RESUMO

Bismuth-iron semiconductor materials have been widely studied in the photocatalytic field due to their excellent light responsiveness. Among them, the potential and mechanism regarding photocatalytic degradation of organic pollutants by Bi2Fe4O9 are seriously ignored. In this research, Bi2Fe4O9/reduced graphene oxide (BFO/rGO) was successfully synthesized for tetracycline (TC) removal. Under visible light irradiation, the TC degradation efficiency reached 83.73% within 60 min, which was much higher than that of pure BFO or rGO. The impacts of crucial factors (TC initial concentration, humic acid concentration, pH value and inorganic anions) were systematically analyzed. The photoelectric performance experiments indicated that the addition of rGO decreased the electron-hole pair recombination efficiency and improved the charge transfer efficiency, thus significantly enhancing the photocatalytic performance. According to quenching experiments and EPR (Electron Paramagnetic Resonance) analysis, superoxide radical (•O2-) and hole (h+) were determined as the main active species during degradation reactions. Eventually, the possible degradation routes of TC were presented by identifying intermediates.

BACKGROUND: B-cell lymphoma 6 (BCL6) is a transcription repressor that plays a tumor suppressor or promoting role in various tumors. However, its function and molecular mechanism in gastric cancer (GC) remain unclear. Ferroptosis, a novel programmed cell death, is closely related to tumor development. In this research, we aimed to explore the role and mechanism of BCL6 in malignant progression and ferroptosis of gastric cancer. METHODS: Firstly, BCL6 was identified as an important biomarker that attenuated the proliferation and metastasis of GC through tumor microarrays and confirmed in GC cell lines. RNA sequence was performed to explore the downstream genes of BCL6. The underlying mechanisms were further investigated by ChIP, dual luciferase reporter assays and rescue experiments. Cell death, lipid peroxidation, MDA and Fe2+ level were detected to determine the effect of BCL6 on ferroptosis and the mechanism was revealed. CHX, MG132 treatment and rescue experiments were used to explore the upstream regulatory mechanism of BCL6. RESULTS: Here we showed that BCL6 expression was significantly decreased in GC tissues, and patients with low BCL6 expression showed more malignant clinical features and poor prognosis. The upregulation of BCL6 may significantly inhibited the proliferation and metastasis of GC cells in vitro and in vivo. In addition, we found that BCL6 directly binds and transcriptionally represses Wnt receptor Frizzled 7 (FZD7) to inhibit the proliferation, metastasis of GC cells. We also found that BCL6 promoted lipid peroxidation, MDA and Fe2+ level to facilitate ferroptosis of GC cells by FZD7/ß-catenin/TP63/GPX4 pathway. Furthermore, the expression and function of BCL6 in GC were regulated by the ring finger protein 180 (RNF180)/ras homolog gene family member C (RhoC) pathway, which had been elucidated to be involved in significantly mediating the proliferation and metastasis of GC cells. CONCLUSIONS: In summary, BCL6 should be considered a potential intermediate tumor suppressor to inhibit the malignant progression and induce ferroptosis, which might be a promising molecular biomarker for further mechanistic investigation of GC.

