RESUMO

An all-fiber orbital angular momentum (OAM) generator enabling direct turning of the fundamental mode (${{\rm HE}_{11}}$HE11) to the second OAM modes (${ l} = {\pm 2}$l=±2) with an efficiency of $\sim90\% $∼90% has been proposed and experimentally demonstrated, which is realized based on utilization of a second-order helical fiber grating written in a few-mode fiber. This is the first time, to the best of our knowledge, that an all-fiber second-order OAM has been achieved with using only one component, i.e., the helical long-period fiber grating. The proposed method opens a new way to efficiently generate an all-fiber higher-order OAM using a conventional multimode fiber.

Despite systematic approaches to mapping networks of genetic interactions in Saccharomyces cerevisiae, exploration of genetic interactions on a genome-wide scale has been limited. The S. cerevisiae haploid genome has 110 regions that are longer than 10 kb but harbor only non-essential genes. Here, we attempted to delete these regions by PCR-mediated chromosomal deletion technology (PCD), which enables chromosomal segments to be deleted by a one-step transformation. Thirty-three of the 110 regions could be deleted, but the remaining 77 regions could not. To determine whether the 77 undeletable regions are essential, we successfully converted 67 of them to mini-chromosomes marked with URA3 using PCR-mediated chromosome splitting technology and conducted a mitotic loss assay of the mini-chromosomes. Fifty-six of the 67 regions were found to be essential for cell growth, and 49 of these carried co-lethal gene pair(s) that were not previously been detected by synthetic genetic array analysis. This result implies that regions harboring only non-essential genes contain unidentified synthetic lethal combinations at an unexpectedly high frequency, revealing a novel landscape of genetic interactions in the S. cerevisiae genome. Furthermore, this study indicates that segmental deletion might be exploited for not only revealing genome function but also breeding stress-tolerant strains.

An H3/H4 histone chaperone, Asf1, plays an essential role in maintaining genomic stability in many species, including fission yeast. Here, we showed that overexpression of a CENP-A chaperone Sim3 suppressed the temperature sensitive phenotype of asf1-33 and asf1-30 mutants and the defect in chromatin structure, and prevented the accumulation of DNA damage in asf1-33 mutants at high temperatures. Furthermore, asf1-33 and Δsim3 were synthetic lethal. Consistent with this, shutdown of sim3 expression in asf1-33 Δsim3 double mutants that contained extragenic sim3 resulted in growth retardation. In addition, the Δsim3 mutant displayed sensitivity to thiabendazol and hydroxyurea, which suggests that Sim3 plays a general role in maintaining chromatin structure. Our results suggest a possibility that Sim3 functions as a histone chaperone.

RESUMO

In stereotactic radiosurgery we can irradiate a targeted volume precisely with a narrow high-energy x-ray beam, and thus the motion of a targeted area may cause side effects to normal organs. This paper describes our motion detection system with three USB cameras. To reduce the effect of change in illuminance in a tracking area we used an infrared light and USB cameras that were sensitive to the infrared light. The motion detection of a patient was performed by tracking his/her ears and nose with three USB cameras, where pattern matching between a predefined template image for each view and acquired images was done by an exhaustive search method with a general-purpose computing on a graphics processing unit (GPGPU). The results of the experiments showed that the measurement accuracy of our system was less than 0.7 mm, amounting to less than half of that of our previous system.

RESUMO

A 33-year-old woman suffered from epistaxis and perforation of the nasal septum. Based on a biopsy of nasal mucosa, Wegener's granulomatosis was suspected initially. Her nasal symptoms improved spontaneously, but tinnitus, hearing loss, and dizziness appeared within 3 months. Laboratory analyses revealed no inflammation, and antineutrophil cytoplasmic antibodies were negative. Audiometry revealed bilateral sensorineural hearing loss. A second biopsy of the nasal septum showed an inflammatory change in the cartilage. Thus we diagnosed early-stage relapsing polychondritis.

OBJECTIVE: The purpose of this study was to evaluate the refractory period of the facial nerve by transcranial magnetic paired stimulation (TMPS) with as short interstimulus interval (ISI) as possible. Also, by applying TMPS, the long latency response obtained at the same time was recorded and its neurophysiological characteristics were studied. METHODS: (Experiment 1) The subjects comprised of 30 normal volunteers and 19 patients with Bell's palsy. The experiments were carried out using two sets of Magstim model 200 and Bistim modules, a large coil measuring 90 mm in diameter with a maximal output of 2.0 tesla (T), and a Neuropack 8 (Nihon Kohden Co., Japan) to control the stimulation and record the electromyographic findings. The amplitude of compound muscle action potentials (CMAP) to the orbicularis oris muscle was studied by TMPS at the parieto-occipital region. The ISI was set at 0.9, 1.1, 1.3, 1.5, 1.7, 1.9 and 2.1 ms in order to measure the refractory periods of the nerve. (Experiment 2) The subjects comprised of ten normal volunteers. The same method as in experiment 1 was carried out. However, this time, the lead electrodes were placed on the orbicularis oculi muscle, similar to that of the blink reflex. The ISI was set at 40, 60, 80, 100, 200, 300, 500, 800 and 1000 ms, and the effects of the facilitation and inhibition were studied. RESULTS: (Experiment 1) In normal subjects, when the ISI was less than 1 ms, a significant decrease in the amplitude was noted. In severe palsy cases with House-Brackmann Grade IV-V TMS yielded no response. In two of the cases of House-Brackmann Grade III, the CMAP was obtained. (Experiment 2) The long latency response with TMPS was most strongly inhibited when ISI was 80 ms. CONCLUSION: We were able to investigate the refractory periods, the reflex pathway of the facial nerve and the trigeminal nerve including the pons and the medulla oblongata by TMPS.
