RESUMO

OBJECTIVE: To investigate the changes in serum homeobox A9 (HOXA9 ), soluble E-cadherin (SE-CAD) and type Ⅲ procollagen (PCⅢ) levels in acute myeloid leukemia (AML) patients after chemotherapy with DCAG regimen and their relationship with prognosis. METHODS: The clinical data of 80 patients with relapsed/refractory AML diagnosed and treated in our hospital from March 2018 to December 2021 were retrospectively analyzed. According to different treatment regimen, the patients were divided into DCAG group (n=40) and CAG group (n=40). The clinical efficacy and changes of HOXA9 , SE-CAD and PCⅢ levels before and after treatment were compared. In addition, all patients were divided into remission group (n=58) and non-remission group (n=22) according to the clinical efficacy. Univariate and multivariate analyses were performed to analyze the risk factors affecting the prognosis of AML patients. The predictive efficacy of the three single indicators, HOXA9 , SE-CAD, and PC III, and their combination on prognosis was analyzed. RESULTS: Compared with before treatment, the levels of HOXA9 , SE-CAD and PCⅢ in both the DCAG and CAG groups were decreased after treatment, and the improvement of each indicator and the clinical efficacy in the DCAG group were significantly better than those in the CAG group (all P < 0.05). Multivariate analysis showed that increased bone marrow blast count, HOXA9 mRNA, SE-CAD and PCⅢ levels were independent risk factors affecting the efficacy of chemotherapy in AML patients (all P < 0.05). ROC curves showed that the combination of HOXA9 mRNA, SE-CAD and PCIII could effectively predict the prognosis of AML patients, with a sensitivity of 84.80% and a specificity of 88.20%. CONCLUSION: DCAG regimen can significantly improve the levels of HOXA9 mRNA, SE-CAD and PCⅢ in AML patients, these three indicators are all independent risk factors affecting the prognosis of AML patients, and the combination of the three indicators can effectively predict the prognosis of the patients.

RESUMO

An electrochemistry-promoted oxidative cleavage of (sp3)C-C(sp3)/H bonds in alkylarenes was developed. Various aryl alkanes can be smoothly converted into ketones/aldehydes under aerobic conditions using a user-friendly undivided cell setup. The features of air as oxidant, scalability, and mild conditions make them attractive in synthetic organic chemistry.

BACKGROUND: With the development of instrument technology, the functions and detection methods of automatic blood cell analyzers have become more complex. To ensure optimal clinical applicability, it is crucial to select an automatic blood cell analyzer with excellent clinical detection performance. This study evaluated the latest Mindray BC-6800Plus automatic blood cell analyzer and assessed its performance in the detection of nucleated red blood cells (NRBCs). METHODS: A total of 490 clinical blood samples were used to assess the performance of the instrument, including parameters such as precision, linearity, conformity rate of manual microscopic examination, carryover, and limit of quantitation. RESULTS: The instrument showed a small carryover (≤0.02) and excellent linearity (R2≥0.9986). The reproducibility of the sample tests was satisfactory, and the coefficient of variation (CV) of the test results [0.98-1.72% and 0.62-6.97% for white blood cells (WBCs) and NRBCs, respectively] were significantly lower than that declared by the manufacturer (2.5% and 20% for WBCs and NRBCs, respectively). Thus, the BC-6800Plus satisfies the requirements of clinical testing. Two separate Mindray BC-6800Plus machines were tested and found to be in good agreement with each other and with manual microscopy methods. Furthermore, WBC and NRBC counts were highly consistent with results obtained using the XN-9100 blood analyzer. CONCLUSIONS: The Mindray BC-6800Plus is an excellent analyzer that can provide timely and accurate reports for clinical laboratory detection of NRBC.

RESUMO

A site-specific oxidation of (sp3)C-C(sp3) and (sp3)C-H bonds in aryl alkanes by the use of NaNO2/HCl was explored. The method is chemical-oxidant-free, transition-metal-free, uses water as the solvent, and proceeds under mild conditions, making it valuable and attractive to synthetic organic chemistry.
