In view of the fact that a specific migration limit (SML) is to be established in the near future for styrene monomer in plastic food contact materials (FCMs), data on the dietary exposure of the European population, as well as sensitive and reliable analytical methodologies to implement compliance testing, are needed. The properties of the substance styrene as well as those of styrenic polymers pose challenges for analysts and their design of experimental migration tests. The aim of this study was to assess the level of styrene in polystyrene (PS)-packed dairy products from supermarkets and compare these values with the results from simulated migration testing. In addition to the conventional food simulant and test conditions described in Regulation (EU) No 10/2011 for refrigerated dairy products (50% ethanol for 10 days at 40 °C), milder simulants and test conditions (10% ethanol and 20% ethanol for 10 days at 40 °C and 20 °C) were investigated. Styrene levels in the investigated foods ranged from 2.8 µg/kg to 22.4 µg/kg. The use of 50% ethanol causes interactions with PS (swelling) that do not occur with dairy products and leads to highly exaggerated migration results. In contrast, testing PS for 10 days at 40 °C with 10% and 20% ethanol leads to higher styrene migration levels than found in real food, which are still conservative but far less extreme. Testing PS for 10 days at 20 °C leads to styrene migration levels that are more comparable to, but still overestimate, those found in real food products stored under refrigerated conditions.

The European Food Safety Authority is re-evaluating styrene for assessing the safety of food contact materials (FCM) such as polystyrene (PS) and started a systematic review of the data on migration levels in food. A restriction for styrene is expected in the near future. The main food contact application of PS is dairy packaging, mainly at refrigerated storage. In this study, seventeen dairy products packed in PS taken from the Italian and German markets were investigated. Styrene concentrations in the refrigerated dairy products (yogurt, cream) ranged from 5 to 30 µg/kg at the best before date, while in single serving portions of coffee creamer, which were stored at room temperature until the best before date of approx. 190 days, 401 µg/kg were measured. Among several parameters, the ratio between the surface contact area of the package and the quantity of the food packed, the time/temperature conditions of production/filling and storage of the products were identified as the main factors influencing styrene migration into food under realistic conditions. Yogurts fermented in the pots for approximately 8 h at 40-50 °C showed higher styrene levels than those fermented in an incubator and filled at 20 °C. The fat content might influence the styrene level but the effect, if any, was too small in relation to the variability of other parameters. Levels of styrene migrating into 50% ethanol food simulant under standardized condition (10 days/40 °C) were found to be much higher than levels in refrigerated foods. This raises the question as to whether compliance testing for PS plastics should be adapted taking into consideration the correlation between migration testing by laboratory simulations and migration into real food.

Migration kinetic data from general purpose polystyrene (GPPS) and high impact polystyrene (HIPS) were generated for a set of model substances as well as styrene monomer and oligomers at different temperatures (20 °C, 40 °C, 60 °C) using food simulants stipulated in the European Regulation (EU) 10/2011 and real foods like milk, cream and olive oil (20 °C, 40 °C). The extent of polymer swelling was characterized gravimetrically and visual changes of the test specimens after migration contact were recorded. Isooctane and 95% ethanol caused strong swelling and visual changes of HIPS, overestimating real migration into foods especially at high temperatures; GPPS was affected by isooctane only at 60 °C. With 50% ethanol, after 10 days contact at 60 °C or 40 °C both polymers were slightly swollen. Contrary, most of the real foods analyzed caused no detectable swelling or visual changes of the investigated polymers. This study demonstrates that the recommendations provided by EU regulations are not always in agreement with the physicochemical properties of styrenic polymers. The critical point remains the selection of adequate food simulants/testing conditions, since the high overestimation of aggressive media can lead to non-compliance of polystyrene materials even if the migration into real food would be of no concern.

Polyethylene terephthalate (PET) is one of the most widely used food contact materials due to its excellent mechanical properties and recyclability. Migration of substances from PET and assessment of compliance are usually determined by experimental testing, which can be challenging depending on the migrants of interest. Low concentrations and missing reference standards, among other factors, have led to inadequate investigation of the migration potential of PET oligomers. Migration modeling can overcome such limitations and is therefore a suitable starting point for exposure and risk assessment. In this study, the activation energy-based (EA) model and the AP model were used to systematically evaluate the migration potential of 52 PET oligomers for 12 different application scenarios. Modeling parameters and conditions were evaluated to investigate their impact and relevance on the assessment of realistic exposures. Obtained results were compared with safety thresholds known from the concept of toxicological thresholds of concern. This allowed the evaluation and identification of oligomers and/or applications where migration or exposure levels may be associated with a potential risk because they exceed these safety thresholds. Overall, this study demonstrated that migration modeling can be a high-throughput, fast, flexible, and suitable approach for comprehensive exposure assessment.

