RESUMO

Although lactation mastitis (LM) has been extensively researched, the incidence rate of LM remains a salient clinical problem. To reduce this incidence rate and achieve a better prognosis, early and specific quantitative indicators are particularly important. It has been found that milk electrolyte concentrations (chloride, potassium, and sodium) and electrical conductivity (EC) significantly change in the early stages of LM in an animal model. Several studies have evaluated EC for the detection of subclinical mastitis in cows. EC, chloride, and sodium content of milk were more accurate for predicting infection status than were other variables. In the early stages of LM, lactic sodium, chloride, and EC increase, but potassium decreases. However, these indicators have not been reported in the diagnosis of LM in humans. This review summarizes the pathogenesis and the mechanism of LM in terms of milk electrolyte concentration and EC, and aim to provide new ideas for the detection of sub-clinical mastitis in humans.

RESUMO

Although gastroesophageal reflux disease (GERD), a common chronic disease in clinical practice, has been widely studied, its potential adverse impact on patients is still a significant clinical concern. It is necessary to understand the pathogenesis of the disease and choose appropriate treatment according to its mechanism. The pathogenesis of GERD is diverse and complex. As the traditional treatment methods are expensive and ineffective in alleviating symptoms in some patients, new treatment options need to be explored. Our previous study suggested that the activation of nuclear factor-kappa beta (NF-κB) in esophageal mucosa may be related to the injury of epithelial barrier function caused by reflux. Based on the literature and our previous study results, it is speculated that inhibition of NF-κB activation may block the insult of GERD on the esophageal mucosal barrier. NF-κB may play an important role in the development of GERD. This article reviews the pathogenesis of GERD and the relationship between NF-κB and GERD, in order to provide new strategies for the treatment of GERD.

RESUMO

Poly-(butylene adipate-co-terephthalate) (PBAT) has become one of the most prevalent biodegradable plastic film materials owing to its good degradability, mechanical properties, and processability. However, the degradation time of this material was too fast and the functional period was short, which limited its application. Herein, three new tropolone-based UV absorbers (UVA-1C, UVA-4C and UVA-6C) were rationally designed and blended into PBAT. The PBAT/UVA films that formed were used against UV aging and prolonged the functional period of PBAT film. The three new absorbers were synthesized by bridging two tropolones using three different organic chains with different flexibility. Among them, the UVA-6C showed the strongest UV absorbance at around 238 nm and 320 nm. Consequently, the PBAT/UVA-6C film showed an extended validity period of 240 h in the Xenon lamp aging machine and a prolonged functional period of 8 d during the field application test when compared to pure PBAT. More importantly, a 7.8% increase in the maize yield was obtained under PBAT/UVA-6C film relative to pure PBAT film. Obviously, the novel prepared UVA-6C compound is a good candidate for UV absorption in PBAT, which makes PBAT/UVA-6C film more advantageous over pure PBAT in practical applications as biodegradable agricultural film.

RESUMO

The mixed hexa-transition metal (hexa-TM) sandwiched arsenotungstate derivative, [CuI3(pz)2(phen)3]2[CuI(phen)2][{Na(H2O)2}{(VIV5CuIIO6)(AsIIIW9O33)2}]·6H2O (1) (pz = pyrazine; phen = 1,10-phenanthroline), has been hydrothermally synthesized and structurally characterized. In compound 1, two {AsW9O33} clusters are connected by mixed hexa-TM ring unit {VVI5CuIIO6} to form a sandwich-type dimer, which are further bonded in "ABAB" mode by the {Na(H2O)2} linker resulting in pure inorganic chains. The unique "L-shaped" trinuclear complex {Cu3(phen)3(pz)2} is supported together via staggered π-π interactions to generate extending waveform two-dimensional supramolecular layers, which are further aggregated with their adjacent analogues by complexes {Cu(phen)2} via H-bonding interaction to yield an unprecedented three-dimensional (3D) metal-organic networks with one-dimensional (1D) cavities. The pure inorganic 1D sandwich chains are implanted in the cavities as guest units via supramolecular interactions to form a POMOF 3D framework. Compound 1, as the electrode of the supercapacitor, exhibits higher specific capacitances (825 F g-1 at a current density of 2.4 A g-1), better rate capability, more durable cyclic stability (91.4% of cycle efficiency after 3000 cycles), and improved conductivity and electroactivity compared to those of parent polyoxometalate (POM) Na9[AsW9O33]·19H2O (2) and 6-Cu-substituted POM [Cu6(imi)6{AsIIIW9O30Cl3}2]·6H2O (3), which may be attributed to the introduction of V4+, the unique host-guest structure, and the rich π electron system. In addition, compound 1 exhibits dual-function electrocatalytic behavior in reducing inorganic salt IO3- and oxidizing the organic molecule dopamine.

