RESUMO

The ubiquitin specific protease 7 (USP7) is a deubiquitinating enzyme with numerous substrates. Aberrant expression of USP7 is associated with tumor progression. This study aims to investigate how a deregulated USP7 expression affects chromosomal instability and prognosis of breast cancer patients in silico and radiosensitivity and DNA repair in breast cancer cells in vitro. The investigations in silico were performed using overall survival and USP7 mRNA expression data of breast cancer patients. The results showed that a high USP7 expression was associated with increased chromosomal instability and decreased overall survival. The in vitro experiments were performed in a luminal and a triple-negative breast cancer cell line. Proliferation, DNA repair, DNA replication stress, and survival after USP7 overexpression or inhibition and irradiation were analyzed. Both, USP7 inhibition and overexpression resulted in decreased cellular survival, distinct radiosensitization and an increased number of residual DNA double-strand breaks in the S phase following irradiation. RAD51 recruitment and base incorporation were decreased after USP7 inhibition plus irradiation and more single-stranded DNA was detected. The results show that deregulation of USP7 activity disrupts DNA repair in the S phase by increasing DNA replication stress and presents USP7 as a promising target to overcome the radioresistance of breast tumors.

Mineral identification using machine learning requires a significant amount of training data. We built a library of 2D hyperspectral images of minerals. The library contains reflectance images of 130 samples, of 76 distinct minerals, with more than 3.9 million data points. In order to produce this dataset, various well-characterized mineral samples were scanned, using a SPECIM Short Wave Infrared (SWIR) camera, which captures wavelengths from 900 to 2500 nm. Minerals were selected to represent all the mineral classes and the most common mineral occurrences. For each sample, the following data are provided: (a) At least one hyperspectral image of the sample, consisting of 256 wavelengths between 900 and 2500 nm. The raw data, the high dynamic range (HDR) image, and the masked HDR image are provided for each scan (each of them in HDF5 format). (b) A text file describing the sample, providing supplementary information for the subsequent analysis (c) RGB images (JPEG files) and automated 3D reconstructions (Stanford Triangle PLY files). These data help the user to visualize and understand specific sample characteristics. 2D hyperspectral images were produced for each mineral, which consist of many different spectra with high diversity. The scans feature similar spectra than the ones in other available spectral libraries. An artificial neural network was trained to demonstrate the high quality of the dataset. This spectral library is mainly aimed at training machine learning algorithms, such as neural networks, but can be also used as validation data for other types of classification algorithms.

The stable isotope composition of waters (delta2H, delta18O) can be used as a natural tracer of hydrologic processes in systems affected by acid mine drainage. We investigated the delta2H and delta18O values of pore waters from four oxidizing sulfidic mine tailings impoundments in different climatic regions of Chile (Piuquenes at La Andina with Alpine climate, Cauquenes and Carén at El Teniente with Mediterranean climate, and Talabre at the Chuquicamata deposit with hyperarid climate). No clear relationship was found between altitude and isotopic composition. The observed displacement of the tailings pore waters from the local meteoric water line toward higher delta18O values (by approximately +2 per thousand delta18O relative to delta2H) is partly due to water-rock interaction processes, including hydration and O-isotope exchange with sulfates and Fe(III) oxyhydroxides produced by pyrite oxidation. In most tailings, from the saturated zone toward the surface, isotopically different zones can be distinguished. Zone I is characterized by an upward depletion of 2H and 18O in the pore waters from the saturated zone and the lowermost vadose zone, due to ascending diffused isotopically light water triggered by the constant loss of water vapor by evaporation at the surface. In zone II, the capillary flow of a mix of vapor and liquid water causes an evaporative isotopic enrichment in 2H and 18O. At the top of the tailings in dry climate a zone III between the capillary zone and the surface contains isotopically light diffused and atmospheric water vapor. In temperate climates, the upper part of the profile is affected by recent rainfall and zone III may not differ isotopically from zone II.