General Purpose Polystyrene (GPPS) and High Impact Polystyrene (HIPS) is used in packaging food as well as for technical products. Knowledge of the diffusion behavior of organic molecules in polystyrene (PS) is important for the evaluation of the diffusion and migration process. Within this study, diffusion coefficients were determined in GPPS and HIPS below and above the glass transition temperature. Diffusion coefficients were determined from desorption kinetics into the gas phase using spiked GPPS and HIPS sheets as well as from permeation kinetics through a thin GPPS film. Overall, 187 diffusion coefficients were determined in GPPS and HIPS at temperatures between 0 °C and 115 °C. From the temperature dependency of the diffusion coefficients 45 activation energies of diffusion EA and the pre-exponential factor D0 were determined. As expected, the activation energies of diffusion EA show a strong dependency from the molecular volume of the investigated substances. At the glass transition temperature, only a slight change of the diffusion behavior were observed. Based on EA and D0, prediction parameters for diffusion coefficients were established.

Food packaging films were coated with polyvinyl acetate (PVA) containing different concentrations of citral or Litsea (L.) cubeba essential oil (EO). Antimicrobial contact trials in style of ISO22916 were performed. Citral coatings achieved bactericidal effects against Escherichia coli (2.1 log) and Staphylococcus aureus (4.3 log) at concentrations of 20%DM. L. cubeba inactivated more than 4 log cycles of both bacteria at a concentration of 20%DM. To determine the antimicrobial activity across the gas phase, a unique method for volatile agents was developed, adapting ISO22196. GC/MS measurements were performed to supplement microbiological tests in a model packaging system with a defined 220 ml headspace (HS). HS-equilibrium concentrations of 1.8 µg/mlAir were found for 20%DM 'citral-coatings, resulting in antimicrobial effects of 3.8 log against of E. coli. Saccharomyces cerevisiae (4.74 log) and Aspergillus niger (4.29 log) were more effectively inactivated by 3%DM and 5%DM coatings. In an application trial with strawberries, simulating a headspace packaging, growth inhibitory effects on the yeast and mold microbiota were found for the 20%DM coatings.

Food contact materials (FCMs) can transfer chemicals arising from their manufacture to food before consumption. Regulatory frameworks ensure consumer safety by prescribing methods for the assessment of FCMs that rely on migration testing either into real-life foods or food simulants. Standard migration testing conditions for single-use FCMs are justifiably conservative, employing recognized worst-case contact times and temperatures. For repeated-use FCMs, the third of three consecutive tests using worst-case conditions is taken as a surrogate of the much shorter contact period that often occurs over the service life of these items. Food contact regulations allow for the use of migration modelling for the chemicals in the FCM and for the partitioning that occurs between the FCM and food/simulant during prolonged contact, under which steady-state conditions are favored. This study demonstrates that the steady-state is rarely reached under repeated-use conditions and that partitioning plays a minor role that results in migration essentially being diffusion controlled. Domains of use have been identified within which partitioning does not play a significant role, allowing modelling based upon diffusion parameters to be used. These findings have the potential to advance the modelling of migration from repeated-use articles for the benefit of regulatory guidance and compliance practices.

Lacking recyclability of multilayer packaging can be overcome by using a thermoreversible crosslinking adhesive consisting of maleimide- and furan-functionalized polyurethane-(PU-)prepolymers, reacting in a Diels-Alder-reaction. Here, the furan-functionalized PU-prepolymer carries furan-side-chains to avoid the usage of an additional crosslinking agent. Thus, N­(2­hydroxyethyl)maleimide and furfurylamine are the only two chemicals contained in the adhesive that are not listed in the appendix of EU Regulation 10/2011. Using migration modelling, it could be shown that, at 23 °C, both chemicals have lag-times of only a few minutes if 45 µm PE is used as a barrier. However, if the residual content is below 30 mg/kg, the legally specified maximum amount of 0.01 mg/kg food is not reached. After determining the diffusion coefficients and the activation energy of diffusion through ethylene-vinyl alcohol copolymer (EVOH), it could be determined that the lag-time of the migrants can be extended to at least 9 years by the use of 3 µm EVOH. From a food law point of view, the use of the described adhesive is possible if the above­mentioned measures are complied.