RESUMO

PURPOSE: Postoperative hemorrhage and hematoma formation is a potentially lethal complication in thyroid surgery, although the patterns and treatment of hemorrhage after total endoscopic thyroidectomy (TET) via breast approach has not been reported previously. We aim to share our experience about postoperative bleeding. METHODS: A retrospective analysis of 1932 patients who underwent TET from April 2008 to May 2018 in our institution was carried out. The patterns of postoperative hemorrhage and hematoma formation that need surgical treatment were summarized and focused on the relation to the source of bleeding and the time interval between first surgery and hemorrhage. Related risk factors were analyzed by univariate or multivariate analysis processes. RESULTS: The overall rate of hemorrhage and hematoma occurrence was only 0.724% (14 in 1932 patients). Of them, 12 occurred in the first 24 h after surgery, and the other two occurred after withdrawal of the drainage tube. The principle independent risk factors for postoperative hemorrhage and hematoma were age (older than 35 years old) and lateral compartment dissection (LCD) revealed by multivariate regression. During re-exploration, obvious bleeding points were detected in 13 patients. Among them, 12 bled from the vessels in the main trocar cavity and another 1 bled from a broken vein located between the two heads of the sternocleidomastoid (SCM) muscle with LCD. CONCLUSIONS: Hemorrhage after TET usually occurs within 24 h, and the main video trocar cavity was the area most likely to bleed. Age and LCD may increase the bleeding risk. Appropriate dissection level is the main solution to prevent postoperative hemorrhage.

RESUMO

Recent studies have indicated that long non-coding RNAs play crucial roles in numerous cancers, including thyroid cancer, while their function in the mechanism of thyroid cancer 131I resistance has not been elucidated to date. The present study identified a functional long non-coding RNA, SLC6A9-5:2, which was involved in the radioactive therapy resistance of thyroid cancer. We demonstrated that SLC6A9-5:2 was remarkably downregulated in 131I-resistant thyroid cancer cell lines and 131I-insensitive patients and was positively correlated with Poly (ADP-ribose) polymerase 1 (PARP-1) expression and its activation. After downregulating SLC6A9 or blocking PARP-1 artificially, the sensitive thyroid cancer cells mostly displayed a tolerant phenotype under 131I exposure. Furthermore, SLC6A9-5:2 overexpression was positively correlated with PARP-1 mRNA and protein levels, which restored the sensitivity of resistant thyroid cancer cells. The present study further revealed that cancer cell death was primarily caused by ATP exhaustion in excessive DNA repair with high PARP-1 activity. In patients with thyroid cancer, a positive correlation between SLC6A9-5:2 and PARP-1 was identified, and low SLC6A9-5:2 expression was associated with a worse prognosis of papillary thyroid carcinoma. Hence, our data provide a new lncRNA-mediated regulatory mechanism implying that SLC6A9-5:2 can be used as a novel therapeutic target for 131I-resistant thyroid cancer.

RESUMO

PURPOSE: The stathmin/oncoprotein 18 (STMN1) is overexpressed in various human cancers. The aim of our study was to investigate its clinical significance and interaction with p53 in oral squamous cell carcinoma (OSCC). MATERIALS AND METHODS: Stathmin expression was assessed by Oncomine, Gene Expression Omnibus (GEO) database, western blotting, and reverse transcription polymerase chain reaction. We investigated the relationship between stathmin expression and clinical characteristics among 109 OSCC patients by immunohistochemical staining. The prognosis factors were analyzed using univariate and multivariate analysis. Immunoprecipitation assay and The Cancer Genome Atlas (TCGA) analysis were used to detect the relationship between mutant p53 and stathmin. RESULTS: Stathmin was overexpressed in OSCC. In immunohistochemical analysis, high stathmin expression correlated with gender (P = 0.040), T stage (P = 0.015), TNM stage (P = 0.045), and pathological differentiation (P = 0.000). We found a correlation between the stathmin expression and overall survival (P = 0.027). Multivariate analysis suggested only lymph node metastasis (P = 0.007) and stathmin expression (P = 0.013) as independent prognostic factors. There was interaction between stathmin and p53 in OSCC cell lines with mutant p53 through immunoprecipitation assay. CONCLUSION: These results suggest that overexpression of stathmin could contribute to cancer progression/prognosis, and that interaction between p53 and stathmin may contribute to the gain-of-function of p53.