The effect of ALDOA, an important regulator of tumor metabolism and immune cell function, on gastric cancer (GC) immune infiltration has not been elucidated. Hence, we explored the feasibility of using ALDOA combined with immune molecular markers as novel prognostic or therapeutic targets for GC patients. Bioinformatic analyses were initially performed in multiple databases to assess the prognostic prediction values of ALDOA expression in GC. Subsequently, both ALDOA expression and the clinicopathological characteristics of a total of 114 GC patients who underwent curative gastrectomy were collected to demonstrate the potential association between ALDOA expression and the biological behaviors of GC. Next, the expression of ALDOA and its effect on prognosis were determined at the mRNA and protein levels, respectively, using tissue microarrays and cellular experiments. Subsequently, several molecular mechanisms were revealed based on elaborate analyses, indicating that ALDOA expression was potentially involved in the progression of GC and could be considered a promising biomarker for evaluating the prognosis of GC. High ALDOA expression was frequently found in GC cells and GC tissues at the mRNA and protein levels. Based on survival analysis, the expression of ALDOA indicated comparatively poor overall survival (OS) in GC and was identified as an independent prognostic predictor of GC. Correlation analysis showed that ALDOA expression had a positive association with lymph node metastasis in GC patients. Additionally, microRNA-1179 was found to play a key role in inhibiting the expression of ALDOA in the metabolic pathways of GC cells, which might disrupt the expression of various immune molecules and be detrimental to the prognosis of GC. ALDOA should be considered a promising molecular target for evaluating the prognosis of GC, owing to its potential role in immune regulation.

Cancer patients undergoing paclitaxel infusion usually experience peripheral nerve degeneration and serious neuropathic pain termed paclitaxel-induced peripheral neuropathy (PIPN). However, alterations in the dose or treatment schedule for paclitaxel do not eliminate PIPN, and no therapies are available for PIPN, despite numerous studies to uncover the mechanisms underlying the development/maintenance of this condition. Therefore, we aimed to uncover a novel mechanism underlying the pathogenesis of PIPN. Clinical studies suggest that acute over excitation of primary sensory neurons is linked to the pathogenesis of PIPN. We found that paclitaxel-induced acute hyperexcitability of primary sensory neurons results from the paclitaxel-induced inhibition of KCNQ potassium channels (mainly KCNQ2), found abundantly in sensory neurons and axons. We found that repeated application of XE-991, a specific KCNQ channel blocker, induced PIPN-like alterations in rats, including mechanical hypersensitivity and degeneration of peripheral nerves, as detected by both morphological and behavioral assays. In contrast, genetic deletion of KCNQ2 from peripheral sensory neurons in mice significantly attenuated the development of paclitaxel-induced peripheral sensory fiber degeneration and chronic pain. These findings may lead to a better understanding of the causes of PIPN and provide an impetus for developing new classes of KCNQ activators for its therapeutic treatment.

Background: Recently, the role of pyroptosis in cancer has attracted people's attention, but its function in colon cancer remains unclear. This study aimed to construct a pyroptosis-related model that can effectively predict the prognosis of patients with colon cancer and explore the potential functions of pyroptosis-related genes. Methods: We identified 40 differentially expressed PRGs between colon cancer and normal colon tissues. The model was established using the least absolute shrinkage and selection operator (LASSO) Cox regression method, and the patients were divided into high- and low-risk groups. Finally, we verified its biological function in vitro based on three PRGs and demonstrated discrepant expression of PRGs within colon cancer and non-tumor tissues at the protein level with immunohistochemistry. Results: A pyroptosis-related prognosis model was constructed, which divided 446 patients with colon cancer into high- and low-risk groups. Kaplan-Meier analysis results showed that the survival of patients with colon cancer in the high-risk group was worse than that in the low-risk group. Finally, we also confirmed that this score is an independent prognostic factor for colon cancer progression. Conclusion: In summary, the model established by three PRGs was a reliable indicator for predicting prognosis, suggesting that pyroptosis might be a noteworthy therapeutic target in CC.