PET beverage bottles have been recycled and safely reprocessed into new food contact packaging applications for over two decades. During recollection of post-consumer PET beverage bottles, PET containers from non-food products are inevitably co-collected and thereby enter the PET recycling feed stream. To explore the impact of this mixing on the safety-in-use of recycled PET (rPET) bottles, we determined the concentrations of post-consumer substances in PET containers used for a range of non-food product applications taken from the market. Based on the chemical nature and amounts of these post-consumer substances, we evaluated their potential carry-over into beverages filled in rPET bottles starting from different fractions of non-food PET in the recollection systems and taking worst-case cleaning efficiencies of super-clean recycling processes into account. On the basis of the Threshold of Toxicological Concern (TTC) concept and Cramer classification tools, we present a risk assessment for potential exposure of the consumer to the identified contaminants as well as unidentified, potentially genotoxic substances in beverages. As a result, a fraction of 5% non-food PET in the recycling feed stream, which is very likely to occur in the usual recollection systems, does not pose any risk to the consumer. Our data show that fractions of up to 20%, which may sporadically be contained in certain, local recollection systems, would also not raise a safety concern.

Solute diffusion in solid polymers has tremendous applications in packaging, reservoir, and biomedical technologies but remains poorly understood. Diffusion of non-entangled linear solutes with chemically identical patterns (blobs) deviates dramatically in polymers in the solid-state (αlin > 1, Macromolecules 2013, 46, 874) from their behaviors in the molten state (αlin = 1, Macromolecules, 2007, 40, 3970). This work uses the scale invariance of the diffusivities, D, of linear probes D(N·Mblob + Manchor,T,Tg) = N-αlin(T,Tg)D(Mblob + Manchor,T,Tg) comprising N identical blobs of mass Mblob and possibly one different terminal pattern (anchor of mass Manchor) to evaluate the amounts of hole-free volume in seven polymers (aliphatic, semi-aromatic and aromatic) over a broad range of temperatures (-70 K ≤T-Tg≤ 160 K). The new parameterization of the concept of hole-free volumes opens the application of the free-volume theory (FVT) developed by Vrentas and Duda to practically any polymer, regardless of the availability of free-volume parameters. The quality of the estimations was tested with various probes including n-alkanes, 1-alcohols, n-alkyl acetates, and n-alkylbenzene. The effects of enthalpic and entropic effects of the blobs and the anchor were analyzed and quantified. Blind validation of the reformulated FVT was tested successfully by predicting from first principles the diffusivities of water and toluene in amorphous polyethylene terephthalate from 4 °C to 180 °C and in various other polymers. The new blob model would open the rational design of additives with controlled diffusivities in thermoplastics.

Microplastics in food is a relatively new research field with only few studies available so far. Scientists have been pointing out that some of these studies apply questionable analytical methods. Nevertheless, media often use such results to gain attention of the readers. It is therefore of particular significance, that only those scientific studies are published, clearly presenting valid data on the content of microplastics in food. Unfortunately, the study by Zuccarello et al. shows very critical aspects regarding analytical methods used and conclusions made. The applied procedure is not described and, therefore, does not allow any assessment by other groups, which is indispensable prerequisite of any scientific publication. Moreover, the analytical method used for the identification and quantification of microplastic particles - SEM-EDX - is not sound and not validated. Therefore, in our opinion the results on the contamination of bottled mineral water with microplastics published by Zuccarello et al. are more than questionable.

Microplastics have been ubiquitously found and identified in aquatic and terrestrial environments for several years. Due to their occurrence in the oceans, microplastics were also found and characterised in seafood products and in other foods and beverages such as beer, honey and table salt. Very recently, microplastic particles were also determined in bottled mineral water. The objective of this publication is to present and discuss a compilation of the currently available literature data on microplastics in bottled mineral water. The related oral exposure of the consumer from substances present in microplastics and from the plastics particles themselves is estimated and toxicological arguments for and considerations on risk assessment from the consumption of bottled mineral water containing microplastics are presented. Exposure estimations based on the reported microplastic amounts found in mineral water and the assumption of total mass transfer of small molecules like additives and oligomers present in the plastic would not raise a safety concern. Available toxicokinetic data suggests that marginal fraction of the ingested low amount of microplastics can be absorbed, if at all, the conclusion is very likely that the reported amounts present in bottled mineral water do not raise a safety concern for the consumer. Considering the use of plastic materials in our daily life, occurrence of microplastics in beverages is likely a minor exposure pathway for plastic particles. Due to recent progress in analytical methods and the public discussion on plastics marine litter, public concern about eating and drinking microplastics with food, and related safety issues was raised. However, a better data basis for exposure estimates and risk assessment would be very helpful to better accommodate consumer concerns. The intention of this paper is to deliver a contribution to this topic taking the bottled mineral water as a case example.