RESUMO

A new quinoline-based fluorescent probe was synthesized and characterized. Probe showed opposite fluorescence responses toward Cd(2+) (turn-on) and Hg(2+) (turn-off) in a 100% aqueous environment. X-ray crystallography analysis and TD-DFT calculations showed the different mechanisms of CHEF-based for -Cd(2+) and PET-based for -Hg(2+) caused by different binding modes and different electronic transfer patterns. The detection limits for the analysis of Cd(2+) and Hg(2+) ions were found to be 3.9 × 10(-8) M and 9.8 × 10(-7) M, respectively. Its in vivo sensitivities to Cd(2+) and Hg(2+) were demonstrated in the EC9706 cell line with the use of a laser scanning fluorescence microscope.

RESUMO

Overglaze decoration porcelain is an important category of ancient Chinese ceramics, which has significant artistic value and scientific value. Nondestructive analysis methods such as Raman spectroscopy and EDXRF were used to analyze the overglaze decorations on the Jingdezhen ceramic samples of Yuan, Ming and Qing Dynasty. The recipe and color mechanism of the overglaze pigments were discussed according to the chemical composition and phase composition analysis. The study found that dark red overglaze decorations of ancient Honglvcai, Wucai and famille rose in Jingdezhen are colored by hematite, yellow color is lead tin yellow, carmine decoration is colored by gold less than 0. 1 % in concentration, and green decorations are colored by bivalent copper ion. The result also indicates that the effective combination of Raman spectroscopy and EDXRF can play an important role in the deep research on ceramic artifacts, especially for the overglaze decoration pigments which are interveined each other.

RESUMO

Color glaze is one of the four famous traditional ceramics of Jingdezhen, especially for the products from Ming and Qing Dynasties' official kilns which have rich connotation of technology and culture. The chemical composition and chromaticity characteristic of glaze and body of purple-gold glaze samples from Jingdezhen Ming and Qing Dynasties' official kilns were analyzed by energy dispersive X ray fluorescence (EDXRF) and colorimeter. Preliminary study on the composition, formula and chromaticity characteristic of glaze of purple-gold glaze samples of different period was carried out and the intrinsic causes of ifferences were discussed. The result shows that the concentration of magnesium and calcium in purple-gold glaze is different from the other glazes in Jingdezhen in the same time, probably due to the addition of auburn or brown limestone which is rich in magnesium. The purple-gold glaze sample of Ming Dynasty is darker chiefly because the concentration of magnesium and calcium is higher than the sample of Ming Dynasty which led to iron crystal separated, reducing the brightness and glossiness of glaze. In addition, the body of purple-gold glaze samples from Jingdezhen Ming and Qing Dynasties' official kilns has the characteristics of high silicon and low aluminum and the molar ratio of silicon to aluminum of samples from Ming Dynasty to Qing Dynasty declined, showing that the concentration of kaolin of sample's body of Ming dynasty was increased. The result of this experiment fill deficiency in the ceramic science and technology research in our country about purple-gold glaze from Ming and Qing Dynasties' official kilns and provides scientific material for comprehensive understanding of porcelain marking technology and intrinsic value of Jingdezhen official kiln.