Colorectal cancer (CRC) is one of the most common cancers of the digestive tract, and patients with advanced-stage cancer have poor survival despite the use of multidrug conventional chemotherapy regimens. Intra-tumor heterogeneity of cancerous cells is the main obstacle in the way to effective cancer treatments. Therefore, we are looking for novel approaches to eliminate just cancer cells including nanoparticles (NPs). PPy@Fe3O4 NPs were successfully synthesized through a portable method. The characterization of transmission electron microscopy (TEM), Fourier-Transformed infrared spectrometer, and X-ray powder diffraction have further proved successful preparation of PPy@Fe3O4 NPs. NIR irradiation was used to test the photothermal properties of NPs and an infrared camera was used to record their temperature. The direct effects of PPy@Fe3O4 NPs on colorectal cancer cell DLD1 were assessed using CCK8, plate clone, transwell, flow cytometry, and western blotting in CRC cell. The effect of PPy@Fe3O4 NPs on neoplasm growth in nude mice was evaluated in vivo. This study demonstrated that PPy@ Fe3O4 NPs significantly inhibit the growth, migration, and invasion and promote ferroptosis to the untreated controls in colorectal cancer cells. Mechanical exploration revealed that PPy@Fe3O4 NPs inhibit the multiplication, migration, and invasion of CRC cells in vitro by modulating the NF-κB signaling pathway. Importantly, Ferroptosis inhibitors Fer-1 can reverse the changes in metastasis-associated proteins caused by NPs treatment. Collectively, our observations revealed that PPy@Fe3O4 NPs were blockers of tumor progression and metastasis in CRC. This study brought new insights into bioactive NPs, with application potential in curing CRC or other human disorders.

Partially cystic thyroid nodules (PCTNs) are a kind of thyroid nodule with both solid and cystic components, and are usually misdiagnosed as benign nodules. The objective of this study was to determine the ultrasound (US) characterizations with a TIRADS Grade-4a or higher partially cystic thyroid nodules (PCTNs) which are associated with being malignant or benign. In this study, 133 PCTNs with a TIRADS Grade-4a or higher were enrolled; 83 were malignant and 50 were benign. TI-RADS classification can detect malignant PCTNs, and its sensitivity, specificity, positive predictive value, negative predictive value, and accuracy are 39.8%, 96.0%, 94.3%, 49.0%, and 60.9%, respectively. Univariate analyses revealed that nodule shape, margin, and structure were related to PCTNs' being benign and malignant, among which nodules taller-than-wide, with an irregular shape, non-smooth margin, eccentric sharp angle, or edge sharp angle were significantly associated with malignancy while ovoid to round nodules, smooth margins, multiple separation, and eccentric obtuse angle structures were significantly associated with a benign nature. For the solid part of PCTNs, its free margin, echo, and calcification are related to benign and malignant PCTNs. Among them, the free margin of the solid part is non-smooth, hypoechoic, and microcalcification, which are related to malignant PCTNs, while the free margin of the solid part is smooth, isoechoic, macrocalcification, non-calcification and are related to benign PCTNs. Calcification of solid part and free margin are important factors for predicting malignant PCTNs. In addition, nodules' composition, blood flow signal, and other factors had nothing to do with PCTNs' being benign or malignant. In the multivariate Logistic regression analysis, solid part calcification (OR: 17.28; 95%CI: 5.14~58.08) and free margin (OR: 3.18; 95%CI: 1.01~10.00) were revealed to be the strongest independent predictors for malignancy (P<0.05). Our study indicated that understanding the ultrasound characteristics of malignant PCTNs, to avoid misdiagnosed PCTNs patients, is important to make a precise diagnosis and prognosis of PCTNs.

This article investigates the robustness issues of a set of distributed optimization algorithms, which aim to approach the optimal solution to a sum of local cost functions over an uncertain network. The uncertain communication network consists of transmission channels perturbed by additive deterministic uncertainties, which can describe quantization and transmission errors. A new robust initialization-free algorithm is proposed for the distributed optimization problem of multiple Euler-Lagrange systems, and the explicit relationship of the feedback gain of the algorithm, the communication topology, the properties of the cost function, and the radius of the channel uncertainties is established in order to reach the optimal solution. This result provides a sufficient condition for the selection of the feedback gain when the uncertainty size is less than the unity. As a special case, we discuss the impact of communication uncertainties on the distributed optimization algorithms for first-order integrator networks.