Polyethylene terephthalate (PET) bottles are widely used as packaging material for natural mineral water. However, trace levels of acetaldehyde can migrate into natural mineral water during the shelf life and might influence the taste of the PET bottled water. 2-Aminobenzamide is widely used during PET bottle production as a scavenging agent for acetaldehyde. The aim of this study was the determination of the migration kinetics of 2-aminobenzamide into natural mineral water as well as into 20% ethanol. From the migration kinetics, the diffusion coefficients of 2-aminobenzamide in PET at 23 and 40°C were determined to be 4.2 × 10(-)(16) and 4.2 × 10(-)(15) cm(2) s(-1), respectively. The diffusion coefficient for 20% ethanol at 40°C was determined to be 7.7 × 10(-)(15) cm(2) s(-1), which indicates that 20% ethanol is causing swelling of the PET polymer. From a comparison of migration values between 23 and 40°C, acceleration factors of 9.7 when using water as contact medium and 18.1 for 20% ethanol as simulant can be derived for definition of appropriate accelerated test conditions at 40°C. The European Union regulatory acceleration test based on 80 kJ mol(-1) as conservative activation energy overestimates the experimentally determined acceleration rates by a factor of 1.6 and 3.1, respectively.

Cyclo olefin polymer (COP) is an amorphous polymer with good optical transparency and barrier properties, which is increasingly used for pharmaceutical packaging applications like pre-filled syringes, plastic vials, nutrition bags and blisters as well as for micro-well plates. For regulatory purposes, it is important to know the amount and quantity of compounds which migrate from the polymer into the pharmaceutical product. Within the study, diffusion coefficients of organic (model) compounds in COP at various temperatures were determined and the activation energies of diffusion were calculated according to the Arrhenius approach. Correlations were established between the molecular volume V of the migrating compound and the activation energy of diffusion EA as well as between the pre-exponential factor in the Arrhenius equation D0 and EA. From these correlations a prediction model was established for the migration of organic compounds in COP. This might be a useful tool supporting the evaluation process of COP packed pharmaceutical products.

Plastic bags and tubes are increasingly used for the storage and application of pharmaceutical formulations. The most common polymer material for drug application sets is plasticized poly(vinylchloride) (PVC). During application of pharmaceutical drug solution through PVC tubes, the polymer and the contact media interact which leads to leaching out of polymer additives or sorption of ingredients of the drug solution. Whereas the discussion of leaching of plasticizers is focussed on the toxicological properties of a drug packaging system, the sorption of drug formulation compounds has an influence on the dosage of the active pharmaceutical ingredient resulting in a reduced drug delivery to the patient. Therefore sorption has an influence on the effectiveness and success of the therapy. Within the study, the concentration profiles of nitroglycerin and diazepam solutions were determined after pumping the solutions through infusion administration sets. The study includes plasticized PVC tubes with different plasticizers (DEHP, DEHA, DEHT, TEHTM, DINCH, poly adipate), PVC (DEHP) tubes with different shore hardness as well as alternative polymer materials like EVA, TPE, PUR, silicone, LDPE and PP. From the experimental concentration curves it could be shown, that in the first minutes of the application of the drug solution the sorption of the active compound is at its maximum, resulting in the lowest concentration in the applied pharmaceutical solution. For a PVC tube with DEHP as plasticizer and a shore hardness of 80 only about 57% of the initial nitroglycerin concentration in the solution is applied to the patient in the first minutes of the application. For PVC tube (DEHP, shore 80) the experimental data were simulated using mathematical diffusion models. The concentration profiles during application could be well simulated using the partition coefficient K=50 (nitroglycerin) and K=300 (diazepam), respectively. However, the experimental results indicate, that the sorption of nitroglycerin into the PVC tube alters the diffusion behaviour of the polymer over flow time, which results in an increase of the diffusion coefficient during application. On the other hand, the other investigated alternative tube materials like PE or PP show a significantly lower sorption compared to PVC plasticizer systems. Due to the fact that the amount of sorption is varying over time, the concentration of the active pharmaceutical compound in the solution after passing the infusion administration set is not constant which makes the application of a constant concentration of a certain active ingredient to the patient very difficult. The simulated partition and diffusion coefficients for given PVC(DEHP) tubes were therefore used to simulate the initial concentrations profile of the feeding drug solution to assure a constant concentration flow profile after passing the administration set. The proposed methodology of this study represents a straight forward approach for the assessment of the drug sorption in dynamic flow systems based on experimental data as well as mathematical diffusion modelling. From the results a non-constant initial concentration profile for the active ingredient in a pharmaceutical drug solution can be established in order to compensate the loss of the pharmaceutical compound by sorption during infusion.