RESUMO

Aseptic joint loosening is a key factor that reduces the life span of arthroplasty. There are currently few effective treatments for joint loosening except surgical revision. We explored the inhibitory effects of p110ß-targeted small interfering RNA (siRNA) and lentivirus on particle-induced inflammatory cytokine expression in the murine macrophage cell line, RAW264.7. siRNA- and lentivirus-targeting p110ß were transfected and infected prior to particle stimulation, respectively. Ceramic and titanium particles of different sizes were prepared to stimulate macrophages. Fluorescence microscopy showed that the efficiency of siRNA transfection and lentivirus infection were 74.2 ± 4.2 and 92.3 ± 2.6 %, respectively. TNF-alpha mRNA in the particle stimulation plus RNA interference (RNAi) groups were significantly lower compared with the particle stimulation-only groups (P < 0.05). Similarly, protein levels of TNF-alpha in RNAi-treated groups were significantly decreased after transfection or infection (P < 0.05). It showed that Phosphor-AKT (Ser473) activation was significantly reduced by RNAi through western blot. As assessed by CT, micro-CT and histological analysis, particle implantation induced a significant osteolysis in mice calvaria, which was limited by p110ß lentivirus addition. These results suggested that p110ß subtype of PI3K, followed by activation of Ser473, may possibly participate in the regulation of macrophages activity by wear particles, ultimately resulting in the TNF-α secretion and osteolysis.

RESUMO

Color glazes of Ming and Qing official kilns are excellent representatives of the famous ancient Chinese porcelains. The study of official ware with Jilan glaze has been an important topic. But it made slow progress due to the rarity of samples with strict production management and using system. The recipes, chemical composition and chromaticity characteristic of the Jilan samples excavated from official kilns in the Ming and Qing dynasties were first discussed by systematical testing with the energy dispersive X-ray fluorescence (EDXRF) and color difference meter. The results showed that the porcelain stone content in Jilan bodies of official kiln in the Ming dynasty is higher than the samples of the Qing dynasty. The manganese content in Jilan glazes of the Ming dynasty is higher than that in the Qing dynasty, while the glaze ash addition and the lightness value in the glaze are opposite.

RESUMO

The southern white porcelain is the rise in our country's pottery, it is later than the development of northern white porcelain, but it is rising stars, blockbuster, well-grounded, keeping continuous. Jingdezhen kiln and Dehua kiln are the most representative kilns among the southern white porcelain kilns. the present paper used EDXRF to test 30 pieces of Song, Yuan and Ming period' Dehua white porcelains and Jingdezhen Yuan dynasty Shufu white porcelains samples to analyze the different characteristics of the bodies and the glaze of these samples in terms of time and space. The results show that in the bodies and the glaze of Jingdezhen Shufu white porcelains the iron content is obviously on the high side, and the Dehua white porcelains' potassium content is obviously on the high side, and along with time development they have been in a rising trend, which should be the main reason for the sudden rise of Dehua white porcelain sculptures.

RESUMO

Early crafts of porcelain making in Jindezhen were an important issue in ceramic history of China. The chemical composition of white porcelain and celadon samples excavated from Xianghu Kiln in the five dynasties was analyzed using energy dispersive X-ray fluorescence (EDXRF). Raw material recipes of the samples were discussed. The results showed that white porcelain bodies of Xianghu Kiln in the Five Dynasties were made from porcelain stone, while celadon bodies were made from porcelain stone and Zijin clay. Glaze ash and glaze stone were both used in the formula of white porcelain and celadon glaze, and the amount of glaze ash in the celadon was higher than that in the white porcelain samples.

RESUMO

Conversion of artificial corn stalk wastewater, which was prepared according to the main composition of the corn stalk fermentation lixivium, into bio-hydrogen gas by mixed culture was performed in a 20 L half-continuous flow fermenter. The influences of several environmental factors on the bio-hydrogen production, such as HRT, C/N ratio, Fe2+ and artificial corn stalk wastewater concentration were discussed in the tests. The experimental results showed that HRT, C/N ratio, Fe2+ and artificial corn stalk wastewater concentration significantly affected the fermentation hydrogen production. The maximum H2 yield of 11.80 mol/kg, H2 concentration of 56% and hydrogen production rate of 8.81 L/(L x d) were obtained at HRT = 10 h, C/N = 100, Fe2+ concentration of 100 mg/L and substrate concentration of 12.5 g/L by mixed culture, respectively. In the fermentation hydrogen-producing process, the conversion efficiency of the substrate was more than 90%, and 39.40% of COD was removed from the reactor. The main by-products in the liquid phase were acetic acid, butyric acid, propionic acid and a little ethanol and butanol throughout this study.