BACKGROUND: Controversy over the issue that No. 12a lymph node involvement is distant or regional metastasis remains, and the possible inclusion of 12a lymph nodes in D2 lymphadenectomy is unclear. As reported, gastric cancer (GC) located in the lower third is highly related to the metastasis of station 12a lymph nodes. AIM: To investigate whether the clinicopathological factors and metastasis status of other perigastric nodes can predict station 12a lymph node metastasis and evaluate the prognostic significance of station 12a lymph node dissection in patients with lower-third GC. METHODS: A total of 147 patients with lower-third GC who underwent D2 or D2+ lymphadenectomy, including station 12a lymph node dissection, were included in this retrospective study from June 2003 to March 2011. Survival prognoses were compared between patients with or without station 12a lymph node metastasis. Logistic regression analyses were used to clarify the association between station 12a lymph node metastasis and clinicopathological factors or metastasis status of other perigastric nodes. The metastasis status of each regional lymph node was evaluated to identify the possible predictors of station 12a lymph node metastasis. RESULTS: Metastasis to station 12a lymph nodes was observed in 18 patients with lower-third GC, but not in 129 patients. The incidence of station 12a lymph node involvement was reported as 12.2% in patients with lower-third GC. The overall survival of patients without station 12a lymph node metastasis was significantly better than that of patients with station 12a metastasis (P < 0.001), which could also be seen in patients with or without extranodal soft tissue invasion. Station 12a lymph node metastasis and extranodal soft tissue invasion were identified as independent predictors of poor prognosis in patients with lower-third GC. Advanced pN stage was defined as independent risk factor significantly correlated with station 12a lymph node positivity. Station 3 lymph node staus was also proven to be significantly correlated with station 12a lymph node involvement. CONCLUSION: Metastasis of station 12a lymph nodes could be considered an independent prognosis factor for patients with lower-third GC. The dissection of station 12a lymph nodes may not be ignored in D2 or D2+ lymphadenectomy due to difficulties in predicting station 12a lymph node metastasis.

ADAMTS9 belongs to the ADAMTS (a disintegrin and metalloproteinase with thrombospondin motifs) protein family, and its expression is frequently silenced due to promoter hypermethylation in various human cancers. However, the underlying mechanisms remain largely unknown. In this study, we investigated the inhibitory effects of ADAMTS9 on gastric cancer (GC) cells. We initially examined ADAMTS9 protein level in 135 GC and adjacent normal tissue pairs, showing that ADAMTS9 was strikingly decreased in the malignant specimens and patients with low ADAMTS9 expression exhibited more malignant phenotypes and poorer outcome. ADAMTS9 expression was restored in AGS and BGC-823 cells, which then markedly suppressed cellular viability and motility in vitro and in vivo. As ADAMTS9 was enriched in the nuclei of gastric mucosal cells, RNA-sequencing experiment showed that ADAMTS9 significantly altered gene expression profile in BGC-823 cells. Additionally, DNA methyltransferase 3α (DNMT3A) was identified to be responsible for the hypermethylation of ADAMTS9 promoter, and this methyltransferase was ubiquitinated by ring finger protein 180 (RNF180) and then subject to proteasome-mediated degradation. In conclusion, we uncovered RNF180/DNMT3A/ADAMTS9 axis in GC cells and showed how the signaling pathway affected GC cells.

A batch fluidized bed reactor (BFBR) with modified biosynthetic crystals (MBC), derived from Pseudomonas sp. HXF1, was investigated for the treatment of the groundwater containing fluoride (F-). Impacts of different hydraulic retention time (HRT), pH, and initial F- concentration on F- removal were examined and the maximum defluorination efficiency was recorded as 95.20%. Moreover, recycling experiments were performed to evaluate the stability of repeated use. BFBR/MBC system showed a long-term effective treatment outcome with low fluctuation in the concentrations of residual Ca2+ and F-. The formed precipitates were characterized by SEM, XPS, XRD, and FTIR. The defluorination mechanisms of BFBR/MBC system were defined as the chemisorption and induced crystallization of Ca5(PO4)3F on the MBC surface. As a feasible, economical, and environment-friendly technique, the method has a long-term value, which suggests promising applications in F- removal and resourceful treatment.

