RESUMO

During a survey of species diversity of Penicillium and Talaromyces in sugarcane (Saccharum officinarum) rhizosphere in the Khuzestan province of Iran [1], 195 strains were examined, from which 187 belonged to Penicillium (11 species) and eight to Talaromyces (one species). In the present study, three strains of Penicillium belonging to section Exilicaulis series Restricta, identified as P. restrictum by Ansari et al. [1], were subjected to a phylogenetic study. The multilocus phylogeny of partial ß-tubulin, calmodulin and RNA polymerase II second largest subunit genes enabled the recognition of one new phylogenetic species that is here formally described as Penicillium rhizophilum sp. nov. This species is phylogenetically distinct in series Restricta, but it does not show significant morphological differences from other species previously classified in the series. Therefore, we here placed bias on the phylogenetic species concept. The holotype of Penicillium rhizophilum sp. nov. is IRAN 18169F and the ex-type culture is LA30T (=IRAN 4042CT=CBS 149737T).

The genus Cercospora contains many devastating plant pathogens linked to leaf spot diseases afflicting various plants. Identification of Cercospora species based on morphology or host plant association has proven unreliable due to simple morphology and wide host range in many cases; hence, multi-gene DNA sequence data are essential for accurate species identification. Considering the complexity and cost involved in application of multi-locus DNA phylogenetic approaches for species delineation in Cercospora; rapid and cost-effective methods are urgently needed for species recognition. In this study, we applied rep-PCR (repetitive-sequence based polymerase chain reaction) fingerprinting methods referred to as BOX-PCR to differentiate species of Cercospora. Cluster analysis of the banding patterns of 52 Cercospora strains indicated the ability of BOX-PCR technique using BOXA1R primer to generate species-specific DNA fingerprints from all the tested strains. Since this technique was able to discriminate between all the 20 examined Cercospora species during this study, which corresponded well to the species identified based on multi-gene DNA sequence data, our findings revealed the efficiency of BOX-PCR system as a suitable complementary method for molecular identification of the genus Cercospora at species level.

Sugarcane is an important crop in Southern Iran for agri-food, energy, and pharmaceutical industries. Among the pathogens that colonize sugarcane, mycotoxigenic Fusarium species are reason of serious concern for both their pathogenicity on plants and ability to produce harmful mycotoxins to humans and animals. We studied 104 Fusarium strains, selected within a wider Fusarium set isolated from sugarcane in Southern Iran, for molecular identification, phylogeny and mycotoxin analyses. Most of Fusarium strains belonged to Fusarium fujikuroi Species Complex (FFSC) and identified mainly as F. proliferatum, at minor extent as F. sacchari, and rarely as F. thapsinum, and F. verticillioides. Moreover, 14 strains identified as FFSC could not be assigned to any known species, although they were phylogenetically closely related to F. andiyazi, likely representing a new phylogenetic species. A subset of FFSC strains were analyzed for in vitro production of fumonisins (FBs), beauvericin (BEA), and enniatins (ENNs). Fusarium proliferatum strains produced FBs at high amount, and, at a lesser extent, BEA, and ENNs; F.sacchari produced only BEA and B ENNs at very low level; Fusarium sp. strains produced only B ENNs. The paper provides new insights on the genetic diversity of Fusarium species and their mycotoxin profile occurring on sugarcane in Iran.

Knowledge of the genetic diversity detected among fungal species belonging to the genus Aspergillus is of key importance for explaining their important ecological role in the environment and agriculture. The current study aimed to identify Aspergillus species occurring in the rhizosphere of sugarcane in the South of Iran, and to investigate their mycotoxin profiles. One-hundred and twenty-five Aspergillus strains were isolated from the soil of eight major sugarcane-producing sites, and were molecularly identified using sequences of partial -tubulin (benA) and partial calmodulin (CaM) genes. Our molecular and phylogenetic results showed that around 70% of strains belonged to the Aspergillus section Nigri, and around 25% of species belonged to the Aspergillus section Terrei. Species belonging to both sections are able to produce different mycotoxins. The production of mycotoxins was measured for each species, according to their known mycotoxin profile: patulin (PAT) and sterigmatocystin (STG) for Aspergillusterreus; ochratoxin A (OTA) and fumonisins for Aspergilluswelwitschiae; and OTA alone for Aspergillustubingensis. The data showed that the production of OTA was detected in only 4 out of 10 strains of A.welwitschiae, while none of the A.tubingensis strains analyzed produced the mycotoxin. Fumonisins were produced by 8 out of 10 strains of A.welwitschiae. Finally, none of the 23 strains of A.terreus produced STG, while 13 of them produced PAT. The occurrence of such mycotoxigenic plant pathogens among the fungal community occurring in soil of sugarcane fields may represent a significant source of inoculum for the possible colonization of sugarcane plants, since the early stages of plant growth, due to the mycotoxin production capability, could have worrisome implications in terms of both the safety and loss of products at harvest.

