RESUMO

Understanding climate variability and stability under extremely warm 'greenhouse' conditions in the past is essential for future climate predictions. However, information on millennial-scale (and shorter) climate variability during such periods is scarce, owing to a lack of suitable high-resolution, deep-time archives. Here we present a continuous record of decadal- to orbital-scale continental climate variability from annually laminated lacustrine deposits formed during the late Early Cretaceous (123-120 Ma: late Barremian-early Aptian) in southeastern Mongolia. Inter-annual changes in lake algal productivity for a 1091-year interval reveal a pronounced solar influence on decadal- to centennial-scale climatic variations (including the ~ 11-year Schwabe cycle). Decadally-resolved Ca/Ti ratios (proxy for evaporation/precipitation changes) for a ~ 355-kyr long interval further indicate millennial-scale (~ 1000-2000-yr) extreme drought events in inner-continental areas of mid-latitude palaeo-Asia during the Cretaceous. Millennial-scale oscillations in Ca/Ti ratio show distinct amplitude modulation (AM) induced by the precession, obliquity and short eccentricity cycles. Similar millennial-scale AM by Milankovitch cycle band was also previously observed in the abrupt climatic oscillations (known as Dansgaard-Oeschger events) in the 'intermediate glacial' state of the late Pleistocene, and in their potential analogues in the Jurassic 'greenhouse'. Our findings indicate that external solar activity forcing was effective on decadal-centennial timescales, whilst the millennial-scale variations were likely amplified by internal process such as changes in deep-water formation strength, even during the Cretaceous 'greenhouse' period.

Rhythmically bedded cherts are observed in both pelagic marine and lacustrine deposits, but the formation mechanism in the latter remains highly uncertain. Our study of alternating chert-dolomite beds in the Eocene Green River Formation, Utah, USA reveals dense accumulations of organic-matter spheres (30-50 µm diameter) of probable algal cyst origin in the chert layers, and centennial- to millennial-scale periodicities in chert layer deposition. A positive correlation between the degree of degradation of the organic spheres and Si distribution implies decomposition of algal organic matter lead to precipitation of lacustrine chert. As both alkalinity and dissolved silica were likely high in the palaeo-lake waters of the Green River Formation, we hypothesize that decomposition of algal organic matter lowered the pH of sediment pore waters and caused silica precipitation. We propose a formation model in which the initial abundance of algal organic matter in sediment varies with productivity at the lake surface, and the decomposition of this algal matter controls the extent of silica precipitation in sediment. The formation of rhythmically bedded chert-dolomite may be linked to centennial- to millennial-scale climatic/environmental factors that modulate algal productivity, which are possibly tied to solar activity cycles known to have similar periodicities.

Isolated spherical carbonate concretions are frequently observed in finer grained marine sediments of widely varying geological age. Recent studies on various kinds of spherical carbonate (CaCO3) concretions revealed that they formed very rapidly under tightly constrained conditions. However, the formation ages of the isolated spherical carbonate concretions have never been determined. Here we use 87Sr/86Sr ratios to determine the ages of these spherical concretions. The studied concretions formed in the Yatsuo Group of Miocene age in central Japan. Some formed post-mortem around tusk-shells (Fissidentalium spp.), while other concretions have no shell fossils inside. The deformation of sedimentary layers around the concretions, combined with geochemical analyses, reveal that Sr was incorporated into the CaCO3 concretions during their rapid formation. Strontium isotopic stratigraphy using 87Sr/86Sr ratios of all concretions indicates an age of 17.02 ± 0.27 Ma, with higher accuracy than the ages estimated using micro-fossils from the Yatsuo Group. The results imply that the 87Sr/86Sr ratio of isolated spherical carbonate concretions can be applied generally to determine the numerical ages of marine sediments, when concretions formed soon after sedimentation. The 87Sr/86Sr age determinations have high accuracy, even in cases without any fossils evidence.

Isolated spherical carbonate concretions observed in marine sediments are fascinating natural objet trouve because of their rounded shapes and distinct sharp boundaries. They occur in varied matrices and often contain well preserved fossils. The formation process of such concretions has been explained by diffusion and rapid syn-depositional reactions with organic solutes and other pore water constituents. However, the rates, conditions and formation process of syngenetic spherical concretions are still not fully clear. Based on the examination of different kinds of spherical concretions from several locations in Japan, a diffusion based growth diagram was applied to define the generalized growth conditions of spherical concretions formed around decaying organic matter. All analytical data imply that the spherical concretions formed very rapidly, at least three to four orders of magnitude faster than previously estimated timescales. The values indicate that spherical concretions are preferentially grown within clay- to silt-grade marine sediments deposited in relatively deep (a few tens of metres) environments dominated by diffusive solute transport, very early in diagenesis.

RESUMO

An atypical urease-negative mutant of Actinobacillus pleuropneumoniae serovar 2 was isolated in Japan. Nucleotide sequence analysis of the urease gene cluster revealed that the insertion of a short DNA sequence into the cbiM gene was responsible for the urease-negative activity of the mutant. Veterinary diagnostic laboratories should be watchful for the presence of aberrant urease-negative A. pleuropneumoniae isolates.