This study investigated the influence of different carbon to nitrogen (C/N) ratios on the bio-removal efficiency of aquatic pollutants like calcium (Ca2+), fluoride (F-), and nitrate (NO3-N) in a quartz sand-filled biofilm reactor (QSBR) to treat the low C/N wastewater using Acinetobacter sp. H12 at pH 6.50. The simultaneous bio-removal rate of Ca2+, F-, and NO3- reached 56.31%, 96.33, and 96.95 respectively. Nitrogen gas (N2) was produced with no evidence of N2O emission. Moreover, the morphological study of strain H12 and biological precipitates through SEM revealed that strain H12 provides the nucleation sites for microbially induced calcium precipitation to remove Ca2+ and F-. Besides, XPS and XRD peak spectra implicated that Ca2+ and F- were removed as CaF2 and Ca5(PO4)3F co-precipitates. The 16S rRNA sequencing analyses revealed that H12 belongs to Acinetobacter and has stronger MICP and denitrification potential as compared with other strains under low C/N conditions.

In this research, an immobilized biofilm reactor was established for the simultaneous removal of calcium (Ca2+), fluoride (F-), nickel (Ni2+), and nitrate (NO3--N) by microbial induced calcium precipitation (MICP). The operating parameters of the reactor, hydraulic retention time (HRT: 4, 8, and 12 h), influent Ca2+ concentration (36.0, 108.0, and 180.0 mg L-1), and influent Ni2+ concentration (0.0, 3.0, and 6.0 mg L-1) were discussed. Under the HRT of 12 h, influent Ca2+ concentration of 180.0 mg L-1, and influent Ni2+ concentration of 3.0 mg L-1, the removal ratios of Ca2+, F-, Ni2+, and NO3--N reached 45.31%, 79.55%, 85.11%, and 55.29%, respectively, which was the reactor stable operation performance. The SEM revealed the morphology of calcium-precipitated bio-crystals. XPS showed the Ca2+ and Ni2+ precipitate components and XRD further revealed the formation of CaCO3, Ca5(PO4)3OH, and NiCO3 precipitation. Nitrogen (N2) was the main gas produced in the reactor. Fluorescence spectroscopy manifested that extracellular polymers played an important role in the organism nucleation. High-throughput sequencing exhibited that Acinetobacter sp. H12 was the dominant bacterial group. This study provided a new insight for simultaneous remediation of Ca2+, F-, Ni2+, and NO3--N in water bodies.

PURPOSE: This study aimed to investigate the effects of a high body mass index (BMI) on the outcomes of radical gastrectomy for gastric cancer. METHODS: We conducted a retrospective cohort study of 1729 patients with stage I to III gastric cancer who received open radical gastrectomy from February 2003 to August 2011. The patients were divided into 3 groups according to their BMI: a low BMI group (BMI < 18.5 kg/m2), normal BMI group (18.5 ≤ BMI < 25 kg/m2), and high BMI group (BMI ≥ 25 kg/m2). RESULTS: A total of 871 patients were included in the final analysis, of which the median BMI was 22.7 kg/m2 (range 13.6-44.9 kg/m2). A high BMI increased the risk of postoperative intestinal fistula but not the risk of a reduced number of examined lymph nodes or hospital death. Furthermore, a high BMI did not negatively affect the overall survival (OS) of gastric cancer patients. CONCLUSIONS: A high BMI increased the operative morbidity after radical gastrectomy for gastric cancer. However, a high BMI did not negatively affect the quality of lymphadenectomy or the OS of gastric cancer patients in experienced high-volume centers. A careful approach during operation and meticulous perioperative management are required for gastric cancer patients with a high BMI.

