RESUMO

To address the shortcomings of traditional reliability theory in characterizing the stability of deep underground structures, the advanced first order second moment of reliability was improved to obtain fuzzy random reliability, which is more consistent with the working conditions. The traditional sensitivity analysis model was optimized using fuzzy random optimization, and an analytical calculation model of the mean and standard deviation of the fuzzy random reliability sensitivity was established. A big data hidden Markov model and expectation-maximization algorithm were used to improve the digital characteristics of fuzzy random variables. The fuzzy random sensitivity optimization model was used to confirm the effect of concrete compressive strength, thick-diameter ratio, reinforcement ratio, uncertainty coefficient of calculation model, and soil depth on the overall structural reliability of a reinforced concrete double-layer wellbore in deep alluvial soil. Through numerical calculations, these characteristics were observed to be the main influencing factors. Furthermore, while the soil depth was negatively correlated, the other influencing factors were all positively correlated with the overall reliability. This study provides an effective reference for the safe construction of deep underground structures in the future.

The surface adsorption of eight binary molten salts, AgNO3-M1NO3(M1 = Li, Na, K, Rb), NaNO3-M2NO3 (M2 = K, Rb), Ca(NO3)2-CsNO3, and Cd(NO3)2-NaNO3, has been investigated. It is found that the surface tension and temperature of molten salts at constant pressure and mole fraction satisfy the same equation as that for pure liquid compounds reported in our previous works. The heats of phase transition from the bulk to the surface phase for eight molten salts are determined. The heats per unit area are all at the order of -10-2 J/m2. The phase transition is exothermic because the entropy in the surface phase is smaller than the entropy in the bulk phase. The ratio of the solute surface concentration to the solute bulk concentration is approximated as the first-order polynomials of the solute bulk concentration. Then, curves of the surface tension vs the solute bulk concentration are well fitted. The ratio (ΔcBs/ΔcBα) is used to interpret the changing trend of the surface tension with bulk concentrations of solute. It is also found that the surface tension of molten salts decreases linearly with the solute surface concentration.

Mastering the creep characteristics of artificial frozen soil and scientifically evaluating the creep model is an important guarantee for the safety of subway tunnel freezing construction. Base on the construction of Nantong metro tunnel, the uniaxial compressive strength tests of the artificially frozen soft soil were carried out to obtain the influence law of temperature on the uniaxial compressive strength, and the uniaxial creep tests were carried out to obtain the influence law of temperature and stress grade on creep, at - 5, - 10 and - 15 °C. The experimental results show that the creep characteristics of frozen soft soil specimens have obvious fuzzy randomness. The traditional ant colony algorithm is improved by optimizing the pheromone fuzzification coefficient, which improves the search efficiency and avoids the local optimum effectively. Subsequently, the improved fuzzy ant colony algorithm is used to invert the flexibility parameters of commonly used permafrost creep models. The fuzzy weight of evaluation index and the fuzzy random evaluation matrix were determined to evaluate the optimal creep model under three different stress levels of frozen soft soil. Finally, the reliability of the fuzzy random evaluation method was verified by engineering measured data.

Cavity solitons are shape-preserving waveforms infinitely revolving around a cavity, which have numerous applications from spectroscopy to telecommunications. Although the cavity solitons have been widely studied for their special time-frequency characteristics over the past decade, the spectral flatness will be a large limitation in some applications without any extra filtering process. In this paper, we report on the generation of a distinct cavity soliton in a cyclic polarization permutation fiber resonator. With the simultaneous excitation of two orthogonal polarization modes with equally opposite dispersion, vectorial cavity solitons possessing broader and flatter spectra can be generated. In order to verify the concept, a numerical model of the polarization-maintaining fiber is proposed and the soliton with a flattened spectrum can be formed. The results enrich the soliton dynamics in the vectorial dissipation system.

The creep characteristics of frozen rock and soil are crucial for construction safety in cases of underground freezing. Uniaxial compression tests and uniaxial creep tests were performed at temperatures of - 10, - 15, - 20, and - 25 °C for silty clay used in Nantong metro freezing construction to investigate the effect law of the stress-strain curves and creep curves. However, owing to the complex effects of factors such as temperature and ground pressure, the mechanical properties of underground frozen silty clay are uncertain. The Burgers creep damage model was established by using an elastic damage element to simulate the accelerated creep stage. The traditional particle swarm optimization algorithm was improved using the inertia weight and the fuzzy random coefficient. The creep parameters of the Burgers damage model were optimized using the improved fuzzy random particle swarm algorithm at different temperatures and pressure levels. Engineering examples indicated that the optimized creep model can more effectively characterize the creep stages of frozen silty clay in Nantong metro freezing construction. The improved fuzzy random particle swarm algorithm has wider engineering applicability and faster convergence than the traditional algorithm.

To evaluate the importance of conserved Arg114 and Arg337 residues of Escherichia coli gamma-glutamyltranspeptidase (EcGGT), Lys, Leu, or Asp-substituted mutants were constructed by site-directed mutagenesis. The wild-type and mutant enzymes were overexpressed in the recombinant E. coli M15 and purified by nickel-chelate chromatography to near homogeneity. With the exception of R114K, all the other mutants significantly lost GGT activity, confirming the importance of these two residues in EcGGT. Kinetic analysis of R114L, R114D, R337K, and R337L revealed a significant increase in K(m) with a minor change in k(cat), leading to more than an 8-fold decrease in k(cat)/K(m) values. Mutations of Arg337 impaired the capability of autocatalytic processing of the enzyme. In vitro maturation experiments revealed that EcGGT precursor mutants, pro-R337K and pro-R337L, could precede a time-dependent autocatalytic process to generate the small and large subunits, while no autocatalytic processing was observed in pro-R337D. Computer modeling showed that the critical bonding distance of Gln390 O-Thr391 HG1 and Gln390 C-Thr391 OG1 are significantly increased in Arg337 replacements, implying that these distance changes might be responsible for the lack of enzyme maturation.

A truncated Escherichia coli Novablue gamma-glutamyltranspeptidase (EcGGT) gene lacking the first 48-bp coding sequence for part of the signal sequence was amplified by polymerase chain reaction and cloned into expression vector pQE-30 to generate pQE-EcGGT. The maximum production of His(6)-tagged enzyme by E. coli M15 (pQE-EcGGT) was achieved with 0.1 mM IPTG induction for 12 h at 20 degrees C. The overexpressed enzyme was purified to homogeneity by nickel-chelate chromatography to a specific transpeptidase activity of 4.25 U/mg protein and a final yield of 83%. The molecular masses of the subunits of the purified enzyme were estimated to be 41 and 21 kDa respectively by SDS-PAGE, indicating EcGGT still undergoes the post-translational cleavage even in the truncation of signal sequence. The optimum temperature and pH for the recombinant enzyme were 40 degrees C and 9, respectively. The apparent K (m) and V (max) values for gamma-glutamyl-p-nitroanilide as gamma-glutamyl donor in the transpeptidation reaction were 37.9 microM and 53.7 x 10(-3) mM min(-1), respectively. The synthesis of L -theanine was performed in a reaction mixture containing 10 mM L -Gln, 40 mM ethylamine, and 1.04 U His(6)-tagged EcGGT/ml, pH 10, and a conversion rate of 45% was obtained.