RESUMO

OBJECTIVE: To study the role of nuclear factor-kappa B (NF-kappaB) and cycloxygenase-2 (COX-2) in the onset of phlegm obstruction due to lung-deficiency in rats and the therapeutic mechanism of Huatan Recipe. METHODS: Twenty-four SD rats were randomly divided into normal group, model group and treatment group, with 8 rats in each group. The rats in the model group and treatment group were exposed to sulfur dioxide and cold wind to establish the rat model of phlegm obstruction due to lung-deficiency, and the rats in the treatment group were also treated with Huatan Recipe, a compound traditional Chinese medicine. The expression of NF-kappaB in the bronchial epithelial cells of the rats was tested with the method of immunohistochemistry, and the COX-2 mRNA in the lung tissues of the rats was measured by using reverse transcription-polymerase chain reaction. RESULTS: The expressions of NF-kappaB and COX-2 mRNA in rats of the model group were higher than those of the normal group (P<0.01), and the expressions of NF-kappaB and COX-2 mRNA in rats of the treatment group were obviously lower than those of the model group (P<0.01). CONCLUSION: The NF-kappaB and COX-2 play an important role in the onset of phlegm obstruction in rats. Huatan Recipe may prevent the development of phlegm obstruction by down-regulating the expressions of NF-kappaB and COX-2 mRNA.

RESUMO

OBJECTIVE: To explore the effects of experimental varicocele on the apoptosis of spermatogenic cells in rats. METHODS: Thirty-two Sprague-Dawley(SD) male rats were randomly divided into four groups: pseudo-operation group, 45 d experimental group, 60 d experimental group and 90 d experimental group. Experimental varicocele was created by partial ligation of left renal vein. The apoptosis number of spermatogenic cells was measured by flow cytometry. RESULTS: No marked apoptosis peak was observed in the control group, but it was observed in all the other groups, and the height of the peak increased with the duration of experiment. CONCLUSIONS: Varicocele can induce apoptosis of large numbers of spermatogenic cells, and the decrease of spermatogenic cells. This may be the mechanism of varicocele causing male sterility.

RESUMO

Classical swine fever virus (CSFV), an enveloped positive-stranded RNA virus in the genus Pestivirus of the Flaviviridae family, is the causative agent of a highly contagious swine disease characterized by symptoms of hemorrhagic fever and immune depression, usually leading to substantial economic losses. The serological methods for detection of CSFV antibody such as ELISA are important means for the diagnosis of CSFV and immune surveillance. It is difficult to obtain CSFV antigen with high quality using traditional method because its titration titer is low in cell culture. CSFV has four structural protein named C, E0, El and E2. The E2 protein contains major antigenic determinants that are conserved between different CSFV strains and involved in neutralization by antibodies. So recombinant E2 protein can be developed as an alternative to the intact viral antigen. So far, CSFV E2 have not been expressed in E. coli with high level. Many factors, such as the secondary structure, the stability of 5' and 3' terminus of gene, the location of SD sequence and the bias of codes, are involved in the expressing level of foreign gene in E. coli . In this study, two sites of the E2 gene sequence were confirmed to be detrimental to its expression efficiency in E. coli through the computer-aided analysis. So they were mutated using recombinant PCR without changing the amino acids sequence of CSFV E2 gene. A plasmid was constructed by inserting the mutated E2 gene into the prokaryotic expression vector pET-28a(+) and named pETE2. The E. coli competent host BL21 (DE3)lysS transformed with pETE2 could express the E2 gene at high level, amounting to 28% of the total protein of the induced recombinant bacteria at the presence of IPTG. Except the hydrophobic transmembrane domain at C terminus, the recombinant E2 protein includes the total aa sequence. So it contains all the potential linear antigen epitopes of E2 protein because hydrophobic aa region can not form epitope. The recombinant E2 protein was CSFV-specific as proved by Western blotting and indirect ELISA. The rabbits immunized with the recombinant E2 can be protect from the challenge of hog cholera lapinized virus. This is the first report that E2 gene is expressed with high level expression in E. coli. In conclusion, it is an effective measure that mutate the CSFV E2 gene to increase its expression level in E. coli. The recombinant CSFV E2 protein possess fine immunonicity and can be used the antigen for the detection of CSFV antibody.