Species of Septoria are commonly associated with leaf spot diseases of a broad range of plant hosts worldwide. During our investigation of fungi associated with leaf spot diseases in northern and northwestern Iran, several Septoria isolates were recovered from symptomatic leaves on different herbaceous and woody plants in the Asteraceae, Betulaceae, and Salicaceae families. These isolates were studied by applying a polyphasic approach including morphological and cultural data and a multigene phylogeny using a combined data set of partial sequences of the 28S nuc rRNA gene (large subunit [28S]), internal transcribed spacer regions and intervening 5.8S nuc rRNA gene (ITS) of the nuc rDNA operon, actin (actA), translation elongation factor 1-α (tef1), calmodulin (cmdA), ß-tubulin (tub2), and DNA-directed RNA polymerase II second largest subunit (rpb2). Four novel species are proposed, namely, Septoria eclipticola on Eclipta prostrata, Septoria firouraghina on Cirsium arvense, Septoria guilanensis on Populus deltoides, and Septoria taleshana on Alnus subcordata. All species are illustrated, and their morphology and phylogenetic relationships with other Septoria species are discussed.

Plasma treatment is recognized as a suitable technology to improve germination efficiency of numerous seeds. The objective of this paper is to demonstrate whether cold air plasma can change the quality and quantity of wheat yield. Effects of cold plasma treatment on wheat (Pishgam variety) yield were studied by a randomized complete block design experiment at the Faculty of Agriculture of Tarbiat Modarres University, Iran, during 2015-17. Seeds were pre-treated with 80 W of cold plasma at four levels of time, 60, 120, 180 and 240 seconds. Plasma effects on yield and quality of wheat were determined by measuring plant photosynthesis, grain yield, biological yield, 1000-grain weight, total chlorophyll, carotenoid, anthocyanin, protein and starch content. Results showed that plasma treatments had positive effects on wheat characteristics, and treatment of 180 s had the highest stimulatory effect. In both years, cold plasma increased grain yield at 180 s, but decreased it at 240 s compared with control. The rate of plant photosynthesis, grain yield, 1000-grain weight, carotenoid and anthocyanin were enhanced at 180 s. The starch content and grain protein were enhanced at 120 s cold plasma application compared with control.

The ending of dual nomenclatural systems for pleomorphic fungi in 2011 requires the reconciliation of competing names, ideally linked through culture based or molecular methods. The phylogenetic systematics of Hypocreales and its many genera have received extensive study in the last two decades, however resolution of competing names in Cordycipitaceae has not yet been addressed. Here we present a molecular phylogenetic investigation of Cordycipitaceae that enables identification of competing names in this family, and provides the basis upon which these names can be maintained or suppressed. The taxonomy presented here seeks to harmonize competing names by principles of priority, recognition of monophyletic groups, and the practical usage of affected taxa. In total, we propose maintaining nine generic names, Akanthomyces, Ascopolyporus, Beauveria, Cordyceps, Engyodontium, Gibellula, Hyperdermium, Parengyodontium, and Simplicillium and the rejection of eight generic names, Evlachovaea, Granulomanus, Isaria, Lecanicillium, Microhilum, Phytocordyceps, Synsterigmatocystis, and Torrubiella. Two new generic names, Hevansia and Blackwellomyces, and a new species, Beauveria blattidicola, are described. New combinations are also proposed in the genera Akanthomyces, Beauveria, Blackwellomyces, and Hevansia.

