versionin other languages, so CAT-C, CAT-G and CAT-V can be used in individuals whose main language is Catalan, Galician or Basque.

RESUMO

BACKGROUND: In the police force, some variables such as occupational rank, sex, age and work-shift are associated with stress in workers. The aim of this paper was to determine possible differences in the perception of occupational stress at work depending on rank, sex, age and work-shift of police agents in the Community of Madrid, Spain. METHOD: A cross-sectional study was conducted in 24 municipalities of the Community of Madrid. A total number of 565 police agents participated. The ranks of the police agents were: constable, corporal, sergeant and police chief. Occupational stress (psychosocial risk factors at work) was assessed with the DECORE Questionnaire. RESULTS: All police agents perceived psychosocial risk factors adversely; especially agents of lesser rank perceived less control, fewer rewards and scarce support. There were significant differences in the perception of insufficient rewards between constables and other categories; and between constables and corporals in the perception of insufficient organisational support. No significant differences were found in the perception of psychosocial risk factors in relation to the rest of variables. CONCLUSIONS: The police rank should be taken into account for the development of preventive measures to reduce stress.

RESUMO

True stress-true strain curves of naturally spun viscid line fibres retrieved directly from the spiral of orb-webs built by Argiope trifasciata spiders were measured using a novel methodology. This new procedure combines a method for removing the aqueous coating of the fibres and a technique that allows the accurate measurement of their cross-sectional area. Comparison of the tensile behaviour of different samples indicated that naturally spun viscid lines show a large variability, comparable to that of other silks, such as major ampullate gland silk and silkworm silk. Nevertheless, application of a statistical analysis allowed the identification of two independent parameters that underlie the variability and characterize the observed range of true stress-true strain curves. The combination of this result with previous mechanical and microstructural data suggested the assignment of these two independent effects to the degree of alignment of the protein chains and to the local relative humidity, which, in turn, depends on the composition of the viscous coating and on the external environmental conditions.

RESUMO

Motivated by the need of poor and rural Mexico, where the population has limited access to advanced medical technology and services, we have developed a new paradigm for medical diagnostic based on the technology of "Volumetric Electromagnetic Phase Shift Spectroscopy" (VEPS), as an inexpensive partial substitute to medical imaging. VEPS, can detect changes in tissue properties inside the body through non-contact, multi-frequency electromagnetic measurements from the exterior of the body, and thereby provide rapid and inexpensive diagnostics in a way that is amenable for use in economically disadvantaged parts of the world. We describe the technology and report results from a limited pilot study with 46 healthy volunteers and eight patients with CT radiology confirmed brain edema and brain hematoma. Data analysis with a non-parametric statistical Mann-Whitney U test, shows that in the frequency range of from 26 MHz to 39 MHz, VEPS can distinguish non-invasively and without contact, with a statistical significance of p<0.05, between healthy subjects and those with a medical conditions in the brain. In the frequency range of between 153 MHz to 166 MHz it can distinguish with a statistical significance of p<0.05 between subjects with brain edema and those with a hematoma in the brain. A classifier build from measurements in these two frequency ranges can provide instantaneous diagnostic of the medical condition of the brain of a patient, from a single set of measurements. While this is a small-scale pilot study, it illustrates the potential of VEPS to change the paradigm of medical diagnostic of brain injury through a VEPS classifier-based technology. Obviously substantially larger-scale studies are needed to verify and expand on the findings in this small pilot study.

RESUMO

We present a rapid (5 min) and controlled sensitisation method for dye sensitized solar cells which gives 6.0% for the Ru dye N719 and 3.7% for the IR absorbing squaraine SQ1. Rapid co-sensitization (N719 and SQ1) gives efficiencies up to 7.9%. Devices have similar or better efficiency to those dyed passively for 18 h.