Carbonate concretions occur in sedimentary rocks of widely varying geological ages throughout the world. Many of these concretions are isolated spheres, centered on fossils. The formation of such concretions has been variously explained by diffusion of inorganic carbon and organic matter in buried marine sediments. However, details of the syn-depositional chemical processes by which the isolated spherical shape developed and the associated carbon sources are little known. Here we present evidence that spherical carbonate concretions (diameters φ : 14 ~ 37 mm) around tusk-shells (Fissidentalium spp.) were formed within weeks or months following death of the organism by the seepage of fatty acid from decaying soft body tissues. Characteristic concentrations of carbonate around the mouth of a tusk-shell reveal very rapid formation during the decay of organic matter from the tusk-shell. Available observations and geochemical evidence have enabled us to construct a 'Diffusion-growth rate cross-plot' that can be used to estimate the growth rate of all kinds of isolated spherical carbonate concretions identified in marine formations. Results shown here suggest that isolated spherical concretions that are not associated with fossils might also be formed from carbon sourced in the decaying soft body tissues of non-skeletal organisms with otherwise low preservation potential.

A total of 159 Thai isolates of Mycoplasma hyopneumoniae isolated from pneumonic lungs of pigs during 2006-2011 were investigated for their in vitro susceptibility to 12 antimicrobial agents. Low activity of chlortetracycline was indicated by the MIC range from 3.12-100 µg/ml and MIC90 of 50 µg/ml. Seventy-six isolates showed resistance to enrofloxacin, whereas 2 isolates showed resistance to macrolides and lincomycin. In addition, a point mutation at A2058G was revealed by sequence analysis of 23S ribosomal RNA in both isolates. The present results confirmed the rapid increase of resistant M. hyopneumoniae isolates against chlortetracycline, enrofloxacin, macrolides and lincomycin in Thailand. Selection of drugs to control swine diseases in Thailand must be done more prudently in consideration of reducing the antimicrobial resistance.

Tritium concentrations in Japanese precipitation samples collected after the March 2011 accident at the Fukushima Dai-ichi Nuclear Power Plant (FNPP1) were measured. Values exceeding the pre-accident background were detected at three out of seven localities (Tsukuba, Kashiwa and Hongo) southwest of the FNPP1 at distances varying between 170 and 220 km from the source. The highest tritium content was found in the first rainfall in Tsukuba after the accident; however concentrations were 500 times less than the regulatory limit for tritium in drinking water. Tritium concentrations decreased steadily and rapidly with time, becoming indistinguishable from the pre-accident values within five weeks. The atmospheric tritium activities in the vicinity of the FNPP1 during the earliest stage of the accident was estimated to be 1.5×10(3) Bq/m(3), which is potentially capable of producing rainwater exceeding the regulatory limit, but only in the immediate vicinity of the source.

A total of 151 strains of Mycoplasma hyorhinis isolated from porcine lung lesions (weaned pigs, n=71, and finishers, n=80) were investigated for their in vitro susceptibility to 10 antimicrobial agents. Thirty-one strains (28 from weaned pigs and 3 from finishers) showed resistance to 16-membered macrolide antibiotics and lincomycin. The prevalence of the 16-membered macrolide-resistant M. hyorhinis strain in weaned pigs from Japanese herds has approximately quadrupled in the past 10 years. Several of the 31 strains were examined for mutations in the 23S ribosomal RNA (rRNA). All field strains tested showed a transition of A to G at position 2059 of 23S rRNA-rendered Escherichia coli. On the other hand, individual tylosin- and lincomycin-resistant mutants of M. hyorhinis were selected in vitro from the susceptible type strain BTS7 by 3 to 9 serial passages in subinhibitory concentrations of each antibiotic. The 23S rRNA sequences of both tylosin and lincomycin-resistant mutants were compared with that of the radical BTS7 strain. The BTS7 mutant strain selected by tylosin showed the same transition as the field-isolated strains of A2059G. However, the transition selected in lincomycin showed mutations in domains II and V of 23S rRNA, G2597U, C2611U in domain V, and the addition of an adenine at the pentameric adenine loop in domain II. The strain selected by lincomycin showed an additional point mutation of A2062G selected by tylosin.

The growth of Salmonella Choleraesuis was examined in Rappaport Vassiliadis broth (RV) and Hajna-tetrathionate broth (HTT) at 37 and 42 degrees C. As the enrichment in RV at 37 degrees C was satisfactory for isolating S. Choleraesuis, we used this enrichment for isolation from the samples collected from 15 asymptomatic pigs reared on a S. Choleraesuis contaminated farm. S. Choleraesuis was frequently isolated from six pigs (40.0%) under field conditions. The isolation of other Salmonella serovars than S. Choleraesuis was attempted by using both RV enrichment at 37 degrees C and HTT enrichment at 42 degrees C. Salmonella organisms were isolated from 156 (44.8%) of 348 fecal samples and more frequently with HTT at 42 degrees C (43.4%) than with RV at 37 degrees C (20.9%). If other serovars in addition to S. Choleraesuis are to be surveyed, HTT enrichment should be used in combination with RV enrichment.

In 1996 and 1997, the seroprevalence against Leptospira in parturient sows with premature birth or stillbirth from two herds was investigated. In three out of four sow serum samples obtained in Gunma Prefecture, the antibody titers to Leptospira interrogans serovar copenhageni (M20) were higher than 10,000 (the reciprocal of the serum dilution). Furthermore, the antibody titers to L. interrogans serovar canicola (Hond Utrecht IV) were significantly high in the three sows and the titers ranged from 1,000 to 3,000. In sows obtained in Chiba Prefecture, significantly high antibody titers to serovar copenhageni (M20) were confirmed in eight out of 40 sows, and antibody titers greater than 10,000 in six of them. Significantly high antibody titers to L. interrogans serovar icterohaemorrhagiae (RGA) and L. canicola (Hond Utrecht IV) were confirmed in four and 18 out of the 40 sows, respectively, compared with the titers to the other serovars. These findings may indicate the prevalence of leptospirosis in pig herds in both Gunma and Chiba Prefectures.