Ophiocordycipitaceae is a diverse family comprising ecologically, economically, medicinally, and culturally important fungi. The family was recognized due to the polyphyly of the genus Cordyceps and the broad diversity of the mostly arthropod-pathogenic lineages of Hypocreales. The other two cordyceps-like families, Cordycipitaceae and Clavicipitaceae, will be revised taxonomically elsewhere. Historically, many species were placed in Cordyceps, but other genera have been described in this family as well, including several based on anamorphic features. Currently there are 24 generic names in use across both asexual and sexual life stages for species of Ophiocordycipitaceae. To reflect changes in Art. 59 in the International Code of Nomenclature for algae, fungi, and plants (ICN), we propose to protect and to suppress names within Ophiocordycipitaceae, and to present taxonomic revisions in the genus Tolypocladium, based on rigorous and extensively sampled molecular phylogenetic analyses. When approaching this task, we considered the principles of priority, monophyly, minimizing taxonomic revisions, and the practical utility of these fungi within the wider biological research community.

Species of Botryosphaeriaceae are important pathogens and endophytes associated with woody plants. Botryosphaeria and Neofusicoccum are two well known genera of the family. In this study 125 isolates morphologically resembling members of this family were collected from about 20 different fruit and forest trees in Iran. Based on morphology, MSP-PCR profile and DNA sequence data (ITS and tef1-a), four species were identified. Of these, Botryosphaeria dothidea, Neofusicoccum mediterraneum and N. parvum are known while Botryosphaeria scharifii is described here as new. N. mediterraneum is a new record for Iran and is reported here for the first time on mango trees. High diversity within Iranian population of N. parvum suggests the need to revise and reassess the morphological species description of N. parvum and closely related species.

Twenty-one species of Chaetomium known from Iran were compared on the basis of morphological and molecular characters. Six new species are recognized, five isolated from cereals and one from nematode cysts. A combined sequence dataset of the ITS region, partial LSU rDNA, and ß-tubulin gene sufficiently resolved five species groups of Chaetomium that are largely concordant with combined features of peridium structure, ascospore shape and germ pore position. Among the new species C. undulatulum is a close relative of C. globosum, C. rectangulare is close to C. elatum, C. interruptum and C. grande are close to C. megalocarpum, altogether forming the C. globosum species group. Chaetomium iranianum and C. truncatulum are members of the C. carinthiacum species group, characterized by spirally coiled ascomatal hairs and fusiform ascospores. A chrysosporium-like anamorph is newly described for C. acropullum.

Morphological and molecular analyses demonstrate that Coniolariella gamsii and Coniolaria murandii are distinct species. The latter species is validated here as Coniolariella macrothecia. A key to the five species of the genus is provided.

Among fungi, species of the genus Pochonia Batista & O.M. Fonseca are considered as promising biological control agents with high potential to reduce root-knot nematode (RKN) and nematode populations. In this research we investigated Fars province of Iran for the presence of Pochonia spp., compared pathogenicity of different Pochonia species on eggs of RKN in vitro, and selected the best isolates for further studies. During 2004-2006, 128 soil samples of fields infested with cyst nematodes and 18 soil samples infested with RKN were collected from Fars province of Iran. In vitro pathogenicity tests were carried out on 36 isolates of Pochonia spp. obtained from CBS and IRAN culture collections. The seven best isolates of this experiment were selected for greenhouse test and their ability in controlling RKN was examined in natural soil. In greenhouse test fresh weight of plant's tops and roots, gall index, nematode multiplication, second-stage juveniles' population in soil, reproduction rate (P(f)/P(i)), proportion of infected eggs, control efficacy, root colonization and soil colony forming units were determined. In vitro pathogenicity of Pochonia on RKN eggs varied between 39% and 95% eggs infected. In greenhouse experiment, three isolates are promising for control of RKN and selected isolates are subjected to more extensive testing to determine their effectiveness in a range of conditions before being developed as commercial biological control agents.