The barrier effect of a silicon oxide (SiOx) coating on the inner surface of PET bottles, in terms of the ability to reduce the migration of post-consumer compounds from the PET bottle wall into food simulants (3% acetic acid and 10% ethanol), was investigated. The barrier effect was examined by artificially introducing model substances (surrogates) into the PET bottle wall to represent a worst-case scenario. Test bottles with three different spiking levels up to approximately 1000 mg kg(-1) per surrogate were blown and coated on the inner surface. The SiOx-coated bottles and the non-coated reference bottles were filled with food simulants. From the specific migration of the surrogates with different bottles wall concentrations, the maximum surrogate concentrations in the bottle wall corresponding to migration of 10 microg l(-1) were determined. It was shown that the SiOx coating layer is an efficient barrier to post-consumer compounds. The maximum bottle wall concentrations of the surrogates corresponding to migration of 10 microg l(-1) were in the range of 200 mg kg(-1) for toluene and approximately 900 mg kg(-1) for benzophenone. Consequently, the SiOx coating allows use of conventionally recycled post-consumer PET flakes (without a super-clean recycling process) for packaging aqueous and low alcoholic foodstuffs (under cold-fill conditions) and protects food from migration of unwanted contaminants from post-consumer PET.

The aim of the study was to investigate and evaluate the cleaning efficiency of a new recycling concept for post-consumer poly(ethylene terephthalate) (PET). The so-called Flake To Resin (FTR) recycling process produces PET pellets or preforms from conventionally recycled PET flakes for the application in new PET packaging in direct food contact. The investigated process can be considered as 'super-clean' recycling process and was developed to introduce conventional recycled post-consumer (PCR) PET flakes up to an amount of 50% into the pellet and preform production. Within the study the cleaning efficiency of the investigated FTR process was determined by a challenge test. The experimental results obtained from three challenge tests with different input concentrations of the surrogates and different amounts of post-consumer PET flakes show that all applied surrogates are very efficiently removed by the investigated recycling process. The cleaning efficiencies for all surrogates are above 99.9%. In the final product of the process no surrogates could be determined above the detection limits (0.5 mg kg(-1)) even if the initial concentrations were in the percentage range. From a migrational point the final product, which are either PET pellets or preforms, was similar to virgin PET. Only the PET typical substances acetaldehyde and ethylene glycol had slightly higher concentrations than found in a conventional PET virgin sample used as reference. The generally accepted migration limit of 10 microg kg(-1) for the surrogates in the final products of the challenge tests is established for all kinds of foodstuffs.

RESUMO

ESR spectra of the captodative alpha-amino-alpha-carbonylmethyl radicals 8 have been recorded. No coalescence temperature for the rotation of the two NMe groups was found at temperatures below the decomposition temperature of the radicals. From known coalescence temperatures and rotational barriers of substituted methyl radicals the rotational barrier of >/=17 kcal mol(-)(1) was estimated for the (*)C-N bond in the radicals 8. Enthalpies DeltaH(diss) and entropies DeltaS(diss) of the homolytic dissociation of 7a,c,d into 8a,c,d have been obtained from equilibrium measurements by ESR. By correcting for substituent interaction enthalpies in 7 (steric and geminal), a radical stabilization enthalpy RSE = -20.7 +/- 1.0 kcal mol(-)(1) was obtained for 8. By addition of the known RSEs of dialkylamino- and carbonyl groups, a RSE = -9.9 kcal mol(-)(1) is predicted for 8. The difference between the experimental and predicted values of 10.8 kcal mol(-)(1) is attributed to a synergistic captodative substituent effect. A linear correlation between the radical stabilization enthalpies of the radicals 8 and of other mono- and disubstituted alkyl radicals and their ESR aH(alpha) coupling constants was found. According to this correlation the reduction of aH(alpha) by 1 G corresponds to an increase in RSE of 1.57 kcal mol(-)(1). The large resonance of the captodative alpha-amino-alpha-carbonylmethyl radicals 3, expressed by their high RSE, their small aH(alpha) coupling constant, and their high rotational barrier, can be rationalized by a strong interaction between the alpha-amino and the alpha-carbonyl groups similar to that in amides and expressed in the resonance structures 6.
