OBJECTIVE: The study was undertaken to determine the clinical effectiveness of percutaneous kyphoplasty (PKP) with unilateral balloon infusion of low dose of bone cement for treatment of osteoporotic vertebral compression fractures (OVCFs) in the elderly. PATIENTS AND METHODS: A retrospective study was carried out. A total of 36 patients with OVCFs treated by PKP from August 2019 and August 2020 were included. Patients were divided into two groups according to the amount of bone cement infused into the vertebral body. The amount of cement in conventional-dose group was 3.5-6.0 mL and the amount of cement in small-dose group was 1.8-3.0 mL. Pain relief before and after the operation were evaluated, and the leakage of bone cement in the two groups was also observed. RESULTS: Two groups of patients have obtained a good clinical efficacy. Pain has significant differences before and after the operation (p < 0.05). More importantly, compared with conventional-dose group, small-dose group has lower bone cement leakage rate (p < 0.05). CONCLUSIONS: PKP with small-dose bone cement infusion can obtain the same clinical effects of conventional-dose, but the incidence of bone cement leakage is lower and safe.

OBJECTIVE: To elucidate whether microRNA-374b could participate in the development of lung cancer (LC) through downregulating PTEN (gene of phosphate and tensin homolog deleted on chromosome ten) expression via activating PI3K/Akt pathway. PATIENTS AND METHODS: Expression levels of microRNA-374b and PTEN in LC tissues and adjacent normal tissues were detected by quantitative Real Time-Polymerase Chain Reaction (qRT-PCR). Moreover, the expression level of microRNA-374b in LC cell lines was detected as well. The microRNA-374b inhibitor was constructed and transfected to downregulate microRNA-374b expression in A549 and H358 cells. The regulatory effects of microRNA-374b on migratory and proliferative capacities of LC cells were explored by wound healing and cell counting kit-8 (CCK-8) assay, respectively. After co-transfection of microRNA-374b inhibitor and si-PTEN in LC cells, expression levels of PTEN/PI3K/Akt were determined by qRT-PCR and Western blot. RESULTS: QRT-PCR results showed that microRNA-374b expression was higher, while PTEN expression was lower in LC tissues than adjacent tissues. Identically, microRNA-374b was also highly expressed in LC cell lines. PTEN expression was negatively correlated with microRNA-374b expression in LC. The downregulation of microRNA-374b in A549 and H358 cells inhibited their migratory and proliferative potentials. Subsequently, we verified that microRNA-374b could bind to PTEN through dual-luciferase reporter gene assay. MicroRNA-374b could inhibit PTEN expression and activate the PI3K/Akt pathway. Furthermore, PTEN knockdown enhanced migratory and proliferative abilities of LC cells, which were attenuated by co-transfection of microRNA-374b inhibitor. CONCLUSIONS: MicroRNA-374b promotes the development of LC by downregulating PTEN expression through activating PI3K/Akt pathway.

With suspension cultured cells of tobacco (Nicoliana Tobacum L.), effects of inorganic phosphate, nitrogen sources and carbon source on cell growth and CoQ10 content were investigated. It showed that, When treated with 30 g/L of sucrose, the total amount of CoQ10 was highest (8212 g/L); and the cell biomass and CoQ10 content were 19.8 micrograms/L, 414.7 micrograms/g(dwt) respectively. When treated 340 mg/L of KH2PO4, the cell biomass, the content and total amount of CoQ10 were 21.04 g/L, 514.5 micrograms/g(dwt) and 10824 micrograms/L respectively. When treated with the ratio of NH4+/NO3- was 1:2, the cell biomass, the content and total amount of CoQ10 were 21.04 g/L, 514.3 micrograms/g(dwt) and 10,820 micrograms/L respectively. The high ration of NH4+/NO3- was suitable for the formation of CoQ10, but not for the cell growth.