Pseudomonas sp. HXF1, a strain capable of heterotrophic nitrification, aerobic denitrification (HNAD), and biomineralization was identified and employed for the simultaneous removal of ammonia nitrogen (NH4+-N) and fluoride (F-). It removed 99.2% of NH4+-N without accumulation of nitrous nitrogen (NO2--N) and nitrate nitrogen (NO3--N), while removed 87.3% of F-. Response surface methodology (RSM) was used to study the best removal conditions for NH4+-N and F-. The results of nitrogen balance experiments with NH4Cl, NaNO2, and NaNO3 as single nitrogen sources and amplification experiments of denitrification genes proved that the bacterial strains may remove NH4+-N through HNAD. The experimental results of Scanning electron microscope (SEM), X-ray photoelectron spectroscopy (XPS), and X-ray diffractometer (XRD) indicated that the way of F- removal may be adsorption and co-precipitation. The results demonstrated that the strain HXF1 has great potential in the biological denitrification and F- removal of groundwater.

RESUMO

This paper considers the formation control problem for general linear networked agents constrained with event-triggered communications. We propose four kinds of edge-based event-triggered protocols, each of which can be used to achieve given formation structures and eliminate the unexpected Zeno behavior. Since the whole protocols are designed according to sampled information at event instants rather than real-time information, these protocols efficiently avoid continuous communications, reduce the bandwidth need of communication, and decrease the energy consuming. The distributed static state feedback edge-based event-triggered protocol or the adaptive one is applicable for the occasions with available states. Different from the state feedback protocols, users can choose the distributed static output feedback edge-based event-triggered protocol or the adaptive one no matter whether agents' states are available or not. It is worth emphasizing that the adaptive state (or output) feedback event-triggered protocol requires no global information of the network topology and can be used in a fully distributed and scalable manner. Finally, numerical examples on formation control are offered to testify the feasibility of the proposed protocols.

The removal properties and mechanisms of fluoride (F-) and nickel (Ni2+) were studied by biomineralizing bacteria (Acinetobacter sp. H12). The results showed that the removal ratio of F-, Ca2+ and Ni2+ reached 75% (0.031 mg·L-1·h-1), 84.96% (2.123 mg·L-1·h-1), and 56.67% (0.024 mg·L-1·h-1) after 72 h, respectively. The removal ratio of nitrate (NO3-) reached 100% (0.686 mg·L-1·h-1) after 24 h. SEM and XRD images indicated that bioprecipitation of CaF2, Ca5(PO4)3F, Ca5(PO4)3(OH), NiCO3, CaCO3 and Ni were formed, and some of these precipitation used bacteria as nucleation sites to form biological crystal seeds. N2 was the primary product in gas chromatography analysis. Meanwhile, both the fluorescence spectroscopy and fourier transform near-infrared spectroscopy analysis proved that strain H12 had good ability to remove fluoride and nickel ions simultaneously.

Biosynthetic crystals (BC) were prepared through microbially induced calcium carbonate precipitation (MICP) for fluoride (F-) removal from the groundwater. Batch experiments were conducted to evaluate the fluoride adsorption capacity and the impacts of critical factors (organic matter, pH, initial fluoride concentration and BC dosage) on defluorination efficiency of BC. The maximum adsorption amount and defluorination efficiency were recorded as 5.10 mg g-1 and 98.24%, respectively. The adsorption kinetics and isotherms studies showed that pseudo-second-order kinetic model and Freundlich isotherm model were best fitting to the reaction. Adsorption thermodynamic parameters indicated a spontaneous, endothermic and thermodynamically favorable adsorption process. Moreover, the mechanism of F- removal by BC was further analyzed by SEM, XPS, XRD and FTIR. The method can cope with the problem of applying the external organic substances in MICP, and avoid the microbial safety risk in the effluent. As an economically and environmentally friendly adsorbent, BC can be used for F- removal from groundwater.