Verticillium fungicola, the type species of Verticillium sect. Albo-erecta, and related taxa were studied using morphological and molecular techniques. Sequences of the ITS region and SSU rDNA suggest that V. fungicola is very close to members of the genus Lecanicillium and unrelated to other species that were originally accommodated in the same section. New combinations in Lecanicillium are proposed for Verticillium fungicola and its var. flavidum at species rank, L. f. var. aleophilum is retained as a variety. These taxa can be distinguished from each other by optimum and maximum temperatures for growth, in addition to ITS sequence differences. Morphologically, L. flavidum is also distinct by repeated branching of the conidiophores, whereas the two varieties of L. fungicola have a simple conidiophore axis.

Virulence of 11 native strains of entomopathogenic fungi; Metarhizium anisopliae (three strains), Beauveria bassiana (six strains) and Lecanicillium psalliotae (two strains) collected from different parts of Iran, were studied against different developmental stages of Rhipicephalus (Boophilus) annulatus. After the exposure of ticks to the fungal strains in different concentrations (i.e. 10(3), 10(5), 10(7) conidia/ml), various parameters such as mortality rate and reproductive efficiency of engorged females, mortality of unfed tick larvae and eclosion percentage of infected eggs were evaluated to determine the fungal virulence. Based on the obtained results, five strains including M. anisopliae (IRAN 437 C and DEMI 001), B. bassiana (IRAN 403 C) and L. psalliotae (IRAN 468 C and IRAN 518 C) were found to be virulent to various stages of tick developmental cycle. Mortality rate of engorged females was found to be dose-dependent with regard to the conidial concentration used. Total mortality rates of 90-100%, 70% and 56.6% were observed for M. anisopliae (IRAN 437 C and DEMI 001), B. bassiana (IRAN 403 C) and L. psalliotae (IRAN 468 C), 6-11 days post inoculation (PI) with 10(7) conidia/ml, respectively. Most strains were able to inhibit egg laying by females in the range of 0-26% in different conidial concentrations. The results indicated that the mean egg laying of treated engorged tick females exposed to M. anisopliae (IRAN 437 C) was less than the mean values of those treated with other fungal strains. Results revealed 89.1%, 35.5% and 56.3% decrease in egg hatchability and 88.69%, 78.15% and 59.74% reduction in reproductive efficiency of the ticks using 10(7) conidia/ml of M. anisopliae (IRAN 437 C), B. bassiana (IRAN 403 C) and L. psalliotae (IRAN 468 C), respectively. In general, the entomopathogenic effects of native M. anisopliae and B. bassiana against various developmental stages of R. (B.) annulatus were confirmed in the present work. Likewise, although L. psalliotae, which was introduced for the first time as an entomopathogenic fungus against tick had not more than 13.3% mortality effect against adult females, but its effect on egg hatchability and reproductive efficiency was remarkable.

The monotype species of the genus Verticillium, Verticillium tenerum, is a synonym of the older name Sporotrichum luteo-album. Its purported teleomorph connection with 'Nectria' inventa is refuted and the preserved specimens of that species are considered as probably identical with Stephanonectria keithii (Bionectriaceae). V. luteo-album takes a unique position in the Glomerella clade of ascomycetes, as sister of the Verticillium-Plectosphaerella clade, which comprises plant-pathogenic species. V. luteo-album is not closely related to V. dahliae and its relatives, which are also situated in this clade. Conservation of the name Verticillium with V. dahliae as conserved type will be necessary to retain this generic name for the plant-pathogenic Verticillium species. In anticipation of this conservation, the new combination Acrostalagmus luteo-albus (syn. Sporotrichum luteo-album) is made.

Trichoderma brevicompactum, a new species, was isolated from soil or tree bark in North, Central and South America, including the Caribbean Islands, and southwestern and southeastern Asia. Morphological and physiological characters, the internal transcribed spacer regions of the rDNA cluster (ITS1-5.8SrDNA-ITS2) and partial sequences of translation elongation factor 1-alpha (tef1) are described. Trichoderma brevicompactum is characterized by a pachybasium-type morphology, morphologically resembling other small-spored species referable to Trichoderma section Pachybasium but with essentially subglobose conidia. It is most closely related phylogenetically to Hypocrea lutea, from which it differs in morphological and physiological characters.
